本発明の化合物は、それらの塩及び溶媒和を含み、既知の有機合成技術を用いて調整され、任意の多数の可能な合成経路に従って合成され得る。
本発明の化合物の合成は、化学式(A)によって示される構造を有するボロン含有フラグメント(F1)を含み得る。
本発明の化合物の中央部は、F2−F1中間体形成のためのペプチド結合構造によってフラグメントF1と結合したフラグメントF2によって表され得る。ペプチド結合又はアミド結合を介したカップリング化合物のための方法は当業者に周知であり、例えば、The Peptides:Analysis,Synthesis,Biology,Vol.I.,eds.Gross,et al.,Academic Press,1979において記載されている。F2フラグメントの実施例は化学式(E)で提供されている(Pgはアミノ保護基、Yとmは本明細書で定義されている)。さらに、Bocを用いたアミノ酸のアミノ基の保護又は別のアミノ保護基は、当業者に周知である。
更にフラグメント(F3)は、ペプチド結合形成によって、F2−F1中間体のF2フラグメントと結合され得る。ペプチド結合又はアミド結合を介したカップリング化合物のための方法は当業者に周知であり、例えば、The Peptides:Analysis,Synthesis,Biology,Vol.I.,eds.Gross,et al.,Academic Press,1979などにおいて記載されている。F2フラグメントの実施例は化学式(F)(R2はH、及びZとnは本明細書で定義されている)において提供される。
[実施例1]
工程1:2−(2−メチルプロピル)−(3aS,4S,6S,7aR)−ヘキサヒドロ−3a,5,5−トリメチル−4,6−メタノ−1,3,2−ベンゾジオキサボロール。
[実施例2]
無水DMF(100ml)中、BocNH(NO2)ArgOH(15.7g,49.3mmol)の溶液に、HATU(O−(7−アザベンゾトリアゾール−1−イル)−1,1,3,3−テトラメチルウラニウムヘキサフルオロリン酸塩;18.7g、49.3mmol)及びHOAt(1−ヒドロキシ−7−アザベンゾトリアゾール;6.71g,49.3mmol)を加えた。前記混合物を0℃まで冷却し、N−メチルモルホリンを加えた(13.6ml,0.123mol)。10分後、実施例1の(1R)−1−[(3aS,4S,6S,7aR)−ヘキサヒドロ−3a,5,5−トリメチル−4,6−メタノ−1,3,2−ベンゾジオキサボロール−2−イル]−3−メチルブチルアミン塩酸塩(12.4g,41.1mmol)を加えた。冷却槽を除去し、さらにこの混合物を室温で4.5時間撹拌した。前記混合物を酢酸エチル(800ml)で希釈し、2%クエン酸溶液(2×150ml)、2%NaHCO3溶液、及び2%NaCl溶液(2×150ml)で洗浄した。水層をさらに酢酸エチル(150ml)で抽出した。一つに集めた有機層を硫酸ナトリウムで乾燥し、濃縮した。得られた油の残渣は酢酸エチル(500ml)に再溶解し、この溶液を冷水(200ml)で洗浄した。水層をさらに酢酸エチル(500ml)で抽出した。一つに集めた有機層を硫酸ナトリウムで乾燥し、濃縮した。前記残渣をジエチルエーテル(100ml)に再溶解し、この溶液を撹拌しながらヘキサン(600ml)にゆっくり加えた。濾過して白色固体(43.4g)を集め、カラムクロマトグラフィーで精製し、初期は50:50ヘキサン:酢酸エチル混合物、その後酢酸エチルを用いて溶出した。生成物を含む画分を濃縮し、残渣をジエチルエーテル(100ml)に再溶解し、得られた溶液を撹拌しながらヘキサン(600ml)にゆっくり加えた。濾過によって白色固体が得られた(15.2g、収率66%)。
