Abstract
Description
(a) タクロリムスおよびシクロスポリンよりなる群から選択される少なくとも1種類の免疫抑制剤に特異的に結合可能な少なくとも1種類の抗体またはタンパク質と、(b) 少なくとも約1.0Mの濃度の塩を含む測定用希釈剤と、(c)(i) 約4:1から約1:2の範囲の比のプロピレングリコールとエチレングリコールと、(ii)(a)の少なくとも1種類の免疫抑制剤を含むコントロール組成物を含む溶解試薬と、を備える。
別に定めない限り、特許請求の範囲および明細書に使用される用語は以下に記載の通り定義される。
疫抑制剤に特異的に結合する抗体はこの免疫抑制剤に対し「特異性がある」と言われる。
本発明の測定法方法は、一般に動物、好ましくは哺乳動物、より好ましくはヒトに由来する検査試料に基づいて実施する。
A.概説
本発明による免疫学的測定法は、検査試料における免疫抑制剤の定性および/または定量に用いることができる。例えばこれらの方法は、タクロリムス、エベロリムス、ゾタロリムス、シクロスポリンおよびこれらの化合物の任意のアナログの免疫学的測定法に適用できる。ある実施形態では、免疫抑制剤に対する免疫学的測定法は、抽出試薬組成物を検査試料および水と組み合わせ、検査試料を形成することで実施できる。抗体を免疫抑制剤(存在する場合)に結合させるのに適した条件下で、検査試料または検査試料抽出物を少なくとも1種類の免疫抑制剤に対して特異的な抗体と接触させ、測定混合物を形成し、抗体の免疫抑制剤への結合を検出する。
検査試料中の免疫抑制剤の定性的検出のための免疫学的測定法では、免疫抑制剤に結合する少なくとも1種類の抗体を、薬剤を含む疑いのある少なくとも1点の検査試料または検体抽出液に接触させて抗体−薬免疫複合体を形成する。本発明による免疫学的測定法においては、特定の免疫抑制剤に結合する任意の適当な抗体が使用できる。タクロリムス、ゾタロリムス、シクロスポリンおよびエベロリムスのそれぞれへの抗体は当該分野で公知および/または市販されており、これらはいずれも使用できる。

本発明の免疫学的測定法は、当分野で知られる任意のフォーマットを使用して実施できる。例えばサンドイッチフォーマット、競合阻害フォーマット(順または逆競合的阻害測定法の両者を包含する)または蛍光偏光フォーマットがあるが、これらに限定されるものではない。
本発明はまた、免疫抑制剤について検査試料を測定する検査キットを提供する。本発明による検査キットは、本発明による1以上の免疫学的測定法を実施するために有用な1以上の試薬を含む。検査キットは、1以上の容器を備えたパッケージを有し、1以上の分離された組成物として、また場合により、試薬が併存可能な場合は混合物として、試薬を保持する。検査キットは、他の素材(複数可)を含んでもよく、これらはユーザーの観点から望ましい可能性のある緩衝液(複数可)、希釈剤(複数可)、標準(複数可)および/または検体処理、洗浄または測定法の他の任意の工程に有用であるものなどである。

本実施例は、タクロリムスについての自動免疫学的測定法において測定用希釈剤に塩を添加した効果を説明する。
この実施例は、タクロリムスの自動免疫学的測定法において微粒子試薬への塩の添加の効果を説明する。これらの測定法については、NaClを種々の濃度でIMx(登録商標)Tacrolimus II測定法(Abbott Laboratories,Abbott Park,IL)の微粒子試薬成分に加える。
この実施例では、タクロリムスの自動免疫学的測定法によって測定用希釈剤における種々の塩の添加の効果を説明する。
Claims (30)
- 検査試料中の免疫抑制剤の存在または濃度を評価する方法であって、
検査試料または検査試料抽出物を免疫抑制剤に特異的な抗体に、免疫抑制剤(もし存在するのであれば)に抗体が結合するのに適した条件下で接触させて測定混合物を形成すること(前記条件は約0.4Mを超える塩濃度を含む。)、および
免疫抑制剤に対する抗体の結合を検出することを含む、方法。 - 免疫抑制剤が、タクロリムス、エベロリムス、ゾタロリムス、シクロスポリンまたはこれらの任意の化合物のアナログよりなる群から選択される、請求項1の方法。
- 免疫抑制剤がタクロリムスを含む、請求項1の方法。
- 免疫抑制剤がシクロスポリンを含む、請求項1の方法。
- 検査試料がヒト血液試料を含む、請求項1の方法。
- 塩濃度が約4.0M以下である、請求項1の方法。
- 塩濃度が約2.0Mである、請求項1の方法。
- 塩が1価アニオンを含む、請求項1の方法。
- 1価アニオンが塩化物を含む、請求項9の方法。
- 塩がアルカリ金属の塩化物である塩を含む、請求項9の方法。
- 塩が塩化ナトリウムを含む、請求項10の方法。
- 測定混合物がさらに、
エチレングリコール、プロピレングリコールおよびこれのアナログよりなる群から選択されるグリコール、および
炭素原子数が5個以下の少なくとも1種類のアルコール
を含む、請求項1から12のいずれかの方法。 - アルコールがメタノール、エタノールおよびプロパノールよりなる群から選択される、請求項12の方法。
- グリコール対アルコールの比が、約4:1から約1:4の範囲である、請求項12の方法。
- グリコール対アルコールの比が、約4:1から約1:2の範囲である、請求項14の方法。
- 検査キットであって、
(a)少なくとも1種類の免疫抑制剤に特異的に結合可能な少なくとも1種類の抗体、
(b)少なくとも約1.0Mの濃度の塩を含む測定用希釈剤、および
(c)エチレングリコール、プロピレングリコールおよびこのアナログよりなる群から選択されるグリコール、ならびに
炭素原子数が5個以下の少なくとも1種類のアルコール
を含む溶解試薬、
を含む検査キット。 - 免疫抑制剤が、タクロリムス、エベロリムス、ゾタロリムス、シクロスポリンおよびこれらの任意の化合物のアナログよりなる群から選択される、請求項16の検査キット。
- 免疫抑制剤がタクロリムスを含有する、請求項16の検査キット。
- 免疫抑制剤がシクロスポリンを含有する、請求項16の検査キット。
- 塩濃度が約10.0M以下である、請求項16の検査キット。
- 塩濃度が約4.5Mである、請求項16の検査キット。
- 塩が1価アニオンを含む、請求項16の検査キット。
- 1価アニオンが塩化物を含む、請求項22の検査キット。
- 塩がアルカリ金属の塩化物である塩を含む、請求項23の検査キット。
- 塩が塩化ナトリウムを含む、請求項24の検査キット。
- (a)の少なくとも1種類の免疫抑制剤を含むコントロール組成物をさらに含む、請求項16から25のいずれかの検査キット。
- アルコールがメタノール、エタノールおよびプロパノールよりなる群から選択される、請求項26の検査キット。

- 検査キットであって、
(a)タクロリムスおよびシクロスポリンよりなる群から選択される少なくとも1種類の免疫抑制剤に特異的に結合可能な少なくとも1種類の抗体またはタンパク質、
(b)少なくとも約1.0Mの濃度の塩を含む測定用希釈剤、
(c)(i)約4:1から約1:2の範囲の比のプロピレングリコールとエタノール
を含む溶解試薬、ならびに
(d)(a)の少なくとも1種類の免疫抑制剤を含むコントロール組成物
を備える検査キット。
