Description
SLLMWITQCペプチドは、一連の腫瘍により発現されるNY-ESO-1タンパク質に由来する(Chenら, (1997) PNAS USA 94 1914〜1918)。癌性細胞のクラスI HLA分子は、SLLMWITQCを含むこのタンパク質からのペプチドを提示する。よって、SLLMWITQC-HLA-A2複合体は、例えば癌細胞に細胞毒性作用剤又は免疫増強剤を送達する目的のために、TCRが標的できる癌マーカーを提供する。しかし、この目的のためには、TCRが、天然型TCRがペプチド-HLA複合体に対して有するよりも、より高い親和性及び/又はより遅い解離速度を有することが望ましい。
本発明は、SLLMWITQC-HLA-A*0201複合体について1μM以下の高い相互作用の親和性(KD)及び/又は1×10-3 S-1又はそれより遅い、より遅い解離速度(koff)を有するTCRをはじめて利用可能とする。このようなTCRは、該複合体を提示する癌細胞の標的のために、単独で又は治療剤と併用して、有用である。
本発明は、SLLMWITQC-HLA-A*0201複合体についてのKDが1μMに等しいか又はそれより低く、及び/又はSLLMWITQC-HLA-A*0201複合体についての解離速度(koff)が1×10-3 S-1又はそれより遅いことを特徴とする、SLLMWITQC-HLA-A*0201への結合特性を有しかつ少なくとも1つのTCRα鎖可変ドメイン及び/又は少なくとも1つのTCRβ鎖可変ドメインを含むT細胞受容体(TCR)に関する。KD及び/又は(koff)の測定は、いずれの既知の方法により行うことができる。好ましい方法は、実施例5の表面プラズモン共鳴法(Biacore)である。
SLLMWITQC-HLA-A*0201複合体に特異的な天然型1G4 TCRは、次のVα鎖及びVβ鎖遺伝子使用(usage)を有する。

のその相補性決定領域(CDR)及び/又は可変ドメイン枠組み構造領域において変異されたTCRを含む。本発明のTCRの可変ドメインのその他の超可変領域、例えば超可変領域4 (HV4)を、高親和性変異体を産生するために変異させることも想定される。

多価TCR複合体の作製において用い得る多量体化ドメインを含むいくつかのヒトタンパク質が存在する。例えば、モノマーscFVフラグメントに比較して血清残存率が増大し、解離速度が大幅に低減されたscFv抗体フラグメントのテトラマーを作製するのに用いられるp53の四量体化ドメイン(Willudaら (2001) J. Biol. Chem. 276 (17) 14385〜14392)。ヘモグロビンも、この種の適用に用い得る可能性がある四量体化ドメインを有する。
ある態様において、本発明のTCRは、治療剤又は検出可能な部分(detectable moiety)と結合できる。例えば、上記の治療剤又は検出可能な部分は、TCRと共有的に連結できる。
本発明のある実施形態において、上記の治療剤又は検出可能な部分は、一方又は両方のTCR鎖のC-末端に共有的に連結する。

Structural biology 7 463〜469)は、タンパク質中に新規な結合部位を作り出すための骨格の使用のレビューを提供する。このレビューは、次のタンパク質を骨格の起源として言及している:CP1ジンクフィンガー、Tendamistat、Zドメイン(プロテインAのアナログ)、PST1、高次コイル、LACI-D1及びシトクロムb562。その他のタンパク質骨格の研究は、フィブロネクチン、緑色蛍光タンパク質(GFP)及びアンキリン反復の使用を報告している。
癌の治療のために、腫瘍又は転移の近傍での局在化は、トキシン又は免疫増強剤の効果を増強する。ワクチンの送達のために、ワクチン抗原を抗原提示細胞の近傍に局在化させることができ、よって、抗原の有効性を増大させることができる。上記の方法は、撮像目的にも適用できる。
本発明のscTCR又はdTCR (ヒト配列に対応する定常配列及び可変配列で構成されるのが好ましい)は、実質的に純粋な形で、又は精製若しくは単離された調製物として提供できる。例えば、これは、その他のタンパク質を実質的に含まない形で提供できる。
(a) 1G4 TCRのα及びβ鎖可変ドメインを含むTCRを作製し、ここでα及びβ鎖可変ドメインの一方又は両方は、請求項7及び8で同定される1又は複数のアミノ酸に変異を含み;
(b) 上記の変異TCRを、SLLMWITQC-HLA-A*0201と、TCRのSLLMWITQC-HLA-A*0201への結合を可能にする適切な条件下で接触させ、
相互作用のKD及び/又はkoffを測定する
ことを含む。

