Description
用語“分離マトリクス”は、本明細書中において、事実上いずれかの分子または化合物の分離のために有用な材料を意味し、リガンド(その他の分子と相互作用することができる官能基)が結合した支持体(担体)から構成される。
本発明は、リガンドが支持体の薄いシェルとして提供される、新規な分離マトリクスに関する。リガンドは、支持体に対して直接的に結合することができ、または増量剤を介して結合することができる。

図1は、本発明に従う増量剤-リガンドビーズの説明図である。球状粒子を取り囲むシェルとほとんど同様に、ビーズに対して増量剤層がどの程度の薄さであるのかが、この図から明らかである。
イントロダクション
生体分子のラージスケールの精製のために通常は使用されるクロマトグラフィー媒体を適切なリガンドと置換して、分離を達成する。リガンドは、多量のサンプルを吸着するため、通常はビーズ中で均質に置換される。しかしながら、本発明者らは、ビーズの非常に薄い外部層が、リガンドにより置換される場合にのみ、高いタンパク質容量を得ることができることを見いだした(図1)。この種のシェル-ビーズの高いタンパク質容量を得るための必要条件は、リガンドを増量剤に対して結合することである。さらに、複合体[リガンド-増量剤]をマトリクスに結合されることが、重要である。2つの試作品が、非常に薄いシェル(2〜3μmの厚さ)に結合させたDEAE-デキストランをセファロース6 Fast Flowに対して用いて、製造された。これらの2種の試作品を、Q-基(-N(CH3)3)を薄いシェルと結合させたが増量剤を含まない試作品と比較した。さらに、この結果を、商業的に利用可能な媒体であるDEAEセファロースFast Flow(GE Healthcare, Uppsala, Sweden)とも比較したが、この場合、DEAE基がビーズ全体に均質的に分布し、そして精製時には増量剤を何も使用しない。
マトリクスの容量は、確定された床容量(settled bed volume)のことをいい、そしてグラムで示されるマトリクスの重量は、吸引乾燥重量のことをいう。ラージスケールの反応のため、マグネティックバースターラーの使用が、ビーズを損傷しやすいため、攪拌は、懸架性モーター-駆動性スターラーのことをいう。スモールスケールの反応(20 mLまたはゲル20 gまで)を、閉鎖バイアル中で行い、そして攪拌は、震盪テーブルを使用することを意味する。従来の方法を、機能性の分析、およびアリル化の程度またはビーズ上のアミン含量の程度の決定のために使用した。
セファロース6 Fast Flowのアリル活性化
セファロース6 Fast Flowを、グラスフィルタ上で蒸留水を用いて洗浄した。ゲル、250 mL、をフィルター上に注ぎ、そして3首の丸底フラスコ中で秤量した。NaOH(115 mL、50%溶液)を添加し、そして機械的攪拌を開始した。ホウ化水素ナトリウム、0.5 g、および硫酸ナトリウム、31 g、をフラスコに添加し、そしてスラリーを水浴中で50℃まで加熱した。およそ1時間後、250 mLのアリルグリシジルエーテル(AGE)を添加した。次いで、スラリーを、一晩強く攪拌した。約20時間後、スラリーをグラスフィルタに移し、そして蒸留水(×4)、エタノール(×4)および蒸留水(×4)を用いて洗浄した。
試作品D1のシェル活性化
アリル化されたゲル、25 mL、をフラスコ中で秤量し、そして250 mLの蒸留水および2.5gの酢酸ナトリウムを添加した。0.21等量の臭素、75μL、を、キャップ付きバイアル中、20 mLの蒸留水中に溶解した。臭素のこの量は、約2μmのシェル厚に対応する。この臭素溶液を、強く攪拌する間、アリルゲルスラリーに対して瞬間的に添加した。およそ2分後、ゲルをグラスフィルタ上で蒸留水を用いて洗浄した。
アリル化されたゲル、40 mL、を、フラスコ中で秤量し、そして400 mLの蒸留水および4 gの酢酸ナトリウムを添加した。0.31等量の臭素、175μL、を、キャップ付きバイアル中、30 mLの蒸留水中に溶解した。臭素のこの量は、約3μmのシェル厚に対応する。この臭素溶液を、強く攪拌する間、アリルゲルスラリーに対して瞬間的に添加した。およそ2分後、ゲルをグラスフィルタ上で蒸留水を用いて洗浄した。
23.5 mLの部分的に臭化されたゲルを、グラスフィルタ上で乾燥吸引して18.5 gにし、そして各ゲルをフラスコに移し、そして1時間室温(約20℃)にて、32 mLの蒸留水中19 g DEAE-デキストランの溶液と混合した。DEAE-デキストラン(500 000 g/molの分子量)を、GE Healthcare, Uppsala, Swedenから入手した。
