Abstract
【解決手段】ウロキナーゼ型プラスミノーゲンアクチベーター遺伝子を導入し、肝臓で該ポリペプチドを発現するヒト-マウスキメラ肝からなり、非ヒトの免疫無防備状態にある異種トランスジェニック動物に基づくモデルからなる。また、トランスジェニック動物を用いて、候補治療薬(例えばHCV感染に対する抗ウイルス活性を有する物質)を同定する方法からなる。

Description
本発明は一般に感染症の分野、特にウイルス性病原体に対するモデルに関する。
C型肝炎ウイルス(HCV)により引き起されるヒトの肝疾患は、この十年間で世界中の医療現場が直面する極めて困難な問題の一つとなっている。1989年にウイルスの配列が明らかにされた(Chooら、Science 244、359〜361 (1989)(非特許文献1))ことで、HCVに対する研究の時代が大きく開かれた。現在、多く見積もって175百万人が感染していると推定されている(Sarbahら、Cell 62、447〜456(1990)(非特許文献2))。HCVは慢性ウイルス性肝炎のなかで最も一般的な型であり、先進国における推定羅患率は1〜2%である。慢性のHCV肝炎は羅患患者の少なくとも25%で肝硬変に至り、肝硬変の発症後、年に1〜4%の割合で患者に肝細胞癌が発症すると推定されている。北米では、現在HCVは肝移植の最も一般的な適応となっている。
本発明は、ヒトの向肝性病原体、特にヒトC型肝炎ウイルス(HCV)の感染に感受性のある非ヒト動物モデルを特徴とする。このモデルは、導入遺伝子が肝臓内でのウロキナーゼ型プラスミノーゲンアクチベーターの発現をもたらす、ヒト-マウスキメラ肝を有する非ヒトの免疫無防備状態のトランスジェニック動物を元にしている。本発明はまた、候補治療薬、例えばHCV感染に対する抗ウイルス作用を有する物質を同定する方法を特徴とする。

本明細書で使用される「キメラ」(例えば「キメラ動物」または「キメラ肝」)は、異種の組織または細胞を含む臓器または動物体を意味する。動物が、動物の肝臓に移植されたヒトの肝細胞が存在するためにキメラであるキメラ動物は特に重要である。
本発明は、C型肝炎ウイルス(HCV)に対するネズミ科動物モデルの開発を基礎としている。このようなネズミ科動物モデルは一般に、トランスジェニックマウスの肝臓への、宿主の適切な発生段階、好ましくは宿主の出生直後、におけるヒト肝細胞の移植を含む。理論に拘束されるわけではないが、モデルの開発が成功するか否かは、少なくとも部分的には以下の条件による:(1)免疫不全の背景を有する宿主を使用することで、導入された異種(ヒト)細胞の、免疫系による攻撃を避けること;(2)ホモ接合状態で存在する、アルブミンプロモーターに結合したウロキナーゼの導入遺伝子を含むトランスジェニック動物を使用することで、肝細胞の成長および細胞の分裂に対する強力な継続的刺激を提供すること;および(3)生育可能なヒト肝細胞を、肝細胞のライフサイクルの適切な時期、および宿主動物の発生の初期段階で宿主動物に導入して、宿主における大多数の、および/または高率のヒト細胞の長期生存をもたらすこと。
宿主動物は一般に、ウロキナーゼ型プラスミノーゲンアクチベーター(uPA)の肝臓における産生が上昇し、ヒト肝細胞の移植または維持が可能な、非ヒトの免疫無防備状態にある哺乳類である。本発明の動物モデルの元となりうる例示的な非ヒト動物には、マウス、ラット、モルモット、ハムスター、ヒツジ、ブタ、霊長類などがあるが必ずしもこれらに限定されることはない。ある態様では、宿主動物はげっ歯類の動物、好ましくはマウスである。好ましい態様では、宿主動物は、免疫無防備状態のマウスであり、好ましくはウロキナーゼ型プラスミノーゲンアクチベーター(uPA)遺伝子が導入された免疫無防備状態のトランスジェニックマウスであり、より好ましくはuPAの肝臓特異的な産生を提供する導入遺伝子(例えば、Alb-uPA導入遺伝子、例えばHeckelら Cell 62: 447 (1990)を参照)を含む免疫無防備状態のマウスである。本発明の用途に適したマウスは、系列C.B-17、C3H、BALB/c、C57131/6、AKR、BA、B10、129などを含むが必ずしもこれらに限定されない任意の多様なバックグラウンドの系統から作製することができる。宿主動物は雄でも雌でもよい。
上述の通り、宿主動物は好ましくは免疫無防備状態にある。移植に適した、および所望の免疫不能状態を有するのに適した免疫無防備状態の哺乳類宿主を利用することができる。あるいは、それほど好ましくはないものの、免疫無防備状態の動物を、免疫応答性の動物から、例えば1種または複数の化合物(例えばシクロスポリン)の投与、または当技術分野で周知の他の方法で作製することができる。一般には、免疫無防備状態の宿主は異種の組織または細胞に対する完全な免疫応答を開始することができない。免疫グロブリンおよびT細胞の抗原受容体をコードする座位において生殖系列のDNA再編成が生じなくなる遺伝的欠損のために免疫無防備状態である動物は特に重要である。また正常動物と比較して、機能性のT細胞、B細胞、およびナチュラルキラー(NK)細胞の数の大きな低下または非検出に至る1種または複数の遺伝的欠損をもつ免疫無防備状態の動物も重要である。