無水ジクロロメタン(100ml)中、BocNH(NO2)ArgOH(5.82g、18.2mmol)の懸濁液に、N−メチルモルホリン(2.0ml、18.2mmol)を加えた。前記混合物を−15℃まで冷却した後、イソブチルクロロギ酸エステルを加えた(2.37ml,18.2mmol)。この混合物を−15℃で10分間撹拌した後、実施例1において得られる(1R)−1−[(3aS,4S,6S,7aR)−ヘキサヒドロ−3a,5,5−トリメチル−4,6−メタノ−1,3,2−ベンゾジオキサボロール−2−イル]−3−メチルブチルアミン塩酸塩(5.0g,16.6mmol)を加え、さらに直ちにN−メチルモルホリン(2.0ml,18.2mmol)を加えた。前記反応混合物を−15℃で1.5時間撹拌した後、室温まで温め、酢酸エチル(150ml)、水と0.1N塩酸(10ml)に分配した。前記有機層を飽和NaHCO3溶液で洗浄し、無水硫酸ナトリウムで乾燥し、濃縮した。油の残渣(9.25g)を酢酸エチルからの再結晶で精製し、十分純粋な生成物の3つの集団が得られた(5.03g,収率54%)。
[実施例3]
ジオキサン(15ml)中、4Nの塩酸溶液を、0℃に冷却しながらジオキサン(40ml)とジエチルエーテル(7ml)の混合物中、実施例2のN−[(1S)−1−[[[(1R)−1−[(3aS,4S,6S,7aR)−ヘキサヒドロ−3a,5,5−トリメチル−4,6−メタノ−1,3,2−ベンゾジオキサボロール-2−イル]−3−メチルブチル]アミノ]カルボニル]−4−[[イミノ(ニトロアミノ)メチル]ブチル]のカルバミン酸1,1−ジメチルエチルエステル(4.04g,7.06mmol)の溶液に加えた。前記反応混合物を室温まで温めて、更に4時間撹拌した。前記溶媒をロータリーエバポレータで除去し、残渣をジエチルエーテル(50ml)で処理し、この混合物を室温で3日間撹拌した。得られた個体を濾過で集めることにより、純粋な生成物が3.18g(90%収率)得られた。
実施例2のN[(1S)−1−[[[(1R)−1−[(3aS,4S,6S,7aR)−ヘキサヒドロ−3a,5,5−トリメチル−4,6−メタノ−1,3,2−ベンゾジオキサボロール−2−イル]−3−メチルブチル]アミノ]カルボニル]−4−[[イミノ(ニトロアミノ)メチル]アミノ]ブチル]のカルバミン酸1,1−ジメチルエチルエステル(3g,5.3mmol)をEt2O(40mL)に溶解し、Et2O(20mL)中約10%HCl溶液を窒素雰囲気下0℃で滴下して加えた。前記反応混合物は室温まで温め、更に5時間撹拌した。

無水テトラヒドロフラン(30ml)中の10−ウンデセン−1−オール(4.23g,24.8mmol)、フタルイミド(3.65g,24.8mmol)、及びトリフェニルホスフィン(6.51g,24.8mmol)混合物に対して、無水テトラヒドロフラン(10ml)中のDEAD(3.9ml,24.8mmol)溶液を温度を8〜10℃以下に保ちながらゆっくり加えた。2時間後、更にDEAD(1.0ml,6.37mmol)とトリフェニルホスンフィン(1.3g,4.96mmol)を添加し、この混合物を室温で一晩撹拌した。前記反応混合物を濃縮し、残渣をジエチルエーテル(50ml)を用いて倍散した。前記固体を濾過して除去し、更にジエチルエーテル(2×50ml)で洗浄した。一つに集めた濾液を濃縮し、残渣を40℃でヘキサン(50ml)で倍散した。得られた個体を濾過によって除去し、ヘキサン(2×50ml)で洗浄した。一つに集めた濾液を濃縮し、残渣を、ヘキサン:酢酸エチル混合物10:2を溶出に用いたカラムクロマトグラフィーによって精製した。前記生成物は低融点の白色固体として得られた(4.9g、収率66%)。
ヘキサン(20ml)と酢酸(6ml)の混合物中の工程1の2−ウンデセ−10−エニル−1,3−ジオキソ−1,3−ジヒドロイソインドール(2g,6.68mmol)とAliquat(登録商標)336(0.2g)溶液を、水(28ml)中の過マンガン酸カリウム(2.76g,20mmol)溶液に0℃に冷却しながら加えた。前記反応混合物は室温で7時間撹拌した後、紫色がなくなるまで亜硫酸ナトリウム水溶液を加えた。その後前記混合物を酢酸エチルで抽出し、有機相を硫酸ナトリウムで乾燥して濃縮した。前記残渣はシリカゲルクロマトグラフィーで精製し、ヘキサン:酢酸エチル2:1を用いて溶出した。前記生成物は白色固体として得られた(1.29g,収率61%)。M.p.58〜60℃。
[実施例6]
