Abstract
【解決手段】 AAV及び哺乳動物神経系標的細胞の双方に外因性であるDNAを含むように修飾されたAAV由来ベクターを供給する工程及び前記ベクターを前記細胞に導入させる工程を含む方法。外因性DNAは、神経系障害の治療に有効な遺伝子産物をコードする遺伝子を含んでいることが好ましい。AAVベクターが組込まれるため、安定な長期発現が達成される。

Description
生体内の方法は、始めは主に向神経性単純ヘルペスウイルス(HSV−1)の使用に対するものであったが、HSVベクターは発現の不安定性及び野生型への復帰を含むいくつかの問題がある(下記参照)。最近の開発は、アデノウイルスベクターの使用であった。アデノウイルスベクターの実験は、マーカー遺伝子のラット脳への発現を示し、2ヵ月間持続したが、発現は劇的に低下した(Davidsonら(1993)Nature Genetics 3:219-2223)。ウイルスベクターの方法のほかに、他の研究者らは、カチオンリポソーム:プラスミド複合体の直接注入を用いてマーカー遺伝子の低レベル及び一過性の発現を得た(Onoら(1990)Neurosci.Lett.117:259-263)。
単純ヘルペスウイルス1型(HSV−1)のゲノムは、約70個の遺伝子を特徴とする約150kbの直鎖状二本鎖DNAである。多くのウイルス遺伝子は、ウイルスがその増殖能を失うことなく欠失される。“即時型”(IE)遺伝子が最初に転写される。他のウイルス遺伝子の発現を調節するトランス作用因子をコードしている。“初期”(E)遺伝子産物は、ウイルスDNAの複製に関与する。後期遺伝子は、ビリオン及びタンパク質の構造成分をコードし、IE及びE遺伝子の転写を止めるか又は宿主細胞タンパク質の翻訳を中断させる。
毒性及び複製能の低下したHSVが示されたが、なお危険な形に変異するか又は潜在ウイルスを活性化することができ、HSVが組込まないので長期発現を得ることは困難である。
アデノウイルスゲノムは、約36kbの二本鎖DNAからなる。アデノウイルスは、気道上皮細胞を標的にするが、神経細胞に感染することができる。
組換えアデノウイルスベクターは、遺伝子移入の伝達体として非分裂細胞に用いられた。これらのベクターは、アデノウイルスEla即時型遺伝子が除去されるがたいていの遺伝子が保持されるので、組換えHSVベクターと同じである。Ela遺伝子が小さく(約1.5kb)かつアデノウイルスゲノムがHSVゲノムのサイズの1/3であるので、アデノウイルスゲノム内に異種遺伝子を挿入するために他の必要でないアデノウイルスが除去される。
実際に、アデノウイルス感染に起因する疾患は、HSV感染によって誘発されるものより重篤ではなく、これはHSVベクターと比べて組換えアデノウイルスベクターの主な利点である。しかしながら、多くのアデノウイルス遺伝子の保持及び発現は、HSVベクターと共に記載されたものと同じ問題、特に受容細胞に対する細胞毒性の問題がある。

急性アデノウイルス感染中で他の病気中でない糞、眼及び呼吸試料から非病原性同時感染物質としてAAVが単離された。
同様に、潜在的AAV感染がヒト及び非ヒト細胞の双方に同定された。全体的には、ウイルス組込みは、細胞増殖又は形態に明らかな影響がないと考えられる。Samulski(1993)Curr.Op.Gen.Devel.3:74-80参照。
2つの主要なオープンリーディングフレームがある。左のフレームは、少なくとも4つの非構造タンパク質をコードする(Rep群)。2つのプロモーターP5及びP19があり、これらのタンパク質の発現を調節する。微分スプライシングの結果として、P5プロモーターは、タンパク質Rep78及びRep68の生産に対するものであり、P19プロモーターは、Rep52及びRep40に対するものである。Repタンパク質は、ウイルスDNA複製、ウイルスプロモーターから転写のトランス活性化及び非相同エンハンサー及びプロモーターの抑制に関与すると考えられる。
McLaughlinら((1988)J.Viol.62:1963-73)は2つのAAVベクターを調製した:dl52−91はAAVrep遺伝子を保持し、dl3−94はAAVコード配列の全てが欠失された。しかしながら、2つの145塩基末端反復及び更にAAVポリアデニル化シグナルを含む139塩基を保持するものである。制限部位は、シグナルの両側に導入された。
ネオマイシン耐性をコードする異種遺伝子が両方のベクターに挿入された。組換えAAVゲノムとの相補性によりウイルス株を調製し、トランスに消失しているAAV遺伝子産物を供給したが、それ自体大きすぎて詰込まれなかった。
残念ながら、ウイルス株は、おそらく欠陥ウイルスと補完ウイルス間の相同組換えの結果として野生型AAV(dl3−94の場合10%)で汚染された。
このようにしてベクターとヘルパーAAV間の配列相同性が実質的に除去されたので、検出可能な野生型AAVは、相同的組換えにより生じなかった。更に、ヘルパーDNA自体は、AAV末端がこの目的に必要であることから複製及び包膜されなかった。即ち、AAV系では、HSVと異なり、ヘルパーウイルスが完全に除去され、ヘルパーを含まないAAVベクター株が残った。
外因性DNAは、神経系障害の治療に有効な遺伝子産物をコードする遺伝子を含んでいることが好ましい。ある実施態様においては、この遺伝子は、神経系内の特定の種類の細胞又は領域に特異的なプロモーターに操作上結合される。AAVベクターが組込まれることから、安定な長期発現(例えば、7ヵ月より長い)が達成される。
本発明のベクターは、外因性DNAが導入されたアデノ随伴ウイルスの誘導体である。
野生型アデノ随伴ウイルスは既に欠損しており、即ち、溶菌感染にヘルパーウイルスの存在が必要であるが、ベクターが送達される被検者にベクターを補足することができるヘルペスウイルス又はアデノウイルス感染が隠れている可能性がある。この可能性を守るために、ベクターが救済の可能性を減じるように修飾されることは非常に好ましい。
