Abstract
Description
本発明は、癌の治療に使用するための修飾サイトカイン類に関する。さらに詳細には、本発明は、腫瘍血管および抗原提示細胞を「ホーミング(homing)」することができるサイトカイン誘導体に関する。

幾種かのサイトカインの抗腫瘍活性は周知であり、記述されている。幾種かのサイトカイン類は、ヒトでも既に治療に使用されている(29)。たとえば、インターロイキン−2(IL−2)およびインターフェロンa(IFNa)等のサイトカイン類は、異なる型の腫瘍、たとえば、腎転移性癌、毛様細胞白血病、カポジ肉腫、黒色腫、多発性骨髄腫、等々を有する患者で、確然たる抗腫瘍活性を示した。IFNB、腫瘍壊死因子(TNF)、TNFB、IL−1、IL−4、IL−6、IL−12、IL−15およびコロニー刺激因子(CFS)のような、他のサイトカイン類は、幾つかの型の腫瘍に対して、ある一定の抗腫瘍活性を示し、従って、さらなる研究対象である。

一般に、サイトカイン類の治療的使用は、全身性毒性のため、強度に限定される。たとえば、TNFは、当初、幾種かの腫瘍の出血性壊死を誘導できる力(1)、および、異なる腫瘍系に対するin vitro細胞傷害作用(2)を求めて発見されたが、その後、過剰産生状態の場合には、人体に対して危険な影響を及ぼすこともある、プロ炎症活性を有することが証明された(3)。

全身性毒性が、ヒトにおける薬理学有効量のサイトカイン類の使用に伴う基本的な問題であるため、治療効果を維持しながら、この部類の生物学的エフェクターの毒作用を減少させるために、新規な誘導体および治療戦略を現在評価中である。

幾つかの新規なアプローチは、以下のことに関する。
a)TNFを腫瘍に送達し、局所濃度を高めることができる融合タンパク質の開発。たとえば、TNFおよび腫瘍特異的抗体からなる融合タンパク質が産生される(4)。
b)抗腫瘍活性を維持し、且つ全身性毒性を低減されたTNF突然変異体の開発。従って、唯一の受容体(p55またはp75)を選択的に認識することができる突然変異体が既に作製されている(5)。
c)TNFの抗腫瘍活性を損なわずに、毒作用を幾らか減少させることができる、抗TNF抗体の使用。このような抗体は、既に、文献に記述されている(30)。
d)より高い半減期を有するTNF誘導体(たとえば、ポリエチレングルコールとコンジュゲート(conjugate)した、TNF)の使用。

腫瘍部位を選択的に標的とすることができるTNF誘導体の作製について、最近報告された。たとえば、抗トランスフェリン受容体mAbの重鎖の遺伝子とTNF遺伝子とを融合することによって得られる融合タンパク質(4)、または、TNFと腫瘍関連TAG72抗原に対するモノクローナル抗体の「ヒンジ」領域との融合タンパク質(6)、またはFv−TNF融合タンパク質(6)が記載されている。

欧州特許EP 251 494号には、アビジンまたはストレプトアビジンとコンジュゲートした抗体、コンジュゲートした抗体を複合体化することができる薬剤、および、抗体により認識される標的細胞に対するビオチン−ストレプトアビジン相互作用によって、治療薬または診断用薬を集中させることができるように、逐次的に且つ十分に遅らせて投与されるビオチンと結合した診断用薬または治療薬からなる化合物を含む、診断用薬または治療薬を投与するためのシステムが開示されている。記載されている治療薬または診断用薬は、金属キレート、特に、シスプラチナム、ドキソルビシン等々の、放射性核種および低分子量抗腫瘍薬のキレートを含む。

欧州特許EP 496 074号には、ビオチニル化した抗体、アビジンまたはストレプトアビジンおよびビオチニル化した診断薬または治療薬を逐次投与できる方法が開示されている。リシンのような細胞傷害剤が総括的に記述されているものの、放射標識化合物に関連のある利用が、主として開示されている。

WO95/15979号には、リガンドまたは抗リガンドとコンジュゲートした特異的標的分子を含む第1のコンジュゲートを投与し、その後、抗リガンドまたはリガンドにコンジュゲートした有毒な薬剤からなる第2のコンジュゲートを投与することに基づく、極めて有毒な薬剤を、細胞標的上に集中させる方法が開示されている。