ジオキサン(10ml)中、エタンスルホニルクロライド(3.9ml,41.1mmol)の溶液を、0〜5℃で攪拌しながら、1N NaOH(56ml)及びジオキサン(10ml)中の6−アミノヘキサン酸(2g,15.2mmol)の溶液に加えた。前記反応混合物のpHは、25%水酸化ナトリウム溶液を添加することによって8〜9に調整した。前記混合物を室温まで温め、30分間攪拌した。さらに25%NaOH溶液を加えてpHを約11に調整した。3.5時間後,1N塩酸(15ml)と酢酸エチル(60ml)を加えた。前記有機相を硫酸ナトリウムで乾燥して濃縮した。残渣をジエチルエーテル(5ml)とヘキサン(15ml)の混合物で倍散した。前記個体を濾過して集め、1.3gの乾燥した標題化合物が得られた(収率40%)。1HNMR(DMOSO−d6):11.9(1H,s);6.97(1H,t,J=5.7Hz);2.97(2H,q,J=7.1);2.88(2H,q,J=6.6);2.2(2H,t,J=7.3);1.47(4H,m);1.29(2H,m);1.18(3H,t,J=7.3)。
[実施例7]
ジオキサン(5ml)中、エタンスルホニルクロライド(1.5ml,15.7mmol)溶液を、0〜5℃で攪拌しながら、1N NaOH(22ml)及びジオキサン(5ml)中の8−アミノオクタン酸(1g,6.28mmol)溶液に加えた。前記混合物を室温まで温め、3.5分間攪拌した。この間、1時間間隔で、25%NaOH溶液を添加することにより前記pHを7〜8に調整した。この反応混合物をジエチルエーテル(30ml)で洗浄した。1N HClを添加して前記pHを1〜2に調整し、この混合物を酢酸エチル(70ml)で抽出した。前記有機相を硫酸ナトリウムで乾燥して濃縮した。前記残渣をジエチルエーテル混合物で倍散した。前記個体を濾過して集め、真空下で乾燥し、600mgの標題化合物を得た(収率38%)。1HNMR(DMSO−d6):11.9(1H,s);6.96(1H,t,J=6Hz);2.96(2H,q,J=7.1);2.88(2H,q,J=6.6);2.2(2H,t,J=7.3);1.45(4H,m);1.26(6H,m);1.18(3H,t,J=7.3)。

無水ジクロロメタン(10ml)中、実施例5に従って調整した10−(1,3−ジオキソ−1,3−ジヒドロ−イソインドール−2−イル)−デカン酸(353mg,1.11mmol)の溶液に対して、N−メチルモルホリン(122μl,1.11mmol)を加えた。前記混合物を−15℃に冷却した後、クロロギ酸イソブチル(144μl,1.11mmol)をゆっくり加えた。15分後、実施例3の(2S)−2−アミノ−5−[[イミノ(ニトロアミノ)メチル]アミノ]ペンタンアミド、N−[(1R)−1−[(3aS,4S,6S,7aR)−ヘキサヒドロ−3a,5,5−トリメチル−4,6−メタノ−1,3,2−ベンゾジオキサボロール−2−イル]−3−メチルブチル]−塩酸塩(508mg,1.01mmol)、及びさらにN−メチルモルホリン(122μl,1.11mmol)を加えた。この反応混合物を−15〜10℃で4時間攪拌した後、小容量に濃縮し、酢酸エチル(20ml)と水(10ml)に分配した。前記水相をさらに酢酸エチル(10ml)で抽出した。一つに集めた有機相を硫酸ナトリウムで乾燥し、濃縮した。前記残渣を酢酸エチル(3ml)に溶解し、この溶液を室温で攪拌しながらヘキサン(120ml)に滴下した。デカンテーションにより固体を集め、真空下乾燥した(730mg,94%)。

本発明の化合物はプロテアソーム活性を阻害することができる。以下の表F−1は、例えばプロテアソーム活性阻害能力に関する本発明の化合物の幾つかの実施例に関連するデータを提供する。
本発明の化合物はプロテアソームの活性を阻害し、プロテアソームが直接あるいは間接的に関連する細胞内の様々な機能を阻害或いは遮断する。例えば、プロテアソーム阻害剤は、細胞内アポトーシスを誘導するなどの調節をし得る。一部の実施形態において、本明細書の化合物は、アポトーシスの誘導により腫瘍細胞を壊す。従って、本発明の化合物は癌、腫瘍、又は他の増殖障害に使用され得る。