理論では、そのような修飾は、1つ以上のウイルス遺伝子に対して点変異の形を取ることができ、その変異は遺伝子の発現を全く防止するか或いは非機能性の修飾遺伝子産物の発現を生じる。しかしながら、点変異は可逆的である。従って、所望されない各遺伝子が単純に欠失されることは好ましく、異種DNAのウイルスパッケージ内の部屋を生じる利点がある。
試験管内でベクターを増殖するために、感受性のある細胞はAAV由来ベクター及び適切なAAV由来ヘルパーウイルス又はプラスミドで同時トランスフェクトされる。好ましくは、ベクターは、AAVから実質的に複製及びパッケージングの認識シグナルのみ残る。
ベクターは、更に、パッケージング及び複製を妨害することなく異種DNAがクローン化される1つ以上の制限部位を含むことができる。好ましくは、少なくとも1つのユニークな制限部位が与えられる。ベクターは、また、遺伝子操作を容易にする1つ以上のマーカー遺伝子を含むことができる。適切なマーカー遺伝子としては、ネオマイシン及びヒグロマイシン耐性遺伝子、細菌lacZ及び蛍ルシフェラーゼ遺伝子が挙げられるが、これらに限定されない。
AAV由来ヘルパーウイルス又はプラスミドは、ベクター株を生産するために担持AAV遺伝子の発現時に、適切な宿主細胞中試験管内でベクターの複製及びパッケージングに必要なタンパク質を与えることができるウイルス又はプラスミドである。
好適実施態様においては、ヘルパーウイルス又はプラスミドは、ヘルパーDNAとベクターDNA間の組換えの危険を減じるように操作されたものである。最も望ましくは、ベクターDNAのAAV配列とヘルパーDNAのAAV配列間に実質的に配列相同性がない。例えば、ヘルパーDNAは、AAV逆方向末端反復がアデノウイルスのような他のウイルスの対応する配列(例えば、アデノウイルス5型DNA)で置き換えられるAAVでもよい。SamulskiらJ.Virol.63:3822-28参照。
また、他の好適実施態様においては、ヘルパーアデノウイルスは、56℃で30分間加熱不活性化することにより除去されるか又は塩化セシウム勾配の遠心分離により詰込まれたAAVベクターから分離される。
本発明に従って組換えDNA及びRNA分子を構築する基本的な手順は、Sambrook,J.ら,Moleular Cloning: A Laborarory Manual,Sec.Edit.,Cold Spring Harbor Press,Cold Spring Harbor,NY(1989)に開示されており、参考として本明細書に引用する。
本発明の“外因性DNA”は、AAV及び標的細胞の双方に外因性でなければならない。DNAは、合成DNA、相補的DNA、ゲノムDNA又はその組合わせであることができる。DNAは、ベクターに取込まれかつ標的細胞に送達されるならば、任意の配列又は長さを有することができる。典型的には、AAVのパッケージング制限があるために、外因性DNAは約10〜5,000塩基の長さを有する。好ましくは、DNAは100〜4,000塩基である。
パーキンソン病(PD)に対する現在の方法は、尾状核(CP)におけるドーパミン作動性神経刺激伝達を促進することに基づいている。治療の主力は、経口L−ドーパ(及び末梢脱炭酸酵素インヒビター)であり、神経支配のないCPにおいて内在性AADCによってドーパミンに変換される。別の薬理的方法としては、ブロモクリプチン及びアポモルフィンを含む直接ドーパミン作動薬及びドーパミン代謝酵素(例えば、モノアミンオキシダーゼ)インヒビター(MAOI)、例えば、デプレニルが挙げられる。これらの治療は、PD患者の短期の生活に著しい改善を示したが、病気は進行し、全ての患者が結局5〜10年間にわたる経口治療に対して不応性になる。
本発明は、HSV−1欠損ベクター方法より顕著な利点を有する。詳細には、欠損HSV−1ウイルスの復帰頻度は約10-5であり、生体内実験に必要とされたウイルスの量と共に十分な野生型ヘルペス感染を生じ実験動物の毒性及び死滅をもたらす。更に、最初の2週間の発現は高いが、2週間を超えるベクター遺伝子発現のレベルは、おそらく初期発現の5〜20%まで低下する。
生体内で線条体細胞をドーパミン作動性表現型に導入する上記の説明は、PDに対する遺伝子治療方法の最初の段階である。しかしながら、PDは、ドーパミンニューロンの80%が失われると症状を示すようになる。変性は進行的であり、神経支配のない患者はますます現在の治療全てに不応性になりかつ凍え及び完全な不動化を増すにつれて“オン−オフ”現象を示す。ドーパミン合成酵素を発現するウイルスベクターを用いて線条体細胞を導入することは純粋に改善する方法であり、基礎をなす病気の過程は衰えないで続くであろう。これを目的として、ドーパミン作動性ニューロンの変性を更に防止しかつ再生を促進する“神経防御又は向神経”因子を発現するベクターが構築された。
従って、PDに対する遺伝子治療は、GDNF(遺伝子バンクHUMGDNF02;受託番号第L19063号)、脳由来向神経因子(BDNF)、神経成長因子(NGF)(EMBL HSNGF2;受託番号第X53655号)及び/又はニュートロフィン(NT)−3(遺伝子バンクHUMBDNF;受託番号第M37762号)及びNT−4(遺伝子バンクHUMPPNT4P;受託番号第M86528号)を含むニュートロフィン因子ファミリーの他の因子の遺伝子のAAVによる送達を含むことができる。
従って、スーパーオキシドジスムターゼ(SOD1又はSOD2)(SOD−1に対して遺伝子バンクHUMCUZNDI,受託番号第M12367号; SOD−2に対してEMBL HSSOD2G,受託番号第X65965号)、カタラーゼ(EMBL HSCATR,受託番号第X04076号)及び/又はグルタチオンペルオキシダーゼ(MBL HSGSHPX,受託番号第Y00433号)の遺伝子を送達するためにAAVベクターを用いることが望まれる。