RNA単離
全RNAを、10000のクローン性T細胞から、100μlのTRI Reagent (Sigma)に再懸濁し、溶解物を製造業者の使用説明に従って処理することにより単離した。最後の沈殿の後に、RNAを12.5μlのRNアーゼフリーの水に再溶解した。
上記のRNAのサンプルに、2.5μlの10 mM オリゴ-dT15 (Promega)を加え、サンプルを60℃で2分間インキュベートし、次いで氷上に置いた。OmniscriptRTキット(Qiagen)を用いて、2μlのRTバッファー(10×)、2μlの5 mM dNTP、1μlのOmniscript逆転写酵素を加えることにより逆転写を行った。サンプルを混合し、37℃で1時間インキュベートした。次いで、cDNAを-80℃で貯蔵した。
α鎖精製PCR産物をClaI及びSalIIで消化し、ClaI 及びXhoIで切断したpEX954 (図9を参照)にライゲーションした。
β鎖精製PCR産物をAseI及びAgeIで消化し、NdeI/AgeIで切断したpEX821 (図10を参照)にライゲーションした。
切断したPCR産物及び切断したベクターは、製造業者の使用説明に従ってRapid DNA Ligationキット(Roche)を用いてライゲーションした。
ライゲーションしたプラスミドを、コンピテント大腸菌XL1-blue株の細胞に形質転換し、100mg/mlアンピシリン含有LB/アガープレートに播種した。37℃で一晩インキュベーションした後に、単独のコロニーを採取し、100mg/mlアンピシリン含有10 ml LB中で振とうしながら37℃で一晩増殖させた。クローン化されたプラスミドを、Miniprepキット(Qiagen)を用いて精製し、挿入断片を、自動DNAシーケンサー(Lark Technologies)を用いて配列決定した。
図5a及び5bは、それぞれ、図4a及び4bのDNA配列から産生されるNY-ESO TCRα及びβ鎖細胞外アミノ酸配列を示す。
実施例1で記載されるようにして作製した可溶性ジスルフィド結合連結天然型1G4 TCRは、SLLMWITQC (配列番号125) -HLA-A*0201複合体に対する親和性が増大した本発明のTCRを産生するための鋳型として用いることができる。
geneからのQuickChange (商標) Site-Directed Mutagenesis Kit)。
簡単に、このことは、所望のコドン変更を組み込むプライマーと、突然変異誘発の鋳型として適切な1G4 TCR鎖を含有するプラスミドとを用いることにより達成される。