上述の通り、本発明のキメラ動物は、ウロキナーゼ型プラスミノーゲンアクチベーター(uPA)の発現について「ノックイン」トランスジェニック動物でもある。ある態様では、このような導入遺伝子はAlb-uPA導入遺伝子であり、これはネズミ科動物アルブミンエンハンサー/プロモーター、ネズミ科動物uPA遺伝子コード領域、および成長ホルモン遺伝子の3'非翻訳かつ隣接配列から構成される(Heckelら、Cell 62: 447〜56 (1990);Sandgrenら、Cell 66:245〜56 (1991))。好ましくはこのような動物は、ウロキナーゼ型プラスミノーゲンアクチベーター導入遺伝子についてヘテロ接合ではなくホモ接合である。Alb-uPA導入遺伝子は、これをもつ肝細胞に致死的な障害(lethal insult)を生じるほか、ウロキナーゼの濃度を局所的(肝内)に高める。これをきっかけとして肝細胞成長因子が処理されて肝臓内で活性型となる。理論に拘束されるわけではないが、Alb-uPAトランスジェニック動物の発生の適切な時期に導入された生育可能な同種または異種の細胞は、このような環境で刺激されて複製を開始する。したがってドナー細胞は、導入遺伝子による致死的な障害の結果死滅する内因性肝細胞に「置き換わる」ように成長する。
宿主動物に移植するヒト肝細胞は、ヒトの肝臓組織から当技術分野で周知の任意の従来の方法で単離される。一般にヒト肝細胞は、新鮮な組織(例えば、死後数時間以内に得られる組織)、または新鮮な状態で凍結された組織(例えば、新鮮な状態で凍結処理されて約0℃以下で維持されたもの)であることができる。使用する細胞は、ヒトの肝臓組織から新鮮な状態で回収された単離直後(すなわち2〜4時間以内)のものであることが望ましい。特定の凍結保存溶媒中に保持されたヒト肝細胞は、長期にわたって(例えば液体窒素中で)保存することが可能であり、必要に応じて解凍して、保存された肝細胞の発生を可能とすることができる。一般に、単離手法ならびに取り扱いおよび保存のプロトコールは、肝臓に向かう血流の停止による温虚血時間を最短化すること(例えば、通常は約30分〜60分未満、好ましくは約20分〜約40分未満)、および保存に起因する寒冷虚血時間を最短化すること(例えば、一般的には約12時間未満、通常は約1時間〜2時間未満)が通常重要である。ある態様では、ヒト組織は正常である(例えば検出可能な病原体を含まず、形状および組織学的所見が正常であり、実質的に異常が認められない)。通常、暖虚血曝露時間は約20〜50分を超えない。
ドナー肝細胞の免疫無防備状態のトランスジェニック宿主への導入のタイミングは、キメラ肝を向肝性病原体感染に感受性とし、また、病原体の複製を可能とするために十分ないくつかのヒト肝細胞が集合したキメラ肝を作るために重要な場合がある。感染性が低く、および/または複製速度が遅い肝向性病原体(例えばHCV)の場合に特にこれがあてはまる。動物がネズミ科動物(murine)(例えばマウス(mouse))の場合、移植の時期は、宿主が望ましくは10日〜2週齢未満であり、好ましくは約7〜10日齢であり、または約1週齢またはそれ未満(すなわち約5〜7日齢未満)である。一般的には、移植は好ましくは約8〜10日齢と15日齢の間に実施される。移植の時期は、良好な結果を保ちながら柔軟性を得るために約7〜18日齢まで延長可能である。理論に拘束されるわけではないが、本明細書に記載された移植のタイミングは、極めて早い時期の移植に関連する過度の技術面に起因する死亡率(すなわち、動物のサイズが小さいことに起因する)と、最大の複製刺激を得るまでの時間(例えば、移植前に生じるレシピエント肝における細胞分裂の回数がドナーとなるヒト細胞の移植の成功率および程度に影響することがある)との間の妥協の産物である。また移植のタイミングは、導入遺伝子によってもたらされる肝臓細胞の再増殖に及ぼす刺激が時間の経過とともに弱まり、約6週齢以降のレシピエントでは一般に消失してしまうことからも重要である(Rhimら、(1994) Science 263:1149〜52;ホモ接合の場合は約10〜12週)。
本発明のキメラ動物は、多様な他のスクリーニングアッセイ法で使用することができる。例えば肝疾患を引き起したり寄与したりすることが疑われる任意の多様な候補薬剤、ならびに適切な拮抗物質および阻害性治療薬をキメラ動物に投与して、対象薬剤が移植ヒト細胞の機能に及ぼす作用を評価することでスクリーニングすることができる。本発明のキメラ動物モデルを用いて、肝疾患の治療、または任意の肝臓特異的でないヒト疾患の治療に用いる低分子量の分子を用いる療法を含む、肝細胞に対する毒性について化合物のスクリーニングを行うこともできる。一般的には、任意の化合物を投与して、肝細胞に対する毒性を評価することができる。例えば癌に対する重要な推定療法の評価は、本発明の動物モデルにおける肝毒性に対して最初にスクリーニングを行うことができる。移植ヒト肝細胞の機能は、上述の通りに(例えば、宿主の血清に含まれるヒト血清アルブミンまたはα-1アンチトリプシンの量を評価することで)評価可能である。肝細胞の損傷は、血清に含まれる肝臓特異的な酵素(ALT-アラニンアミノトランスフェラーゼ)のアッセイ法と、肝臓中のヒト細胞に対する損傷を明らかにする組織学的評価を組み合わせることで評価することができる。手短に説明すると、肝毒性を評価するアッセイ法は、機能に着目したものでも、組織学変化に着目したものでも、またはその両方であってもよい。
ある態様では、本発明の動物モデルを用いて、ウイルス性肝炎、より具体的にはHCV感染によって引き起された症状を改善する薬剤を同定し、および/または、より直接的には、感染ウイルスの病原性機構に影響を与える。例えば、ウイルス感染を抑制したり、ウイルスの複製を低下させたり、またはそれ以外ではウイルスの増殖サイクルを破壊したりする。一般的には、候補薬剤を本発明の動物モデルに投与し、候補薬剤が及ぼす作用を、対照に対して(例えば、未感染動物に対して、抗HCV作用が知られている薬剤(例えばIL-2α)を投与するHCV感染動物に対して等)評価する。例えば候補薬剤を本発明のHCV感染動物に投与することが可能であり、投与前の動物のウイルス量および/または対照である未投与HCV感染動物のウイルス量と投与動物のウイルス量を(例えば血清試料を対象としたRT-PCR法による測定で)比較することができる。一般に、候補薬剤投与後における感染動物でウイルス量が検出可能な低下、また有意な低下がみられれば、対象薬剤に抗ウイルス作用があることを意味する。