複雑部分発作及び詳細には側頭葉てんかん(TLE)は、てんかんの最も不応性形態の1つである。ある患者には抗てんかん薬(AED)が発作を制御するが、40%が多重AED治療にもかかわらず制御されないままである。これらの患者に対する本方法は、切除手術を検討するための段階的評価を行うことである。
典型的には、側頭葉てんかんは発作起点の位置(てんかん発作領域)として定義され、前方の海馬を含む内側の側頭葉が切除される。TLEは遺伝病ではなく、病因が確立されていないが、この病気は、興奮を亢進させるか又は抑制を遮断する介在による抑制に対する興奮の失調に起因して発作を生じ、逆に興奮の拮抗と抑制の亢進は抗てんかん作用を有する。TLEに対する遺伝子治療方法は、抑制を改善するものである。これを目的として、アデノシンA−1レセプター(遺伝子バンクS56143; 受託番号第S56143号)cDNAがAAVベクターに挿入された。アデノシンが脳の天然抗痙攣薬(A−1レセプターを介して作用する)であることが示されかつレセプターのレベルがてんかん発作領域で低下するので、この方法は抑制を亢進させ発作を防止すると思われる。
抑制を亢進させる関連方法は、ニューロン興奮性を変えるイオンチャンネル、詳細には、カルシウム活性化カリウムチャンネル及びATP感受性カリウムチャンネルを含む活性依存性チャンネルの発現を増強するものである。
補足的な方法は、興奮性レセプター、詳細には、NMDAR、mGluRを含むグルタミン酸塩レセプター及びAMPA及びカイニン酸塩の双方を含む向イオン性グルタミン酸塩レセプターに対するアンチセンスを発現するものである。我々はHSV−1ベクターを用いるGluR6の発現がてんかんを誘発することを示したので、これらの種類のレセプターに対するアンチセンスを発現するベクターは発作を抑制することができることを予想することは妥当なことである。
脳腫瘍は、子供及び中年の成人に非常に影響を及ぼす治りにくく一般には致死疾患である。現在、たいていの脳腫瘍の原因は不明であり、治療法は全く効果がなかった。最近、脳腫瘍の治療に可能な方法として遺伝子治療を用いる2つの方法があった。第1の方法では、分裂細胞でのみ複製することができる変異株HSVが用いられた。これは、健康な脳の非分裂細胞を回避しながら分裂腫瘍細胞の破壊をもたらさねばならない。これらのウイルスが実験動物における脳腫瘍増殖速度を減じるというある実験証拠があったが、結果は変化しうるものでありかつ所望されるほど印象的でなかった。この方法のヒト試験はなかった。
第2の方法は、単純ヘルペスウイルス1型からのチミジンキナーゼ(TK)遺伝子を複製欠損レトロウイルスベクターに挿入するものである。このレトロウイルスベクターは、TK遺伝子を分裂腫瘍細胞に伝達しただけで、遺伝子を非分裂腫瘍細胞或いは健康な脳組織に伝達できなかった。
非分裂細胞は、活性化薬剤によってさえ影響されない。腫瘍治療に対するこの方法は、現在NIHで臨床試験中である。
この治療による結果も動物実験において変化しうるものであったので、最近の実験は、複製応答能のあるレトロウイルスベクターの使用により動物における応答を改善することができることが示された。しかしながら、この方法は、大量の野生型レトロウイルスを直接ヒト患者に入れることが非常に危険なためにヒト疾患に適用できなかった。これは、野生型レトロウイルスを受容細胞のゲノムにランダムに組込んだ後に細胞毒性及び新規腫瘍の誘導の可能性のために以前には決してヒト患者に可能でなかった。
まず、非分裂腫瘍細胞及び正常細胞内で薬剤をリン酸化する能力は、腫瘍内で活性化薬剤の長いプールを生じなければならない。傍観者効果の観察を生じると、HSVTK遺伝子を含む非分裂腫瘍細胞は薬剤をリン酸化しなければならず、次に、これにより、ウイルス遺伝子を導入されなかった近くの分裂細胞に入る。
即ち、導入された非分裂細胞が逆効果を及ぼさないとしても、非分裂細胞は近くの非導入細胞の破壊を可能にする。この方法で、分裂細胞の大きな集団が破壊される。
本発明のベクターの標的細胞は、哺乳動物の中枢又は末梢神経系の細胞である。1実施態様においては、細胞は、試験管内で培養した細胞である。
他の実施態様においては、細胞は、ベクターが細胞に送達される時に生存している哺乳動物の一部である。哺乳動物は、送達の時に発達の段階、例えば、胚、胎児、乳児、幼若、成体であってもよい。
ベクターは、中枢神経系の細胞、末梢神経系の細胞又は双方に送達される。ベクターが中枢神経系の細胞に送達されると、脊髄、脳幹(髄質、脳橋及び中脳)、小脳、間脳(視床、視床下部)、大脳(線条体、大脳皮質又はこの皮質内の後頭葉、側頭葉、頭頂葉又は前頭葉)又はその組合わせの細胞に送達される。
同様に、末梢神経系内の感覚系及び/又は奏効器伝導路の細胞に送達される。
ベクターを末梢神経系に向けるためには脊髄に注入されるか又は更に限定されたPNS分布が探索される場合には、末梢神経節又は問題の身体部分の肌(皮下又は筋肉内)に注入される。
特定の状態において、ベクターは、脈管内方法により投与される。例えば、ベクターは、血液脳関門が妨害する状態において動脈内(頸動脈)に投与される。
そのような症状としては、脳梗塞(卒中)及び脳腫瘍が挙げられる。更に、全体的な送達の場合、ベクターは、マンニトールを含む高張液の注入によって達成された血液脳関門の“開放”中に投与される。静脈内送達と共に、使用者は、神経系以外の細胞にベクターを送達することを許容しなければならないことは当然のことである。
投与経路は、更に、直接可視化によるベクターの局所適用、例えば、表在性皮質適用又は他の非脳定位適用である。