構築物は、PCRステッチング(stitching)により作製した。

得られたPCR産物を、制限酵素AseI及びXmaIを用いて消化し、NdeI及びXmaIで切断したpEX205にライゲーションした。
実施例1、2又は3のようにしてそれぞれ作製した変異α鎖及びβ鎖を含む発現プラスミドを、大腸菌BL21pLysS株に別々に形質転換し、単独アンピシリン耐性コロニーを37℃にてTYP (アンピシリン100μg/ml)培地で0.4のOD600まで増殖させ、その後、0.5mM IPTGを用いてタンパク質の発現を誘導した。誘導の3時間後に、Beckman J-6B中で、4000rpmにて30分間の遠心分離により細胞を回収した。細胞ペレットを、50mM Tris-HCl、25% (w/v)スクロース、1mM NaEDTA、0.1% (w/v) アジ化Na、10mM DTT、pH 8.0を含有するバッファーに再懸濁した。一晩の凍結−融解工程の後に、再懸濁した細胞を、標準的な直径12mmのプローブを用いるMilsonix XL2020超音波破砕機で合計約10分間、1分間のバーストにて超音波破砕した。インクルージョンボディのペレットを、Beckman J2-21遠心分離機で13000rpmにて30分間の遠心分離により回収した。3回の洗浄剤での洗浄を行って細胞破片及び膜成分を除去した。各回においてインクルージョンボディのペレットをTritonバッファー(50mM Tris-HCl、0.5% Triton-X100、200mM NaCl、10mM NaEDTA、0.1% (w/v) アジ化Na、2mM DTT、pH 8.0)中でホモジナイズし、その後、Beckman J2-21で13000rpmにて15分間の遠心分離によりペレット化した。洗浄剤及び塩を、次のバッファー中での同様の洗浄により除去した:50mM Tris-HCl、1mM NaEDTA、0.1% (w/v) アジ化Na、2mM DTT、pH 8.0。最後に、インクルージョンボディを30 mgの一定量に分配し、-70℃で凍結した。インクルージョンボディタンパク質収率は、6M グアニジン-HClでの可溶化及びBradford 色素結合アッセイ(PerBio)での測定により定量した。
オン交換カラムにロードし、結合したタンパク質を、Akta Purifier (Pharmacia)を用いて50カラム容量の0〜500mM NaClのグラジエントで溶出した。ピークフラクションを4℃にて貯蔵し、クーマシー染色SDS-PAGEで分析し、その後、プールして濃縮した。最後に、sTCRを精製して、HBS-EPバッファー(10 mM HEPES pH 7.4、150 mM NaCl、3.5 mM EDTA、0.05% Nonidet p40)で予め平衡化したSuperdex 200HRゲルろ過カラムを用いて特性決定した。約50 kDaの相対分子量で溶出するピークをプールし、濃縮し、その後、BIAcore表面プラズモン共鳴分析法により特性決定した。
表面プラズモン共鳴バイオセンサ(Biacore 3000(商標))を用いて、sTCRのそのペプチド-MHCリガンドへの結合を分析した。このことは、半配向された様式(semi-oriented fashion)でストレプトアビジン被覆結合表面に固定化された単一pMHC複合体(以下に記載)を作製することにより促進され、同時に4つまでの異なるpMHC (異なるフローセルに固定化された)への可溶性T細胞受容体の結合を効率的に試験することを可能にした。HLA複合体を手動で注入することにより、固定化されたクラスI分子のレベルを簡便に精密に処理することが可能となる。
薬混合物をロードし、カラムを、PBSを用いて0.5 ml/分にて展開した。ビオチン化pHLA-A*0201分子を、約15 mlにてシングルピークとして溶出した。タンパク質含有フラクションをプールし、氷上で冷却し、プロテアーゼ阻害剤カクテルを添加した。タンパク質濃度を、クーマシー結合アッセイ(PerBio)を用いて決定し、ビオチン化pHLA-A*0201分子の一定量を-20℃で凍結保存した。ストレプトアビジンを標準的なアミンカップリング法により固定化した。
WT 1G4 sTCRの段階希釈を調製し、2つの異なるフローセルに、5μl min-1の一定の流速で注入した。該フローセルの一方は約1000 RUの特異的SLLMWITQC-HLA-A*0201複合体で被覆され、第二のものは約1000 RUの非特異的HLA-A2-ペプチド複合体で被覆されていた。コントロールセルからの測定を用いて、各濃度についての応答を標準化した。標準化したデータ応答を、TCRサンプルの濃度に対してプロットし、平衡結合定数KDを測定するために、双曲線に適合させた(Price & Dwek, Principles and Problems in Physical Chemistry
for Biochemists (第2版) 1979, Clarendon Press, Oxford)。
高親和性TCRについて、KDを、解離速度定数kd及び会合速度定数kaを実験的に測定することにより決定した。平衡定数KDは、kd/kaとして算出された。
TCRを、一方が約300 RUの特異的HLA-A2-nyesoペプチド複合体で被覆され、第二のものが約300 RUの非特異的HLA-A2-ペプチド複合体で被覆された2つの異なるセルに注入した。流速は、50μl/分に設定した。典型的には、250μlのTCRを約3μMの濃度で注入した。応答がベースラインに戻るまでバッファーを流した。動力学的パラメータは、Biaevaluationソフトウェアを用いて算出した。解離相を、半減期の計算を可能にする指数減少等式(single exponential decay equation)にも調和させた。
可溶性ジスルフィド連結天然型1G4 TCR (それぞれ配列番号9及び10に示されるα及びβTCR鎖からなる)とSLLMWITQC-HLA-A*0201複合体との間の相互作用は、上記の方法を用いて分析し、15μMのKD及び1.28×10-1 S-1のkoffを示した。
次の表に記載するTCRは、1μM以下のKD及び1×10-3 S-1又はそれより遅いkoffを有する。
T2リンパ芽球細胞にNY-ESO由来SLLMWITQC、NY-ESO-アナログSLLMWITQVペプチド又は無関係のペプチドを、一連の濃度(10-5〜10-10M)で180分間、37℃にて適用した。NY-ESO-アナログSLLMWITQVペプチド(V-変異形ペプチド)を用いた。なぜなら、このペプチドは、天然型NY-ESO由来SLLMWITQCペプチドよりも高い親和性をHLA-A*0201複合体の結合裂溝(binding cleft)について有することが知られているからである。適用の後に、細胞を、血清フリーRPMIで洗浄し、5×105細胞を高親和性c58c61 NY-ESO TCR- IL-2融合タンパク質と10分間、室温にてインキュベートし、その後、PE (Serotec)とコンジュゲートした二次抗IL-2 mAbと15分間、室温にてインキュベートした。洗浄後、結合したTCR-IL-2を、FACSVantage SE (Becton Dickinson)を用いるフローサイトメトリにより定量した。ペプチドを適用したT2細胞を用いるコントロールも含めたが、TCR-IL-2は省略した。