所望の薬理活性を有する化合物を、生理学的に許容される担体に含まれた状態で治療目的で宿主に投与することができる。このような治療薬は、さざまなな方法で、経口的に、局所的に、非経口的に(例えば皮下、腹腔内、血管内、吸入など)投与することができる。導入方法に基づいて、化合物をさまざまな方法で製剤化することができる。製剤化したときに治療的に活性のある化合物の濃度は、約0.1〜100重量%の範囲を変動する場合がある。
一部変更することで、本発明の動物モデルを用いて、候補ワクチンが、向肝性病原体による感染を予防する能力または改善する能力をもつか否かについてスクリーニングを行うこともできる。一般的に「ワクチン」は、投与されることで、標的となる病原体に対する免疫応答を宿主が開始するのを促す物質である。誘発される液性、細胞性、または液性/細胞性の免疫応答は、ワクチン開発対象の病原体による感染抑制を促進可能である。本発明では、向肝性病原体(例えば、細菌病原体、ウイルス性病原体、または寄生虫病原体、特に例えばHCVなどのウイルス性病原体)の肝内での複製および/または向肝性病原体による感染を抑制する、防御的な免疫応答を誘発する予防的ワクチンが特に重要である。受動免疫、または速やかに促進される特異的な能動免疫(例えば、抗HCV免疫グロブリンなど)の供給により防御をもたらす治療用ワクチンも重要である。
上述の、または上述に加えて変形形態である本発明のキメラ動物の用途は、本明細書を読むことで当業者には容易に明らかになる。例えばキメラ動物には、向肝性薬剤を感染、好ましくは慢性的に感染させることが可能であり、単離されうる薬剤の供給源として使用することが可能である。本発明のキメラ動物のこのような用途は、例えば、病原体の単離がヒト検体の生検を必要とする場合、または有用な量の入手が困難な場合、病原体がインビトロで容易に培養できない場合、インビトロにおける病原体の培養(例えば、ブロス培養または細胞培養における成長)が病原性および/または臨床的な意義などに影響を及ぼす可能性のある病原体を変化させる場合に特に有用である。一般的には、単離された病原体を、適切な経路で(例えば静脈内投与、筋肉内投与、腹腔内投与、または経口投与)、好ましくはヒト疾患の天然の感染経路と最も良好に相関する感染経路でキメラ動物に接種する。病原体のヒト肝細胞への感染を確立した後、また、病原体を複製させるのに十分な時間が経過した後に、病原体を感染キメラ動物から適切な方法(例えば、肝臓に由来する血液試料の単離など)で単離する。
キメラ動物はまた、ヒトの肝疾患の診断の一貫として使用することもできる。例えば原因不明の肝疾患患者の場合、または病原体を培養することなく下した診断が確定的でない場合、原因物質を含むことが疑われる試料を、患者から(例えば、患者の血液または肝生検から)単離する場合がある。試料は、疑われる物質について濃縮し、分画し、またそうでない場合は処理して投与可能な状態として提供し、キメラ動物に投与することができる。次にキメラ動物は、試料の投与が、移植されたヒト肝細胞に及ぼす作用を調べるために評価される場合がある。ヒト肝細胞に及ぼす作用は、例えばキメラ動物の血清試料を単離、調査する(例えば移植されたヒト肝細胞の機能を調べることで、および/または、動物の血清に含まれる病原体を検出することで、例えばHCVまたは他の微生物病原体の存在を検出する)ことによって達成することができる。ヒト肝細胞を組織学的に調べて、患者試料の作用を決定することもできる。
本発明を適応させて、個々の患者に基づく診断法および理論的な治療法を提供することもできる。例えば、患者の生検(例えば経皮針生検)で得られたヒト肝細胞を用いてキメラのネズミ科動物宿主を作製することができる。次にこのキメラのネズミ科動物宿主を用いて、患者に感染する向肝性病原体を評価し、治療薬に対する病原体の感受性を調べ、また治療に対する患者の肝細胞の潜在的な毒性を明らかにすることができる。したがって本発明は、個々の特異的な向肝性病原体の相補物に対して(例えば、1種または複数の感染性の向肝性病原体に対して)最も有効な治療法の目的に合わせることを促すように設計することができる。
本発明は、ヒトのアテローム硬化性血管疾患(心血管疾患を含む)の治療法を評価する系として適応させることもできる。アテローム動脈硬化は、西洋社会で心臓病と脳卒中の主要因であり、カナダでは死亡率のほぼ半分を占めるに至っている(Ross (1993) Nature 362:801〜809)。高量の低密度リポタンパク質(LDL)とアテローム動脈硬化との正の相関が明らかにされてから数十年が経過している(Brownら、Ann. Rev. Biochem. 52:223〜261 (1983))。LDLは、ヒドラーゼ、ならびにリポタンパク質内における脂質およびアポタンパク質の転移がかかわる一連の複雑な反応ために、循環系において超低密度リポタンパク質(VLDL)に由来する(Fieldingら、(1996)「脂質、リポタンパク質、および膜の生化学(Biochemistry of Lipids、Lipoproteins and Membranes)」、(D.E. 下記の実施例は、当業者に、本発明の製造および使用の方法の完全な開示および説明を提供するために提示しているが、本発明者が自身の発明とみなす範囲を限定することを意図するものではなく、下記の実施例が全てまたは実行された実験のみを表すことを意図しているものでもない。使用した数値(例えば、量、温度など)に関しては、正確さを確実にするために努力を払っているが、ある程度の実験誤差および偏差は考慮されるべきである。特に記さない限りは、部は質量部であり、分子量は質量平均分子量であり、温度は摂氏であり、かつ圧力は大気圧またはその近傍である。
