Abstract
Description
2種の増殖因子BLyS(Bリンパ球刺激因子)及びAPRIL(増殖誘発リガンド)に関する独自の結合親和性を有する、3種の受容体TACI(膜貫通アクチベーター及びCAML−インターアクター)、BCMA(B細胞成熟抗原)及びBAFF−R(B細胞活性化因子の受容体)が同定されている(Marstersら、2000;Thompsonら、2001)。
アタシセプト(atacicept)(INN)は、受容体TACI(膜貫通アクチベーター及びカルシウムモジュレーター及びシクロフィリンリガンド(CAML)−インターアクター)の細胞外リガンド結合部分及びヒトIgGの修飾されたFc部分を含む、組換え融合タンパク質である。アタシセプトは、BLyS(Bリンパ球刺激因子)及びAPRIL(増殖誘発リガンド)に対するアンタゴニストとして作用し、これらは両方共腫瘍壊死因子(TNF)スーパーファミリーの一員である。BLyS及びAPRILは、B細胞成熟機能及び生存の重要な調節因子であることが示されている。

方法1
サンプルをPBS1×pH=7.2で0.5 mg/mLまで希釈し、40μL(20μg)を、TSKゲルG3000SWXL、5μm、7.8×300 mmにロードした。全てのランについて、溶離液は、0.05 Mリン酸ナトリウム、0.5 M硫酸アンモニウム、pH=6.0とした。
サンプルを移動相で0.25 mg/mLまで希釈し、40μL(20μg)を、TSKゲルSAXLガードカラム6 mm×4 cmを連結した、TSKゲルG3000SWXL、5μm、7.8×300 mmにロードした。全てのランについて、溶離液は、0.05 Mリン酸ナトリウム、0.5 M硫酸アンモニウム、pH=6.0とした。
サンプルをPBS1×pH=7.2で0.5 mg/mLまで希釈し、40μL(20μg)を、71%の緩衝液A(0.1%TFA添加水)、及び29%緩衝液B(0.1%TFA添加アセトニトリル)で平衡化した、PLRP 4000 Aカラム、 8 μm, 50×4.6 mmにロードした。サンプルを、流速2 mL/分の直線グラジエントを用いて溶出した。異なる量の標準(IRS TACI-Fc5 2002/2001)を注入することにより、キャリブレーション曲線を作成した。
サンプルを、37℃2時間、酵素的分解(Lys−C)に供し、ガードカラム付のVydac C18 (4.6×50 mm)カラムで逆相クロマトグラフィを行った。

TACI−Fc薬物製品サンプルを、タンパク質濃度が4 mg/mLとなるよう、精製水で希釈した。その後、10μLの希釈サンプルを、200μLの変性還元溶液中(グアニジン 6 M中、0.15 MDTT)で希釈し、ボルテックスし、最後に60℃±2℃で90分インキュベートする。開始条件(71%溶離液A、0.05%トリフルオロ酢酸添加水、及び29%溶離液B、0.04%トリフルオロ酢酸添加アセトニトリル)で事前に平衡化したカラム(広孔ブチル、5mm、4.6 mm i.d.×50 mm、cod. 71 16-05、J. T. Baker製)に、75〜150μL(15〜30μg)を注入する。
TACI−Fc薬物製品サンプルを、タンパク質濃度10 mg/mLとするために、10 mMリン酸ナトリウム緩衝液、pH4.0中の、100 mg/mLのポロキサマー188溶液中で希釈した。TACI−Fc濃度が100 mg/mLより高い場合、計量によりサンプルの希釈を行うべきである。
TACI−Fc薬物サンプルのペプチドマッピングを行い、酸化状態の定量についての、MALDI−ToF検出の利用可能性を確認した。
12mmの光路長を有する2チャンネル・チャコール−エポン・センターピースを用いて、細胞にサンプルをロードした。HPLC試験の条件を模倣するように、サンプルを、希釈剤としてSE−HPLC溶離緩衝液を用いて希釈した。対応する緩衝液を、参照チャネルにロードした(当該装置は、デュアルビーム分光光度計のように機能する)。その後、ロードした細胞を、Beckman Optima XL-I分析超遠心のAN-50Ti分析用ローターに入れた。以下の試験設定に従って、この分析を行った。

0.5 mg/mLのTACI−Fc溶液を調製し(水での希釈により)、その濃度(c)を、ランベルト・ベールの等式により得た:OD=εbc(ε=モル吸光係数;b=光学細胞厚み)、ε(280 nm)=1.56(mL/mg)。出発溶液の濃度を、希釈係数により計算されたこれらの値を乗じることにより決定した。