WO99/13329号には、上記分子と、NGR受容体のリガンドとのコンジュゲートに基づいて、分子を、腫瘍血管形成性血管に向けて送る方法が開示されている。多数の分子が可能な候補であると示唆されているが、ドキソルビシンのみについて具体的に記述されている。NGR受容体のリガンドを、免疫応答を誘導するサイトカイン媒体として使用することについては、全く開示されていない。

アミノペプチダーゼ−N受容体(CD13)のリガンドと結合させることによって、ある特定のサイトカインの治療係数を顕著に改善できること、また、それらの免疫療法特性を増強できることが、意外にも判明した。CD13は、様々な種で高度に保存されている150kDaの貫膜糖タンパク質である。CD13は、正常細胞上、ならびに骨髄性腫瘍系統、血管形成性内皮および一部の上皮で発現される。CD13受容体は、そのペプチドリガンドが、アミノ酸性「NGR」モチーフを共有するという理由で、通常、「NGR」受容体として同定される。

第1の態様によれば、本発明は、TNFおよびIFN7から選択されるサイトカインと、CD13受容体のリガンドとのコンジュゲーション(conjugation)生成物を提供する。上記リガンドは、抗体であってもよく、そのフラグメント、たとえば、Fab、Fv、一本鎖Fv、任意に、修飾された、天然ではないアミノ酸を含む、CD13受容体に結合することができる、ペプチドまたはペプチド擬似物、すなわち、ペプチド様分子であってもよい。リガンドは、CNGRCVSGCAGRC、NGRAHA、GNGRG、シクロCVLNGRMECまたはシクロCNGRC等の、NGRモチーフを含む直鎖状または環状のペプチドであることが好ましく、ペプチドCNGRCであることがさらに好ましい。このペプチドは、サイトカインに直接結合させてもよく、またはアミノ酸1個、アミノ酸配列または有機残基、たとえば、6−アミノカプリル−N−ヒドロキシスクシンイミドであってもよい、スペーサーを介して、間接的に結合させてもよい。結合手順は、当業者に周知であり、遺伝子工学または化学合成技術を含む。

ペプチドリガンドは、サイトカインN末端に連結している、従って、修飾サイトカインが、その受容体に結合する際の妨害を最小限に抑えることが好ましい。あるいは、ペプチドは、分子上の天然の、または、遺伝子工学技術で人工的に挿入された、アミド結合またはカルボン酸結合アクセプターである、アミノ酸残基に連結されていてもよい。修飾サイトカインは、このペプチドをコードする5’隣接配列を含むcDNAを使用して作製することが好ましい。

好ましい実施形態によれば、TNFとCNGRC配列との間にコンジュゲーション生成物が提供される。さらに好ましくは、TNFのアミノ末端は、スペーサーG(グリシン)を介して、CNGRCペプチドに連結されている。

結果として得られる生成物(NGR−TNF)は、RMA−Tリンパ腫動物モデルで、TNFより活性であった。さらに、NGR−TNFを投与した動物は、RMA−T細胞またはRMA細胞の、さらなる腫瘍形成性用量を排除することができた。免疫適格動物では、通常のTNFと比較して、抗腫瘍活性の増大が確認できたが、免疫不全動物では確認できなかった。このことから、「NGR」ペプチドと結合したTNFの抗腫瘍活性の増大は、結合体の直接的な細胞傷害活性よりむしろ、免疫応答増強に起因することがわかる。

NGR−TNFによって誘導されるin vivo免疫作用は、CD13受容体に直接関係していることも証明されている。たとえば、CD13受容体に対する抗体ならびにGNGRCリガンドは、in vivoでNGR−TNFと競合することが観察されていて、従って、NGR−TNFによる受容体ターゲティングの機序が示唆される。

2つのTNF受容体、p75TNFRおよびp55TNFRの1つに選択的に結合することができるTNFの突然変異体形を使用することによって、TNF/CD13リガンド結合体の治療係数をさらに向上させることができる。上記TNF突然変異体は、位置指定突然変異誘発によって得ることができる(5;7)。

本発明のさらなる実施形態は、CD13リガンドで修飾されたサイトカインが、腫瘍抗原または他の腫瘍血管形成性マーカー、たとえば、αvインテグリン、メタロプロテアーゼまたは血管成長因子に対する抗体またはそれらのフラグメント、または細胞外マトリックスの成分に対する抗体またはそれらのフラグメント、たとえば、抗テネイシン抗体または抗フィブロネクチンEDBドメインとコンジュゲートしている、二官能性誘導体によって実現される。TNFと、胃および卵巣の腺癌によって発現される腫瘍関連TAG72抗原に対するmAbのヒンジ領域との間の、融合生成物の作製が最近報告された(6)。

