Description
本発明は、被検体、特にヒト被検体の免疫応答を誘発または誘導する方法に関し、さらには、投与で針を用いない経路に有用な方法に関し、それによって皮下および筋肉経路のようなワクチンの投与の従来の注入経路に代わる方法を提供できる。
長年にわたって、非生ワクチン抗原を送達する手段として気道を利用したさまざまな試みがなされている。この経路は、ワクチン受容性に関する長所を示し(「針恐怖症」を避ける)、局所免疫応答を誘導する機会、および粘膜免疫系の想定される統一性を考えれば、遠位粘膜サイトでの応答を誘導する可能性を有する。ヒトおよび動物において鼻腔内経路によるワクチン送達は多くの配慮が注がれる。現在までの結果によれば、この経路は、抗原接種と免疫誘導とを確保するために、非常に高い抗原投与量および/または特別な送達技術の利用が必要であることが分かっている。同様に、従来、肺内または肺経路を通じるワクチン送達は、免疫応答を最適化するために、概して、マイクロカプセル化(たとえば、オヤ アルパ(Oya Alpa)ら、2005参照)などの特別な送達の技術が要求された。

本発明の背景として、発明者は、カニューレ処置されたヒツジをモデルとしてワクチン送達の形態について調べた。カニューレ処置は、詳細に調査すべき流入領域リンパ節の流出を可能とする。この研究は、ヒツジに対して、ワクチンモデルとしてISCOMATRIXTM(イスコマトリックス)アジュバントインフルエンザウイルスワクチンを用いて行なった。この研究の重要な発見は、鼻腔内投与においては、高い抗原投与であったとしても、あまり効率的でなく、弱い局所免疫を誘導することである。
これらの発見は、ワクチンを肺葉深くに送達することによる肺内経路によって、カニューレ処置されていないヒツジに対するワクチン送達の効果を調査する方向への研究の変化を促進した。
血液採取
ヒツジを拘束し、10mLシリンジと18G針を用いて頚動脈から血液10mLを採取した。
本研究では、スクロースグラディエント精製A/NEW Cledonia 20/99H1N1ウイルスをインフルエンザ抗原として用いた。該ウイルスは不活化されており、界面活性剤で破壊しておいた(Coulterら、1998)。抗原濃度は、血球凝集素含有量をベースにして一元放射免疫拡散法で決定した。GMPグレードISCOMATRIXTMアジュバンドは、以前説明されているプロセスに従ってCSLリミテッドより準備した(PearseとDrane、2005)。免疫付与の前に、ISCOMATRIXTMアジュバンドと好ましい量の抗原とを混合したものをワクチン処方物とした。
肺内免疫付与において、ヒツジを慎重にハーネスに拘束した。そして、左鼻腔から左肺の下葉に気管支鏡を注入した。
皮下免疫付与のために、ワクチン処方物を1mLシリンジと25G針を用いて大腿部内部に200μlの全容量を送達した。
気管支肺胞洗浄液(BAL)サンプルを採取するために、免疫のためと同様に拘束しているヒツジに気管支鏡を挿入し、気管支鏡に取り付けたシリンジを用いて肺葉にPBS(リン酸バッファーpH7.2)10mLを送達した。その際、同じシリンジを用いて気管支鏡によってBAL流体を回収した。
BALにおける抗インフルエンザ抗体と血清サンプルは、EIA(エンザイム イムノエッセイ)によって二連で評価した。つまり、濃度10μl/mLでインフルエンザ抗原を含む炭酸塩バッファー(pH9.6)50μLで、96ウェルマキシソープフラットボトムプレート(NUNU、ロスキルド(Roskilde)、デンマーク)の表面を被覆して一晩おいた。プレートは1%カゼイン酸ナトリウムでブロッキングしてから、二連でサンプルの1/5系列希釈の100μL添加した。特異的抗インフルエンザ抗体の結合は、ホースラディッシュペルオキシダーゼと抱合するウサギ抗ヒツジ全Ig、または、ウサギ抗マウスホースラディッシュペルオキシダーゼによって結合させた抗ウシ/ヒツジIgA(セロテック(Serotec)、オクスフォード)を用いて検出した。色度はTMB物質(ザイムド(Zymed)、サンフランシスコ)を添加することで発色させ、2MのH2SO4を添加することで停止させた。450nm光学濃度をBiO−Tel ELx800プレートリーダーで検出し、抗体エンドポイントタイターを計算した。