コンフォメーション解析
CDは、一般的にペプチド及びタンパク質のコンフォメーションを研究するために使用される。遠紫外線(180〜250 nm)、及び近紫外線領域(250〜350 nm)の両方において、複数の因子が、CDスペクトルにおける特徴的ピークの出現に影響を及ぼす可能性があり、例えば、タンパク質濃度、温度、pH及びイオン強度がある。遠紫外線で観察される一般的なバンド位置は文献で報告されており、特定の二次構造の種類を表す(α−ヘリックス、β−シート、ランダムコイル)。近紫外線領域で観察されるCDバンドは、主に、Trp、Tyr、Phe及びジスルフィド結合によるものである。

固定波長でのCDにより観測される温度スキャンは、異なる温度での、タンパク質の2次及び3次構造の両方を調べる価値ある手法である。当該測定により、異なる形態における、タンパク質のアンフォールディング温度(Tunf)を評価することが可能となる。Tunfは、タンパク質のアンフォールディングの自由エネルギー(タンパク質の安定性の指標である)と直接的な関係を有するわけではないが、Tunfの任意の増加は、タンパク質安定性の増加に比例するはずであることが、広く認められている。したがって、Tmの変化は、特定の組成物が任意の安定化又は不安定化効果を有するかを示唆する。熱的変性は、温度と共にタンパク質コンフォメーション変化に関連するTrp(トリプトファン)のシグナル変動を観測することにより調べた。当該薬物物質調製物を、55〜70℃の範囲で、加熱した(1℃/分)。3次構造に対する温度の効果を、292.5 nmでの最小に対するCD楕円率の変化により検出した。遷移温度を計算するために、第四級多項式フィッティングを使用した。

動的光散乱は、粒子のブラウン運動により誘導される散乱を測定し、それを粒子サイズと関連付ける。これは、粒子をレーザービームにさらし、散乱光の強度ゆらぎを解析することが必要となる。より正確には、ブラウン運動により動く粒子のスピードを、粒子の大きさと関連付ける(ストークス−アインシュタイン等式)。デジタル式相関器が、2つのシグナル(この場合、強度シグナル)間の類似度を経時で測定し、それが、粒子の直径に関連する散乱強度ゆらぎの性質及び程度に関連する情報を与える。相関関数の決定後、それをサイズ分布の計算に用いることができる。
RALSは、蛍光検出器を用いて測定され、ここで励起及び発光波長は同一に設定されている。この立体配置において、当該蛍光検出器は、非常に感度の高いRALS検出器となる。RALSの増加は、サンプルにおける凝集/沈殿の指標である。
内在蛍光
タンパク質は、内在蛍光に寄与する可能性のある3つの芳香族アミノ酸残基(トリプトファン:Trp;チロシン:Tyr;フェニルアラニン:Phe)を含有する。フォールディングしたタンパク質の蛍光は、個々の芳香族残基由来の蛍光の組み合わせである。タンパク質蛍光は、一般的に、280 nm以上の波長、通常は295 nmで励起される。ほとんどの発光は、トリプトファン残基の励起により、チロシン及びフェニルアラニンによる発光はわずかである。トリプトファン最大蛍光発光の強度、量子収量及び波長は、非常に溶媒依存性である。トリプトファン残基を取り巻く溶媒の極性の減少により、蛍光スペクトルが低波長側へのシフトし、蛍光強度が増加する。タンパク質の疎水性中心に埋め込まれているトリプトファン残基は、タンパク質表面のトリプトファンと比較して、10〜20 nmシフトする。さらにトリプトファン蛍光を、近傍のプロトン化された、Asp又はGlu等の酸性基により消光させることができる。すなわち、蛍光は、芳香族残基が存在する微小環境における変動を反映する、強力な観測用ツールとして使用することができる。
RALSの測定は、FluoroMax蛍光分光計で、500〜800 nmの範囲で同期スキャン(λexc=λem)することによって行われる。これらの条件下(サンプルによる吸収はなく、光源による影響もない)、暴露強度は、主に溶液中に存在する散乱現象による(入射光/タンパク質)。総散乱強度は、タンパク質容量(dimension)の増加と共に増加するため、この補法は、凝集、サブユニット解離、分解等の事象の発生を観測するのに有用である。