ベクターを特定の種類の細胞、例えば、ニューロンに標的とするために、ベクターを細胞の表面レセプターに特異的に結合する回帰剤と結合することが必要である。即ち、ベクターは、ある種の神経系細胞がレセプターを有するリガンド(例えば、エンケファリン)に結合される。結合は、共有結合、例えば、グルタルアルデヒドのような架橋剤、又は非共有結合、例えば、アビジン化リガンドのビオチニル化ベクターへの結合とすることができる。共有結合の他の形はベクター株を調製するために用いたヘルペスウイルスを操作することにより得られるので、コードされたコートタンパク質の1つは未変性AAVコートタンパク質とリガンドが表面に曝されるようなペプチド又はタンパク質リガンドのキメラである。
標的細胞は、ヒト細胞又は他の哺乳動物の細胞、特に非ヒト霊長類及びげっ歯類(マウス、ラット、ウサギ、ハムスター)、食肉類(ネコ、イヌ)及び偶蹄類(ウシ、ブタ、ヒツジ、ヤギ、ウマ)目の哺乳類とすることができる。
外因性DNAが発現可能な遺伝子を含む場合、遺伝子は、天然に存在する遺伝子、天然に存在するポリペプチドをコードするにもかかわらず本来天然に存在しない遺伝子又はそのようなポリペプチドの認識可能な変異体をコードする遺伝子とすることができる。また、宿主細胞に見られるDNAに対して“アンチセンス”のmRNA又は宿主細胞でふつうに転写されたmRNAをコードすることができるが、アンチセンスRNA自体は機能タンパク質に翻訳できない。
遺伝子発現に必要とされる調節領域の正確な種類は、生物によって異なるものであるが、一般にはRNA転写を開始するプロモーターが含まれる。そのような領域は、TATAボックスのような転写の開始と関係した5’非コード配列を含めることができる。プロモーターは、構成性又は調節可能とすることができる。
プロモーターの調節可能性は、しばしば“オペレーター”と呼ばれる特定の遺伝子因子と結合され、インデューサー又はリプレッサーが結合する。オペレーターは、その調節を変えるように修飾される。1つのプロモーターのオペレーターが他のプロモーターに移されるハイブリッドプロモーターを構築することができる。
従って、プロモーターは、主として中枢神経系内又は主として末梢神経系内で活性なものとすることができ、両者の中で著しく活性とすることもできる。CNSで活性である場合には、脊髄、脳幹(髄質、脳橋、中脳又はその組合わせ)、小脳、間脳(視床及び/又は視床下部)、大脳(線条体及び/又は大脳皮質及び後者の場合、後頭葉、側頭葉、頭頂葉及び/又は前頭葉)又はその組合わせに特異的とすることができる。特異性は、絶対であっても相対的であってもよい。
一般に、組織特異プロモーターを見出すためには、その組織でだけ(又は主として)発現されるタンパク質を同定し、次に、そのタンパク質をコードする遺伝子を単離する。(遺伝子は、正常な細胞遺伝子又はその細胞に感染するウイルスのウイルス遺伝子とすることができる)。その遺伝子のプロモーターは、他の遺伝子に結合される際に所望の組織特異活性を保持すると考えられる。
プロモーターの組織特異性は、特定の遺伝子因子と結合され、修飾されるか又は第2プロモーターに移される。
(1)全ての種類の細胞における発現:
強力なウイルス(例えば、即時型CMV、Invitrogen,Inc.、カリフォルニア州、サンジエゴからプラスミドpCDNAlで入手)及び相対的に非特異的細胞プロモーター(例えば、β−アクチン、遺伝子バンクHUMACTBET,K00790)の両者が全ての種類の細胞において発現させるために用いられる。
(2)ニューロン特異的発現:
方法としては、ニューロン特異的プロモーター、例えば、ニューロン特異的エノラーゼ(EMBL HSENO2,X51956)、AADC、神経フィラメント(遺伝子バンクHUMNFL,L04147)、シナプシン(遺伝子バンクHUMSYNIB,M55301)及びセロトニンレセプター(遺伝子バンクS62283)プロモーター並びにニューロンに対する発現を制限するサイレンサー因子と共に広範囲に活性なプロモーターの組合わせの使用が含まれる。
方法としては、グリア線維酸タンパク質(GFAP)プロモーター(遺伝子バンクHUMGFAP,J04569)、S100プロモーター(遺伝子バンクHUMS100AS,M65210)及びグルタミンシンターゼ(EMBL HSGLUS,X59834)プロモーターの使用が含まれる。
(4)発現は下記の遺伝因子を用いて特定のニューロン分集団まで制限される:
ペプチド生産プロモーター:例えば、エンケファリン(遺伝子バンクHUMENKPH1,K00488)、プロジノルフィン、ソマトスタチン(遺伝子バンクPATSOMG,JOO787; 遺伝子バンクHUMSOM1,J00306); モノアミン生産プロモーター:チロシンヒドロキシラーゼ(遺伝子バンクM23597)、ドーパミンβ−ヒドロキシラーゼ(遺伝子バンクRATDBHDR,M96011)、PNMT(EMBL HSPNMTB,X52730); コリン作動性ニューロン:コリンアセチルトランスフェラーゼプロモーター(遺伝子バンクHUMCHAT1,M89915; EMBL HSCHAT,X56585)。
場合によっては、所望のRNA産物をコードする遺伝子配列に対する非コード領域3’が得られる。この領域は、終結及びポリアデニル化を与えるような転写終結調節配列に保持される。即ち、コード配列に自然に相接する3’領域を保持することにより、転写終結シグナルが与えられる。コード配列と自然に結合した転写終結シグナルが発現宿主細胞で巧く機能しない場合には、宿主細胞で機能する別の3’領域が置き換えられる。
