Abstract
Description
a)前記液体の第1の試料を、ホモシステインおよびシステインの両方を基質として利用するデスルフラーゼで処理し、前記デスルフラーゼの少なくとも1つの生成物のレベルを測定してシステイン(tCys)およびホモシステイン(tHcy)の濃度の和を決定すること;
b)前記液体の第2の試料を、ホモシステインを特異的に基質として利用するデスルフラーゼで処理し、少なくとも1つの生成物のレベルを測定してホモシステインの総濃度(tHcy)を決定すること、および
c)ステップb)で得られたtHcyをステップa)で得られたtCysプラスtHcyから差し引いて、総システイン濃度を得ること。
Claims (10)
- 生物学的液体において総システイン濃度(tCys)を測定する方法であって、
a)前記液体の第1の試料を、ホモシステインおよびシステインをいずれも基質として利用する、非特異的デスルフラーゼで処理すること、および前記デスルフラーゼの少なくとも1つの生成物のレベルを測定してシステイン濃度(tCys)およびホモシステイン濃度(tHcy)の和を決定すること;
b)前記生物学的液体の第2の試料を、ホモシステインを特異的に基質として利用する、ホモシステイナーゼで処理すること、および少なくとも1つの生成物のレベルを測定して総ホモシステイン濃度(tHcy)を決定すること;および
c)ステップb)で得られたtHcyをステップa)で得られた(tCys+tHcy)から差し引いて総システイン濃度を得ること、
を含んでなる方法。 - ステップa)およびステップb)で測定される生成物が硫化水素である、請求項1に記載の方法。
- 酸化剤およびジアルキルフェニレンジアミンで処理することによって硫化水素が測定される、請求項2に記載の方法。
- 前記酸化剤が鉄(III)イオンである、請求項3に記載の方法。
- 前記非特異的デスルフラーゼがP. putidaから分離可能である、請求項1に記載の方法。
- 前記ホモシステイナーゼがT. vaginalisから分離可能である、請求項1に記載の方法。
- 前記生物学的液体が血清もしくは血漿である、請求項1に記載の方法。
- 適当な容器内に、システインおよびホモシステインをいずれも基質として利用する非特異的デスルフラーゼ、ならびに、ホモシステインを特異的に基質として利用するホモシステイナーゼを、アッセイを実施するための使用説明書とともに含んでなる、総システイン濃度を測定するためのキット。
- ジスルフィド結合を還元するために有効な還元剤をさらに包含する、請求項8に記載のキット。
- 少なくとも1つの生成物を検出するための試薬をさらに包含する、請求項8に記載のキット。