ヒト組織移植片に対し寛容性があるAlb-uPAトランスジェニックマウスを作出するために、導入遺伝子に関するマウスのヘテロ接合体(TgN(AlblPlau)144Bri系(The Jackson Laboratory社))を、C.b-17/SCID-beige系(C.b-17/GbmsTac-scid-bgN7系(Taconic Farms社)、ホモ接合体)動物と交配した。一連の戻し交配を通じて、当技術分野において周知の方法に従いマウスIgGを検出するためにサンドイッチELISA法を用い、総血清IgGの定量により確認されるような、SCID-beige形質を、ホモ接合状態で繁殖した。IgGの定量は、マウスIgG標準(Cappel社)を用い各プレートについて作成した標準曲線から算出した。SCID-beige形質の「漏出(leakiness)」は、正常血清IgGの>1%と定義し(Bosmaら、Ann. Rev. Immun.、9:323(1991))、このカットオフ値を上回る血清IgGレベルを有する動物は安楽死させた。各工程において、Alb-uPA導入遺伝子を保持する動物は、導入遺伝子構築物の3' UTR由来の151bp産物を増幅する2種の18-merプライマーを使用する、尾部生検標本から抽出したゲノムDNAのPCR分析により同定した(Jackson Laboratorie社技術サポート)。ホモ接合Alb/uPA形質はこれまでに、出血性合併症および肝不全に随伴する高い周産期死亡率と関連づけられてきたが(Heckelら、Cell、62:447(1990))、本発明者らは、自分たちのscid/bg/Alb-uPA動物コロニーにおいて、新生児死亡率が約30%であることを発見した。この動物を提供するコロニーは、最初にヘテロ接合体繁殖業者により開発され、ホモ接合マウスにおいて中程度の新生児死亡率を伴うが、このコロニーにより、完全なホモ接合体が導き出された。動物を、ウイルス/抗原を含まない条件下で飼育し、カナダ動物管理評議会(Canadian Council on Animal Care)により制定された指針(1993)に従い飼育した。本明細書に記された全ての動物実験は、アルバータ大学動物の福祉に関する委員会(University of Alberta Animal Welfare Conunittee)の承認の下で行われた。
2種の異なる血清を基にしたアッセイ法を用い、本発明者らのキメラマウスにおいて、ヒト肝細胞移植片を評価した。第一のアッセイ法は、ヒト血清アルブミンを測定するドットブロットアッセイ法であり;第二のアッセイ法は、ヒトα-1アンチトリプシン(hAAT)を測定するELISAアッセイ法である。同マウスを、移植後6週目および12週目にアッセイした。

ホモ(HOMO)は、動物が導入遺伝子についてホモ接合体であることを意味する;ヘテロ(HETERO)は、動物が導入遺伝子についてヘテロ接合体であることを意味する。両アッセイ法では、概して傾向が同じであることが示され、uPA導入遺伝子について、ホモ接合体において、ヘテロ接合体と比べてより高いヒト由来のタンパク質の産生が示される。
本実施例は、免疫無防備状態のscid/bg/Alb-uPAマウスへのヒト肝細胞の移植が成功したことを明らかにするものである。
最初に成功した生着および増殖の長期にわたる結果を決定するために、8匹の動物の第二同腹子を同様の様式で移植した。この実験時点で利用可能な肝細胞は、B型肝炎ウイルスの慢性キャリヤである患者から入手した。この患者は、陽性血清HBsAgレベルおよび陰性血清HBV DNAの両方を示し、従って、活発なウイルス複製を伴わない慢性キャリヤ状態を示す(Davis、South. Med. J.、90:866(1997))。

キメラマウスにおけるHBV複製を支持するキメラ動物の作出における前述の成功は、HBVモデルとしての該動物の用途を裏付けている。しかし、先に論じた(背景)HBVとHCVの間の大きな差異は、動物モデルが通常の感染経路(例えば、経静脈伝播)を通じHCV感染に対し易感染性であること、または特にHCV動物モデルおよびHCVの珍しい細胞系の開発のための他者の失敗を考慮し、キメラ肝が活動的HCV感染を支持することについては、理にかなった予想はできないことを意味している。HBV動物モデルによるかなりの成功およびHCV動物モデルによる他者の失敗の繰り返しは、HBVからHCVへ単純に外挿することができないことを示している。従って、ウイルスに感染したヒト血清を用い、キメラ肝を有するマウスにおける一次HCV感染の確立を試みた。
本実施例において、本発明の動物モデルはHCV複製を支持することができることを明らかにするために、前記作業を更に拡大した。
下記の方法および材料を、本実施例において使用した。
動物を、ウイルス/抗原非含有条件において飼育し、カナダ動物管理評議会(CCAC)により制定された指針(1993)に従い管理した。動物実験は、アルバータ大学動物の福祉に関する委員会(UAAWC)により承認を受けた。
ヒト組織の使用に関する倫理的承認を、アルバータ大学医学部倫理委員会(UAFMREB)から得て、インフォームドコンセントを、肝細胞ドナー全員から入手した。ヒト肝臓組織のセグメント(15〜20cm3)を、通常は病理試験後には廃棄されるような肝切除標本の領域から入手した。この手術の大部分は、肝内悪性疾患のために行われた。