試作品D2の塩素イオン能力は、126μmol/mLであった。220μmol/mLの残留アリル含量は、3μmの理論シェル厚に対応する。
4 mLの各ゲル(試作品D1およびD2)を、オーバーヘッドスターラーを備えたビーカー中で蒸留水と混合した。スラリーが、残留する深いオレンジ色/黄色を有するまで、臭素を添加した。3分間の攪拌後、スラリーが完全に退色されるまで、ナトリウムホミエート(sodium formiate)を添加した。次いで、ゲルをグラスフィルタ上で蒸留水を用いて洗浄した。
セファロース6 Fast Flowのアリル活性化
セファロース6 Fast Flowをグラスフィルタ上で蒸留水を用いて洗浄した。ゲル、250 mL、をフィルター上に注ぎ、そして3つ首丸底フラスコ中で秤量した。NaOH(115 mL、50%溶液)を添加し、そして機械的攪拌を開始した。ホウ化水素ナトリウム、0.5 g、および硫酸ナトリウム、31 g、をフラスコに添加し、そしてスラリーを水浴中で50℃まで加熱した。およそ1時間後、250 mLのアリルグリシジルエーテル(AGE)を添加した。次いで、スラリーを、一晩強く攪拌した。約20時間後、スラリーをグラスフィルタに移し、そして蒸留水(×4)、エタノール(×4)および 蒸留水(×4)を用いて洗浄した。
試作品のシェル活性化、Q-試作品
アリル化されたゲル、40 mL、をフラスコ中で秤量し、そして400 mLの蒸留水および4 gの酢酸ナトリウムを添加した。0.31等量の臭素、175μL、を、キャップ付きバイアル中、30 mLの蒸留水中に溶解した。臭素のこの量は、約3μmのシェル厚に対応する。この臭素溶液を、強く攪拌する間、アリルゲルスラリーに対して瞬間的に添加した。およそ2分後、ゲルをグラスフィルタ上で蒸留水を用いて洗浄した。
15 mLの部分的に臭化されたゲル(上記を参照)を、フラスコに移し、そして10 mLの水および6 mLの塩化トリメチルアンモニウム溶液と混合した。pHを50%NaOH を用いて12.5に調整し、そしてスラリーを45℃にまで加熱し、そして一晩強く攪拌した。およそ18時間後、ゲルをグラスフィルタ上で蒸留水を用いて洗浄した。
ビーズコアにおけるアリル排除
4 mLのゲル(試作品Q)を、オーバーヘッドスターラーを備えたビーカー中で蒸留水と混合した。スラリーが、残留する深いオレンジ色/黄色を有するまで、臭素を添加した。3分間の攪拌後、スラリーが完全に退色されるまで、ナトリウムホルミエート(sodium formiate)を添加した。次いで、ゲルをグラスフィルタ上で蒸留水を用いて洗浄した。
イントロダクション
3種のシェル媒体試作品(試作品:D1、D2およびQ)のクロマトグラフィーのふるまいを試験するため、オボアルブミンの貫流容量(breakthrough capacity)を調べた(実験の項を参照)。これらの試作品の結果を、リガンドがビーズの全体にわたり均質に分配されている、確立された媒体であるDEAEセファロースFast Flowと比較した。D-試作品とQ-試作品との比較から、増量剤を使用して、複合体[増量剤-リガンド]をシェルと結合させることについての高い能力および利点を得ることの重要性が示される。
貫流容量に関して調べる対象となるシェル媒体(試作品:D1、D2およびQ)を、Tricorn 5/100カラム中にパッキングし、そしてサンプル溶液を、バッファー溶液を用いて平衡化した後、1.0 mL/minの流速でカラムを通してポンプで流した。貫流容量を、最大UV検出器シグナル(280 nm)の10%で評価した。最大UVシグナルは、試験溶液を検出器内部に直接的にポンプで送り込むことにより推定された。最大吸光度の10%での貫流容量(Qb10%)を、以下の式に従って算出した:

式中、TR10%は、最大吸光度の10%での保持時間(分)であり、TRDは、システム中の空隙容量(void volume)時間(分)であり、Cは、サンプル濃度(4 mgタンパク質/mL)であり、そしてVCはカラム容量(mL)である。貫流容量測定にて使用される吸着バッファーは、50 mM Tris(pH 8.0)であった。
サンプルは、50 mM Tris(pH 8.0)中に溶解したオボアルブミンであり、そして供給濃度は、3.7 mg/mlに調整された。

下流のクロマトグラフィー精製プラットフォームを改良して、プロセス生産性およびタンパク質製造を大幅に上昇させるため、分離媒体のタンパク質容量(capacity)を上昇させることが非常に重要である。[増量剤-リガンド]複合体を非常に薄いシェル中に置換する場合のシェルビーズにより、高い貫流容量を達成することができることを証明するため(図1)、3種の試作品を構築した。All 3種の試作品全ては、セファロース6 Fast Flowに基づくものであった(表1)。