ELISAによる抗原特異的抗体応答を示した血清およびBALサンプルは、血球凝集抑制(HAI)についても調べた。このアッセイは、インフルエンザウイルスによる赤血球凝集の阻害状態を測定することで、機能的抗体のタイターを決定するものである。サンプルは、七面鳥の赤血球を用いてタマゴで増殖させたA/NEW Cledonia 20/99ウイルス(H1N1)に対してHAIテストした。HAIタイターは、赤血球のインフルエンザ凝集を阻害したエンドポイント希釈物によって決定した。HAIアッセイは、オーストラリアのメルボルンにあるWHOとのインフルエンザ委託研究共同研究所(WHO Collaborating Center for Reference and Research on Influenza)で行なった。
シリンジおよび18G針を用いて頸静脈から採取された血液サンプル(50mL)を5000U/mLのヘパリン100μLが入った50mLチューブに入れた。該細胞が壊れないように800gで20分間遠心分離し、バフィーコートを15mLチューブに入れ、PBSで8mLになるよう希釈した。トランスファーピペットを用いてフイコール−パック(Ficoll−paque)の3.5mLを細胞懸だく液の底に加えて、そして、サンプルが壊れないように1000gで30分間遠心分離した。次に、末梢単核球細胞は、フイコール−PBSの界面から採取され、PBSで洗浄し、完全培地(10%FCS、L−グルタミン、ペニシリン(100U/mL)、ストレプトマイシン(100μg/mL)および50μMの2−メルカプトエタノールを補充したダルベッコ 修正イーグル培地)に5×106/mLで再度懸だくした。
抗原再刺激アッセイに擱いて、細胞100μL(5×105/ウェル)を96ウェル組織培養プレートに分注した。該プレートには、培地のみ、または、A/NEW Cledoniaインフルエンザ抗原を10または20μg/mL含む培地いずれかを3連で100μL加えた(最終濃度5または10μg/mL抗原)。加湿インキュベーターにて37℃で5日間培養した後に、すべてのウェルを1μCiのトリチウム化チミジン20μLでパルスラベルした。細胞は、パッカードハーベスターを用いてグラスファイバフィルタの上に播種した。そして、細胞はマイクロスキント(Microscint)シンチレーション液とカセットに回収され、β放射線を自動マイクロプレートスキントカウンターを用いて測定した。
上部肺への送達、および下部肺への送達による免疫付与の有効性の評価は、以下に説明するように行なった。図1(Nickelら、1979)は、上部および下部肺への送達に用いられたヒツジ肺の場所(部位)を示す。
ワクチンは「上部肺」に送達した。−ワクチンは、ファイバ光学気管支鏡を用いて上部肺に送達された。ワクチンは、主気管分岐点の1cm先の主気管支に導入された。
ワクチンは、「下部肺」に送達した。−ワクチンは、ファイバ光学気管支鏡をもちいて下葉に深く送達された。ワクチンは、尾部気管支(caudal bronchus)が尾部区気管支(caudal segmental bronchi)とつながる10cm先の気管分岐点に導入された。
免疫付与、採血、BAL採取およびELISAアッセイは、ほかの実験で説明されるように行なった。ヒツジは、100μgISCOMATRIXTMアジュバンドに0.04μgインフルエンザ抗原の3回投与した。
肺内経路への免疫付与による抗体誘導
3回の別々の実験で、ヒツジに肺内経路によって免疫した。採血は、1回(°)投与から2週間後、そして、2回および3回投与から1週間後に行なわれた。
