Abstract
Description
本発明の方法を具現するために用いることのできる装置の実施形態は、米国仮出願60/423,368,米国出願番号10/358445、およびCordemansらに付与された米国特許6,540,922に見つけることができ、これらのそれぞれは、その全体を参照により本願に組み込む。高増殖性細胞の処理方法は、本発明の装置を用いて実施することができる。水性媒体(例えば生理学的流体)などの液体媒体を処理するために使用可能な装置の1つの特定の実施形態を図1に示す。ある実施形態において、処理すべき流体は高増殖性細胞を含む。他の実施形態において、処理すべき流体は、例えば診断後などに、高増殖性細胞を含む疑いのある生理学的液体であってもよい。ウィルスおよび/またはウィルス感染細胞を含んだ溶液のみならず、幹細胞などの完全に分化していない細胞も処理することができる。処理可能なウィルスの例としては、HIV、HCV、HBV、ヘルペスウィルス、ハンタウィルス、インフルエンザ、およびエボラなどが挙げられる。

(実施例1)
American Type Culture Collection(ATCC、Rockville、MD、USA)から入手したヒト白血病細胞系列(K562、Nalm−6、KGla、およびHL−60)を、10%仔牛血清(Gibco、Grand Island、NY、USA)および1%L−グルタミン(Gibco)を添加したRPMI−1640(BioWhittaker、Walkersville、MD、USA)中で培養した。白血病細胞を回収し、リン酸緩衝生理食塩水(PBS、pH=7.2、Gibco)中に再懸濁し、迅速に実験に使用した。ヘパリン添加静脈血は、健常ボランティアおよび白血病患者から得た。単核球を、Ficoll−Hypaque密度勾配遠心分離(International Medical Products、Brussels、Belgium)により分離した。
(実施例2)
DNA分断化はアポトーシスに関連している。分解したDNAによる細胞の定量化を、Nicoletti、IらによってJ Immunol Methods 139 (2):271(1991)に記載される方法、およびApotarget迅速DNAラダー検出キット(Biosource)を用いて行った。細胞ペレット(106細胞)を20Lの溶解バッファに再懸濁し、製品説明書に従ってDNAを抽出した。ゲル電気泳動(1%アガロース)によって分離した後、DNAを解析した。正のコントロールとして、プレートをUVトランスイルミネータの直下に10分間(5mW/cm2の強度)置くことにより、細胞にUV光を照射した。その後、細胞を37°Cで5時間および18時間インキュベートしてから、アポトーシスを調べた。
(実施例3)
細胞アポトーシスのいくつかのモデルにおいて、ミトコンドリア膜電位(ΔYm)の初期崩壊に続いて、DNA分断化の進行が観察された。ΔYmに対する高周波数超音波処理の影響を測定するために以下のアッセイを行った。
(実施例4)
アポトーシスの間にグルタチオンの減少がおこることが判っている。超音波処理後の細胞グルタチオン含量を測定するためのアッセイを行った。D.W.Hedleyら、Cytometry 15:349(1994)に記載されているように細胞内グルタチオンのレベルを測定するために、細胞トラッカーグリーンCMFDA(5−クロロメチルフルオレセインジアセテート;Molecular Probes)を用いた。
(実施例5)
カスパーゼ3は、化学療法誘導性アポトーシスにおいて重要な役割を果たすことが判っている。具体的には、カスパーゼの活性化は、アクチン、ゲルソリン、またはPARPなどの細胞性基質の開裂によって細胞死をもたらす。超音波誘導性アポトーシスにおけるカスパーゼ3の関与を直接調べるために、フローサイトメトリーおよび比色分析を用いてカスパーゼ活性を測定した。
(実施例6)
bcl−2ファミリーの様々なタンパク質が、アポトーシスの開始または阻止に関わっている。bcl−2ファミリーの2つの主要メンバーであるbcl−2とbaxとが超音波によるアポトーシスの誘導に関与しているかどうかを判定するために以下のアッセイを行った。浸透化処理後、細胞をアイソタイプが一致するネガティブコントロールであるFITC標識マウス抗ヒトbcl−2(Dako、Glostrup、Denmark)およびbaxに対するポリクローナルウサギ抗体とともにインキュベートした。次に、FITC標識二次抗体(Dako)をbaxに加えた。bcl−2およびbax発現を定量化するために、既知量の蛍光色素(Dako)で標識したビーズ混合物を用いて、血球計数器をキャリブレーションした。平均蛍光強度(MFI)の値を、キャリブレーション曲線を用いて相当する可溶蛍光色素分子(MESF)に変換した。
(実施例7)
アポトーシスの間、原形質膜の内側から外側へのホスファチジルセリン残基の反転、およびこの変化は、PS残基に結合するアネキシンFITCを用いて検出することができる。以下、実施したアネキシンV結合アッセイについて説明する。アネキシンVフルオレセインイソチオシアネート(FITC)およびヨウ化プロピジウム(PI)染色細胞のフローサイトメトリー分析は、Biosource International(Camarillo、CA、USA)から購入したキットを用いて、業者が推奨するようにして行った。データは、アネキシンV−FITC/ヨウ化プロピジウム(図示せず)の平均蛍光強度の変化を示す点プロットによって表した。
(実施例8)
細胞生存性への影響に対する重要な証拠は、細胞が増殖してコロニーを形成することができないことである。以下に、クローニング効率への高周波数超音波照射の影響を測定するためにK562細胞に対して行ったクローン形成法について説明する。簡単にいうと、培養培地は、20%FCSと最終濃度4%のメチルセルロースを添加したIMDMから成る。培養を5%CO2空気中、37°Cでインキュベートし、5日後にコロニー(>20細胞)を評定した。K562細胞系列のクローン形成効率は16%であった。図5に示すように、1回および3回の処理後、K562細胞のクローニング効率には有意な低下(それぞれ25%および42%の阻止)が見られたことから、高周波数超音波に対する白血病細胞の感受性が確認された。
(実施例9)
高周波数超音波によるアポトーシスの誘導に対する活性酸素スカベンジャーの影響を測定するために以下の手順を実施した。K562細胞をLヒスチジン(10mM)および/またはマンニトール(100mM)とともにインキュベートした。いくらかの細胞は高周波数超音波によって処理し、他のものは処理しなかった。細胞アポトーシスは、アネキシンV/PIアッセイによって検出した。表1に提供するような結果から、超音波によって誘導された細胞損傷は、ヒスチジンおよびマンニトールの存在下において著しく減少する(3回の連続処理によって誘導したアポトーシスをそれぞれ43%および47%阻止)ことが実証された。
(実施例10)
超音波処理後、0.5、2、および5時間培養したK562細胞に対してフローサイトメトリーによる追跡を行った。1回の処理後、アポトーシス細胞のレベルはコントロール細胞の3倍であった(5時間培養後、処理済みおよび未処理細胞に対して、それぞれ26%および8%)。5〜10%の壊死効果も観察され、これは薬物または光力学的処理(PDT)を用いた場合に見られるよりもはるかに下回る。同一条件(7mW/mL、20秒)かつ異なる間隔での連続した照射により、K562細胞のアポトーシスは、1回および3回の連続処理後に、それぞれ37±3%(p<0.02)および49±5%(p<0.02)に増加した(図6)。連続処理後には、形態学的変化(例えば、細胞収縮、膜気泡化、クロマチン濃縮)も観察された(結果示さず)。図7は、細胞ホスファチジルセリンの量が、アネキシンVアッセイによって検出されるように連続高周波数超音波処理後に増加していることを実証するものである。
(実施例11)
K562に加えて、KGla(未成熟の最小分化急性骨髄性白血病芽球)、HL−60 (前骨髄球性白血病)、およびNalm−6(すべての細胞系列)を含む他の正常および悪性細胞系列に対しても、超音波処理の影響を調べた。図8に示した結果は、超音波に対する感受性は細胞の種類に依存するが、連続処理により、評価したすべての細胞系列に対するアポトーシス細胞の数が有意に増加することが実証された。

