Claims (4)
- ａ）宿主細胞を、宿主細胞により分泌可能な形態のプロペプチド配列内にエンテロキナーゼ認識部位を有するトリプシノーゲンをコードする組換え核酸で形質転換する工程、
ｂ）組換え核酸の発現および培養培地への発現産物の分泌を可能にする条件下で宿主細胞を培養する工程、ここで該条件は、プロペプチド配列の自己触媒切断が実質的に妨げられるように選択される、
ｃ）培養培地から発現産物を単離する工程、
ｄ）プロペプチド配列を切断して、活性トリプシンを形成させる工程、および
ｅ）任意に、非切断トリプシノーゲン分子と活性トリプシンとを分離する工程を含むトリプシンの組換え製造方法。 - プロペプチド配列が天然のプロペプチド配列と比べて短いことを特徴とする、プロペプチド配列内にエンテロキナーゼ認識部位を有するトリプシノーゲンをコードする組換え核酸。
- 請求項２記載の核酸で、または該核酸の少なくとも１つのコピーを含有してなる組換えベクターで形質転換されていることを特徴とする組換え細胞。
- 天然のプロペプチド配列と比べて短いプロペプチド配列を有し、エンテロキナーゼ認識部位を含むことを特徴とする組換えトリプシノーゲン。