他の実施態様においては、核酸分子は、標的生物(ウイルス及び他の病原体)のゲノム又は他のDNA配列に対して又は生物の細胞で転写されたメッセンジャーRNAに対して“アンチセンス”であり、十分にハイブリッド形成して標的ゲノムDNAの転写又は標的メッセンジャーRNAの翻訳を阻害する。そのようなハイブリッド形成の効率は、ハイブリッド形成配列の長さ及び構造の関数である。配列が長いほど及び完全化に対する相補性が近いほど、相互作用が強い。塩基対の不適正の数が増加するにつれて、ハイブリッド形成効率は低下する。更に、パッケージング配列DNA又はアンチセンスRNAのGC含量もAT(又はAU)塩基対と比べてGC塩基対に存在する水素結合のためにハイブリッド効率に影響する。即ち、GC含量が多い標的配列は標的として好ましい。
別の実施態様においては、遺伝子はリボザイム、即ち、好ましい酵素活性を有するRNAをコードする。
遺伝子を神経系に伝達するための本方法は、ウイルス遺伝子を保持する組換えウイルスベクター或いは残余の及び潜在的に危険なヘルパーウイルスを含む欠損ベクターを用いる。アデノ随伴ウイルス(AAV)ベクターは、ウイルス遺伝子の全部が除去され(ウイルスゲノムの96%)ヘルパーウイルスが完全に除去されているDNAを組込んでいる非病原性のベクターである。β−ガラクトシダーゼを発現するAAVベクターを、線条体、海馬及び黒質を含むラット脳領域に定位脳手術で注入した。ベクターDNA及び導入された遺伝子発現は、注入後1日から3ヵ月まで検出した。ヒトチロシンヒドロキシラーゼ(TH)を発現する第2ベクターを作成した。このベクター(AAVth)を一側性6−ヒドロキシドーパミン障害ラットの神経支配のない線条体に注入し、グリア及びニューロンの双方を含む線条体細胞でTH免疫反応性が4ヵ月まで得られた。AAVベクターで治療したラットに病変又は毒性の証拠はなかった。初期データは、AAVベクターを介した線条体におけるTH導入がAAVlac対照と比べて障害の生じたラットに顕著な行動に関する回復を生じることを示している。
プラスミド:プラスミドpSub201(Samulskiら(1989)J.Virol.63:3822-28)をXbalで消化してほぼ全体のAAVゲノムを除去し、末端反復のみ残した。CMVプロモーター−lacZ遺伝子−SV40ポリAシグナルカセットを、SpeI及びXbaIで消化することによりプラスミドpHCL(Kaplittら(1991)Mol.Cell.Neurosci.2:320-30)から単離し、これをZbaI消化p Su201に挿入してpAAlacを作成した。第2プラスミドを、pAAVlacをHindIII及びXbaIで消化することにより作成(pAAV−CMV−ポリA)してlacZ遺伝子及びポリAシグナルを除去し、続いてポリリンカー及びSV40ポリAシグナルを含むpREP4(Invitrogen)からのHindIII−XbaI断片を挿入した。次に、このプラスミドをHindIII及びBamHIで消化し、続いてpAAVthを作成するためにヒトチロシンヒドロキシラーゼ(hTH)cDNAを挿入した(O'Malleyら(1987)Biochemistry 26:6910-14)。
AAVベクターを作成するために、プラスミド(pAAlac又はpAAVth)をリン酸カルシウム法(Grahamら(1973)Virology 52:496-67)により、アデノウイルスElaタンパク質及びSV40T抗原の双方を構成的に発現する293T細胞、293細胞の変異体(Grahamら(1977)J.Gen.Virol.36:59-74),(D.Baltimoreから入手)に移した。ベクタープラスミドを、必要な複製及び構造タンパク質を与えるヘルパープラスミドpAd8と共に同時トランスフェクトした。翌日、細胞にアデノウイルス株dl309(Jones & Shenk(1978)Cell 13:181-88)(Thomas Shenkから入手、プリンストン大学)を感染させた。完全な細胞変性作用に続いて、凍結/解凍多サイクルによってウイルスを回収した。次に、残余アデノウイルスを不活性化するために、ウイルス株を56℃で30分間加熱した(Samulskiら1989)。ベクター力価は、X−galの組織化学分析(Kaplittら(1991)Mol.Cell.Neurosci.2:320-30)又はモノクローナル抗hTH抗体(Boehringer Mannheim)及びABCエリート検出システム(Vector Labs)を用いる、連続希釈のベクター株を感染させた293T細胞におけるhTH発現の免疫細胞化学同定により得た。
AAVlacを感染させた動物からの脳切片の分析に対して、組織を0.1M HEPES(pH7.3)中2%パラホルムアルデヒド/5mMEGTA/2mMMgCl2を用いて心臓内灌流で固定した。EGTAを添加すると内在性細胞酵素によるバックグラウンド染色が除去される。組織培養細胞をPBS(pH7.2)中2%ホルムアルデヒド/0.2%グルタルアルデヒドで固定した。β−ガラクトシダーゼ発現の検出用X−gal組織化学を以前に記載されたように行った。
脳切片を清浄剤バッファー(PBS中0.01%デオキシコール酸ナトリウム/0.02%NP−40)中に1時間入れた。PBSの洗浄に続いて、切片をアルコール中で脱水し、200μlのPCR反応バッファーを各スライドに加えた(PCR反応バッファー:1×PCRバッファー/1μM各プライマー/1M MgCl2/10μlジゴキシニン−dUTP(Boehringer))。lacZに特異的なプライマーは次の通りである。
N−末端 - 5' → 3'CCGACTGATGCCTTCTGAACAA (配列番号1)