実験1 ヒツジに100μgISCOMATRIXTMアジュバンドを加えた抗原をそれぞれ1、5または15μg免疫した。3回免疫付与は、3週間間隔をあけて行なった。血清サンプルは、始まる前、および、それぞれワクチン接種した後採取した。
肺内に送達されたアジュバントインフルエンザ抗原の1μgは15μgと同様に有効であるという驚くべき発見に続いて、2番目の実験(実験2)では、抗原投与量のさらなる減少について調べた。
実験2 ヒツジに100μgISCOMATRIXTMアジュバンドを加えた抗原をそれぞれ0.04、0.2、1、5または15μg免疫した。3回免疫付与は、3週間別々に行なった。血清サンプルは、始まる前、および、それぞれ免疫付与した後採取した。
これらの実験ですべての抗原投与量(たとえ0.04μg抗原ほど低くても)が受容性動物に十分な抗体応答を誘導することが示された。低い抗原投与量は1回目の投与後ごくわずかな抗体を誘導する。しかしながら、受容性動物での免疫は、次の2および3回目の投与後は、高い抗原投与量の免疫と同様である。これらの抗体応答は、機能的に血球凝集抑制(HAI)を含む。
0.04μg抗原および100μgISCOMATRIXTMアジュバンドを有するヒツジ免疫化肺内における血清IgGおよびIgAレベルは、抗原単独で15μg皮下免疫付与して得られたものと同様であった(現在のワクチン投与量)。低い投与量の肺内免疫付与は、肺における特異的IgAおよびIgGの非常によいレベルをつくりだす(BAL−気管支肺胞洗浄)。低い投与量の肺内免疫付与は、皮下注入で高い投与量による誘導より優れている。
第3の実験により:
(a)第1、2による知見が再現された。
(b)アジュバントがとても低い抗原投与量に対して免疫の誘導に有効であることが示された。
(c)肺内にアジュバント添加した低い抗原投与量を送達することは、皮下に送達された同じ抗原の低い投与量ワクチンまたは現在のワクチン投与量とアジュバント添加と同様な血清抗体および優れた肺抗体を誘導することが見出された。
(d)皮下注入したとても低い投与量の抗原を2回以上投与するとトレランスを誘導することを示すようであり、または、第3の投与後の血清抗体としての阻害効果は減少したようである。この阻害は、以下の肺送達では起こらない。
(e)100μgISCOMATRIXTMアジュバントと0.04μg抗原とから誘導されるより少ないが、100μgISCOMATRIXTMアジュバントと0.008μg抗原でさえ肺内免疫付与は十分な抗体誘導を誘導したことが分かった。
実験1〜3の結合データは、図2および表1にまとめた。

これらの結果は、3回の投与の後、上部肺への送達は血清とBAL応答を誘導したけれども、IgGおよびIgA双方の血清およびBAL双方の有意によい応答は、下部肺送達によって誘導されたことを示した。
100μgISCOMATRIXTMアジュバントとインフルエンザウイルス抗原とを用いて肺内免疫付与することで、全身性および粘膜の双方の抗体誘導においてHAI活性を伴う高い有効性が証明された。
強い血清抗体応答は、抗原の極めて低いレベル(100μgISCOMATRIXTMアジュバントおよび0.04μgインフルエンザ抗原)でさえ観測することができる。これらの応答は、インフルエンザ抗原単独で15μgの皮下注入(現行のワクチンと同様に)によって誘導される応答よりも強いものであり、ISCOMATRIXTMアジュバントと皮下0.04μgインフルエンザ抗原とによって誘導される応答と同等であった。
ISCOMATRIXTMアジュバントと0.04μgインフルエンザ抗原との肺内送達によって誘導される粘膜抗体応答は、皮下にインフルエンザ抗原15μg注入する場合、または皮下にISCOMATRIXTMアジュバントとインフルエンザ抗原0.04μgとを注入する場合よりも、高く評価された。後者のいずれもが、検出できる粘膜(肺)応答を誘導しなかった。
≪肺内経路を通じたワクチンによる細胞免疫誘導≫
