スレオニンインポーターをクローニングするための宿主株としてスレオニンインポーター欠損株を使用するため、欠損株をコリネバクテリウム・グルタミカムから作製した。この目的のために、高スレオニン要求性株を低スレオニン要求性株から作製した。
実施例1で作製された、高スレオニン要求性株のコリネバクテリウム・グルタミカムCJ L-11を、コリネバクテリウム・グルタミカム野生株のATCC 13032からスレオニンインポーターをクローニングするために、宿主株として使用した。
実施例2で得られた低スレオニン要求性クローンの遺伝子を調べるため、適切なプライマーを合成し、オーバーラップさせてDNAシーケンスを行った。この方法で、クローニングされたDNAフラグメントの塩基配列を決定した(配列番号1を参照されたい)。
スレオニン移入におけるクローニングされたthrYの関与を確認するため、本発明者らは、低スレオニン要求性株が高スレオニン要求性株に形質転換されるかを調べる目的で、問題のコリネバクテリウム・グルタミカムCJ L-1株(低スレオニン要求性株)の遺伝子を破壊した。
スレオニン産生株のthrYの破壊がどのようにスレオニンの収率に影響を与えるかを見出すため、本発明者らは以下の実験を行った。
Claims (3)
- 配列番号1のDNA配列のうち、1,772番目の塩基から3,025番目の塩基の連続したDNA配列により発現される、コリネバクテリウム・グルタミカム(Corynebacterium glutamicum)由来のスレオニンインポーター。
- コリネバクテリウム・グルタミカムにおいて、請求項1に記載のスレオニンインポーターを欠損することによりスレオニンの収率を上げる方法。
- 請求項2に記載の方法により作製されたスレオニン産生株。