雄のスプラグ−ダウレイラットを全ての実験に用いた。動物は、動物管理と使用のNIHガイドラインに従って取り扱った。手術手順として、動物をエンフルランとNO2の混合物で麻酔した。脳定位マイクロインジェクションを全ての脳領域注入に用い、Paxinos & Watsonの図、脳定位座標におけるラット脳、(Academic Press,オーストラリア、シドニー:1982)に従って座標を求めた。免疫細胞化学用の組織を切除し、速やかに封入剤を凍結した。5μm切片をクリオスタットと共に用い、切片を緩衝ホルマリン中に固定した。X−gal組織化学用組織を上記のように調製した。
以前に記載されたPereseら(1989)Brain Res.494:285-93,Duringら(1992)Exp.Neurol.115:193-99の方法を用いて一側性黒質障害を作成した。簡単に述べると、雄のスプラグダウレイラット290〜310gをキシラジン/ケタミンで麻酔し、コフ脳定位枠に入れた。頭骨を露出させ、左の黒質の上にバードリルで穴をあけた、ラムダ+3.5、L2.15。新たにつくった6−ヒドロキシドーパミン(PBS中2mlの0.1%アスコルビン酸中4mg)をハミルトン注射器に充填し、2分間かけて2つの位置に下げた。中間位置の座標は側方1.9及び腹側7.1mmで針の角度は吻側に面するが、外側の位置は側方2.3mm及び背側の腹面6.8mmで針の角度は外向きであった(Grahamら,(1977)J.Gen.Virol.36:59-74)。各々の位置に2mlを5分かけて注入し、針を更に5分間そのままにした後、更に5分かけて取り出した。
6−OHDA注入の10〜16日後にラットを試験した。半球状のロータメーターに入れ、Heftiら(1980),Brain Res.,195:123-27に記載されたようにアポモルフィン(1mg/kg)を投与した15〜20分後に体が完全に回転する総数を記録した。少なくとも2週間離した3回の試験の最低を用いて基準回転率とした。1分あたり10回転を超える安定な(25%未満の変動)非対称性回転挙動を一貫して示したラットをAAVlac或いはAAVth注入のためにランダムに選んだ。
ほぼ完全な病変の上記行動基準にかなったラットをケタミン/キシラジン(1kgにつき70mg/7mg)で麻酔し、コフ脳定位枠に入れた。頭骨を露出させ、Paxinos & Watson座標AP0.2、L2.6及びAP1.5、L2.0及びL3.0で神経支配のない線条体(左)の上をドリルで穴をあけた。AAVlac或いはAAVthをハミルトン注射器を用いてDV深さ5mmの3つの位置の各々に徐々に注入した。各注入容量は2mlであった。ラットについて手術の1及び2ヵ月後にアポモルフィン誘導回転行動を試験した。
脳切片の免疫組織化学(IHC)分析用に、ラットをクロラール水和物で深く麻酔した後に1M PBS(pH=7.3)、次に4%パラホルムアルデヒド(PF)で心臓内灌流を続けた。脳を切除し、4%PF、次に凍結保護剤として上昇方式のスクロース溶液(PBS中10/15/30%)に固定(3〜4時間)した。切片(7〜30mm)をクリオスタット(Reichert-Jung)内で切断し、ポリリシン被覆スライド上に取り付けた。我々は、特定のニューロン又はグリア分集団におけるTH陽性細胞の同時検出用二重標識プロトコールを用いた。切片をまず阻止バッファー(1M リン酸塩バッファー食塩水(PBS)中5%ヤギ血清(GS)/5%正常ウマ血清(NHS)中でインキュベートした。次に、切片を阻止バッファー(マウス抗TH[Boehringer Mannheim,1:200]、マウス抗NF[Sigma,1:400]、ウサギ抗TH[ChemiconInternational,1:3000]、ウサギ抗GFAP[メモリアルホスピタルの病理部からの寄贈、1:800]に希釈した一次抗体中室温で(2〜4時間)インキュベートし、阻止バッファーですすぎ、二次抗体(マウス抗テキサスレッド[ベクター、1:75又はビオチニル化ウサギ抗IgG[ベクター、1:400]中室温で(1時間)インキュベートした。切片をPBS中で洗浄し、アビジン−ニュートライトFITC[Molecular Probes Inc,1:400]中室温でインキュベートした。スライドをPBS/グリセロール(0.05:1)を用いてカバーガラスをかぶせ、−20℃で保持した。
脳に遺伝子移入するためのアデノ随伴ウイルス(AAV)ベクターの作成
細菌lacZ遺伝子を、AAVゲノムの末端間にプラスミドpsub201(Samulskiら(1989)J.Virol.63:3822-28)に挿入した。これらの末端は、AAVベクターへの切断及びパッケージングの認識シグナルを含んでいる。lacZ遺伝子は、細菌酵素β−ガラクトシダーゼをコードし、適切な基質との反応の際に不溶性の青色沈殿を生産する。ヒトサイトメガロウイルス(CMV)即時型プロモーターを用いて遺伝子発現させ、SV40ポリアデニル化シグナルをlacZ遺伝子の下流に入れた(図1)。細胞にpAAVlac及びAAV構造タンパク質を与えるがAAV末端を欠いてウイルスに詰込むことができない第2プラスミド、pAd8(Samulskiら1989)をトランスフェクトした。次に、同時トランスフェクトした細胞をアデノウイルス5型(Jones & Shenk(1978)Cell 13:181-188)に感染させて残りの複製及びパッケージング機械を与えた(図2)。得られた株は、詰込まれたAAVlacベクター(AAVlac)及び子孫ヘルパーアデノウイルスから構成され、ヘルパーウイルスは、次に、56℃で30分間加熱することにより除去した。アデノウイルスの完全除去は、このウイルス株に感染した1週間後に培養細胞中にウイルスプラークを検出できないことにより確認した。次に、AAVベクターを培養293細胞の感染、β−ガラクトシダーゼ発現の組織化学染色及び青色細胞の計数により力価測定した。感染の1日及び5日後に観察された細胞数に差がなく、ベクターの複製及び拡散がないことを示した。AAV認識シグナルのないlacZプラスミドを用いてこの工程を繰り返すと、得られた株を感染した後に陽性細胞は観察されなかった。これは、lacZ遺伝子が自律複製できないAAVウイルスに詰込まれ残余のアデノウイルスが完全に除去されたことを示している。