R10アッセイ培地:10% FCS (熱不活化, Gibco, cat# 10108-165)、88% RPMI 1640 (Gibco, cat# 42401-018)、1%グルタミン(Gibco, cat# 25030-024)及び1%ペニシリン/ストレプトマイシン(Gibco, cat# 15070-063)。
ペプチド:(種々の起源から入手) 最初にDMSO (Sigma, cat# D2650)中に4mg/mlで溶解し、凍結した。
PBS (Gibco, cat#10010-015)。
50000の標的細胞を、96ウェルELISPOTプレート(Diaclone)のウェル当たり50μlのR10培地中に播種した。
上記の標的細胞培養に、次のものを加えた。
50μlのR10培地中に1×10-7 M高親和性c58c61 TCR、又は無関係のTCR
50μlのR10培地中に600のSLLMWITQC-HLA-A*0201特異的T細胞(クローン1G4)。
これらの培養は、37℃、5%CO2にて、24時間インキュベートした。ELISPOTプレートは製造業者の使用説明に従って処理した。
可溶性高親和性c58c61 1G4 TCRは、IFN-γ産生により測定して、黒色腫細胞に対して、1G4 T細胞クローンの活性化を強く阻害した。一方、無関係の高親和性TCRは阻害効果がなかった(MEL-624癌細胞系の結果について図17を参照。SK-MEL-37 癌細胞系の結果について図18を参照)。
可溶性高親和性c58c61 1G4 TCRがSLLMWITQC-HLA-A*0201特異的CTLクローン(1G4)の活性化を阻害できることを証明するために、次のアッセイを行った。IFN-γ産生を、CTL活性化の読み取り値として用いた。

