Description
ビタミンD又はその誘導体の安定性を改善するために、本発明の固体分散体はシクロデキストリンとともに、有効成分としてビタミンD又はその誘導体を含む。
また、本発明はビタミンD又はその誘導体の固体分散体とビスホスホネートとを含む、骨粗鬆症の予防又は治療用複合製剤を提供する。
R1は、クロロ、メチル、2−アミノエチル、3−アミノプロピル、4−アミノブチル、4−クロロフェニルチオ、2−(N−メチル−N−n−ペンチル)アミノエチル、3−ピリジルメチル、シクロヘプチルアミノ、(1−イミダゾリル)メチル、又は1−ピロリジニルエチルであり;R2は、水素、クロロ、又はヒドロキシであり;Mは水素又はナトリウムであり;zは正数である。
また、本発明は下記の段階を含む前記複合製剤の製造方法を提供する:
1)シクロデキストリン及びビタミンD又はその誘導体を溶媒に溶解させた後、得た混合物から溶媒を除去して固体分散体を得る段階;
2)段階1)で得た固体分散体を圧縮して粉末を得る段階;及び
3)段階2)で得た粉末をビスホスホネートと混合した後、乾燥して得られる混合物を複合製剤の形態で製剤化する段階。
表1に記載された量によって、2−ヒドロキシプロピル−β−シクロデキストリン(アルドリッチ社製)をエタノール/水の混合液に入れて溶液が透明になるまで攪拌した後、これにコレカルシフェロール(Fluka社製、Chemie GmbH、Buchs)を添加した。得られた混合物を噴霧乾燥器(Buchi社製、 Mini Spray Dryer、 B-191、スイス)を用いて噴霧乾燥させて固体分散体を得た。前記噴霧乾燥は入り口温度50℃及びポンプ速度は30rpmで行い、固体分散体は噴霧後2時間50℃で乾燥させた。
d,l−α−トコフェロール(BASF社製)0.02mgをさらに添加することを除いては実施例1と同一な方法によってビタミンD又はその誘導体の固体分散体を製造した。
アスコルビン酸(BASF社製)0.05mgをさらに添加することを除いては実施例1と同一な方法によってビタミンD又はその誘導体の固体分散体を製造した。
d,l−α−トコフェロール(BASF社製)0.02mg及びアスコルビン酸(BASF社製)0.05mgをさらに添加することを除いては実施例1と同一な方法によってビタミンD又はその誘導体の固体分散体を製造した。
2−ヒドロキシプロピル−β−シクロデキストリン56.00mgをコレカルシフェロール0.07mgと均質に混合して固体分散体を得た。
表4に記載された量によって、実施例1で製造されたビタミンD又はその誘導体の固体分散体を低置換されたヒドロキシプロピルセルロース及びブチル化ヒドロキシトルエン(BHT)と均質に混合し、得られた混合物を圧縮した後、粒子に粉砕し、30メッシュの篩いを通過させて均質した粉末を得た。得られた粉末をアレンドロネート(Medichem社製、スペイン)、マンニトール、低置換されたヒドロキシプロピルセルロース、クロスカルメロースナトリウム及び二酸化チタンと均質に混合した後、ここにステアリン酸マグネシウムを添加し、乾燥−混合物を錠剤に製剤化した。
表4に記載された量によって、実施例1で製造されたビタミンD又はその誘導体の固体分散体をブチル化ヒドロキシトルエン(BHT)とのみ均質に混合することを除いては前記製剤例1と同一な方法によって錠剤を得た。
表5に記載された量によって、コレカルシフェロール粉末(乾燥ビタミンD3 100CWS;Roche社製)を80メッシュの篩いにかけて、マンニトールと均質に混合してから、低置換されたヒドロキシプロピルセルロース及びBHTと均質に混合した後、アレンドロネート、クロスカルメロースナトリウム及び二酸化チタンと均質に混合し、ここにステアリン酸マグネシウムを添加して得られた乾燥混合物を錠剤に製剤化した。
製剤例1で製造した複合製剤325mgをフタル酸ヒドロキシプロピルメチルセルロース(HP−55)、アセチルモノグリセリド(Myvacet 9−40)、酸化チタン、及びタルクをアセトン及びエタノールの混合物に溶解させて製造した腸溶性コーティング液でコーティングした。1錠のコーティング工程に用いられたコーティング物質の量はそれぞれ表6に記載されている。
製剤例1で製造した複合製剤の代りに製剤例2で製造した複合製剤を用いたことを除いては、製剤例3と同一な方法によって腸溶性コーティングされた複合製剤を得た。
製剤例1で製造した複合製剤の代りに比較製剤例1で製造した複合製剤を用いたことを除いては、製剤例3と同一な方法によって腸溶性コーティングされた複合製剤を得た。
製剤例1で製造した複合製剤の代わりに比較製剤例2で製造した複合製剤を用いたことを除いては、製剤例3と同一な方法によって腸溶性コーティングされた複合製剤を得た。
ビタミンD又はその誘導体を含む固体分散体の安定性試験
試験物質として比較例2(コレカルシフェロール0.07mg)及び実施例1〜4で製造した固体分散体を用い、前記試験物質を60℃の乾燥オーブンで保管しながら、経時による各試験物質のコレカルシフェロール含量の変化を分析した。
−移動相:(クロロホルム:n−ヘキサン:テトラヒドロフラン=650:350:10)(v/v)
−注入量:100μL
−流速:1.0mL/分
−検出機:紫外吸光光度計(波長254nm;L−7400、HITACHI社製、日本)。