微小分子の回転は、そのサイズ、形及び局所環境(すなわち溶媒)に依存する。極性化発光測定は、分子サイズの小変化(凝集、結合、開裂)及び環境変化(局所粘度、相転移等)を検出するためによく使用される。これらの測定における第一のステップは、フルオロフォアの選択群(光選択)の励起である。垂直極性化光は、典型的には、その吸収双極子が垂直方向に向く、分子群を励起するために使用する。この相において、垂直極性化励起光は、経路上での励起の際に、偏光子を用いて作製される。励起されると、当該分子は、励起状態の寿命の間(〜10-9秒)回転する可能性がある。当該化合物移転は、蛍光発光を脱分極することになる。当該分極化発光成分の測定により、分子の回転運動の種類及び程度の計算をすることができる。蛍光発光の分極化成分は、発光経路における偏光子を用いて測定する。垂直(V)及び水平(H)の発光成分の大きさから、回転挙動の程度及び種類を計算できる。異方性(A)は、総光強度で除した、直線分極化成分の強度の間の差異として定義される比率である。
Gは、補正因子、G=IHV/IHHである。
TACI−Fcインビトロバイオアッセイは、Jurkat(ヒト急性T細胞リンパ球)形質移入細胞(Jurkat pKZ142)に基づく。この細胞系列は、2つのプラスミドを形質移入されている。第一のプラスミドは、CMVプロモーターの制御下の全長TACI cDNA、そして第二のプラスミドは、ルシフェラーゼレポーター遺伝子を駆動するNF−kB/AP−1をコードする。この方法は、zTNF4が細胞表面TACIレポーターに結合し、シグナル伝達カスケードを誘因し、形質移入したNF−kB/AP−1ルシフェラーゼレポーター遺伝子を刺激する能力に基づく。可溶性TACI−Fcの存在は、TACI受容体の結合によるxTNF4を阻害し、ルシフェラーゼ発現を低下させる。
pH、緩衝剤種類、及び賦形剤のタンパク質安定性への影響を評価した。70又は100 mg/mLの濃度でTACI−Fcの溶液を調製し、以下の変数について予備的に調べた。
−pH(4、5、6、7)
−緩衝剤(酢酸塩、リン酸塩、コハク酸塩、クエン酸塩、ヒスチジン)
−糖(マンニトール、ソルビトール、グルコース、スクロース、トレハロース)
−賦形剤(塩化ナトリウム、塩化マグネシウム、塩化カルシウム、グリシン)
−凍結−融解(F−T)サイクル(1、3、5回のF−T)、20 mM緩衝液中(ヒスチジン、リン酸塩、コハク酸、クエン酸)pH5−6−7。
−40℃でのインキュベーション(振とう及び非振とう条件)
−2〜8℃での保存、20 mM緩衝液中(酢酸塩、ヒスチジン、リン酸塩)pH5−6−7。
予備的期間の間になされた選択に基づき、実験計画を設定し、タンパク質安定性に関する異なるレベルでの事前に調べられた因子の影響を評価した。酢酸塩及びヒスチジン緩衝液中の調製物を、以下の界面活性剤:ポロキサマー188(Lutrol(登録商標) F-68)及びTween20pと共に、及び以下の賦形剤:アルギニン、グリシン、リジン、マンニトール及びトレハロースと共に試験した。これらの調製物をガラスバイアル中、2〜8℃、25℃及び40℃で保存し、凝集(SE−HPLC及びAUCにより)、pH、外観及びオスモル濃度について試験した。生物物理学的解析手法(例えば、円偏光二色性、第二UV微分分光法、内因性蛍光等)も適用した。
予備調製期間の終わりに、いくつかの候補調製物が確認され、それらは、70又は100 mg/mLいずれかのTACI−Fc、10 mM酢酸緩衝液、賦形剤としてマンニトール(51 mg/mL)又は無水トレハロース(80又は96 mg/mL)、ポロキサマー188(Lutrol(登録商標) F-68)(0.05 mg/mL)含有又は未含有のいずれか、を含む調製物であった。pH値は、4.8、5.0、5.2及び5.4で試験した。

25℃では、70 mg/mLの調製物は、一般的に100 mg/mLのものよりも、凝集の比率(凝集/月(%))に関して緩やかな傾きを示した。後者の群においては、調製物21D及び21Eが、最も緩やかな傾きを示すものであった。40℃では、調製物21Eが、100 mg/mL液体調製物の群において最も緩やかな値の傾きを呈した。
AUCプロファイルは、調製物21Eについて2〜8℃及び25℃では、変化は観察されなかった。安定時間にわたるモノマー増加傾向が、70 mg/mL調製物について検出された。