レシピエント(5〜14日齢)に、ハロタン/O2で麻酔し、左側腹部を小切開した。手術のための拡大操作下で、1x106個の生存肝細胞を27gバタフライ注入セット(Abbott社)により脾下端へ注入し、血流途絶のために、1個の滅菌チタンクリップを注入部位を横断するように配置した。この脾臓を腹部に戻し、かつ側腹部切開を2層で閉鎖した。
マウス血清(20μl)を、モノクローナル抗HA抗体(Clone HSA-9、Sigma社)およびプロテインG-アガロースビーズ(Boehringer-Mannheim社)により収集した抗原-抗体複合体と共にインキュベーションした。還元条件下で、免疫沈降物を、SDS-PAGEにより分離し、かつニトロセルロースに転写した。ウェスタンブロットを、ビオチン化したモノクローナル抗HA抗体(Clone HSA-11、Sigma社)を用いて調製し、シグナル検出のためにストレプトアビジン-HRP複合体および化学発光基質(Pierce社)を用いた。
マウスDNA(3μg)をPvuIIで消化し、0.7%アガロースゲル上でサイズ分画し、Hybond-N+膜(Amersham Life Science社)に転写し、uPA遺伝子の最終イントロン(7312位から7920位、GenBankアクセッション番号M17922)由来の[32P]標識プローブにハイブリダイズした。2.88kbのバンドは、uPA導入遺伝子(T)に由来し、2.53kbのバンドは、内因性uPA遺伝子(E)に由来した。ハイブリダイゼーションは、フジ(Fuji)ホスホイメージャーおよびイメージゲージ(Image Gauge)ソフトウェアにより定量した。
マウス肝生検標本を、10%ホルマリンで固定し、パラフィン包埋した。厚さ5μの切片を、常法によりヘマトキシリン・エオシン(H&E)で染色した。選択した切片を、内因性アビジン/ビオチンブロッキングキット(Zymed Laboratories社)により処理し、モノクローナル抗ヒト肝細胞抗体(DAKO、1:20希釈)で染色し;結合した抗体を、スーパーセンシティブ-イムノディテクションシステム(BioGenex社)を用いて検出した。
マウス血清試料(2μl)を、還元緩衝液40μl中で100℃で5分間インキュベーションし、2μlアリコートを、ニトロセルロース上に3つ組でブロットした。乾燥した膜を、転写緩衝液中に浸し、3%PBS-Tweenでブロッキングし、ウェスタンブロットとして調製した。化学発光を、STORMホスホイメージャーを用いて各ブロットについて調製した標準曲線から定量した。
定量HCV分析を、Alberta Provincial Laboratory of Public Health社(エドモントン、アルバータ、カナダ)、またはCanadian Center for Disease Control社(ウィニペグ、マニトバ、カナダ)により、盲検法で行った。血清試料についての分析は、コバス-アンプリコアHCVモニターシステム(Roche Diagnostics社)を、製造業者の指示に従って行った。
総RNAを、マウス肝生検標本、または感染したヒト血清から、TRIZOL(Gibco BRL社)を用いて単離した。RT-PCRは、熱安定性rTth逆転写酵素RNA PCRキット(Perkin Elmer社)を、製造業者の指示に従って行った。逆転写14について、陽性鎖RNAをアンチセンス

プライマーにより検出した。鎖特異的cDNAを、他のプライマーの添加により増幅し、5'非コード領域(NCR)の240塩基対(bp)領域を標的化し、95℃で30秒、66℃で45秒、および70℃で90秒を35サイクル、その後、70℃で5分間処理した。反応産物を、2%アガロースゲルに負荷し、Hybond-N+ナイロン膜(Amersham Pharmacia Biotech社)に転写し、HCV 5' NCRを、α-32P標識DNAプローブと42℃で一晩ハイブリダイズした。
総RNAを、マウス肝からトリゾル(Trizol)試薬(GIBCO/BRL社)を用い、およびHCVを感染したヒト血清からQIAamp Viral RNAミニキット(Qiagen社)を用いて、各々製造業者のプロトコールに従い単離した。抽出したRNAは、32Pで標識し、ゲル精製したアンチセンスリボプローブ((+)鎖の検出)、センスリボプローブ((-)鎖の検出)、および/またはβ-アクチンアンチセンスリボプローブによりプロービングした。
3種のプラスミド構築物を、切断されたHCV RNAのインビトロ転写のために個別に調製した。HCVPfix/KS+は、HCVセリンプロテイナーゼの発現試験のために本発明者らの実験室において初めて開発された構築物である。HCV感染患者から得た新鮮な血清から調製した(Chomczynskiら、Anal Biochem、162:156-159(1987))総RNAは、95℃で5分間変性した。cDNA合成は、AMVスーパー逆転写酵素(Molecular Genetic Resources社)を反応容量20μlで42℃にて90分間行った。使用したアンチセンスオリゴヌクレオチドプライマーは、
(3'プライマー)であった。PCRは、総容量100μlで行い、かつ最終cDNA反応混合液2μlは、20mM Tris-HCl(pH8.4)、50mM KCl、1.5mM MgCl2、200μMの各デオキシリボヌクレオシド三リン酸、0.5μMの3'プライマーおよび5'プライマー
、および2.5U Taq DNAポリメラーゼ(GIBCO/BRL社)を含んだ。エンテロキナーゼ切断部位を追加した5'プライマー、および全623bp断片を、pBluescript KS+(Stratagene社、PDI)のEco RIおよびSalI部位にクローニングした。
