Description
ここに、本発明者等は副作用がなく成長を効果的に促進し得る活性を有する食品を開発する為に長年研究した結果、哺乳動物の初乳又は乳汁由来の乳清から分離した、1KD〜30KDの分子量を有する乳清分離分画が成長を促進させる活性を有することを確認することにより、本発明を完成した。
本発明による食品組成物に含まれる乳清分離分画は、哺乳動物の乳清から分離されたものであって、α−ラクトアルブミン及びβ−ラクトグロブリンが除去され、1KD〜30KDの分子量を有することを特徴とする。
(b)前記(a)段階の乳清からα−ラクトアルブミンとβ−ラクトグロブリンを除去する段階;及び
(c)前記(b)段階のα−ラクトアルブミン及びβ−ラクトグロブリンが除去された乳清から、1KD〜30KDの分子量を有する分画を分離して収得する段階を含む方法により製造できる。
(ii)前記(i)段階の脱脂乳のpHを4〜5に調整してカゼインを凝固させ、遠心分離して上澄液を収得する段階でなされたものが好ましい。
(ii)前記(i)段階で収得した上澄液のpHを4.5〜7.0に調整し、β−ラクトグロブリンを沈澱させ、上澄液を収得する段階でなされた塩析法により行われる。
さらに、前記にて投与は経口投与が好ましい。
前記乳清分離分画は前記に記載されたような方法で製造することができる。
但し、下記実施例は本発明を例示するものにして、本発明の内容がこれに限定されるものではない。
本発明の乳清分離分画の製造
分娩後48時間の間分泌されるウシの初乳を収集後遠心分離して、クリーム層を除去することにより、脂肪が除去された脱脂初乳を製造した。前記脱脂初乳に1N塩酸を添加してpHを4.6に調整することにより、カゼインを凝固させ、遠心分離して上澄液である初乳乳清を収得した。分離された乳清に1.5Mクエン酸三ナトリウム(trisodium citrate)を添加してpHを5.5に調整し、さらに、1.5Mクエン酸(citric acid)を添加してpHを3.9に調整して、35℃で150分間加熱し、α−ラクトアルブミンを沈澱させた。これを遠心分離してα−ラクトアルブミンを除去して上澄液を回収した。上澄液に1N NaOHを添加してpH4.5に調整することにより、β−ラクトグロブリンを沈澱させ遠心分離して上澄液を収得した。収得した上澄液を30KD限外濾過膜を通過させ、濾過液を得た後前記濾過液を再度1KD限外濾過膜を通過させて残留物を得た。前記残留物を凍結乾燥して粉末状態の1KD乃至30KDの分子量を有する乳清分離分画を収得した。
動物モデルを利用した乳清分離分画の成長促進効果調査
<1−1>乳清分離分画の供給
成長期のSD(Sparague−Dawley)雄白鼠(3週齢、体重50〜65g)28匹を1週間の間適応させ、7匹ずつ4グループに分けて実験に使用した。実験の際、飼育条件としては、温度23±1℃、湿度40〜60%及び明暗周期を12時間とした。適応期間である1週間は一般食餌と水を制限なく供給し、実験期間である6週間は各実験群別に製造された食餌を水と共に制限なく供給した。
2)310025 AIN−VX ビタミンミキス(vitamin mix)
実験例<1−1>の各グループの食餌投与前体重と6週間食餌投与後、体重を測定した。測定値は統計処理して平均値と標準偏差を計算し、測定された体重から下記式を利用して体重増加量を計算した。
平均体重増加量=(実験完了時最終平均体重−実験開始時平均体重)
対照群と各実験群間の有意性検定はスチューデントT−テスト(Student T−test)を利用して統計処理した後、信頼区間(P value)が0.05より小さい場合、統計学的に有意な意味があるものと判定した。
**P<0.01において有意な差がある。
前記実施例<1−1>で6週間の実験が完了した後、本発明の分離分画を摂取させた実験群及び対照群の左右大腿骨を摘出して、骨組織に付着している筋肉、脂肪、靭帯等を全て除去し、75℃の乾燥天火で3時間乾燥した後大腿骨の長さを測定した。

本発明の乳清分離分画がIGF−Iの分泌を促進し、成長を促進させる効果があるか否かを確認する為に、前記実験例<1−1>において6週間の実験を完了した対照群及び実験群の血液を採取し、OCTEIA Rat IGF−1キット(IDS Co., USA)を使用して血清内IGF−Iの濃度を測定した。