ELISAプレートは、製造業者の使用説明に従って準備した(Diacloneキット, Immunodiagnostic systems, UK)。
T2細胞系標的細胞を洗浄し、種々の濃度(100nM〜10pM)のSLLMWITQCペプチドの存在下又は不在下でR10培地に再懸濁し、37℃、5%CO2にて1時間インキュベートした。
ウェル当たり10,000の標的細胞を、96ウェルELISAプレートに播種した。
50μlのR10培地中に1×10-6 M〜3×10-12 Mの高親和性c58c61 1G4 TCR又は野生型1G4 TCR。
50μlのR10培地中に5000の1G4エフェクター細胞。
次いで、プレートを37℃、5% CO2にて48時間インキュベートした。次いで、製造業者の使用説明に従ってELISAを行った。
可溶性高親和性c58c61 1G4 TCRは、IFN-γ産生により測定されるように、ペプチド適用標的細胞に対して1G4 T細胞クローンの活性化を強く阻害した。一方、野生型1G4 TCRは、阻害効果を有さなかった(高親和性c58c61 1G4 TCRについては図19を、野生型1G4 TCRについては図20を参照されたい)。
この研究は、実施例6に記載の高親和性c58c61 1G4 TCR-IL-2融合タンパク質の、ヌードマウスに移植されたヒト腫瘍細胞の増殖を阻害する能力を探索するために行った。
50匹の雌性ヌードマウス(HARLAN, France)をこの試験で用いた。
全ての動物に、細胞表面での適切なクラスI-ペプチド標的の発現を増強させるためにNY-ESOペプチド/ユビキチンミニ遺伝子構築物で安定的にトランスフェクションしたヒト黒色腫形成細胞系(SK-MEL-37)を皮下注射した。腫瘍は、動物で5日間増殖させ、処置の開始の前に腫瘍を発生させた。
用量は、PBS中の0.02〜1.0 mg/kgの範囲の高親和性1G4 TCR/IL-2融合タンパク質であり、腫瘍移植後5、6、7、8、11、13、17、20、24、28及び30日に投与した。全ての実験において、コントロール処理群は、PBS単独でTCR/IL-2イムノコンジュゲートを置き換えたものを含んでいた。
次いで、腫瘍のサイズをカリパスを用いて測定し、腫瘍の容積を、次の式:

に従って決定した。
腫瘍増殖阻害についてのTCR/IL-2イムノコンジュゲートの治療効果を図21に示す。
TCR/IL-2イムノコンジュゲートは、図21に示す腫瘍増殖曲線により示されるように、明確な用量依存的抗腫瘍効果を示した。
癌細胞(Mel 526、Mel 624及びSK-Mel-37細胞系)上のいくつかのSLLMWITQC-HLA-A*0201抗原を、高親和性c58c61 1G4 TCRを用いる単一分子蛍光顕微鏡法により決定した(1つの蛍光シグナルは、標的細胞の表面上の同起源のpMHCリガンドに結合した単一の標識TCRと関連するという仮定に基づいて)。このことは、抗原発現癌細胞を標的するビオチン化TCRを用い、その後、細胞に結合したTCRをストレプトアビジン-R フィコエリスリン(PE)コンジュゲートにより標識することにより促進された。個別のPE分子を、次いで、三次元蛍光顕微鏡法により撮像した。
(Zeiss)顕微鏡を用いて行った。300Wキセノンアークランプ(Sutter)を備えたラムダLS光源を照明に用い、光路内に0.3及び0.6の中性濃度フィルタを配置することにより最適レベルまで光強度を下げた。励起及び放射スペクトルを、TRITC/DiIフィルタセット(Chroma)を用いて分離した。細胞を、z-スタック取り込みにより三次元で撮像した(21面、1μm間隔)。像の取得及び分析は、Metamorphソフトウェア(Universal Imaging)を、記載されたようにして行った(Irvineら, Nature (419), p845〜9,及びPurbhooら, Nature Immunology (5), p524〜30)。
図22に示すように、上記の方法により、Mel 526、Mel 624及びSK-Mel-37癌細胞の表面上のSLLMWITQC-HLA-A*0201抗原に結合した高親和性1G4 TCRを撮影することに成功した。