20Sヒト赤血球プロテアソーム(HEP)のキモトリプシン様活性試験
本発明の化合物のプロテアソームキモトリプシン様活性は、以下の方法に従って評価した。
α−キモトリプシン活性試験
96ウェルマイクロタイタープレートにおいて、Sigma Inc.から購入したウシα−キモトリプシンを0.5M NaCl 50mM Hepes緩衝液中10ng/mLでまいた(約2pM触媒部位)。Sigma Inc.,St.Louis,MO,USAから購入した蛍光物質Suc−AAPF−AMC(スクシニル−Ala−Ala−Pro−Phe−7−アミド−4−メチルクマリン)をジメチルスルホキシド中、10mMの保存溶液から最終濃度25μMになるように加えた。反応容積は1ウェルあたり100μlであった。室温で種々時間インキュベートした後、遊離AMC濃度をPerkin Elmer HTS 7000Plusマイクロプレートリーダーを用いて370nM励起、465nM発光で測定した。α−キモトリプシン活性は、加水分解基質が時間と直線で増加し、蛍光シグナル変化が遊離AMC濃度と比例する条件で測定した。
HEPのIC50値とα−キモトリプシン阻害の測定
IC50値は一般的に、酵素の活性が50%の阻害を引き起こすのに必要な化合物の濃度として定義される。IC50値は、その設計用途に対する化合物の活性指標として有用である。本発明のプロテアソーム阻害剤は、ヒト赤血球プロテアソーム(HEP)の約1μモルの阻害より少ないIC50を有する場合に活性であるとみなされ得る。一部の実施形態において、前記阻害剤はHEPに対して特異性を示し、HEPの阻害IC50に対するウシα−キモトリプシンの阻害IC50の比、すなわちIC50(αキモトリプシン)/IC50(HEP)が約100より大きい。
Molt−4細胞下部におけるプロテアーゼのキモトリプシン様活性の細胞性アッセイ
Molt−4細胞(ヒト白血病)におけるプロテアソームのキモトリプシン様活性は、以下の方法Aに従って試験した。この方法の簡単な解説は以前に公開されている(Harding et al.,J.Immunol.,1995,155,1767)。
MOLT−4細胞株におけるプロテアソーム阻害剤のEC50値の測定
EC50値は一般的に、最少反応と最大反応(本試験に対してはそれぞれ0%と85〜90%)の中間の酵素活性を引き起こすのに必要な化合物の濃度として定義される。EC50値は、化合物活性のその設計用途に対する有用な指標である。本発明の化合物は、約10マイクロモルより少ないEC50値を有する場合に活性であるとみなされ得る。
プロテアソームのトリプシン様活性試験
ヒトプロテアソームのトリプシン様活性は、以下の変更とともに、上述に記載したように試験しても良い。反応は、1mMの2−メルカプトメタノールを補足したトリス−グリセロール緩衝液(pH9.5)において実行し、基質は、例えばベンジルオキシカルボニル−Phe−Arg−AMC(100μM)のような蛍光基質であっても良い。
細胞筋破壊in vivo阻害
幼若ラットにおけるヒラメ筋の減量(unweighting)萎縮における阻害効果は、例えば、Tischler,Metabolism,1990,39,756において記載されている方法などによって測定しても良い。例えば幼若雌Sprague−Dawleyラット(80〜90g)の尻尾にギブスをし(tail−cast)、Jasper,et al.,J.Appl.Physiol.,1984,57,1472に記載したように、後肢をつるした。前記動物の後肢を、各動物を個別に収容するケージの床の上に持ち上げる。動物は食物と水に自由に近づくことができ、吊り下げ時と終了時に体重を測定することができる。吊り下げ期間中、前記動物はそれらのつま先がケージの床に接触しておらず、前記ギブスのために尻尾が腫脹していないことを確実にするために毎日点検する。