本発明の乳清分離分画が成長ホルモンの分泌を促進して成長を促進させる効果があるか否かを確認する為、前記実験例<1−1>で6週間の実験を完了した実験群及び対照群の血液を採取し、DSL−1900 ACTIVEキット(DSL Co.,USA)を利用して血清内成長ホルモン濃度をIRMA(immuno radio metric assay)法で測定した。

乳清分離分画を含む成長促進用食品組成物の製造
実施例1で製造した乳清分離分画を含む成長促進用食品組成物を、下記表6のような組成比で配合して通常の方法により打錠及びコーティングして錠剤に製造した。
Claims (14)
- 哺乳動物の乳清から分離され、α−ラクトアルブミン及びβ−ラクトグロブリンが除去された1KD〜30KDの分子量を有する乳清分離分画を含む成長促進用食品組成物。
- 前記乳清分離分画が(a)哺乳動物の乳汁から脂肪とカゼインを除去して乳清を収得する段階;
(b)前記(a)段階の乳清からα−ラクトアルブミンとβ−ラクトグロブリンを除去する段階;及び
(c)前記(b)段階のα−ラクトアルブミン及びβ−ラクトグロブリンが除去された乳清から1KD〜30KDの分子量を有する分画を分離して収得する段階を含む方法により製造されたものであることを特徴とする第1項記載の食品組成物。 - 前記(a)段階が(i)乳汁を遠心分離し、クリーム層を除去して脱脂乳を収得する段階;及び
(ii)前記(i)段階の脱脂乳のpHを4〜5に調整してカゼインを凝固させ、遠心分離して上澄液を収得する段階からなることを特徴とする第2項記載の食品組成物。 - 前記哺乳動物の乳汁がウシの初乳であることを特徴とする第2項又は第3項記載の食品組成物。
- 前記(b)段階のα−ラクトアルブミンとβ−ラクトグロブリンの除去が、電気泳動法、塩析法及びイオン交換クロマトグラフィー法からなる群より選ばれた方法により行われるものであることを特徴とする第2項記載の食品組成物。
- 前記(b)段階のα−ラクトアルブミンとβ−ラクトグロブリンの除去が、
(i)前記(a)段階の脂肪とカゼインが除去された乳清のpHを5.0〜7.0に調整し、pHを再度2.5〜4.2に調整することにより、α−ラクトアルブミンを沈澱させ、上澄液を収得する段階;及び
(ii)前記(i)段階の上澄液のpHを、4.5〜7.0に調整してβ−ラクトグロブリンを沈澱させ、上澄液を収得する段階からなる塩析法により行うことを特徴とする第2項記載の食品組成物。 - 前記(c)段階における1KD〜30KDの分子量を有する分画の分離が、限外濾過又はゲル濾過方法を利用して行うものであることを特徴とする第2項記載の食品組成物。
- 前記(c)段階で収得された1KD〜30KDの分子量を有する分画を乾燥して粉末化する段階を追加して含むことを特徴とする第2項記載の食品組成物。
- 第1項の乳清分離分画0.1〜50重量%、食用乳清カルシウム0.5〜60重量%、アルコキシグリセロール粉末0.01〜30重量%、レシチン粉末0.1〜80重量%、乳蛋白加水分解物粉末0.01〜35重量%、クロレラ抽出物粉末0.01〜20重量%、フラクトオリゴ糖0.1〜50重量%、ヘム鉄0.01〜35重量%、ビタミン類0.001〜20重量%及び乳糖0.1〜99重量%からなるものであることを特徴とする成長促進用食品組成物。
- 第1項の食品組成物を、それらを必要とする哺乳動物に有効量で投与することを含む成長促進方法。
- 前記成長促進が、体重増加、骨の長さ増加、内在性IGF−I水準の増加及び内在性成長ホルモン水準の増加によるものであることを特徴と第10項記載の方法。
- 前記投与が、経口投与であることを特徴とする第10項記載の方法。
- 前記哺乳動物が、ヒトであることを特徴とする第10項記載の方法。
- 第1項乃至第9項のいずれか1項に記載の成長促進用食品組成物を製造する為の、哺乳動物の乳清から分離され、α−ラクトアルブミン及びβ−ラクトグロブリンが除去された1KD〜30KDの分子量を有する乳清分離分画の用途。