- SLLMWITQC-HLA-A*0201複合体についてのKDが1μM以下であり、及び/又はSLLMWITQC-HLA-A*0201複合体についての解離速度(koff)が1×10-3 S-1又はそれより遅いことを特徴とする、SLLMWITQC-HLA-A*0201への結合特性を有しかつ少なくとも1つのTCRα鎖可変ドメイン及び/又は少なくとも1つのTCRβ鎖可変ドメインを含むT細胞受容体(TCR)。
- α鎖可変ドメインとTCRβ鎖可変ドメインとの両方を含む請求項1に記載のTCR。
- αα又はββホモダイマーである請求項1に記載のTCR。

- 1又は複数の表現型サイレント置換を任意に含む配列番号11〜83に示すα鎖可変ドメインアミノ酸配列の1つを含む請求項1〜6のいずれか1つに記載のTCR。
- 1又は複数の表現型サイレント置換を任意に含む配列番号84〜99及び117〜121に示すβ鎖可変ドメインアミノ酸配列の1つを含む請求項1〜6及び11のいずれか1つに記載のTCR。
- 配列番号49に示すα鎖可変ドメインと配列番号94に示すβ鎖可変ドメインとを含み、1又は複数の表現型サイレント置換を任意に含む請求項2に記載のTCR。
- 配列番号100に示すα鎖定常領域アミノ酸配列、及び/又は配列番号101及び102に示すβ鎖定常領域アミノ酸配列の1つをさらに含み、1又は複数の表現型サイレント置換を任意に含む請求項1〜14のいずれか1つに記載のTCR。
- ダイマーT細胞受容体(dTCR)又は単鎖T細胞受容体(scTCR)である請求項1〜15のいずれか1つに記載のTCR。
- TCRα鎖可変ドメインに相当するアミノ酸配列により構成される第一セグメントと、
TCRβ鎖定常ドメイン細胞外配列に相当するアミノ酸配列のN-末端に融合したTCRβ鎖可変ドメイン配列に相当するアミノ酸配列により構成される第二セグメントと、
第一セグメントのC-末端を第二セグメントのN-末端に連結するリンカー配列と
を含むscTCRである請求項4〜16のいずれか1つに記載のTCR。 - TCRβ鎖可変ドメインに相当するアミノ酸配列により構成される第一セグメントと、
TCRα鎖定常ドメイン細胞外配列に相当するアミノ酸配列のN-末端に融合したTCRα鎖可変ドメイン配列に相当するアミノ酸配列により構成される第二セグメントと、
第一セグメントのC-末端を第二セグメントのN-末端に連結するリンカー配列と
を含むscTCRである請求項4〜16のいずれか1つに記載のTCR。 - 第一の鎖と第二の鎖との間にジスルフィド結合を含み、該ジスルフィド結合が天然型αβT細胞受容体中に等価物を有さず、リンカー配列の長さ及びジスルフィド結合の位置が、第一セグメント及び第二セグメントの可変ドメイン配列が天然型αβT細胞受容体中と実質的に同様に互いに配向されるようなものである請求項17又は18に記載のTCR。
- 結合部分において、リンカー配列が、第一セグメントのC-末端を第二セグメントのN-末
端に連結する請求項17〜19のいずれか1つに記載のscTCR。 - 結合部分において、リンカー配列が式-PGGG-(SGGGG)5-P- (配列番号103)又は-PGGG-(SGGGG)6-P- (配列番号104) (式中、Pはプロリンであり、Gはグリシンであり、Sはセリンである)を有する請求項17〜20のいずれか1つに記載のscTCR。
- TCRα鎖可変ドメイン配列に相当する配列がTCRα鎖定常ドメイン細胞外配列に相当する配列のN-末端に融合した第一ポリペプチドと、
TCRβ鎖可変ドメイン配列に相当する配列がTCRβ鎖定常ドメイン細胞外配列に相当する配列のN-末端に融合した第二ポリペプチドと
を含むdTCRであり、
第一ポリペプチドと第二ポリペプチドとが、天然型αβT細胞受容体中に等価物を有さないジスルフィド結合により連結している請求項1、2及び4〜16のいずれか1つに記載のTCR。 - 前記ジスルフィド結合が前記定常ドメイン配列のアミノ酸配列同士を連結し、前記ジスルフィド結合が天然型TCR中に等価物を有さない請求項22に記載のTCR。
- 前記ジスルフィド結合が、天然型TCRにおいてβ炭素原子が0.6 nm未満離れているアミノ酸残基に相当するシステイン残基同士の間にある請求項23に記載のTCR。
- 前記ジスルフィド結合が、TRAC*01のエキソン1のThr 48から置換されたシステイン残基と、TRBC1*01若しくはTRBC2*01又はそれらの非ヒト等価物のエキソン1のSer 57から置換されたシステイン残基との間にある請求項23に記載のTCR。
- dTCR又はscTCRの結合部分が、天然型TCR中でジスルフィド結合により連結されるものに相当する残基間のジスルフィド結合を含む請求項16〜25のいずれか1つに記載のTCR。
- dTCR又はscTCRの結合部分が、天然型TCRの膜貫通配列又は細胞質配列に相当する配列を含まない請求項14〜24のいずれか1つに記載のTCR。
- 配列番号122のα鎖アミノ酸配列と配列番号123のβ鎖アミノ酸配列とからなる可溶性TCR。
- 配列番号122のα鎖アミノ酸配列と配列番号124のβ鎖アミノ酸配列とからなる可溶性TCR。
- 少なくとも1種のポリアルキレングリコール鎖と結合している請求項1〜29のいずれか1つに記載のTCR。
- 前記ポリアルキレングリコール鎖がTCRと共有的に連結している請求項30に記載のTCR。
- 前記ポリアルキレングリコール鎖が少なくとも2つのポリエチレングリコール反復単位を含む請求項30又は31に記載のTCR。

