Description
[1]炭素原子数が4〜24のケトール脂肪酸の、カルボニル基を構成する炭素原子と水酸基が結合した炭素原子が、α位の位置にある、炭素原子数が4〜24のケトール脂肪酸をストレス曝露後の菌子体に適用する工程を含んで成るキノコ類の栽培方法。
[2]菌床栽培における菌掻き工程によりストレス曝露される、[1]に記載のキノコ類の栽培方法。
[3]適用工程が菌掻き工程終了後から原基が形成するまでの間に実施される、[1]又は[2]に記載のキノコ類の栽培方法。
[4]適用工程が菌掻き工程終了後から24時間以内に実施される、[1]〜[3]のいずれか1つに記載のキノコ類の栽培方法。
コメ胚芽350gを石油エーテルで洗浄、脱脂及び乾燥したもの(250g)を、0.1M酢酸緩衝液(pH4.5)1.25Lに懸濁し、この懸濁物をホモジナイズした。
アマ種子は一丸ファルコスから購入した。このアマ種子200g に、アセトン250mlを添加してホモジナイズ(20s ×3)し、得られた沈澱物を目皿ロートで濾取し、溶媒を除去した。次いで、沈澱物を再びアセトン250mlに懸濁してホモジナイズ(10s ×3)し、沈澱物を得た。沈澱物をアセトン及びエチルエーテルで洗浄後、乾燥して、アマ種子のアセトン性粉を得た(150g )。
出発原料とするα−リノレン酸は、水における溶解性が著しく低いので、酵素基質として働くことを容易にするために、α−リノレン酸をナトリウム塩化した。すなわち、炭酸ナトリウム530mgを、精製水10mlに溶解して55℃に加温し、これにα−リノレン酸(ナカライテスク社)を278mg滴下して、3時間攪拌した。反応終了後、Dawex50wxk(H+ form) (ダウケミカル社製)で中和すると、沈澱物が生成した。これを濾過して樹脂を除き、MeOHで洗浄後、減圧下で溶媒を留去した。これにより得られた生成物をイソプロパノールで再結晶し、所望するα−リノレン酸のナトリウム塩(1250mg,83%)を得た。
上記3により得られたα−リノレン酸のナトリウム塩(15mg:50μmol )を、0.1Mのリン酸緩衝液(pH7.0)30mlに溶解した。次いで、この溶液に、酸素気流下、25℃で上記1により得たコメ胚芽由来のリポキシゲナーゼの粗酵素液を3.18ml添加した後、30分間攪拌した後、さらに同じくコメ胚芽由来のリポキシゲナーゼの粗酵素液を3.18mlを添加して、30分間攪拌した。この攪拌終了後、このリポキシゲナーゼ反応物に、窒素気流下で上記2で得たアレンオキサイドシンターゼの粗酵素液を34.5ml添加して、1時間攪拌した後、氷冷下希塩酸を添加して、反応溶液のpHを3.0に調整した。
上記特定ケトール脂肪酸は、所望の濃度となるように純水中で適宜希釈することにより噴霧剤へと調製される。
ヒラタケとブナシメジを用いて、特定ケトール脂肪酸を栽培期間のうち、いつ噴霧するのがもっとも効果的であるかを調べた。上記噴霧剤中、9−ヒドロキシ−10−オキソ−12(Z),15(Z)−オクタデカジエン酸の濃度は、カーネーションに対する特定ケトール脂肪酸の実験において本発明者が使用した濃度に従い、30ppmとした(横山峯幸、植物の生長調節 40:90-100, 2005)。上記噴霧剤を噴霧しない対照区以外に、菌糸接種後、3、10、14、21、28日後のいずれかに当該噴霧剤を噴霧した。当然のことながら、上記の噴霧時期は菌糸が増殖している時期で子実体は現れていない。
1.ヒラタケの実用生産における9−ヒドロキシ−10−オキソ−12(Z),15(Z)−オクタデカジエン酸の適用例
ヒラタケは温帯の世界各地に分布する木材腐朽菌であり、自然界では主に秋から春にかけて広葉樹の枯れ木や倒木、切り株などに多数重なり合って発生する。日本では年間を通じて主に菌床ビンを用いた施設栽培が広く行われている。上述の通り調製した噴霧剤を施設栽培現場で適用し、その作用を調べた。
エリンギはもともとアフリカ北部、中央アジア、ヨーロッパに自生するきのこであるが、近年、人気が出て日本でも盛んに施設栽培されるようになってきた。そこで上述のヒラタケの培養と同様な培養法で生育したエリンギの菌糸について菌掻きを行った後、菌掻き直後の菌糸に対し上記噴霧剤を噴霧した。試験に用いた本数はいずれの試験区とも60本であった。図4から明らかなように9−ヒドロキシ−10−オキソ−12(Z),15(Z)−オクタデカジエン酸は0.2ppmの時に有意な生産量増加効果を示した。また、エリンギについては生産量が増大するだけでなく、子実体培養期間が10日から8日に早まった。これは、きのこの周年栽培を考えた場合、栽培回数を増やすことが出来るので、このような観点からも栽培の効率化を図ることができるものである。
きのこの菌糸体培養は主に健康食品用のきのこで汎用されている方法である。菌糸増殖における特定ケトール脂肪酸の作用性を検討した。ヒラタケ、ウスヒラタケ、エリンギ、シイタケ、ナメコ、マイタケ、ハタケシメジの菌糸をSMY培地(1%ショ糖、1%麦芽エキス、0.4%酵母エキス)を20mL入れた100mlトールビーカーに植えつけた。9−ヒドロキシ−10−オキソ−12(Z),15(Z)−オクタデカジエン酸は無菌水に30nM、100nM、130pMを溶解させ、菌糸植え付け後に上部からシャーレ当たり0.1mLずつ噴霧した。100nMは0.03ppmにあたる。26℃で1ヶ月培養した後、菌糸体だけを取り出し、重量を測定した結果を図5に示した。