(5'プライマー)および
(3'プライマー)であった。この両方向の245bp産物を、pCR2.1 TOPOへ、市販のTOPO TAクローニングキット(Invitrogen社)を用いてTA-クローニングし、pCR 2.1/NCRセンスプラスミドおよびpCR2.1/NCRアンチセンスプラスミドを作成した。全てのクローンは、DNA配列決定により確認した(University of Alberta DNA Core Facility)。
リボプローブは、T7 RNAポリメラーゼ(Promega社)を製造業者の指示に従い使用し、[32P]UTPの存在下で、インビトロ転写により調製した。(+)鎖HCV RNAの検出については、383-ヌクレオチドのアンチセンスリボプローブを、KpnI消化したpCR 2.1/NCRアンチセンスから転写し、(-)鎖については、636-ヌクレオチドのセンスリボプローブを、SalI消化したHCVPfix/KS+から転写した。β-アクチンの検出のためには、直鎖状にしたpTRI-Actin-マウス(AMBION)を用い、304ヌクレオチドのリボプローブを作出した。HCV RNAの鎖特異的検出の特異性を明らかにするために、更に非放射性(radioinert)センスおよびアンチセンスリボプローブも調製した。簡単に述べると、T7 RNAポリメラーゼを使用するインビトロ転写の前に、pCR2.1/NCRセンス/アンチセンスを、KpnIで消化し、HCVPfix/KS+をSalIで消化した。加えて、HCVPfix/KS+を、EcoRIにより直鎖状にし、T3 RNAポリメラーゼ(Promega社)を用いてインビトロ転写した。全てのインビトロ転写反応は、37℃で1時間行い、その後、RNase非含有DNaseI(RQl DNase、Promega社)により37℃で30分間処理した。標識したリボプローブをゲル精製し、非放射性HCV RNAリボプローブは、0.5容量の7.5M酢酸アンモニウムを添加後に、無水エタノール2.5容量で沈殿させた。インビトロ転写したRNAにおける非放射性の強度は、1%アガロースゲルを使う電気泳動により評価した。RNA試料を変性し、一晩42℃でハイブリダイズし、およびRNaseプロテクションアッセイキット(AMBION RPA III Kit)を用い、RNase消化を行った。産物は、8M尿素を含有する5%ポリアクリルアミドゲル上で分離し、Kodak X-Omat ARフィルムに露光した。
Alb-uPAを有するマウスを、C.b-17/scid-bg系の動物と交配し、選択的に戻し交配を行い、scid形質をホモ接合状態で繁殖した。最初の実験において、ヘミ接合様式でAlb-uPA導入遺伝子を保持するホモ接合scid動物を交配し、4〜12日齢の後代の同腹子に、0.5〜1x106個の新たに単離した生存能のあるヒト肝細胞を、経脾的に移植した。ヒト肝細胞により独占的に生成されたヒトアルブミンを、移植片機能の指標として使用した。
延長されたヒト生着の証拠を有するマウスを、HCV感染ヒトドナー由来の血清で感染した。非感染ヒト肝細胞を、ヘテロ接合交配の後代27匹に移植し、移植の6週間後、全てのマウスに、経静脈的±経腹腔的に2名の無関係のHCV陽性ドナーの一方から得たヒト血清0.25mlを接種した(ウイルスの遺伝子型1aおよび6a)。接種後3〜40週の間に選択された血清試料を、RT-PCRにより陽性鎖のHCV RNAの存在について分析した。この実験結果を表2にまとめた。移植片の持続期間は、免疫沈降法/ウェスタンブロット法によるHA検出可能な期間として定義した。

陽性鎖HCV RNAは、ホモ接合動物において永続的に明らかにされたが、HCV RNAはヘミ接合動物において検出不能であった。本発明者らは、ヘミ接合体は、消滅された最初の生着およびより早期の移植片喪失の結果として、検出可能なレベルでHCV複製を支持することに失敗したものとして仮定した。この仮定を検証するために、化学発光およびホスホロイメージングを用いたタンパク質ドット-ブロットアッセイ法を開発して、より正確にHA産生を定量した。ヒトドナー1名由来の1 x 106個の凍結保存したヒト肝細胞を21匹のレシピエント(15匹ホモ接合体および6匹はヘミ接合体)へ移植後、無作為に選択した動物から、定量的HA分析のために試料採取し、および/または免疫組織化学的分析のために屠殺した。この実験結果を図8に示した。
ヒトにおけるウイルス力価の長期持続は、進行中の活発な増殖の結果である。しかし免疫無防備状態のキメラ動物において、HCVの持続を真の感染および複製よりもむしろ緩徐なウイルスの排除によって生じたとすることができる。5匹のホモ接合移植片レシピエントに、感染ヒト血清250μlを接種し(遺伝子型3a;2.95x106ウイルスRNAコピー/ml);その結果、各動物は、7.38x105RNAコピーの接種を受容した。この実験の結果を図9に示す。3/5レシピエントにおいて、ウイルス力価は、接種後5週までに、最初の接種の16倍、27倍および36倍増加し、残りの2匹のレシピエントの力価は、5週間にわたって穏やかに増加した(1.6倍および4.3倍)。
移植したマウス肝におけるHCVの局在を確認するために、ヒト肝細胞が移植され、次にHCV感染したヒト血清が接種されたホモ接合マウスから採取した肝切片を、該ウイルスのポリタンパク質のNS3-NS4領域に対するモノクローナル抗体で免疫染色した。ヒト肝の対照切片は、顆粒状の細胞質の外観を示し、核は染色されなかった(図11A)。繊維症領域および門脈三分岐構造は、NS3-NS4について陽性に染色されなかった。肝細胞の大部分は陽性に染色されたが、低レベルな領域であった。移植していないマウス肝からの対照切片は、全ての染色時において如何なる証拠も示さなかった(示さず)。移植し、感染したマウスから採取した実験切片(図11B)は、対照ヒト切片に類似した細胞質顆粒状外観を持つ肝細胞染色領域を示したが、染色強度はわずかに低下していた。この免疫組織化学的知見は、HCVは、移植されたAlb-uPAマウスのキメラ肝内で真にヒト肝細胞に感染していることの証拠を提供している。
