Abstract
Description
1.発明の分野
本発明は、ボツリヌス毒素(BoNT)などの神経毒の、軽鎖(LC)として知られている活性部分である一部即ちフラグメントの治療的に効果的な量を標的細胞膜の透過化により送達し、体内の種々の部位で臨床的有益性を生じさせることを含む自律神経機能制御の方法および装置に関する。
本発明によると、ボツリヌス神経毒(BoNT)などの神経毒の活性グラグメント(触媒部分と呼ばれる場合もある)、好ましくはBoNTの軽鎖(LC)部分は、細胞膜の透過化により標的細胞に送達される。そのような送達は、様々な臨床的有益性を引き出すための無毒送達スキームを提供する。ボツリヌス毒素は種々の臨床的用途に広く使用されているが、本発明は、改良された送達方法および該毒素の単離された活性フラグメントまたは単離された活性部分の使用を提供する。本発明の方法およびシステムで使用してもよい他の神経毒には、リシンおよびその活性フラグメント、外毒素Aおよびその活性フラグメント、ジフテリア毒素およびその活性フラグメント、コレラ毒素およびその活性フラグメント、破傷風毒素およびその活性フラグメント等が挙げられる。
本発明は、神経毒のフラグメントがその標的細胞に無毒で送達された時点でその触媒効果または毒性効果を依然として維持しながら、神経毒のフラグメントの無毒送達を標的に向ける方法および装置に関する。本明細書の目的のために、「無毒の」、「無毒で」等の用語は、標的位置への送達前のフラグメント分子の状態を指す。本明細書では、フラグメント神経毒は、その触媒環境即ち標的細胞の細胞内基質即ち細胞質ゾル、に送達された時点でその毒性効果を留めていることが意図されている。
Claims (53)
- 哺乳類細胞の標的部位の神経原性機能により影響を受ける状態を治療するための方法であって、
活性神経毒フラグメントを該標的部位の細胞に送達するステップと、
細胞膜に可逆ポレーションを引き起こして該活性フラグメントの送達を可能にする条件下で、該細胞にエネルギーを付与するステップと
を含む、方法。 - 前記活性フラグメントがボツリヌス毒素フラグメント、リシンフラグメント、外毒素Aフラグメント、ジフテリア毒素フラグメント、コレラ毒素フラグメント、および破傷風毒素フラグメントからなる群から選択される、請求項1に記載の方法。
- 前記活性フラグメントが前記ボツリヌスの軽鎖(BoNT−LC)を含む、請求項2に記載の方法。
- 前記活性フラグメントが、前記神経原性機能に関連する神経原性経路を阻害するために治療的に効果的な量のBoNT−LCを含む、請求項3に記載の方法。
- 前記活性フラグメントが、前記ボツリヌス毒素A、B、C、D、E、F、またはGの少なくとも1つから誘導されたBoNT−LCである、請求項4に記載の方法。
- 前記神経原性経路が喘息の少なくとも1つの症状に関連し、その一因となる、請求項1に記載の方法。
- 送達するステップが、カテーテルを近傍に前進させること、および該カテーテル上のアプリケータからエネルギーを付与しながら前記活性神経毒フラグメントを該カテーテルから放出することを含む、請求項1に記載の方法。

細胞膜のポレーションを引き起こして該毒素の送達を増強する条件下で、該標的細胞にエネルギーを付与するためのエネルギー・アプリケータと、
該カテーテルからの導入に適した毒素源と