ヒツジは、0.04μgインフルエンザ抗原単独、または0.04μgインフルエンザ+100μgISCOMATRIXTMアジュバントのいずれかで4回肺内免疫付与された。最後の免疫付与の後1週間後、末梢血を採取し、培地のみ(ネガティブコントロール)で、または5または10μgインフルエンザ抗原と一緒に(再刺激)単細胞を96ウェルプレートで培養した。5日後、ウェルは、24時間トリチル化チミジンでパルスした。その際、放射線を測定して、細胞増殖を数えた。刺激インデックスは、培地単独のコントロール群の平均cpmで再刺激された群の1分間あたりの平均カウント(cpm)を割ることで、計算された。
細胞増殖調査の結果は、表3に示す。ヒツジを抗原単独で肺内にワクチン接種した場合、末梢血において増殖応答は検出することができなかった。しかしながら、そのような応答は、ISCOMATRIXTMアジュバントと抗原とを受けたヒツジにおいては、検出された。これらのデータは、アジュバントの必要性、次に肺ワクチン送達することで、末梢循環における細胞増殖記憶応答を誘導することが示された。
≪微小流動化水中油アジュバントを含む肺内経路による投与のための抗原処方物≫
好ましい濃度の抗原(結果として0.01〜500μgの範囲の単位用量を生成する最終抗原濃度)は、0.05%アジ化ナトリウム、pH7.2のPBS中における5%v/vスクアラン、0.5v/vトゥイーン80、0.5%v/vスパン85と混合した。次に、この混合液は、1平方インチ(6.452cm2)あたり(psi)19〜20,000ポンド(8.618〜9071.849kg)の圧力で高圧流動化を行なった。この処方物は、上述された方法による肺内経路を通じて送達された。
≪ISCOMATRIXTMアジュバントと処方した組換え型サイトメガロウイルスgB抗原で肺内経路を通じた免疫付与によって誘導された全身性および粘膜免疫≫
インフルエンザ抗原とISCOMATRIXTMアジュバントから得られた結果を確証し、拡張するために、組換えウイルス抗原の肺内送達によって誘導された免疫応答をヒツジで調べて研究した。ISCOMATRIXTMアジュバントと処方されたサイトメガロウイルスgBグリコタンパク質の短縮形態を肺内、および皮下経路で送達し、EIAによって血清および肺における抗体応答を評価した。加えて、機能的抗体の誘導を、血清におけるCMV中性抗体アッセイで評価した。肺内にT細胞応答の誘導をもたらした処方の能力は、末梢血単細胞のgB特異的細胞増殖を評価することでも調べられた。
短縮組換えヒトCMVgBタンパク質(△gB)の製造
1.CMV△gBタンパク質をコードしたDNAを含むプラスミドの作製
タンパク質の膜貫通領域を取り除いて設計されたCMVgBの短縮形態をコードしたDNAをオーストラリアのブリズベンのクイーンズランドメディカルリサーチ研究所の准教授ラージ ハーナ(Rajiv Khanna)からいただいた。このDNAは、gBの細胞外、および細胞質内ドメイン(NCBI P06473)がインフレームで融合した組織プラスミノーゲン活性化因子(TPA)のシグナル配列を含むタンパク質をコードしている。なお、そのDNAは、発現タンパク質の開裂を防止するためのアミノ酸460/461周辺の領域をコードしたヌクレオチドにポイントミューテーションを導入した。そのDNAを、KpnI/NotIフラグメントとしてpCEP4ベクターにクローン化した。得られたプラスミド(pCEP4△gB)は、大腸菌(E.Coli)で増殖させ、キアゲン(Qiagen)マキシキットを用いて精製プラスミドを調製した。
振とうフラスコにおいて37℃でFreeStyleTM発現培地(Expression Medium)中で増殖させたFreeStyleTM293−Fの培養物を293フェクチントランスフェクションリージェント(インビトロゲン)を用いてpCEP4△gBでトランスフェクションした。培養中ハイグロマイシンBを用いてセレクションをかけたタンパク質△gBの分泌発現は細胞上澄みサンプルのSDS−PAGEゲル分析で決定した。細胞培養上澄みを採取し、2500rpmで遠心分離機によって浄化した。次に、0.45μmフィルターを通した後にクロマトグラフィにかけた。