これらの個別で、独立して行われたアッセイ法は、接種後2〜5週に試料採取したキメラ肝内の陰性鎖HCV RNAの存在を明確に示している。定量的RT-PCRによる連続した毎週の分析による実験(図9)は、接種後2〜4週でのHCV血清力価の急激な上昇を明らかにし、これは肝内のウイルス複製の最大速度に相当し、これは、(-)鎖ウイルスのRNAの最大量と平行しているものと予想される。これは、(-)鎖が感染の後期(12〜13週に0/2動物試料採取)よりもむしろ、比較的早期(2〜5週に5/6動物試料採取)に検出可能である理由を説明することができる。これらのデータを組み合わせると、結果的にこの動物モデルにおける活発なウイルス複製を裏付けている。更に、HCV感染は慢性である。本発明者らは、最近、機能性ヒト肝細胞移植片(移植後35週での高アルブミン血清レベルの検出により決定(ドットブロットにより約800ユニット以上))および移植後35週での血清中の高力価HCV(1.7x105コピー/ml)を有するキメラAlb/uPAトランスジェニックマウスを基にした本発明の動物モデルを明らかにした。
複製を確認した後、マウスからマウスへのHCV感染の連続継代を試みた。ヒトドナー由来の新鮮な血清(250μl;4.75x105ウイルスRNAコピー)を、未処置のキメラマウスへ経腹腔的に接種した。接種後4週目のウイルス力価は、1.76x106コピー/mlであった。このマウスから採取した血清(125μl;〜2.19x105RNAコピー)を、経腹腔的に第二の未処置のキメラマウスへ接種し、これは接種後4週目に力価1.75x104コピー/mlを示した。この第一継代レシピエント由来の血清を、次に第三の未処置のキメラマウスへ接種した(100μl;〜1.75x103RNAコピー)。接種後5週目に、この第二継代レシピエントは、ウイルス力価3.42x106コピー/mlを有した。複製は生じないが、最初のヒト接種物が持続しているという帰無仮説をたてるならば、この第二継代レシピエントは、最初の接種物からウイルスの〜6000コピーを受け取ることとなると思われる(4.75x105ウイルスコピー x 1:8希釈 x 1:10希釈、マウス血清容積〜1OOOμlと仮定);第二継代レシピエントは、当初のヒト接種物から受け取っているものよりも、576倍より多く測定されたウイルスRNAを有した。この第二継代レシピエントからの血清(30μl)を、2匹の追加の未処置のマウスへ接種し、これら両方を引き続きHCV感染させた(第三継代レシピエント;定量は保留)。こうして連続伝播は、3代継代後の2匹の動物を含む、7匹の動物において明らかにされた。このヒト→マウス→マウス→マウス→マウスの伝播は、HCVゲノムの複製が行われ、および完全に感染性の粒子が産生されることの両方を表している。
現在、文献において、ヒト幹細胞が多能性であることが証明されている。これらは、条件および刺激の適切な組合せにより、造血細胞に加え肝細胞を再生する能力を有する。ヒト幹細胞は、NOD/SCIDマウスにおいて造血細胞を再集合する能力を有することが示されている(例えば、Bhatiaら、J. Exp Med、186(4):619-24(1997);Bhatiaら、Proc Natl Acad Sci USA、94(10):5320-5(1997);Larochelleら、Nat Med、2(12):1329-37(1996)参照)。しかし、肝再集合に関し、小児の肝芽細胞腫における肝を再生するための臨床試験では、幹細胞移植を使用している。
本明細書に説明したプロトコールに従い、10〜14日齢のSCID.bg/Alb-uPaマウスに、ヒト肝細胞を移植した。ヒト肝細胞の代わりに、500万個のヒト臍帯血単球(幹細胞給源)を、経脾注入により移植した。移植後4週目にドットブロットによりヒトアルブミンの存在についてマウス血清を試験するプロトコールは、本明細書に記したものに従った。
本発明のHCV動物モデルを用い、候補となる化学療法薬の抗HCV活性をスクリーニングすることができる。インターフェロンα-2bは、抗HCV活性を有することが既知である。従って、組換えインターフェロンα-2bによる本発明のHCV感染したマウスの処置は、HCV RNAレベルの顕著な減少を生じると思われる。
動物:手術室から得たヒト肝臓組織小片から、コラゲナーゼによる連続灌流を用い、ヒト肝細胞を単離した(リベラーゼHI、Boehringer Mannheim社)。10〜15日齢のホモ接合SCID/uPAマウスに、0.5x106〜1.0x106個の新鮮なヒト肝細胞を、経脾注入により移植した。移植後4週目に、採血し、ヒトアルブミン(HA)濃度について、定量的ドットブロットアッセイ法を用いてアッセイした。>200μg/mlのHAを示すマウスを、移植が成功しているとみなし、本実験に使用した。
本明細書に記したHCVモデルは、具体例として、SCID.bg/Alb-uPaマウスのような、免疫無防御状態の動物におけるキメラマウス/ヒト肝の存在を基にしている。このモデルは、C型肝炎ウイルスの複製を支持するのみならず、更にB型肝炎感染も支持している。現在承認されたC型肝炎に対するワクチンはないので、HBV-感染マウスを用い、ワクチン試験におけるこの動物モデルの価値を検証した。現在HBVに関して利用できる受動および能動の両免疫処置がある。
