Abstract
【解決手段】β−ケト化合物類を、2,3−ビス(ジアルキルホスフィノ)ピラジン誘導体を配位子として有する遷移金属錯体化合物からなる触媒の存在下に不斉水素化反応に付することを特徴とする光学活性β−ヒドロキシカルボン酸誘導体の製造方法。ピラジン誘導体がキノキサリン誘導体であり、遷移金属がルテニウムであることが好ましい。キノキサリン誘導体としては、(S,S)−2,3−ビス(tert−ブチルメチルホスフィノ)キノキサリン、(R,R)−ビス(tert−ブチルメチルホスフィノ)キノキサリン、(S,S)−2,3−ビス(アダマンチルメチルホスフィノ)キノキサリン、(R,R)−ビス(アダマンチルメチルホスフィノ)キノキサリンなどが好ましい。

Description
(S)−3−ヒドロキシ−3−フェニルプロピオン酸エチルの合成
(R,R)−t−Bu−QuinoxP*(1)(4.9mg、15μmol)と[RuCl2(η6−C6H6)]2(3.4mg、6.7μmol)を、乾燥し脱気したDMF(0.5mL)に窒素雰囲気下に溶解させた。混合物を100℃で10分加熱した。50℃まで冷やし、溶媒を減圧留去し、赤〜紫色の固体として触媒を得た。この触媒をグローブボックス内で脱気エタノール(3mL)に溶解させた。この溶液に、3−オキソ−3−フェニルプロピオン酸エチル(129mg、0.67mmol)のジクロロメタン(1mL)溶液を添加し、混合物をステンレスのオートクレーブに移した。オートクレーブ内を水素で4回パージした後に水素圧を20atmとした。オートクレーブ内を50℃で24時間撹拌し、室温まで冷やした後に水素を排気した。溶媒を除去し、残留物をジエチルエーテルに溶解させた。この溶液を水、次いで食塩水で洗い、有機層を無水硫酸ナトリウムで乾燥した。揮発物を留去後、残留物をシリカゲルカラムクロマトグラフィーにて精製した。このようにして、目的物である(S)−3−ヒドロキシ−3−フェニルプロピオン酸エチルを合成した。
基質、水素圧、温度など種々変更した以外は実施例1と同様に合成を行った。結果を実施例1の結果と併せてまとめて表1に示した。