gB特異的モノクローナル抗体58−15(シング(Singh)とコンプトン(Compton)、2000)で捕獲するアフィニティークロマトグラフィを用いて△gBタンパク質を精製した。抗体は、Prosep−A(ミリポア)タンパク質Aカラムで精製した。△gBタンパク質の精製のため、精製された抗体を、平衡バッファー(0.2M NaHCO3/0.5M NaCl,pH8.5)中でHiTrap NHS−活性化HPカラムとカップリングした。浄化した細胞培養上澄みは、100mMホウ酸塩/150mM NaCl pH8.5で、1:1に希釈した。希釈した上澄みを、HiTrapカラムにアプライし、平衡バッファーで平衡化し、そして、まず、0.1Mグリシン−HCl pH2.5、続いて平衡バッファーで溶出し、今度は、0.15M水酸化アンモニウムで再び溶出した。溶出した画分は、3MトリスpH8または1MグリシンpH2.5のいずれかを用いて中和させた。タンパク質精製はクーマシー−ストレインSDS−PAGEゲル電気泳動法で調べた(図5)。タンパク質濃度は、280nmのODを決定することで測定した。
血液採取
方法は、実施例1で説明した方法と同じとした。
免疫付与
ヒツジは、ISCOMATRIXTMアジュバントと処方された△gB
タンパク質とで免疫付与された(表4)。サイトメガロウイルスの△gBタンパク質は上述したように調製した。
すべてのヒツジは、3回ワクチン投与を受けた。血液採取スケジュールを含む詳細は、実施例1で説明したとおりである。
BALの採取は、実施例1で説明したとおり行なった。
EIAによる抗体応答の評価
ヒツジIgGおよびIgAを検出する方法は、以下の例外を含めて実施例1の方法と同様とした。プレート(96ウェル)に、1ウェルあたり100μlの2μg/mL精製△gBタンパク質を含むPBSで一晩コートした。固定したヒツジIgGは、ホースラディッシュペルオキシダーゼと結合したウサギ抗ヒツジ全IgG(H+L)を用いて検出した。IgGおよびIgA双方の場合において、TMBペルオキシダーゼ基質は、それぞれKPLキークガード(Kirkegaard)およびペリー(Perry)ラボラトリーズから供給された。1ウェルあたり、0.5MのH2SO4を50μl添加することで反応をとめた。
機能的血清抗体を誘導するワクチンの能力は、MRC−5細胞の組換えヒトサイトメガロウイルス感染を基礎とした中和アッセイを用いて評価した。アッセイは、オーストラリアのブリズベンのクイーンズランドメディカルリサーチ研究所の准教授ラージ ハーナ(Rajiv Khanna)と同業者とが、緑色蛍光タンパク質を発現する組換えCMVAd169wt86−EGFPを用いて行なった(マースチェル(Marschall)ら、2000;ワング(Wang)ら、2004)。血清希釈物にCMVAd169wt86−EGFPをさらした後にヒト繊維芽細胞(MRC−5細胞)に感染するウイルスの能力をフローサイトメトリー(FACS)を用いて細胞核蛍光の割合を定量化することで評価した。
2. ヒツジの血清(ストレート、100μlDMEMの2倍、4倍希釈物)を96ウェルトレイに分注した。
3. 25μl(2.5×104PFU)Ad169wt86−EGFPウイルスをそれぞれの血清サンプルに添加した。
4. 血清/ウイルスサンプルを空気中7%CO2の雰囲気で2時間37℃インキュベートした。
5. あらかじめ、空気中7%CO2の雰囲気において2時間37℃でインキュベートした10%胎児ウシ血清(DMEM−10)を含むDMEMで生育させたMRC−5細胞を含む48ウェル平板底トレイを準備し、使用するときまで細胞密集度80〜90%とした。