各試験は、5動物ずつ4群に無作為に分けた20匹のラットを吊り下げすることから始める。2日間吊り下げたA群は、より長い時間吊り下げた他の動物におけるヒラメ筋のおおよその大きさの基本データを提供する。この研究に着手した時点における前記群の平均体重を比較し、身体の大きさの違いに対する補正率として使用した。B群は、減量(unweighting)の2日後にマーサリル水溶液で1つの肢を処置したヒラメ筋を有する第二の対照群であり、各群の動物に対して減量中に筋肉萎縮を遅延させる能力を示す。減量開始後2日目、マーサリル水溶液(200nM;4.mu.l/100g開始体重)を1つのヒラメ筋に注入する。対側筋肉は同じ容積の0.9%生理食塩水("溶媒")を注入した。動物はin situにおける注入手法の間、Innovar−vet(10μl/100g体重)精神安定作用の元で維持した。注入後、前記動物を更に24時間吊り下げ、前記ヒラメ筋を除去した。各試験におけるCとD群は、本公開化合物の2つの異なる実施形態それぞれを試験するために使用する。動物は、ジメチルスルホキシド(DMSO)中に含まれる1mMプロテアソーム阻害剤を1つ肢のヒラメ筋に注入し、対側ヒラメ筋にDMSOのみを注入することを除き、B群のように処置する。従って、各試験は2つの対照群と本発明のプロテアソーム阻害剤試験とから成る。異なる対の阻害剤を用いたこのような5つの試験を完了することによって、各阻害剤の試験に対して10の"n"値を提供し、それぞれ2つの異なる出荷動物において試験をする。
動物を犠牲にした後、ヒラメ筋を摘出し、脂肪と結合組織を取り除き、注意深く重量を測る。その後、前記筋肉を10%トリクロロ酢酸(TCA)中でホモジナイズし、遠心分離によって沈殿タンパク質をペレット化する。その後、前記ペレットを10%TCAで1回、エタノール:エーテル(1:1)で1回洗浄する。この最終ペレットを4mlの1N水酸化ナトリウム中に可溶化する。その後、前記サンプルを標準アルブミンを用いたビウレット法によってタンパク質含量を分析する。
全筋肉タンパク質含量における阻害効果は主に、無処置対側筋の対比較によって検討する。含量の比を計算した後、分散分析("ANOVA")によって統計学的に分析する。前記タンパク質含量比を未処置対照動物とも比較できるように、左肢は常に処置した足にする。このような方法において、有意差は2つの肢のタンパク質含量の比較として示されると同時に、試験した阻害剤の相対的な効果も示される。対応のあるスチューデント検定もまた各別々の処置の効果に対して行う。未処置群のデータもまた2日のタンパク質含量の推定値を提供する。これはB、C、及びD群それぞれに対して処置後24時間を越えたタンパク質変化の近似値を許容とする。
各試験は、1つの阻害剤のタンパク質合成の効果を試験した5匹の動物を伴う10匹の動物から成る。対側のDMSO−処置筋肉が阻害剤処置筋肉の対照対となるので、本研究のこの観点においては対照動物は必要ではない。in situ処置24時間後、タンパク質合成の配分率は両方のヒラメ筋において分析する。各筋肉は、3H−フェニルアラニン(50mM;1μCi/ml)を含む0.9%生理食塩水(3.5μl/100g最終体重)を用いて注入する。15分後、中間の3分の2の筋肉を摘出し、筋肉を以下のように処理する。
前記筋肉はまず、タンパク質合成を終結させるために、0.5mMシクロヘキシミドを含む0.84%生理食塩水中で10分間洗浄し、さらに細胞中のフェニルアラニンを補足するために20mMのシクロロイシンで洗浄する。その後、前記筋肉を2.5mLの氷冷2%過塩素酸中でホモジナイズする。沈殿タンパク質を遠心分離によってペレット化する。前記上清の1つのアリコートを液体シンチレーションカウンターに用い、別のアリコートを溶解フェニルアラニン濃度を蛍光分析で測定するために、フェニルアラニンをフェニルアミンに変換する工程に用いる。例えば、Garlick,et al.,Biochem.J.,1980,192,719を参照。これらの値は、細胞内の特異的な活性を提供する。筋肉タンパク質におけるフェニルアラニンの前記特異的な活性は、6N HClにおいて加熱することにより、タンパク質を加水分解した後測定する。アミノ酸は緩衝液中で可溶化する、1つのアリコートはシンチレーションカウンターのために用い、別のものは上清画分のフェニルアラニン分析のために用いる。前記タンパク質合成の配分率は、タンパク質特異的活性/細胞内特異的活性、時間として計算する。
