Abstract
Description
エタノールを含浸させたヒトの胆嚢、結腸、および神経の標本を、1リットルの反応槽に入れた。反応槽の温度を40℃に維持した。反応槽を閉じ、40℃の二酸化炭素で150バールまで加圧した。制御弁により圧力を150バールに維持しながら、新鮮な二酸化炭素をポンプにより10 kg/時間の速度で反応槽に流入させた。45分後、ポンプを停止させ、温度を60℃まで上昇させた。圧力を約230バールまで上昇させた。圧力を210バールに維持しながら、60℃の溶融パラフィンを、2リットル/時間の速度でゆっくり反応槽に流入させて、標本に含浸させた。反応槽がパラフィンでほぼ完全に満たされた後、10分間かけて反応槽を減圧した。余剰のパラフィンを反応槽から排出し、反応槽を40℃まで冷却した後、包埋済みの標本を反応槽から回収した。標本を、従来の手順に基づいて組織学的に分析した。
Claims (12)
- 組織学的分析用に生物学的試料を処理する方法であって、超臨界流体または近超臨界流体を含む組成物に該試料を接触させて、1バールを超える圧力下で好ましくはパラフィンである包埋剤を該試料に含浸させる段階を含む方法。
- 超臨界流体または近超臨界流体が二酸化炭素である、請求項1記載の方法。
- 生物学的試料が、新鮮な組織試料、凍結組織試料、または固定組織試料であり、好ましくは新鮮な未固定の試料である、請求項1または2記載の方法。
- 生物学的試料が、臓器またはその一部を含む、請求項1〜3のいずれか一項記載の方法。
- 試料が、含浸前に、好ましくは超臨界流体を含む組成物を用いることによって、脱水、脱脂、および/または脱石灰される、請求項1〜4のいずれか一項記載の方法。
- 組成物が、好ましくはアルコールである脱水剤をさらに含む、請求項5記載の方法。
- 組成物が、好ましくは酸である脱石灰剤をさらに含む、請求項5または6記載の方法。
- 組織学的分析用に少なくとも1つの試料(10)を調製するための処理装置(1)であって、該少なくとも1つの試料(10)のための少なくとも1つの処理反応槽(9)を含み、少なくとも1つが超臨界相または近超臨界相にある少なくとも1つの物質を該反応槽(9)に供給するための供給手段(4)と、コンジット(8)を介して該反応槽(9)に包埋剤を添加するための少なくとも1つの供給手段(7)とを含むことを特徴とする処理装置(1)。
- 物質を必要な圧力および/または温度にするための加圧手段および/または加温手段(5、6)をさらに含む、請求項8記載の処理装置(1)。
- 反応槽(9)から出る物質の混合物から物質を分離するための分離手段(11)をさらに含む、請求項8または9記載の処理装置(1)。
- 反応槽(9)から排出された物質を再循環させるための再循環手段(13)をさらに含む、請求項8〜10のいずれか一項記載の処理装置(1)。
- 組織学的分析用に生物学的試料を処理するための、請求項8〜10のいずれか一項記載の処理装置の使用。