本発明のさらなる実施形態は、ビオチン/アビジン系を用いた腫瘍のプレターゲティングによって実現される。このアプローチによれば、1)ビオチニル化mAb、2)アビジン(またはストレプトアビジン)および3)CD13リガンドおよびビオチンで修飾された二価サイトカインによって形成される、異なるステージの、腫瘍の抗原性部位上に、三元複合体が得られる。免疫結合体を用いた従来のターゲティングと比較して、プレターゲティングアプローチは、標的にホーミング(自動誘導)された活性分子と自由活性分子との比率を実際に高めることができ、従って、投与毒性を減少できることが、多数の論文で証明されている(11,10,9,8)。このアプローチは、in vitroで細胞傷害性を誘導することができ、また、正常なTNFが不活性な条件で腫瘍細胞成長を低減させることができる、ビオチニル化TNFで、良好な結果をもたらした(14,26)。腫瘍抗原および修飾サイトカインに同時に結合する二重特異性抗体を使用することによって、2相法で、プレターゲティングアプローチを実行することもできる。最近、癌胎児抗原に対する二重特異性抗体およびTNFの使用が、TNF腫瘍プレターゲティングのための手段として記述された(31)。

さらなる実施形態によれば、本発明は、異なるTNFサブユニット上において、CD13リガンドと抗体またはそのフラグメント(抗体またはそのフラグメントは、腫瘍細胞上に発現される抗原、または、腫瘍間質の他成分、たとえばテネイシンおよびフィブロネクチンEDBドメインに対する)との両者に、(直接的に、またはビオチン−アビジン架橋を介して間接的に)結合したTNF分子を含む。この結果、修飾サイトカインの腫瘍誘導特性がさらに向上し、後者は、三量体−単量体−三量体転移を介して、腫瘍微小環境にゆっくり放出される。事実、先の研究で示した通り、TNF結合体の修飾サブユニットは、非修飾三量体TNF分子を形成するために、標的複合体から解離して再会合することができ、次いで、腫瘍微小環境に拡散する。生物活性なTNFの放出は、ターゲティング後24〜48時間以内に起きることが証明されている(21)。

治療で使用するために、本発明の修飾サイトカイン類を、経口または非経口投与向けの医薬品に、適当に製剤化する。非経口投与向けの製剤が好ましく、注射可能な溶液または懸濁液および注入用液を含む。非経口向け製剤の場合、任意に、可溶化剤、等張化剤、保存料、安定剤、乳化剤または分散剤等の賦形剤を加えて、有効成分の有効量を無菌担体中に溶解または懸濁させ、続いて、密閉したバイアルまたはアンプルに分配する。

サイトカイン類をリポソームの形態で製剤化することにより、その生物活性を向上させることができる。事実、TNFアミノ基のアシル化によって、in vitro生物活性を失わずに、疎水性が上昇することが確認されている。さらに、脂質に結合したTNFは、影響を受けないin vitro細胞傷害性、免疫調節作用、および低減したin vivo毒性を有することが報告されている(12,13)。

ヒトでのボーラスTNFの最大耐量は、動物での有効用量より約10倍低い、218〜410μg/m2である(32)。ネズミモデルのデータに基づけば、ヒトで抗腫瘍作用を達成するためには、少なくとも10倍高い用量が必要であると考えられる(15)。高温摘出肢灌流を用いた第1の臨床試験において、メルファランおよびインターフェロンγと併用したTNF 4mgという独特の用量で高い応答率が得られた(16)。その他の研究で、インターフェロンγを省略できること、および治療応答を誘導するのに、さらに低用量のTNFで十分なことが証明されている(17,18)。2種のサイトカインは、内皮細胞に対して相乗効果を発揮するため、それらを組合せた、選択的ターゲティングは、より強い抗腫瘍活性を示す公算が高く、従って、同じサイトカイン類を併用した癌療法で遭遇する全身毒性の問題を克服することが可能である。さらに、TNFは、内皮裏打血管のバリヤー機能を低下させ、従って、それらの巨大分子透過性を高められることが判明している。本発明による修飾TNF分子を用いた治療が低毒性であること、およびそれらの腫瘍血管ホーミング特性を利用して、代替用途は、治療向けまたは診断向けの、他の化合物の腫瘍血管透過性を高めるためのそれらの使用である。たとえば、腫瘍のラジオイムノシンチグラフィーまたは放射免疫療法で、修飾TNFを使用して、腫瘍による放射標識抗体またはホルモン類(腫瘍画像化用化合物)の取り込みを増加させることができる。あるいは、化学療法薬、免疫毒素、薬剤または遺伝子を含むリポソーム、または他の抗癌薬の取り込みも増加させることができ、それらの抗腫瘍作用が増強される。