タンパク質合成の分析は各阻害剤に対して対基準である。対側筋に関する対応のあるスチューデントt検定比較によって、タンパク質合成における阻害剤の効果があるかどうかを測定する。タンパク質破壊は、タンパク質合成の配分率として(パート2より)、さらにタンパク質付着の前記配分率はとして(パート2)、近似的に計算でき、ここでタンパク質の損失はタンパク質付着に対しては負の値が得られる。
in vivoにおける抗腫瘍活性の検討
in vivo試験において用いるプロ田ソーム阻害剤は、静脈内投与(iv)又は経口(po)投与に対して適する媒体において処方する。例えば、iv投与に対しては、化合物は0.9% NaCl、又は0.9% NaCl、solutol HS15、及びジメチルスフホキシドの混合物87:10:3(v:v:v)中にそれぞれ溶解させて投与する。
異なる病理学的起源の以下のヒト及びマウス腫瘍細胞株を本発明の化合物の抗腫瘍活性試験に使用する:H460(ヒト、肺)、A2780(ヒト、卵巣)、PC−3(ヒト、前立腺)、LoVo(ヒト、結腸)、HCT116(ヒト、結腸)、BXPC3(ヒト、膵臓)、PANC−1(ヒト、膵臓)、MX−1(ヒト、乳房)、MOLT(ヒト、白血病)、多発性骨髄腫(ヒト、骨髄腫)、YC8(マウス、リンパ腫)、L1210(マウス、白血病)、3LL(マウス、肺)。
例えばHarlan(Correzzana,Mi Italy)から、5〜6週の免疫応答性又は免疫欠損マウスを購入する。CD1nu/nuマウスは無菌条件、すなわち無菌のケージ、床、餌で維持し、さらに酸性水を使用する。
異なる細胞組織タイプ(肺、卵巣、乳房、前立腺、膵臓、結腸)の固体腫瘍モデルを、免疫応答性マウス(マウスモデル)又は免疫不全マウス(ヒトモデル)の腋窩部の皮下に移植する。もともとはATCCから得たヒト腫瘍細胞株を、"in vitro培養"からの固体腫瘍として、"in vivo"で増殖するために適用させる。
固体(段階的な)又は血液学的腫瘍を有するマウスは、試験群(10マウス/群)において無作為化する。固体腫瘍に対しては、処置を開始するために、各群80〜100mgの平均腫瘍重量とみなし、すなわち、最少及び最大腫瘍を有するマウスを処分する。
以下のパラメーターは、抗腫瘍活性の査定のために評価する:
第一固体腫瘍の増殖;各マウスは週に2回のカリパス測定によって監視する;
対照マウスと比較した処置マウスの生存時間
週に2回の各マウスの体重評価。
毒性は全体の剖検所見と体重損失をもとに毎日評価する。マウスは、溶媒処置した対照動物の死亡以前に死亡した場合、又は有意な体重損失(>20%)、及び/又は脾臓と肝臓の大きさの減少が観察される場合に、毒性で死亡したとみなす。
細胞のin vitro生存率
試験化合物の存在下、in vitroにおける細胞の生存率のIC50値の測定は、以下の方法に従って測定することができる。細胞は種々密度で96ウェルプレートに播種した後、24時間後Calcein−AM生死判別試験を用いて測定し、各細胞種に対して最適最終密度を測定することができる。その後、細胞は当業者が周知の適切な細胞培地100μL中、測定した濃度で96ウェルプレートに播種する。
in vitroの速度論的試験
本発明の化合物は、Rock,et al.,Cell,1994,78,761に記載された手順を用いて、プロテアソーム阻害活性を試験することができる。この方法に従って、プロテアソームと試験化合物が相互作用して複合体を形成する場合に、その平行解離定数(Ki)が確立される。前記反応はウサギの筋肉からのSDS−活性化20Sプロテアソームを用いて行うことができ、プロテアソーム基質はSuc−LLVY−AMCである。
NF−κB活性化阻害剤
本発明の化合物は、Palombella,et al.,Cell,1994,78,773に記載された測定法を実行することによって、NF−κBの阻害活性のために試験することができる。例えば、MG63骨癌細胞は、指定した期間、TNF−αを用いた