100 mg/mL調製物の群においては、21Eが、最も高いTunfを示すものであった。5.0以外のpH値は、より低いTunfをもたらした(すなわち、より高い安定性)。
40℃では、最大発光波長における少数の変動が、70 mg/mL、及び100 mg/mL候補では21Eで検出された。
40℃で保存後、「最適な」pH5以外のpHでは、RALSが顕著に増加する。マンニトールを含む調製物の散乱は、他のものよりもかなり高かった。調製物21A、21B、21D及び21Eは、より低い値の散乱を示した。2〜8℃で保存後、これらは全く散乱光の増加を示さなかった。
調製物21A及び21Bについて、40℃で(1ヶ月)保存後、異方性の変動は観察されなかった。調製物21F及び21Hについて、明らかな変動があった。他のものについては、中間の変動が観察された。
2〜8℃では、サイズ分布の明らかな変動は検出されなかった。25℃での保存後、より大きな種においていくらかの減少%が観察された。40℃で保存後、マンニトール含有のものと、pHが5.0以外のものの両方を除く全ての調製物について、高分子量の種は、劇的に増加しなかった。
100 mg/mLの群において、調製物21D及び21Eについて、より高い熱力学的安定性が観察された。
25℃及び40℃で3ヶ月にわたって、生物活性の減少は観察されなかった。
液体調製物の予備スクリーニング研究は、70 mg/mLのTACI−Fc5溶液の安定性について最適なpHがpH5付近であることが示されている。pHが高くなればなるほど、凝集現象(SE−HPLCにより評価)及び濃度低下の発生(RP−HPLC及び最適密度から推定)がより強かった。塩(NaCl、CaCl2及びMgCl2等)の存在は、同様に凝集体の増加をもたらした。pHが5未満の値も最適ではなく、異なる緩衝液において、pH=5.0と比較した4.0での、円偏光二色性でのコンフォメーション研究によっても示される通りである。予備的なDSC実験は、トレハロース及びスクロースが分子の安定性にいくらかの正の影響をもたらすことを示した(すなわち、より高いアンフォールディング温度)。
目的:
本研究の目的は、反復投与調製物の観点から、異なる静菌剤とTACI−Fcとの適合性を評価することであった。以下の静菌剤を試験した。ベンジルアルコール0.9%;m−クレゾール0.3%;フェノール0.5%;クロロブタノール0.5%;フェニルエタノール0.5%;ベンジルアルコール0.3%+塩化ベンザルコニウム0.001%。
薬物物質+静菌剤:
−40℃で2週間保存後、参照サンプル(静菌剤無添加)と比較して、防腐剤の存在下での薬物物質に対して、総凝集体の増加(15〜70%)が観察された(SECによる)。同様の分解速度が、25℃及び2〜8℃で観察された。
−(SEC及びCD分析により)負の影響が最小であることが判明した静菌剤は、ベンジルアルコール+塩化ベンザルコニウムの混合物であった。
−TACI−Fc液体候補物(酢酸、pH5、トレハロース)への防腐剤の添加により、薬物物質で観察されたものより、それほど顕著でない凝集体の増加がもたらされた(約10〜25%、40℃で2週間後)。これは、凝集の阻止及び負の二次構造の低減(遠紫外線CD実験により証明される)に、トレハロースが有効であることを実証する。
−ベンジルアルコール+塩化ベンザルコニウムの連携は、防腐剤の存在下の調製物群の中で、凝集体について、最良の結果を提供した。
タンパク質の完全性に与える複数の静菌剤の影響を、TACI−Fc薬物物質(未処理バルク)、及びNa−酢酸及びトレハロースpH5中、100 mg/mLで調製した調製TACI−Fcで評価した。
目的:
本研究の目的は、TACI−Fcの液体調製物(Na−酢酸、トレハロース、pH5)を、100 mg/mLで1mLのHypakシリンジに充填し、当該シリンジを2種類のゴム製プランジャー(W4023/50及びW4023/50G FluroTec)で栓をしたものの安定性を評価することであった。
−被覆(W4023/50G FluroTec)及び未被覆(W4023/50G)プランジャーで栓をした1mLのガラスシリンジに充填した、100 mg/mLのTACI−Fcについて、6ヶ月まで試験した結果は、以下の通りに要約できる。