本発明に従う分離マトリクスを調製するため、初めにアガロース支持体粒子を、US 6,602,990(Berg)に記載される様に、調製した。デキストラン増量剤をアリル化し、そしてその後アガロース粒子に対して結合させた。次いで、スルホプロピル(SP)リガンドを、従来技術(1171039A)を介して、アリル基に対して結合させた。
分離マトリクスを、SepharoseTM 6FF(GE Healthcare)を支持体として使用し、そして1.3μmol/gのアリル化度での150 kDの分子量(Mw)を有するデキストランを使用して、上述の実施例4に従って調製した。以下の表4から明らかなように、この分離マトリクスは、タンパク質IgGについて、予期せず高い動的タンパク質容量を示した。
Claims (19)
- 支持体;支持体の外部部分に結合された1またはそれ以上の増量剤;および増量剤に結合されたリガンド;を含む分離マトリクスであって、増量剤が結合する支持体の部分は、マトリクスの全量の50%未満を構成する、前記分離マトリクス。
- 増量剤がマトリクスの内部部分に何も位置しない、請求項1に記載の分離マトリクス。
- 増量剤が結合する支持体の部分がマトリクスの全量の1〜17%を構成する、請求項1または2に記載のマトリクス。
- 増量剤が分岐分子である、請求項1〜3のいずれか1項に記載のマトリクス。
- 増量剤の分子量が、1000〜20×106 Daの範囲である、請求項1〜4のいずれか1項に記載のマトリクス。
- 増量剤が、アガロース;デキストラン;デキストリン;およびそれら2種または3種の混合物、からなる群から選択される、請求項1〜5のいずれか1項に記載のマトリクス。
- 支持体が、実質的に球状粒子から構成される、請求項1〜6のいずれか1項に記載のマトリクス。
- 支持体が、多糖などの天然のポリマーから調製される、請求項1〜7のいずれか1項に記載のマトリクス。
- リガンドが、アニオン交換体;カチオン交換体;疎水性相互作用クロマトグラフィー(HIC)リガンド;逆相クロマトグラフィー(RPC)リガンド;固定化金属アフィニティークロマトグラフィー(IMAC)リガンド;親和性リガンド;および多モードリガンド、からなる群から選択される、請求項1〜8のいずれか1項に記載のマトリクス。
- リガンドが、アニオン交換体またはカチオン交換体である、請求項9に記載のマトリクス。
- リガンドが、プロテインA、プロテインAの1またはそれ以上の部分、またはプロテインA模倣物、を含む親和性リガンドである、請求項9に記載のマトリクス。
- (a)支持体を提供し;
(b)リガンドおよび/またはリガンド結合基が結合される1またはそれ以上の増量剤を提供し;そして
(c)増量剤を支持体の外部部分に対して結合させる;
を含む、分離マトリクスを調製する方法であって、ここで増量剤が結合する支持体の部分がマトリクスの全量の50%未満を構成する、前記方法。 - 工程(b)において提供される増量剤が、支持体に対する増量剤の結合に続く、その後のリガンドの結合を可能にするかどうかについて評価される、請求項12に記載の方法。
- (a)支持体、支持体の外部部分に結合された1またはそれ以上の増量剤、および増量剤に結合されたリガンドを含む分離マトリクスであって、全てのリガンドが増量剤を介して支持体に対して結合されるものを提供し;
(b)液体を分離マトリクスに接触させて、少なくとも一つの標的化合物をリガンドに対して結合させ;そして場合により
(c)(1または複数の)標的化合物を液体から放出させる溶出液を添加することにより、少なくとも一つの標的化合物を溶出する;
を含む、液体から少なくとも一つの標的化合物を分離するプロセス。 - 増量剤が結合する支持体の部分が、1〜17%など、マトリクスの全量の50%未満を構成する、請求項14に記載のプロセス。
- 分離マトリクスを、請求項12または13の方法に従って調製する、請求項14または15に記載のプロセス。
- 分離マトリクスがイオン交換リガンドを含み、濃度勾配溶出により少なくとも一つの標的化合物を溶出する工程を含む、請求項14〜16のいずれか1項に記載のプロセス。
- 少なくとも一つの標的化合物が、組換えタンパク質または核酸である、請求項14〜17のいずれか1項に記載のプロセス。
- 少なくとも一つの標的化合物が、イムノグロブリンまたはその融合タンパク質のフラグメントである、請求項14〜17のいずれか1項に記載のプロセス。