従って、癌の治療または診断で、本発明のサイトカイン類を、複合製剤で、分離製剤で、または、逐次的製剤で、他の診断用物質または治療用物質とも一緒に、使用することができる。

ネズミTNFおよびNGR−TNFの作製
ネズミ組換えTNFおよびCys−Asn−Gly−Arg−Cys−Gly−TNF(NGR−TNF)は、大腸菌における細胞質cDNA発現によって産生した。ネズミMet−TNFl〜156をコードするcDNA(20)は、
5’−CTGGATCCTCACAGAGCAATGACTCCAAAG−3’および
5’−TGCCTCACATATGCTCAGATCATCTTCTC−3’を、
3’および5’プライマーとして使用し、リポ多糖刺激ネズミRAW−264.7単球−マクロファージ細胞から単離されたmRNAを用いて、逆転写酵素−ポリメラーゼ連鎖反応(RT−PCR)によって作製した。

この増幅フラグメントを、Nde IおよびBam HI(New England Biolabs,Beverley,MA)で消化し、同一酵素で予め消化したpET−11b(Novagen,Madison,WI)にクローニングした(pTNF)。

5’プライマーとしての5’−GCAGATCATATGTGCAACGGCCGTTGCGGCCTCAGATCATCTTCTC−3’、および上記3’プライマーを使用し、pTNFを用いて、PCRにより、Cys−Asn−Gly−Arg−Cys−Gly−TNF1〜156をコードするcDNAを増幅した。上述の通りに、この増幅フラグメントを消化し、pET−11bにクローニングして、BL21(DE3)大腸菌細胞(Novagen)の形質転換に使用した。pET11b製造業者の説明書に従って、イソプロピル−β−D−チオガラクトシドで、TNFおよびNGR−TNFの発現を誘導した。細菌を2mMエチレンジアミン四酢酸、20mM Tris−HCl、pH8.0中で超音波処理し、続いて遠心分離(15000×g、20分、4℃)することにより、2リットル培養から、可溶性TNFおよびNGR−TNFを回収した。両抽出液を硫酸アンモニウム(飽和の25%)と混合し、4℃で1時間放置し、さらに、上述の通りに遠心分離した。次いで、上澄中の硫酸アンモニウムを飽和の65%とし、4℃で24時間放置し、さらに遠心分離した。各ペレットを、200mlの1M硫酸アンモニウム、50mM Tris HCl、pH8.0に溶解し、Phenyl−Sepharose 6 Fast Flow(Pharmacia−Upjohn)を用いた疎水性相互作用クロマトグラフィで精製した(勾配溶出、緩衝溶液A:1M硫酸アンモニウムを含む、50mMリン酸ナトリウム、pH8.0;緩衝溶液B:20%グリセロール、5%メタノール、50mMリン酸ナトリウム、pH8.0)。TNF免疫反応性物質を含む分画(ウエスタンブロッティングによる)をプールし、2mMエチレンジアミン四酢酸、20mM Tris−HCl、pH8.0に対して透析し、DEAE−セファロースFast Flow(Pharmacia−Upjohn)を用いたイオン交換クロマトグラフィでさらに精製した(勾配溶出、緩衝溶液A:20mM Tris−HCl、pH8.0;緩衝溶液B:1M塩化ナトリウム、20mM Tris HCl、pH8.0)。TNF−免疫反応性を含む分画をプールし、150mM塩化ナトリウム、50mMリン酸ナトリウム緩衝溶液、pH7.3(PBS)で予め平衡化し、同緩衝溶液で溶出する、Sephacryl−S−300HR(Pharmacia−Upjohn)を用いたゲル濾過クロマトグラフィで精製した。40000〜50000Mr生成物に対応する分画をプールし、等分して−20℃で保存した。クロマトグラフィステップで使用した溶液は全て、無菌で且つ内毒素を含まない水(Salf,Bergamo,Italy)で調製した。最終収量は、TNF 45mgおよびNGR−TNF 34.5mgであった。

