−脱水フェーズ:サンプルに含有された水が、無水の試薬によって置き換えられる、
−清澄化フェーズ:無水の試薬が、無水の試薬及び封入媒体の両方と混合される成分(清澄化媒体)によって置き換えられる、
−含浸フェーズ:清澄化媒体が、含浸媒体(又は封入媒体)によって置き換えられる。
被検査サンプルをプロセスした後、鋼製の型によって、これらを、融点56〜58℃のパラフィンブロック内に封入する。
マイクロトームを使用することによってカットして、厚さ3〜5μmの切片を得る。組織のこれら切片をブラシにて集め、35℃の水を収容する浴に入れて、これらを膨張させる。
被染色組織切片のためには、各種の試薬が組織に対して直接に作用できるように、組織を封入するパラフィンの層を除去しなければならない。
各種の染色プロトコルを行った後、樹脂性マウンティング媒体によるカバーの適用のため、切片をさらに脱水しなければならない。これは、保存された切片の長い寿命を保証する。
本発明による組成物は、例えば、アルコール及びエーテル中において固定され、パラフィン又は他の封入剤中には封入されていない、尿、スミアテスト、痰及び他の液体の如き組織学的サンプル又は他のタイプのサンプル調製においても使用される。
1.洗浄全体の時間を同一に維持したまま、又は必要であれば、短い時間で、要求に応じて、異なった濃度のアルコールでの全ての洗浄プロセス、
2.最終のディアファナイゼーション
を置き換える。
Claims (5)
- 組織学的、解剖学的及び細胞学的サンプルを調製する方法であって、該サンプルを、室温及び大気圧下で、オクタン60容量%、イソプロピルアルコール10容量%及びエチルアルコール30容量%を含む組成物で処理することを特徴とする方法。
- 中間試薬によるサンプルの洗浄を、前記組成物による前記サンプルの処理の前に行うことを特徴とする請求項1に記載の方法。
- 前記中間試薬が95%エチルアルコールであることを特徴とする請求項2に記載の方法。
- 組織学的、解剖学的及び細胞学的サンプルを処理するために使用する組成物であって、オクタン60容量%、イソプロピルアルコール10容量%及びエチルアルコール30容量%を含み、室温及び大気圧下で使用することを特徴とする組成物。
- 組織学的、解剖学的及び細胞学的サンプルを処理するために、請求項4に記載の組成物を使用する方法。
