Claims (1)
- 【特許請求の範囲】 1.ポリカチオン性ブロックおよび少なくとも1つの非組織結合性ブロックを含 むブロックコポリマーまたはグラフトコポリマーであって、該ポリカチオン性ブ ロックが、100,000ダルトン以上の分子量を有する実質的に線状のポリカチオン 性ブロック、または少なくとも8カチオン性チャージを提供するのに十分な分子 量を有する樹状ポリカチオン性ブロックのいずれかである、コポリマー。 2.前記非組織結合性ブロックが、5,000ダルトンを超える分子量を有する、請 求項1に記載のコポリマー。 3.前記非組織結合性ブロックが、50,000ダルトンを超える分子量を有する、請 求項1に記載のコポリマー。 4.前記非組織結合性ブロックが、ポリエチレングリコール、水溶液中で少なく とも1g/Lの溶解度を有する混合ポリアルキレンオキシド、中性水溶性多糖類 、ポリビニルアルコール、ポリ-N-ビニルピロリドン、非カチオン性ポリ(メタ) アクリレート、およびこれらの組み合わせからなる群から選択される、請求項1 に記載のコポリマー。 5.前記非組織結合性ブロックがポリエチレングリコールを含む、請求項5に記 載のコポリマー。 6.前記ポリカチオン性ブロックが、中性pHで正味の正電荷を有する天然および 非天然ポリアミノ酸、正荷電多糖類、ならびに正荷電合成ポリマーからなる群か ら選択される、請求項1に記載のコポリマー。 7.前記ポリカチオン性ブロックが、リジン、ヒスチジン、アルギニン、および オルニチンから選択されるモノマーユニットを含む、請求項1に記載のコポリマ ー。 8.前記正荷電多糖類が、キトサン、部分的に脱アセチル化されたキチン、およ び中性多糖類のアミン含有誘導体からなる群から選択される、請求項7に記載の コポリマー。 9.前記正荷電合成ポリマーが、ポリエチレンイミン、ポリアミノ(メタ)アクリ レート、ポリアミノスチレン、ポリアミノエチレン、ポリ(アミノエチル)エチレ ン、ポリアミノエチルスチレン、およびこれらのN-アルキル誘導体からなる群か ら選択される、請求項6に記載のコポリマー。 10.薬学的に受容可能なキャリアをさらに含む、請求項1に記載のコポリマー 。 11.生物活性試剤をさらに含む、請求項1に記載のコポリマー。 12.前記生物活性試剤がポリマーに化学的に連結されている、請求項11に記 載のコポリマー。 13.ポリカチオン性物質およびポリアニオン性物質の層を含む、巨視的表面上 のポリマーコーティング。 14.前記コーティングが、ポリカチオシとポリアニオンとの交互層として塗布 される、請求項13に記載のコーティング。 15.少なくとも4層である、請求項13に記載のコーティング。 16.少なくとも6層である、請求項13に記載のコーティング。 17.前記ポリカチオンが、中性pHで正味の電荷を有する天然および非天然ポリ アミノ酸、正荷電多糖類、および正荷電合成ポリマーからなる群から選択される 、請求項13に記載のコーティング。 18.前記ポリカチオンが、リジン、ヒスチジン、アルギニン、およびオルニチ ンからなる群から選択されるモノマーユニットを含む、請求項17に記載のコー ティング。 19.前記ポリアニオンが、アルギネート、カラギーナン、ファーセレラン、ペ クチン、キサンタン、ヒアルロン酸、ヘパリン、ヘパラン硫酸、コシドロイチン 硫酸、デルマタン硫酸、デキストラン硫酸、ポリ(メタ)アタリル酸、酸化セルロ ース、カルボキシメチルセルロース、クロスマーメロース(crosmarmelose)、ペ ンダントカルボキシル基を含む合成ポリマーおよびコポリマー、主に負荷電のポ リアミノ酸、ならびに生体適合性ポリフェノール物質からなる群から選択される 、請求項13に記載のコーティング。 20.前記ポリアニオンが、アルギネート、ペクチン、カルボキシメチルセルロ ース、ヘパリン、およびヒアルロン酸からなる群から選択される、請求項19に 記載のコーティング。 21.生物活性試剤をさらに含む、請求項13に記載のコーティング。 22.前記生物活性試剤が少なくとも1つのポリマーに化学的に連結されている、 請求項21に記載のコーティング。 23.前記表向が組織表面である、請求項13に記載のコーティング。 24.前記表向が医用デバイス表面である、請求項13に記載のコーティング。 25.少なくとも1層のポリアニオン性またはポリカチオン性ポリマーが、共有 結合された非組織結合性ブロックをさらに含む、請求項13に記載のコーティン グ。 26.