精製したTNFおよびNGR−TNFの分子量を、エレクトロスプレー質量分析法で測定した。市販のタンパク質アッセイキット(Pierce,Rockford,IL)を使用して、タンパク質含量を測定した。NGR−TNFおよびTNFの内毒素含量は、定量的色素生産性Lymulus Amoebocyte Lysate(LAL)試験(BioWhittaker)で測定したとき、それぞれ、0.75単位/μgおよび1.38単位/μgであった。

以下の通りに、可溶性p55−TNF受容体(sTNF−Rl)−セファロースを用いたアフィニティクロマトグラフィで、少量のTNFおよびNGR−TNFをさらに精製した:組換えsTNF−R1 5mgを、記載の通りに作製し(22)、製造業者の説明書に従って、Activated−CH−Sepharose(Pharmacia)2mlに結合させた。別個のカラム2個(それぞれ1ml)を、無菌で且つ内毒素を含まない溶液で徹底的に洗浄し、PBS中の、精製TNFまたはNGR−TNFを充填し、勾配溶出(1時間、緩衝溶液A:PBS;緩衝溶液B:0.5M塩化ナトリウム、0.2Mグリシン−HCl)で脱着させた。TNF−抗原含有分画を中和し、無菌生理学的溶液に対して透析した。膜上にタンパク質が吸着するのを防止するために、内毒素を含まないヒト血清アルブミンを加えてから透析した(0.5mg/ml)。各分画中のTNF含量を、ELISAおよび細胞溶解アッセイで測定した。

TNFの非還元SDS−PAGEは、単量体TNFの予想値、17〜18kDaの単一バンドを示した(表示せず)。相反して、NGR−TNFの非還元SDS−PAGEおよびウエスタンブロット分析は、単量体、二量体および三量体に対応すると思われる、18、36および50kDaという異なる免疫反応形を示した。還元条件では、50kDaおよび36kDaのバンドのほとんどが、18kDa形に転換されており、鎖間ジスルフィド架橋を有するNGR−TNF分子の存在が示唆される。18kDaの バンドが、物質総量の約2/3を占め、36kDaのバンドが、残りの部分のほとんどであった。これらの電気泳動パターンから、NGR−TNFは、適切な鎖内ジスルフィドを有する3つの単量体サブユニットで構成される三量体(少なくとも50%)と、主として1つ以上の鎖間ジスルフィドを有する三量体による残りの部分との混合物であったことが示唆される。還元SDS−PAGEでも確認された36kDaのバンドから、NGR−TNFが不可逆的変性二量体(総量の約10%)も含んでいたことが示唆される。

ヒト組換えTNFおよびNGR−TNF(CNGRCGのC末端と融合したヒトTNF1〜157からなる)を、組換えDNAテクノロジーによって作製し、本質的にネズミTNFおよびNGR−TNFに関して記載した通りに精製した。ヒトNGR−TNFをコードするcDNAは、以下のプライマー:

CNGRCGと融合した組換えネズミインターフェロン(IFN)γ(NGR−IFNγ)を、本質的に、NGR−TNFに関して記載した通りに、組換えDNAテクノロジーで作製した。CNGRCドメインを、IFNγのC末端と融合させた。さらに、大腸菌細胞で発現させるために、134位のシステインを、セリンと取り替え、メチオニンを−1位に導入した。NGR−IFNγ cDNAの産生に使用したPCRプライマーは、以下の通りであった:5’−A TAT CTA CAT ATG CAC GGC ACA GTC ATT GAA AGC C (センス)および5’−TC GGA TCC TCA GCA ACG GCC GTT GCA GCC GGA GCG ACT CCT TTT CCG CTT CCT GAG GC。このcDNAをpET−11b(Nde I/BamH I)にクローニングし、BL21(DE3)大腸菌細胞(Novagen)の形質転換に使用した。pET11b製造業者の説明書に従って、イソプロピル−β−D−チオガラクトシドで、タンパク質発現を誘導した。標準技術に従って、アガロース上に固定された抗マウスIFNγmAb(AN18)を使用した、免疫アフィニティクロマトグラフィで、この生成物を、大腸菌抽出液から精製した。最終生成物の還元SDS−PAGEおよび非還元SDS−PAGEは、16kDaの単一バンドを示した。エレクトロスプレー質量分析法は、ネズミMet−IFNγ1〜134(Cl34S)CNGRC(NGR−IFNγ)に一致する、16223+3.6Daの分子量を示した(予想値、1625.5Da)。