AAVベクターは成体ラット脳において異種遺伝子を伝達及び安定に発現することができる
AAVlacを、尾状核、扁桃、線条体及び海馬を含む成体ラット脳の種々の領域に定位脳手術で微小注入した。始めにラットを注入の1〜3日後の間に犠牲にし、X−gal組織化学用に切片を処理した。各領域内で陽性細胞を示した。遺伝子移入の脳への効率は、HSV又はアデノウイルスベクターを用いて以前に観察されたものと少なくとも等価であると思われた。
哺乳動物脳内のAAV遺伝子移入及び発現の長期安定性を分析するために、ラットの尾状核にAAVlacを注入し、手術の2〜3ヵ月後に犠牲にした。最初に、ポリメラーゼ連鎖反応(PCR)を脳切片内の使用に適応して原位置でウイルスベクターDNAの増幅及び可視化を可能にした。Nuovoら(1991)Am.J.Pathol.139:1239-44; Nuovoら(1993)PCR Meth,2:305-12; Flotteら(1993)Proc.Nat.Acad.Sci.USA 90:10613-17参照。2ヵ月後に細菌lacZ遺伝子を保持した脳内の多数の細胞を検出した。反対側の脳切片又はTaqポリメラーゼを存在させずに処理した切片に染色はなく、アデノウイルスのみ感染させた脳にも陽性細胞はなかった。次に、ほかのラットを犠牲にし、機能β−ガラクトシダーゼを含む細胞を同定するために、切片をX−gal組織化学用に処理した(Kaplittら(1991)Mol.Cell.Neurosci.2:320-30参照)。ベクター注入の3ヵ月後まで尾状核の注入領域内に陽性細胞を同定した。同時に被検ラットの中に行動に関する又は生理的異常は検出されず、脳切片はAAV遺伝子移入から生じる病変の証拠を示さなかった。
AAVベクターは6−OHDA障害の生じたラットの尾状核においてチロシンヒドロキシラーゼの発現を与える
パーキンソン病(PD)は、黒質線条体路の喪失を特徴とする神経変性疾患であり、尾状核におけるドーパミン作動性伝達を促進する治療に応答する(Yahr & Bergmann,Parkinson's Disease(Raven Press,1987),Yahrら(1969)Arch.Neurol.21:343-54)。実験動物において、チロシンヒドロキシラーゼを発現してジヒドロキシフェニルアラニン(L−ドーパ)を合成する遺伝的に修飾した細胞は、PDのげっ歯類モデルにおいて行動に関する回復を誘導する。(Wolffら(1989)PNAS(USA)86:9011-14; Freedら(1990)Arch.Neurol.47:505-12; Jiaoら(1993)Nature 262:4505)。別の方法は、直接生体内体細胞遺伝子移入の方法であり、新線条体の内在細胞はTHを発現するベクターを導入することによりL−ドーパ産生細胞に変換される。THを発現するHSV−1ベクターは、この方法が組織移植に対して生存可能な代替方法であることができることを示した。(Duringら,(1992)Soc.Neurosci Abstr.18:331-8)。しかしながら、HSV−1ベクターは、上述したように現在いくつかの制限がある。ヒトPD患者に治療上の有用性を有することができるベクターを作成するために、我々はヒトTHcDNA(II型)(O'MalleyらBiochemistry 26:6910-14)を我々のAAVベクター(AAVth)に挿入した。AAVthを詰込み、AAVlacに対して記載されたようにヘルパーウイルスを除去した。
更に、細胞変性効果の徴候もなかった。短期ラット(注入の1週間未満に試験した)に観察された変化は、PBS注入及びAAVlac注入ラットと同様の注入のわずかな針の傷だけであった。長期ラット(2ヵ月より長い)では、残存している針の跡は一貫して見えず、ニューロン損傷又は反応性神経膠症の証拠はなかった。また、被検ラットにおいて脳損傷の行動に関する又は肉眼的病変徴候もなかった。
単一AAVベクターから2つの遺伝子を発現することにより神経伝達物質ドーパミンの新規合成が得られる
ドーパミン合成は、2つの酵素、TH及び芳香族酸脱炭酸酵素(AADC)によって触媒される。THによって触媒された反応はL−ドーパの合成をもたらし、これはドーパミン合成において律速段階である。次に、ドーパミンはAADCによってL−ドーパの変換から生じる。線条体はTHを内在的に生産する細胞を含まないが、AADCを生産する線条体細胞が少しある。従って、AAVth(又はTHのみを用いる他の方法)で治療したラットにおける行動に関する回復は、おそらく得られたL−ドーパのドーパミンへの内在性線条体AADCによる変換に次いで現れる。しかしながら、限られた数の細胞がAADCを生産するので、ドーパミン合成がすべての導入細胞においてTH及びAADC双方の発現によって促進されることは可能である。この方法で、標的細胞は、遺伝子移入後に自律ドーパミン産生細胞になる。実際に最近の証拠は、神経支配のない線条体において両方の遺伝子の発現がTHのみの発現より優れていることが示されている(Kangら1993)。更に、細胞移植後の最も実質的な行動に関する回復は、TH発現筋肉細胞が用いられた場合に現れ、初期の研究からの線維芽細胞と異なり、筋肉細胞が内在性AACD活性を発現する。これにより、TH及びAACDの両方を含むAAVベクターの作成が価値のあることが示された。
パーキンソン病の霊長類モデルにおけるAAVベクター仲介遺伝子治療