−タンパク質含量:保存で、タンパク質含量の減少は観察されなかった。
−短縮形態:短縮形態において、測定値対バイアル中の物質は同程度であった(約40℃及び25℃で5ヶ月後の薬物物質と比較して、2〜8℃では増加しない)。
−生物有効性:生物活性を、3ヶ月保持した(2〜8℃及び25℃)。
−pH:保存で、pH移動は観察されなかった。
1 mLのHypakシリンジに100 mg/mLで充填したTACI−Fc液体調製物(酢酸緩衝液、pH5+トレハロース)は安定であった。この試験において評価された2種類のゴム製プランジャー(W4023/50及びW4023/50G FluroTec)は、同等であり、液体調製物の安定性に影響はなかった。
予備充填型シリンジにおける異なる強度でのアタシセプトの安定性を評価した。方法論は、実施例1に記載した通りに行った。
4種のTACI−Fcアミノ末端切断型が作製された。4種全て、配列番号6のアミノ酸残基番号30に融合された、WO 02/094852に開示されたような修飾されたヒト組織プラスミノーゲン活性化因子シグナル配列(配列番号25)を有する。しかし、これら4種のタンパク質は、「Fc5」が配列番号6のTACIアミノ酸配列に融合される点の位置が異なる。表1は、4種の融合タンパク質の構造を概説している。
前記TACI−Fc発現構築体を使用し、電気穿孔法により、動物タンパク質−非含有培地において増殖した懸濁−適合されたチャイニーズハムスター卵巣(CHO)DG44細胞を、形質移入した(Urlaubら、1986)。CHO DG44細胞は、両方のジヒドロ葉酸レダクターゼ染色体座における欠失のために、機能的ジヒドロ葉酸レダクターゼ遺伝子を欠いている。漸増濃度のメトトレキサート存在下での細胞の増殖は、ジヒドロ葉酸レダクターゼ遺伝子の増幅を生じ、かつ本発現構築体上に連結された組換えタンパク質−コードされた遺伝子を生じた。
メトトレキサート選択プロセスの回収時に、分泌されたTACI−Fcタンパク質を含有する馴化培地を、ウェスタンブロット分析により試験した。

- さらに防腐剤を含んでなる、請求項1〜8のいずれか1項に記載の調製物。
- 前記防腐剤が、ベンジルアルコールと塩化ベンザルコニウムとの組み合わせである、請求項9に記載の調製物。
- 前記TACI−Ig断片が、配列番号1のアミノ酸残基34〜66及び/又はアミノ酸残基71〜104を含んでなる、請求項1〜10のいずれか1項に記載の調製物。
- 前記断片が、配列番号1のアミノ酸残基30〜110、又はそれと少なくとも90%同一であるか、もしくは10個未満の保存的アミノ酸置換を含むその変種であって、BLyS及び/又はAPRILに結合する変種を含む、請求項11に記載の調製物。
- 前記免疫グロブリン−定常ドメインが、ヒトIgG1定常ドメインである、請求項1〜12のいずれか1項に記載の調製物。
- 前記ヒトIgG1定常ドメインが、配列番号2の配列、又は20個未満の保存アミノ酸置換を含むその変種を含んでなる、請求項13に記載の調製物。
- 配列番号3の配列、又はそれと少なくとも90%同一であるか、もしくは30個未満の保存的アミノ酸置換を有するその変種であって、BLyS及び/又はAPRILに結合する変種を含む、請求項1〜14のいずれか1項に記載の調製物。
- 前記TACI−Ig融合タンパク質を、20 mg/mL〜180 mg/mLの濃度範囲で含んでなる、請求項1〜15のいずれか1項に記載の調製物。
- 液状である、請求項1〜16のいずれか1項に記載の調製物。
- 反復投与のための、請求項1〜17のいずれか1項に記載の調製物。
- 請求項1〜18のいずれか1項に記載の調製物を含んでなる、医薬組成物。
- 自己免疫疾患又はリンパ増殖性障害の治療のための、請求項1〜18のいずれか1項に記載の調製物、又は請求項19に記載の医薬組成物。
- TACI−Ig融合タンパク質の溶液を調製するステップ、及び当該溶液のpHを4.5〜5.5、又は4.7〜5.3又は4.9〜5.1、もしくは〜5.0の範囲に調整するステップを含んでなる、請求項1〜18のいずれか1項に記載の調製物の製造方法。
- 滅菌容器に、所定量の前期調製物を入れるステップを含んでなる、請求項21に記載の調製物の製造方法。
- 前記容器が、ガラスバイアル又は予備充填型シリンジから選択される、請求項22に記載の方法。