- 細胞懸濁液の処理装置であって、
高増殖性、未分化またはウィルス感染した細胞の懸濁液からなる流体を収容する区画室と、
100kHzより高い周波数を有する超音波を、有意なキャビテーションを起こさせるまたは流体を有意に加熱するのに不十分な電力と持続時間で放出するように構成された放出器と、
区画室内の超音波場中に、1mm未満の平均径のマイクロバブルを放出するように構成されたマイクロバブル放出器とを含む装置。 - ガスマイクロバブルは、空気および酸素マイクロバブルからなる群より選択されることを特徴とする請求項1に記載の装置。

- ガスマイクロバブルの平均径は30μm未満であることを特徴とする請求項29に記載の方法。
- 区画室に放出される超音波は定常場現象を生まないことを特徴とする請求項29に記載の方法。
- 主として可視領域内の電磁照射線を有する光を超音波場内に放出することをさらに含む請求項29に記載の方法。
- 高増殖性細胞は、腫瘍細胞、骨髄細胞、幹癌細胞、および前癌細胞からなる群より選ばれることを特徴とする請求項29に記載の方法。
- 高増殖性細胞は、白血病細胞であることを特徴とする請求項29に記載の方法。
- 方法は生体外で実施されることを特徴とする請求項39に記載の方法。
- 1W/cm2未満で超音波放出器に電力を供給することをさらに含む請求項29に記載の方法。