6. 細胞培養培地をMRC−5細胞から取り除いた。その際、それぞれのウイルス/血清サンプル50μlを、MRC−5細胞を含む48ウェル平板底トレイにおいて対応するウェルに移動させた。50μlDMEMを加え、48ウェルトレイはゆるやかにゆすって空気中7%中CO2の雰囲気で2時間37℃インキュベートした。
7. 接種物は、48ウェルトレイから吸引され、ウェルは3回DMEM−10で洗浄し、その後それぞれのウェルに500μlのDMEM−10を添加し、空気中7%中CO2の雰囲気で一晩37℃インキュベートした。
8. 翌日、細胞培地と細胞(トリプシンを使用)とを48ウェルトレイから集め、遠心分離によって細胞ペレット化した。
9. ペレットは、PBSに再懸だくし、そして、再度遠心分離によってペレット化した。
10. 1mg/mLの7−アミノクチンマイシン(Aminoctinmycin)Dを0.2μl含む低張バッファー(0.1%クエン酸ナトリウム、0.1%(v/v)トリトンX−100)100μlに、ペレットを激しく再懸だくし、暗所室温で20分間静置した。
11. それぞれのペレットのサンプルは、FACS分析に適したチューブに移され、1%パラホルムアルデヒドを含む100μlのPBSを添加した。混合物は、FACS分析の前30分間氷上で静置させた。
12. サンプルは、ベクトンディッキンソンFACSCantoで分析し、サンプル中の蛍光核の割合を定量化した。
13. それぞれのサンプルの中和百分率は以下の式を用いて計算した。

シリンジおよび18G針を用いて頸静脈から採取された血液サンプル(50mL)を5000U/mLのヘパリン100μLが入った50mLチューブに入れた。該細胞が壊れないように800gで20分間遠心分離し、バフィーコートを15mLチューブに入れ、PBSで8mLになるよう希釈した。トランスファーピペットを用いてフイコール−パック(Ficoll−paque)の3.5mLを細胞懸だく液の底に加えて、そして、サンプルが壊れないように1000gで30分間遠心分離した。その際、末梢単核球細胞は、フイコール−PBSの接触面から採取し、PBSで洗浄し、完全培地(10%FCS、L−グルタミン、ペニシリン(100U/mL)、ストレプトマイシン(100μg/mL)および50μMの2−メルカプトエタノールを補充したダルベッコ 修正イーグル培地)に5×106/mLで再度懸だくした。
抗原再刺激アッセイのため、細胞100μL(5×105/ウェル)を96ウェル組織培養プレートに分注した。該プレートには、培地のみ、または、gBタンパク質を5、10、20または40μg/mL含む培地いずれかを3連で100μL加えた(最終濃度2.5、5、10または20μg/mL)。加湿インキュベーターにて37℃で5日間培養した後に、すべてのウェルを1μCiのトリチウム化チミジン20μLでパルスした。細胞は、パッカードハーベスターを用いてグラスファイバフィルタの上に播種した。そして、細胞をマイクロスキント(Microscint)シンチレーション液とともにカセットに設置し、β放射線を自動マイクロプレートスキントカウンターを用いて測定した。
ISCOMATRIXTMアジュバントと処方した組換えCMV△gB抗原を肺内および皮下に免疫付与して起こる免疫応答
ヒツジは、3週間間隔で皮下または肺内にISCOMATRIXTMアジュバントと処方されたCMV△gB抗原を3回投与量受けた。血清とBALサンプルは、第1投与の1週間前、第2投与の2週間後、ならびに第2および第3投与の1週間後に採取された。抗体EIAデータにおいて、着床前の抗体タイターは、差し引いた。
ISCOMATRIXTMアジュバントと処方された△gBにおけるこれらの実験の結果(図6)は、概して、ISCOMATRIXTMアジュバントと処方された低用量インフルエンザ抗原(0.04μg)と実施例2と同様であった。肺内におけるIgGおよびIgA応答は、3回目の投与後、皮下送達における応答と同様であった。しかしながら、インフルエンザ抗原処方から得られた結果と比較して、肺内経路で送達されたISCOMATRIXTMアジュバントと処方された△gBは、血清IgGおよびIgAを2回投与のみののちの皮下送達と同様に誘導した。肺内経路で送達されたISCOMATRIXTMアジュバントと処方された△gBは、1回投与量後の応答を調べたとき、有意に高い血清IgGを誘導した(図6aおよびb)。