固体分散体の安定性比較試験
実施例1の固体分散体、市販中のコレカルシフェロール粉末である乾燥ビタミンD3 CWS 100(Roche社製)及び乾燥ビタミンD3 100 BHT(BASF社製)を用いて安定性試験を行った。
複合製剤の安定性試験
試験物質、即ち、製剤例1、2及び比較製剤例1、2で製造した複合製剤を60℃の乾燥オーブンで保管しながら、経時による各試験物質中のコレカルシフェロール含量の変化を試験例1に記載された方法と同一な方法によって分析した。その結果は表10に示されている。
腸溶性コーティングされた複合製剤の安定性試験
試験物質、即ち、製剤例3、4及び比較製剤例3、4で製造した複合製剤を60℃の乾燥オーブンで保管しながら、経時による各試験物質中のコレカルシフェロール含量の変化を試験例1と同一な方法によって分析した。その結果は表11に示されている。
Claims (22)
- ビタミンD又はその誘導体とシクロデキストリンとを含む固体分散体。
- 前記ビタミンD又はその誘導体がコレカルシフェロール(ビタミンD3)、エルゴカルシフェロール(ビタミンD2)、カルシトリオール、エルゴカルシトリオール及びこれらの混合物からなる群から選ばれる、請求項1に記載の固体分散体。
- 前記シクロデキストリンが、2−ヒドロキシエチル−β−シクロデキストリン、2−ヒドロキシプロピル−β−シクロデキストリン、2,6−ジメチル−β−シクロデキストリン、スルホブチルエーテル−7−β−シクロデキストリン、(2−カルボキシメトキシ)プロピル−β−シクロデキストリン、2−ヒドロキシエチル−γ−シクロデキストリン、2−ヒドロキシプロピル−γ−シクロデキストリン、及びこれらの混合物からなる群から選ばれる、請求項3に記載の固体分散体。
- 前記シクロデキストリンが、2−ヒドロキシエチル−β−シクロデキストリン、2−ヒドロキシプロピル−β−シクロデキストリン、2,6−ジメチル−β−シクロデキストリン、スルホブチルエーテル−7−β−シクロデキストリン、及びこれらの混合物からなる群から選ばれる、請求項3に記載の固体分散体。
- 前記ビタミンD又はその誘導体に対するシクロデキストリンの重量比が1:1〜1:2,000の範囲である、請求項1に記載の固体分散体。
- 安定化剤、薬剤学的に許容される添加剤、又はこれらの混合物をさらに含む、請求項1に記載の固体分散体。
- 前記安定化剤が、ブチル化ヒドロキシトルエン(BHT)、ブチル化ヒドロキシアニソール(BHA)、エリソルビン酸、アスコルビン酸、トコフェロール及びこれらの混合物からなる群から選ばれる、請求項7に記載の固体分散体。
- 請求項1に記載の固体分散体とビスホスホネートとを含む、骨粗鬆症の予防又は治療用複合製剤。
- 前記ビスホスホネートが、アレンドロネート(Alendronate)、エチドロネート(Etidronate)、クロドロネート(Clodronate)、パミドロネート(Pamidronate)、チルドロネート(Tiludronate)、リセドロネート(Risedronate)、インカドロネート(Incadronate)、ゾレドロネート(zoledronate)、及びこれらの薬剤学的に許容される塩、及びこれらの水和物又は部分水和物からなる群から選ばれる、請求項9に記載の複合製剤。
- 前記ビスホスホネートの量が複合製剤の総重量を基準として0.5〜90重量%の範囲である、請求項9に記載の複合製剤。
- 前記固体分散体の量が前記複合製剤の総重量を基準として0.1〜80重量%の範囲である、請求項9に記載の複合製剤。
- 前記ビタミンD又はその誘導体の量が複合製剤の総重量を基準として0.0005〜20重量%の範囲である、請求項9に記載の複合製剤。
- 安定化剤、薬剤学的に許容される添加剤、又はこれらの混合物をさらに含む、請求項9に記載の複合製剤。
- 前記安定化剤が、ブチル化ヒドロキシトルエン(BHT)、ブチル化ヒドロキシアニソール(BHA)、エリソルビン酸、アスコルビン酸、トコフェロール及びこれらの混合物からなる群から選ばれる、請求項15に記載の複合製剤。
- 前記安定化剤の量が複合製剤の総重量を基準として0.001〜10重量%の範囲である、請求項15に記載の複合製剤。
- 1)シクロデキストリン及びビタミンD又はその誘導体を溶媒に溶解させた後、得られた混合物から溶媒を除去して固体分散体を得る段階;
2)段階1)で得られた固体分散体を圧縮して粉末を得る段階;及び
3)段階2)で得られた粉末をビスホスホネートと混合した後、乾燥して得られて混合物を複合製剤の形態で製剤化する段階を含む、請求項9に記載の複合製剤の製造方法。 - 前記段階1)に用いられる溶媒が水、有機溶媒、又はこれらの混合物である、請求項18に記載の方法。
- 前記有機溶媒がエタノール、イソプロピルアルコール、アセトン、アセトニトリル、ジクロロメタン、クロロホルム、及びこれらの混合物からなる群から選ばれる、請求項19に記載の方法。
- 前記段階3)で得られた複合製剤を腸溶性コーティング物質でコーティングする段階をさらに含む、請求項18に記載の方法。
- 前記腸溶性コーティング物質が複合製剤の総重量を基準として0.5〜30重量%の範囲の量で用いられる、請求項21に記載の方法。