前記の実施例に説明したプロトコールに従い、10〜14日齢のSCID.bg/ALB-uPaマウスに、ヒト肝細胞を移植した。移植後4週目に、マウスを、ヒトアルブミンについてドットブロットにより試験した。次に、強いシグナルを有するこれらの動物を、実験に使用するために選択した。
本発明の動物モデルは、HBVまたはHCVに感染した自己肝細胞の状況において、ヒト免疫応答を調べるための価値のあるツールを提供する。ヒト肝細胞を保持するマウスは、自己末梢血単核細胞(PBMC、2〜3x107細胞/マウス)で免疫再構築することができ、下記の試験を行うためのHBVおよびHCV感染のモデルシステムを提供した。このモデルは、次に下記に例示した方法で使用することができる。
HCV感染に対する様々なサイトカイン類(1型対2型)の役割を試験するために、HCV感染したマウスから血清を収集し、PBMCで再構築し、1型対2型サイトカインの保有率について試験した。これらの実験は、時間経過を追って行い、サイトカイン産生をHCVウイルス負荷と相関させ、かつ対照の感染していないが免疫再構築したマウスと比較した。他方で、これらのマウスにおいて、支配的な1型または2型サイトカイン、例えば、IL-2、γ-IFN、IL-4、IL-10、およびIL-12を注射または排除し(abrogate)(抗サイトカイン抗体の注射により)、ウイルス負荷、入り交じったHCVおよび非HCVペプチドに対するT細胞反応の作用、加えてポリクローン性刺激を評価した。加えて、サイトカインスイッチの進行、サイトカイン産生の欠損またはそれらの調節は、感染直後にまたはより長期間(すなわち、マウスにおいて2〜3ヶ月間)にわたって評価した。インビトロ実験においては、インビトロにおけるT細胞反応に対する追加のある種のサイトカインの役割を調べた。
ヒトの慢性HCV感染において、樹状細胞機能が損なわれていることを示すいくつかの証拠がある。樹状細胞(DC)は、効率的免疫を誘起するためのCD4+ T細胞の最も強力な刺激物質である。従って、HCV感染の状況においてDC機能の試験は、HCV感染に対する免疫応答を理解するために必須である。
慢性HCV感染した個人におけるThおよび細胞傷害性T細胞(CTL)の反応を報告する多くの試験において、HCVの保存された領域からの多くの入り交じったThおよびCTLエピトープが、インビトロにおいて同定され、これはThおよびCTLプライミングがHCV感染した個人において生じることを示唆している。しかし、明らかにこの進行する天然のT細胞反応はそれ自身、ウイルス感染および/または複製を消去するするためには十分ではない。本発明の動物モデルを用いて、強い免疫応答を誘起するために公知の入り交じったThおよびCTLエピトープを用いる様々な免疫処置方法(以下に列記した)を評価することができる。このマウスを、免疫処置後に、CTLおよびTh細胞反応の作成について評価した。平行して、ウイルス負荷を評価した。従って、この動物モデルを用いて、HCV感染に対する免疫を提供するための、ThおよびCTh反応の適当な調節を決定することができる。
HCV慢性キャリヤにおける進行中のHCV感染の寛解に失敗することに関する別の仮説は、これらのエピトープに対するThおよびCTL反応は、実際にウイルス複製を抑制するまたはウイルス感染した細胞を消去することはできず、他のエピトープ決定基に対する免疫応答が、保護的免疫の成立に必要であるという証拠に焦点を当てている。HCVポリタンパク質上の新規T細胞エピトープを決定するために、最初にインシリコ(in silico)試験を行い、保存された構造性に加え、機能性ウイルスタンパク質から推定ThおよびCTLエピトープを同定した。これらの同定したエピトープを、次に改変し、本発明の再構築動物モデルにおいて、それらの強いT細胞反応を作成する能力について評価し、これはこれらが免疫療法のワクチン候補であることを示している。
この方法は、慢性HCV感染における免疫応答を考慮し、本発明の再構築動物モデルを使用し、抗原を基にした療法および低分子を基にした療法の組合せを利用する抗ウイルス療法を同定しており、ここでこれらの低分子はウイルス複製および/または免疫調節の阻害活性を有する。ウイルス複製の効果的抑制またはウイルスクリアランスを提供する組合せは、前述のようにHCV感染した免疫再構築した動物を用いて同定する。ワクチン候補を、サイトカイン(例えばIL-2またはリポソームIL-2)との組合せにおいて評価し、HCV感染を抑制および/または消去する効率的免疫を提供する。
前述のように、Apo B100は、高脂血症から生じるアテローム性動脈硬化症のリスクに関する、当技術分野において受け入れられたマーカーである。高脂血症に対する活性を有する物質のスクリーニングにおける本発明のマウスモデルの使用を評価するために、ヒトApo B100に特異的なマウスのモノクローナル抗体を用い、生着したヒト細胞によるヒトapo B100産生を検出した。血清試料を、キメラAlb/uPA移植した動物から、およびAlb/uPA移植していない動物(陰性対照)から収集した。ヒト血清は、陽性対照として使用した。この血清を、当技術分野において周知の方法に従い抗Apo B100抗体を用いるウェスタンブロットにより分析した。

- 以下を含む、ヒトC型肝炎ウイルス(HCV)を感染させるキメラのネズミ科動物(murine)宿主:
機能的に同系のBリンパ球およびTリンパ球を欠く免疫不全のネズミ科動物宿主;
宿主ネズミ科動物肝臓細胞においてウロキナーゼ型プラスミノーゲンアクチベーターのポリペプチドの発現を提供する、ゲノムに組み込まれた導入遺伝子;および