パーキンソン病に対する治療剤としての二シストロン性ベクターの大きな可能性は、霊長類実験の急速な開始に導いた。パーキンソン病の霊長類モデルは、ヒト臨床試験に入る前の潜在的治療の評価のためのとても貴重な標準モデルであると考えられる。このモデルは、始めは若い人々のグループが特発性パーキンソン病と著しく類似の神経変性疾患を発症していた1980年代の初期に所見から開発された。この疾患の原因は、麻薬の使用までさかのぼり、特定の原因物質が1−メチル−4−フェニル−1,2,3,6−テトラヒドロピリジン(MPTP)であることが判明した(Langston(1985)Trends Pharmacol.Sci.6:375-378)。次に、MPTPを霊長類に投与すると、動物は抗パーキンソン剤を試験する主なモデルになったパーキンソン障害を発症した。末梢に投与されたMPTPは血液脳関門を横切り、そこでモノアミンオキシダーゼB(MAO−B)によりMPP+に変換される。次に、この化合物は、エネルギー依存性前シナプス取込み機序を介して黒質のドーパミン作動性ニューロン内で選択的に濃縮される。これは、黒質ニューロン内に見られるニューロメラミンのMPP+結合能によって促進される(D'Amatoら(1986)Science 231:987-989)。MPP+は、強力な神経毒であり、最後にはパーキンソン病に見られるように黒質ドーパミン作動性ニューロンの退行変性及び黒質線条体のドーパミン経路の喪失を引き起こす(Redmondら(1993)Ann.N.Y.Acad.Sci.695:258-266; Tipton & Singer(1993)J.Neurochem.61:1191-1206)。
AAVベクターからの成長因子の発現によりニューロン病変後の機能回復がもたらされる
パーキンソン病の治療に対する代替方法としてほかのAAVベクターを開発した。いままで、PDに対する大多数の治療方法は、線条体ドーパミンレベルを上げるときに集中した。動物モデルの行動に関する回復が繰り返し示されたが、これは病気の治療でなくむしろ症状の改善である。黒質におけるニューロン変性は疾病過程の症状の結果であり、神経変性の進行は線条体ドーパミンを増加させることによって変わらない。しかしながら、最近、グリア由来向神経因子(GDNF)のような成長因子が黒質のニューロンを保護しかつそれに向性であることができることをいくつか報告された(Linら(1993)Science 260:1130-1132)。従って、AAVベクターは、CMVプロモーターの制御によってGDNF用cDNAを含むAAVベクターが作成された。
Claims (26)
- アデノ随伴ウイルス及び哺乳動物神経系標的細胞の双方に外因性であるDNAの送達方法であって、前記外因性DNAを含むように修飾されたアデノ随伴ウイルス由来ベクターを供給する工程及び前記ベクターを前記細胞に導入させる工程を含む方法。
- 該外因性DNAが発現可能な遺伝子を含み、前記遺伝子が構成的に或いは調節可能な条件下で前記標的細胞において発現される請求項1記載の方法。
- 該発現可能な遺伝子が、前記細胞に対して内在性の遺伝子から転写されたメッセンジャーRNAについてアンチセンスであるメッセンジャーRNAをコードする請求項2記載の方法。
- 該発現可能な遺伝子が、チロシンヒドロキシラーゼ及び芳香族アミノ酸脱炭酸酵素からなる群より選ばれたタンパク質をコードする請求項2記載の方法。
- 該発現可能な遺伝子が、中脳ドーパミン作動性ニューロンに特異的な向神経因子をコードする請求項2記載の方法。
- 該発現可能な遺伝子が、フリーラジカルのレベルを低下させる酵素をコードする請求項2記載の方法。
- 該発現可能な遺伝子が、GABA分泌活性を亢進させるタンパク質をコードする請求項2記載の方法。
- 該発現可能な遺伝子が、ニューロン興奮性を変えるイオンチャンネルを調節するタンパク質をコードする請求項2記載の方法。
- 該アンチセンスRNAが、興奮性レセプターに翻訳可能である該mRNAに対してアンチセンスである請求項3記載の方法。
- 該発現可能な遺伝子がチミジンキナーゼをコードする請求項2記載の方法。
- 該ベクターが発現可能形態のAAV遺伝子を含まない請求項1記載の方法。
- 該ベクターが実質的にAAVの逆方向末端反復のみ保持する請求項1記載の方法。
- 該発現可能な遺伝子がコード配列及び前記コード配列に発現可能に結合した調節配列を含み、もって、前記調節配列が活性化されると、メッセンジャーRNA転写物が前記コード配列から転写される請求項2記載の方法。
- 前記調節配列が該発現を神経系特異的にする請求項13記載の方法。
- 1 該発現が中枢神経系特異的である請求項14記載の方法。
- 該発現が、脊髄、脳幹、小脳、間脳及び終脳からなる群より選ばれた1種以上のCNS成分に特異的である請求項15記載の方法。
- 該発現がニューロン特異的である請求項13記載の方法。
- 該発現がグリア特異的である請求項13記載の方法。
- 該標的細胞が、霊長類、げっ歯類、食肉類及び偶蹄類からなる群より選ばれた哺乳類目の哺乳動物細胞である請求項1記載の方法。
- 該標的細胞がヒト細胞である請求項19記載の方法。
- 該標的細胞が細胞培養内である請求項1記載の方法。
- 該標的細胞が生存している哺乳動物内である請求項1記載の方法。
- 該ベクターが該哺乳動物の実質的に全ての神経系細胞に送達される請求項20記載の方法。
- 該ベクターが該哺乳動物の該神経系の特定の成分に特異的に送達される請求項20記載の方法。
- 神経系の遺伝的に決定された、素因のある又は罹患した疾病の予防、治療又は改善方法であって、請求項1記載の方法で外因性DNAを該神経系の細胞に送達する工程を含み、前記送達が該神経系の前記遺伝的に決定された、素因のある又は罹患した疾病を予防、治療又は改善するように前記外因性DNAが選ばれる方法。
- AAVの複製及びパッケージングシグナルのみ保持しかつチロシンヒドロキシラーゼ及び芳香族アミノ酸脱炭酸酵素を含むAAV由来ベクター。