ビオチン−NGR−TNF結合体は、1M炭酸ナトリウム緩衝溶液、pH6.8中で、NGR−TNFとD−ビオチニル−6−アミノカプロン酸 N−ヒドロキシスクシンイミドエステル(Societd Prodotti Antibiotici S.p.A,Milan,Italy)とを混合する(室温で3時間)ことによって作製した(21)。この反応を、1M Tris−HCl、pH7.5でブロックした。

この結合体を質量分析法で特性決定し、(平均して)1ビオチン/三量体を含むことが判明した。次いで、5×104のRMA−T生細胞を、C57BL/6(Charles River Laboratories,Calco,Italy)の左横腹に皮下注射により(s.c.)投与した。腫瘍面積が40mm2に達したとき、既述の「3日」プロトコール(26)に従って、ビオチニル化抗体、アビジンおよびビオチン−TNFを、マウスに逐次注射した。我々は、ビオチン−mAbl9El2 40μg(i.p.、ステップI)、それぞれ、18および19時間後に、アビジン60μgおよびストレプトアビジン60μg(i.p.、ステップII)、24時間後に、ビオチン−NGR−TNF 3μg(i.p.、ステップIII)を注射した。各化合物は、無菌0.9%塩化ナトリウム溶液で希釈した。対照実験では、アビジンおよびストレプトアビジンを省略した。各実験は、5マウス/群で実施した。腫瘍サイズをカリパスで測定することによって、腫瘍成長を毎日モニタリングした。mAb 19El2−ビオチン/アビジン/ストレプトアビジン/ビオチン−NGR−TNF投与群の、投与前および投与10日後の腫瘍面積は、それぞれ、39±4mm2および8±5mm2であった(動物5匹、平均値±SE)。対照群(mAb 19E12−ビオチン/ビオチン−NGR−TNFのみを投与)では、投与前および投与10日後の腫瘍面積は、それぞれ、40±4mm2および20±6mm2であり、腫瘍誘導抗体およびアビジンを用いたプレターゲティングは、NGR−TNFの活性を増強したことがわかる。

Claims (16)
- TNFまたはIFNγから選択されるサイトカインと、CD13受容体のリガンドとの間のコンジュゲーション生成物。
- 前記サイトカインが、TNFαまたはTNFβである、請求項1に記載のコンジュゲーション生成物。
- 前記CD13受容体のリガンドが、抗体またはそれらの活性なフラグメント、ペプチドまたはペプチド擬似物からなる群から選択される、請求項1または2に記載のコンジュゲーション生成物。
- 前記リガンドが、NGRモチーフを含むペプチドである、請求項3に記載のコンジュゲーション生成物。
- 前記ペプチドが、CNGRCVSGCAGRC、NGRAHA、GNGRG、シクロCVLNGRMEC、線状または環状のCNGRCからなる群から選択される、請求項4に記載のコンジュゲーション生成物。
- サイトカインが、ポリエチレングルコールまたはアシル残基により誘導体化される、請求項1〜5のいずれか1項に記載のコンジュゲーション生成物。
- 前記サイトカインが、腫瘍抗原、腫瘍血管形成マーカーまたは細胞外マトリックス成分に対する抗体またはそのフラグメント、またはビオチンと、さらにコンジュゲートしている、請求項1〜6のいずれか1項に記載のコンジュゲーション生成物。
- 前記サイトカインがTNFであり、前記サイトカインが異なるサブユニットにおいて、CD13リガンドと、抗体、そのフラグメント、またはビオチンのいずれかとにコンジュゲートしている、請求項7に記載のコンジュゲーション生成物。
- CD13リガンドをコードする5’または3’隣接配列を担持するTNFおよびIFNから選択されるサイトカインをコードするcDNA。
- 前記CD13リガンドが、請求項5に記載のペプチドである、請求項9に記載のcDNA。
- 請求項9〜10のcDNAを含む、遺伝子療法用ベクター。
- 薬学的に許容できる担体および賦形剤と一緒に、請求項1〜8に記載のコンジュゲーション生成物の有効量を含む医用組成物。
- 注射可能な溶液または懸濁液、または注入用液の形態である、請求項12に記載の組成物。
- リポソームの形態である、請求項12または13に記載の組成物。
- 癌の診断用または治療用の医薬品を調製するための、請求項1〜8に記載のコンジュゲーション生成物、または請求項9〜10に記載のcDNAの使用。