処置によって刺激することができる。すべての細胞抽出物は、ヒトIFN−β遺伝子プロモーターからのPRDIIプローブを用いた電気移動移動シフト分析によって調整及び分析することができる。
化合物活性
上述の実施例C及び実施例Eの測定法を用いて、以下の表F−1は本発明化合物のプロテアソーム阻害に対する有用性を示す。以下の表において、実施例CのHEP阻害に対して、HEP阻害に対するIC50において"+"を伴う本発明化合物は1000ナノモルより小さく、"++"を伴う本発明化合物は100ナノモルより小さく、"+++"を伴う本発明化合物は10ナノモルより小さい。以下の表において、実施例EのMOLT4の阻害に対して、HEP阻害のEC50において"+"を伴う本発明化合物は10000ナノモルより小さく、"++"を伴う本発明化合物は2000ナノモルより小さく、"+++"を伴う本発明化合物は200ナノモルより小さい。">+"は活性が測定法の限界より大きい場合に生じる。IC50値又はEC50値が示されていない場合、データはまだ測定されていない。
薬学的に使用される場合、化学式(I)の化合物は、薬学的組成物の形態で投与することができる。これらの化合物は、経口、直腸、経皮、皮下、静脈内、筋肉内、及び経鼻を含む様々な経路で投与することが可能であり、薬学的技術者が周知の方法で調整することができる。
Claims (20)
- 化学式(I)の化合物:
R1は、C1〜C4のアルキルであり;
Yは、H、CN、NO2、又はグアニジン保護基であり;
Zは、−CN、−C(=O)OR5、フタルイミド、−NHSO2R6、又はNR3R4であり;
R3は、H又はC1〜C4のアルキルであり、
R4は、アリール−SO2−、アリール−C(=O)−、アラルキル−C(=O)−、アルキル−C(=O)−、アリール−OC(=O)−、アラルキル−OC(=O)−、アルキル−OC(=O)−、アリール−NHC(=O)−、及びアルキル−NHC(=O)−の群から選択され、ここでアリール又はアラルキルは選択的に1若しくはそれ以上のメチル又はメトキシ基で置換されており;
R5は、H、C1〜C6のアルキル、C2〜C6のアルケニル、C2〜C6のアルキニルであって、ここでR5は選択的に1若しくはそれ以上のハロ、C1〜C4のアルキル、アリール、又はヘテロアリールによって置換されており;
R6は、C1〜C8のアルキル、C2〜C8のアルケニル、C2〜C8のアルキニルであって、ここでR6は選択的に1若しくはそれ以上のハロ、C1〜C4のアルキル、アリール、又はヘテロアリールによって置換されており;
Qは、B(OR9)2、ピナンジオールボロン酸エステル、ピナコールボロン酸エステル、1,2−エタンジオールボロン酸エステル、1,3−プロパンジオールボロン酸エステル、1,2−プロパンジオールボロン酸エステル、2,3−ブタンジオールボロン酸エステル、1,1,2,2−テトラメチルエタンジオールボロン酸エステル、1,2−ジイソプロピルエタンジオールボロン酸エステル、5,6−デカンジオールボロン酸エステル、1,2−ジシクロヘキシルエタンジオールボロン酸エステル、ビシクロヘキシル−1,1’−ジオールボロン酸エステル、及び1,2−ジフェニル−1,2−エタンジオールボロン酸エステルから選択されるものであり;
R9は、H、C1〜C4のアルキル、シクロアルキル、シクロアルキルアルキル、アリール、又はアラルキルであり;
mは2、3、又は4であり;さらに
nは3、4、5、6、7、8、9、10、11、12、13、又は14である、化合物。 - 請求項1の化合物において、R1はエチル、プロピル、又はブチルである。
- 請求項1の化合物において、R1は2−プロピルである。
- 請求項1の化合物において、Qはピナンジオールボロン酸エステルである。
- 請求項1の化合物において、Qはビシクロヘキシル−1,1’−ジオールボロン酸エステルである。
- 請求項1の化合物において、QはB(OH)2である。
- 請求項1の化合物において、Zはフタルイミド、NHR4、又はCNである。
- 請求項1の化合物において、nは8、9、10、又は11である。