機能的抗体誘導の場合、血清におけるCMV中和アッセイの結果は、EIAにおいて得られる血清IgG応答の結果と合致した(図7)。ISCOMATRIXTMアジュバントと処方された△gB処方の肺内および皮下の送達の中和抗体応答は、十分に着床前の血清と比較して有意に高かった。中和応答は、3回目の投与の後さらに増加した。この結果は、ISCOMATRIXTMアジュバントと処方された抗原は、肺内および皮下経路によって送達されたとき、中和抗体の誘導における同等性を示している。
肺内を通じたさまざまなアジュバント技術によるワクチン接種によって誘導される全身性および粘膜免疫
これらの実験は、ISCOMATRIXTMアジュバントよりも他のアジュバントが肺内経路による送達をされたときに抗原特異的粘膜および全身性抗体応答を誘導することができるかどうかを調べるために行なわれた。ヒツジ群は、肺内経路で、さまざまなアジュバント処方物と一緒に処方されたインフルエンザ抗原で免疫付与した(表5)。
アジュバント処方物の調製
A.リポソームは以下の方法によって調製した。:
バッファー溶液:
IRバッファー: 0.14M NaCl, 3mM KCl, 8mM
Na2HPO4, 0.05mM CaCl2.2H2O, 1.5mM K
H2PO4 pH7.2
IVDバッファー: 0.14M NaCl, 3.5mM Na2HP
O4, 1.4mM NaH2PO4.2H2O pH7.2
2.Chol/DPPCは、IVDバッファーを30分以上かけて徐々に添加することで1/40に希釈した。
3.希釈溶液は、ST 200 Diafiltration Cartridge (Nephral)を用いてはじめの薄められた体積の1/40に濃縮した。
4.濃縮溶液は、IVDバッファーの25倍体積で限外ろ過することで洗浄した。
5.Chol/DPPCは、さらに限外ろ過することで、2倍に濃縮した。
6.脂質押出しに先立ちChol/DPPCは、少なくとも1時間40℃でインキュベートした。
7.手順どおり、42℃に保った10mLサーモバーレルエクストラダー(Thermobarrel Extruder)を装備した脂質押出し機(T.001 Northern Lipids Inc.,カナダ)を用いてChol/DPPCからリポソームを押出した。
8.1400−2000kPaの圧力を用いて0.1μm PC ディスクフィルターを通してChol/DPPCの10mLロットで押出しを行なった。
9.Chol/DPPC溶液すべてがこの系を10回供するまで、この工程を繰り返した。
10.結果物のリポソームは、コレステロールとDPPC内容物について測定し、そして、ネガティブ染色を用いて電子顕微鏡下で大きさと均質性とを評価した。
脂質Aモノホスホリル(MPL)(Sigma L6895)は、2mg/mLでDMSO(Sigma D2650)に再懸だくされ、使用するまで4℃で保った。
CpG (Sigma−Genosys 1062 4856−011)をpH8の10mM Tris/1mM EDTAに再懸だくし、使用するまで4℃で保った。
すべてのアジュバント処方物は、インフルエンザ抗原の添加前の希釈としてPBSを用いた関連組成物を混合して調製した(表5)。このあとに、リポソームを含む処方物を1/8インチ(0.3175cm)プローブを用いてセッティング4にてVirSonicソニケーターで超音波処理にさらした。処方物は、30秒間空けて5秒2回の超音波処理を施し、すべての操作は氷上で行なった。
実施例1で説明した方法を用いて表5に示すように、ヒツジの群は、インフルエンザ抗原とアジュバントとの処方物で免疫付与し、採血した。
BAL採取
