Abstract
【解決手段】特定なビスアリールアミン誘導体と下記一般式(2)で表されるボロン酸誘導体との炭素―炭素結合形成反応において、パラジウム化合物とtert−ブチル基、アダマンチル基、シクロヘキシル基のうち少なくとも一つを置換基として含むホスフィン化合物からなる触媒を用いること、及び特定の塩基を使用することによって、脱ボロン酸反応、ホモカップリング反応、炭素−窒素結合形成反応などの副反応を抑制しつつ、一工程で、下記一般式(3)で表されるビスアリールアミン誘導体を高収率、高純度で製造する方法。

Description
アリールハライドとボロン酸もしくはボロン酸誘導体との反応はスズキカップリングとしてよく知られており、炭素―炭素結合形成の重要な手段の一つとして用いられている。中でもアリール−アリール間の結合形成において、その収率の高さ、汎用性、温和な条件、使用する試薬の安定性の高さなどから、最もよく使用される反応の一つとなっている(例えば、非特許文献1および2参照)。
窒素雰囲気下、反応容器にビス(4−ブロモフェニル)アミン6.5g(20ミリモル)、フェニルボロン酸6.3g(52ミリモル)、リン酸カリウム12.7g(60ミリモル)を仕込み、アルゴンガス雰囲気下への置換を行った。酢酸パラジウム(II)0.05g(0.2ミリモル)、ブチル−ジ(1−アダマンチル)ホスフィン0.3g(0.8ミリモル)、予めアルゴンガスを充分通気したトルエン80mlを仕込み、100℃に加熱して3時間反応を行った。反応液についてHPLC分析(装置:島津製作所製LC−2010型、カラム:ジーエルサイエンス製Ineril ODS−3 4.6×250mm、流速:0.8ml/min、カラム温度:40℃、検出波長:254nm、溶離液:アセトニトリル/水=95/5(v/v))を行ったところ、HPLCピーク面積比でビス(ビフェニル−4−イル)アミン92.9%、原料であるビス(4−ブロモフェニル−アミン)0%、原料であるビス(4−ブロモフェニル−アミン)の脱ブロモ体6.4%であった。反応液を室温まで冷却した後、不溶分をろ別して、灰色の固形物を得た。得られた固形物からTHF抽出を行った後、THF/メタノールの混合溶媒で晶析による精製を行うことによって、ビス(ビフェニル−4−イル)アミン4.5gを得た。(収率71%)
実施例1において、パラジウム化合物とホスフィン化合物からなる触媒をテトラキス(トリフェニルホスフィン)パラジウム(0)に、塩基を1M−炭酸カリウム水溶液に、溶媒をトルエン/エタノール=4/1(v/v)に、反応温度を73℃に、反応時間を4.5時間にそれぞれ代えた以外は実施例1と同様に反応を行った。反応液について、実施例1と同じ条件でHPLC分析を行ったところ、HPLCピーク面積比でビス(ビフェニル−4−イル)アミン34.8%、原料であるビス(4−ブロモフェニル−アミン)0%、原料であるビス(4−ブロモフェニル−アミン)の脱ブロモ体62.8%であった。
実施例1において、ホスフィン化合物をトリス(2−メチルフェニル)ホスフィンに、塩基をトリエチルアミンに、溶媒をジメチルホルムアミドに、反応温度を105℃に、反応時間を15時間にそれぞれ代えた以外は実施例1と同様に反応を行った。反応液について、実施例1と同じ条件でHPLC分析を行ったところ、HPLCピーク面積比でビス(ビフェニル−4−イル)アミン32.2%、原料であるビス(4−ブロモフェニル−アミン)21.0%、原料であるビス(4−ブロモフェニル−アミン)の脱ブロモ体25.9%であった。
Claims (6)
- 下記一般式(1)で表されるビスアリールアミン誘導体と下記一般式(2)で表されるボロン酸誘導体を原料とし、パラジウム化合物とtert−ブチル基、アダマンチル基、シクロヘキシル基のうち少なくとも一つを置換基として含むホスフィン化合物からなる触媒を使用し、塩基と溶媒を共存させて炭素―炭素結合形成反応を行うことを特徴とする下記一般式(3)で表されるビスアリールアミン誘導体の製造方法。
- 前記tert−ブチル基、アダマンチル基、シクロヘキシル基のうち少なくとも一つを置換基として含むホスフィン化合物が、tert−ブチル基、アダマンチル基、シクロヘキシル基のうち少なくとも一つを置換基として含むトリアルキルホスフィン化合物であることを特徴とする請求項1記載のビスアリールアミン誘導体の製造方法。
- 前記tert−ブチル基、アダマンチル基、シクロヘキシル基のうち少なくとも一つを置換基として含むホスフィン化合物が、ブチル−ジ−1−アダマンチルホスフィン、ビス−tert−ブチル−1−アダマンチルホスフィン、トリスシクロヘキシルホスフィンまたはトリ−tert−ブチルホスフィンであることを特徴とする請求項1または請求項2記載のビスアリールアミン誘導体の製造方法。
- 前記一般式(1)において、Ar1がフェニレン基である、請求項1〜3記載のビスアリールアミン誘導体の製造方法。
- 前記した塩基が炭酸アルカリ金属塩類、リン酸アルカリ金属塩類、水酸化アルカリ金属類、水酸化アルカリ土類金属類、フッ化アルカリ金属塩類または有機塩基であることを特徴とする請求項1〜4記載のビスアリールアミン誘導体の製造方法。
- 前記した塩基が、リン酸カリウム、炭酸カリウムまたは有機塩基であることを特徴とする請求項5記載のビスアリールアミン誘導体の製造方法。