前記非組織結合性ブロックが少なくとも最外層にある、請求項25に記載 のコーティング。 27.巨視的表面を、カプセル化するか、詰めるか、密封するか、または支持す るための方法であって、ポリカチオン性およびポリアニオン性物質の連続層を、 該表面上に堆積する工程を包含する、方法。 28.前記ポリカチオン性およびポリアニオン性物質の層の前記堆積が、組織付 着を最小化もしくは防止するか、手術後の付着を最小化もしくは防止するか、血 栓症を防止するか、癌性細胞の着床を防止するか、感染の治癒もしくは防止を促 進するために組織をコーティングするか、または生物活性試剤の局所送達を向上 させる、請求項27に記載の方法。 29.前記ポリカチオン性物質が、ポリ(D-リジン)、ポリ(オルニチン)、ポリ( アルギニン)、ポリ(ヒスチジン)、ポリ(アミノスチレン)、ポリ(アミノアクリレ ート)、ポリ(N-メチルアミノアクリレート)、ポリ(N-エチルアミノアクリレート )、ポリ(N,N-ジメチルアミノアクリレ−ト)、ポリ(N,N-ジエチルアミノアクリレ ート)、ポリ(アミノメタクリレート)、ポリ(N-メチルアミノメタクリレート)、 ポリ(N-エチルアミノメタクリレート)、ポリ(N,N-ジメチルアミノメタクリレー ト)、ポリ(N,N-ジエチルアミノメタクリレート)、ポリ(エチレンイミン)、第四 級アミン基を含むポリマー、および天然または合成カチオン性多糖類からなる群 から選択される、請求項27に記載の方法。 30.前記ポリカチオン性物質が、ポリリジン、ポリオルニチン、およびポリエ チレンイミンからなる群から選択される、請求項27に記載の方法。 31.前記ポリアニオン性物質が、アルギネート、カラギーナン、ファーセレラ ン、ペクチン、キサンタン、ヒアルロン酸、ヘパリン、ヘパラン硫酸、コンドロ イチン硫酸、デルマタン硫酸、デキストラン硫酸、ポリ(メタ)アクリル酸、酸化 セルロース、カルポキシメチルセルロース、クロスマーメロース、ペンダントカ ルボキシル基を含む合成ポリマーおよびコポリマー、主に負荷電のポリアミノ酸 、ならびに生体適合性ポリフェノール物質からなる群から選択される、請求項2 7に記載の方法。 32.前記ポリアニオン性物質がアルギネートである、詰求項27に記載の方法 。 33.前記物質が生物活性試剤をさらに含む、請求項27に記載の方法。 34.前記生物活性試剤が1つ以上の前記ポリマーに化学的に連結されている、 請求項33に記載の方法。 35.付着が防止されるべき部位が、組織が損傷を受けている領域である、請求 項28に記載の方法。 36.付着が防止されるべき部位が外科的に切られている、請求項28に記載の 方法。 37.血栓症または付着が防止されるべき部位が、損傷を受けている血管の内張 り(lining)である、請求項28に記載の方法。 38.付着を防止されるべき前記組織が、癌性細胞または腫瘍細胞である、請求 項28に記載の方法。 39.前記組織が、器官、もしくはまた損傷を受けた他の器官と接触する身体の 管腔である、請求項28に記載の方法。 40.前記組織が処理されている、請求項28に記載の方法。 41.非生物学的表面をコーティングする方法であって、該表面に、ポリカチオ ン性およびポリアニオン性物質のコポリマーまたは連続層を塗布する工程を包含 する方法であって、ここで: 該コポリマーが、ポリカチオン性ブロックおよび少なくとも1つの非組織結合 性ブロックを含み、ここで該ポリカチオン性ブロックが、100,000ダルトン以上 の分子量を有する実質的に線状のポリカチオン性ブロック、または少なくとも8 カチオン性チャージを提供するのに十分な分子量を有する樹状ポリカチオン性ブ ロックのいずれかである、方法。 42.前記表面が金属表面である、請求項41に記載の方法。 43.医療状態の処置のための、請求項13に記載のコーティングの使用。 44.医用デバイスの表面をコーティングするための、請求項13に記載のコー ティングの使用。 45.医療状態の処置のための、請求項1に記載のコポリマーの使用。 46.前記医療処置が、手術後の付着の最小化または防止、血栓症の最小化また は防止、癌性細胞の着床の最小化または防止、治癒を促進するための組織表面の コーティング、感染を防止するための組織表面のコーティング、および薬物の局 所送達を向上するためのコーティングの塗布からなる群から選択される、請求項 45に記載の使用。 47.医用デバイスの表面を処理するための、詰求項1に記載のコポリマーの使 用。