- 請求項1の化合物において、mは3である。
- 請求項1の化合物において、nは8、9、10、又は11であり、さらにmは3である。

Yは−NO2であり;

QはB(OR9)2、ピナンジオールボロン酸エステル、ピナコールボロン酸エステル、1,2−エタンジオールボロン酸エステル、1,3−プロパンジオールボロン酸エステル、1,2−プロパンジオールボロン酸エステル、2,3−ブタンジオールボロン酸エステル、1,1,2,2、−テトラメチルエタンジオールボロン酸エステル、1,2−ジイソプロピルエタンジオールボロン酸エステル、5,6−デカンジオールボロン酸エステル、1,2−ジシクロヘキシルエタンジオールボロン酸エステル、ビシクロヘキシル−1,1’−ジオールボロン酸エステル、及び1,2−ジフェニル−1,2−エタンジオールボロン酸エステルであり;
R9はH又はC1〜C4のアルキルであり;
mは3であり;
nは4、5、6、7、8、9、10、又は11である、化合物。 - 化学式(I−s)の請求項1の化合物:
R1はC1〜C4のアルキルであり;
YはCN又はNO2であり;
Zは−CN、−C(=O)OR5、フタルイミド、−NHSO2R6、−NR3R4であり;
R3はH又はC1〜C4のアルキルであり;
R4はアリール−SO2−、アルキル−SO2−、アリール−C(=O)−、アラルキル−C(=O)−、アルキル−C(=O)−、アリール−OC(=O)−、アラルキル−OC(=O)−、アルキル−OC(=O)−、アリール−NHC(=O)−、及びアルキル−NHC(=O)−の群から選択されるものであって、ここで前記アリール又はアラルキルは選択的に1若しくはそれ以上のメチル又はメトキシ基で置換されており、
R5はH、C1〜C6のアルキル、C2〜C6のアルケニル、C2〜C6のアルキニルであって、ここでR5は選択的に1若しくはそれ以上のハロ、C1〜C4のアルキル、アリール、又はヘテロアリールで置換されており;
R6はC1〜C8のアルキル、C2〜C8のアルケニル、C2〜C8のアルキニルであって、ここでR6は選択的に1若しくはそれ以上のハロ、C1〜C4のアルキル、アリール、又はヘテロアリールで置換されており:
Qは、B(OR9)2、ピナンジオールボロン酸エステル、1,2−ビシクロヘキシルエタンジオールボロン酸エステル、及びビシクロヘキシル−1,1’−ジオールボロン酸エステルから選択されるものであり;
R9はH又はC1〜C4のアルキルであり;
mは2、3、又は4であり;さらに
nは3、4、5、6、7、8、9、10、11、12、13、又は14である、化合物。 - 請求項1〜13のうちいずれか1つの化合物、及び薬学的に許容可能な担体を含む組成物。
- プロテアソームの活性を阻害する方法であって、請求項1〜13のうちいずれか1つの化合物を前記プロテアソームと接触させる工程を有する、方法。
- 癌を治療する方法であって、請求項1〜13のいずれか1つの化合物の治療的有効量を前記癌を有するか又は前記癌にかかりやすい哺乳類に対して投与する工程を有する、方法。
- 癌を治療する方法であって、請求項1〜13のいずれか1つの化合物の治療的有効量を前記癌を有するか又は前記癌にかかりやすい哺乳類に対して投与する工程を有し、前記癌は、皮膚、前立腺、結腸直腸、膵臓、腎臓、卵巣、乳房、肝臓、舌、肺、及び平滑筋組織から選択されるものである、方法。
- 癌を治療する方法であって、請求項1〜13のいずれか1つの化合物の治療的有効量を前記癌を有するか又は前記癌にかかりやすい哺乳類に対して投与する工程を有し、前記癌は、白血病、リンパ腫、非ホジキンリンパ腫、骨髄腫、多発性骨髄腫から選択されるものである、方法。
- 癌を治療する方法であって、1若しくはそれ以上の抗腫瘍剤又抗癌剤、及び/又は放射線治療と組み合わせて、請求項1〜13のいずれか1つの化合物の治療的有効量を前記癌を有するか又は前記癌にかかりやすい哺乳類に対して投与する工程を有する、方法。
- 転写因子NF−κBの活性を阻害する方法であって、請求項1〜13のいずれか1つの化合物を転写因子NF−κBの阻害剤であるIκBと接触させる工程を有する、方法。