- 非ペプチドポリマー鎖又はペプチドリンカー配列により連結した請求項1〜42のいずれか1つに記載のTCRの少なくとも2つを含む多価TCR複合体。
- 前記ポリマー鎖又はペプチドリンカー配列が、各TCRの、TCRの可変領域配列中に位置しないアミノ酸残基の間に伸びている請求項43に記載のTCR複合体。
- TCRが、ポリアルキレングリコール鎖又はヒト多量体化ドメイン由来のペプチドリンカーにより連結されている請求項43又は44に記載のTCR複合体。
- 二価アルキレンスペーサー残基が、ポリアルキレングリコール鎖と複合体のTCRへのその結合点との間に位置する請求項45に記載のTCR複合体。

- 癌を罹患している患者に、請求項1〜48のいずれか1つに記載のTCR若しくは多価TCR複合体、又は複数の請求項49に記載の細胞の有効量を投与することを含む癌の治療方法。
- 請求項1〜48のいずれか1つに記載のTCR若しくは多価TCR複合体、又は複数の請求項49に記載の細胞の、癌の治療用組成物の製造における使用。
- TCRが、(i) 少なくとも1つのTCRα鎖可変ドメイン及び/又は少なくとも1つのTCRβ鎖可変ドメインを含み、かつ(ii)前記SLLMWITQC-HLA-A*0201複合体についてのKDが1μM未満であり、及び/又はSLLMWITQC-HLA-A*0201に対する解離速度(koff)が1×10-3未満であり
、
(a) α鎖可変ドメイン及びβ鎖可変ドメインの一方又は両方が請求項7又は8で規定されるアミノ酸の1又は複数において変異を含む、1G4 TCRのα鎖可変ドメイン及びβ鎖可変ドメインを含むTCRを作製し;
(b) 前記変異TCRをSLLMWITQC-HLA-A*0201と、TCRのSLLMWITQC-HLA-A*0201への結合を可能にするのに適する条件下で接触させ、
相互作用のKD及び/又はkoffを測定する
ことを含む、SLLMWITQC-HLA-A*0201への結合特性を有する高親和性TCRを作製する方法。