BALの採取は、実施例1で説明したとおり行なった。
ELISAによる抗体応答の評価
血清中およびBALサンプル中のインフルエンザ抗原への抗体応答のアッセイは、実施例1で説明したとおり行なった。
血球凝集抑制の評価
血清中およびBALサンプル中のHAI活性アッセイは、実施例1で説明したとおり行なった。
さまざまなアジュバントとインフルエンザ抗原との処方物の肺内ワクチン接種後における免疫応答の評価
ヒツジは、3週間の間隔で3回投与量を受けた。血清と肺(BAL)サンプルとは、最初の投与量の前と2週間後、そして2回目および3回目の投与量の1週間後に採取した。抗体ELISAデータにおいて、免疫付与前抗体タイターは差し引いた。
これらの実験結果において、さまざまなアジュバント処方物は、アジュバントが抗原と併用され、肺内経路によって送達されたとき、抗体応答を誘導する能力は多様であることが示された(図9および図10)。
特に注目すべきであるのは、2回投与量の後の血清中および3回投与量の後のBAL中双方におけるISCOMATRIXTMアジュバント処方によって、機能的(HAI)抗体の誘導が示されたデータである(図10)。そのデータ(図9)からは、抗原とISCOMATRIXTMアジュバント+MPLとを含む処方物で免疫されたヒツジに1回および2回投与ののち、それぞれ血清IgGおよびIgA抗体においてアジュバント単独に比較して有意な増加が生じたという点でISCOMATRIXTMアジュバントへMPLを添加することによる長所も示される。

Claims (21)
- ヒトまたは動物被検体の免疫応答を誘発または誘導する方法であって、
抗原およびアジュバントを含む組成物を前記被検体に投与することを含み、前記組成物は、肺内経路によって被検体に投与される方法。 - 組成物が、肺内経路用に処方されている請求項1に記載の方法。
- 組成物が、エアロゾルまたは乾燥粉末フォームである請求項2に記載の方法。
- 組成物が、前記被検体に経口投与で送達される請求項1に記載の方法。
- 組成物が、前記被検体の下部肺に送達される請求項1に記載の方法。
- 組成物が、免疫刺激アジュバントを含む請求項1に記載の方法。
- 組成物が、サポニンベースアジュバント、リポソーム、水中油型アジュバント、アルミニウム塩アジュバント、リポポリサッカライドアジュバント、オリゴヌクレオチドアジュバントおよび粘膜アジュバントからなる群から選ばれる少なくとも1つのアジュバントを含む請求項6に記載の方法。
- 組成物が、免疫刺激複合体を含む請求項6に記載の方法。
- 組成物が、イスコマトリックスアジュバントを含む請求項8に記載の方法。
- 組成物が、ほかの免疫刺激アジュバントと組み合わされた免疫刺激複合体を含む請求項6に記載の方法。

- 前記抗原が、粘膜病原体に対する免疫応答を誘発または誘導する抗原である請求項1に記載の方法。
- 前記粘膜病原体が、インフルエンザウイルス、クラミジア肺炎病原体、呼吸器合胞体ウイルス、または、肺炎球菌である請求項13に記載の方法。
- 前記抗原が、非粘膜部の病原体に対する免疫応答を誘発または誘導する抗原である請求項1に記載の方法。
- 前記病原体が、ピロリ菌、サルモネラ菌、大腸菌、コレラ、HIV、または、性感染症生物である請求項15に記載の方法。
- 前記抗原が、腫瘍特異的、または腫瘍関連抗原である請求項1に記載の方法。
- 前記腫瘍が、粘膜部と関連する請求項17に記載の方法。
- 前記腫瘍が、肺腫瘍、消化管の腫瘍、または、生殖管腫瘍である請求項18に記載の方法。
- 被検体に免疫応答を誘発または誘導するためのヒトまたは動物被検体に肺内投与における抗原およびアジュバントを含む組成物の使用、または、
被検体に免疫応答を誘発または誘導するためのヒトまたは動物被検体に肺内投与用の医薬製造のための抗原およびアジュバントを含む組成物の使用。 - 肺内経路によってヒトまたは動物被検体の免疫応答を誘発または誘導するための医薬品であって、
