Description

妨げる。水中油型エマルションの1つの利点は、水で希釈して容易に所望の組成
物とすることができることである。

て許容できない悪影響なしに必要な腸管細菌と接触するように経口投与してもよ

れかの装置(例えばFrench Pressまたは工業用高剪断力ミキサー)を用いて油相
および水相を混合できる。

度(50-250μM)で用い、種々のグラム陰性菌とともに混合物を15分間インキュベ ートした。次いで、混合物の殺菌効果をトリプチケースソイブロス上で評価した
。結果を以下の表4に示す。1/100希釈液においてBCTPを用いて細菌総数に99%以
上の減少が認められた。250μMのEDTA単独で15分で細菌総数を減少させることが
できなかった対照群からわかるように、この総数の減少はEDTA殺菌効果のみによ
るものではなかった。

質的に完全に死滅させることができる。
のように、胞子を効果的に死滅させるには、通常、ホルムアルデヒドまたは次亜
塩素酸ナトリウム(すなわち漂白剤)などの有毒で刺激性のある化学製品の使用
が必要である。従って、胞子形態のB.セレウスを用いて同様の実験を行った。表
6に示すように、両培地とも37℃での10分間の処理はB.セレウスの胞子を死滅さ せるのに十分ではなかった。

実験を行った。便宜には、胞子調製物を1:100の希釈率でBCTPで処理し、非処理 の対照と比較した。0.5、1、2、4、6および8時間後にミリリットルあたりのコロ
ニー形成単位(CFU/ml)数を数量化した。図1に示すように、非処理の対照におけ るCFU/mlは最初の4時間のインキュベーションの間増加し、次いでプラトーに達 した。細菌塗抹標本を0、1、2、4および6時間の時点に調製し、胞子構造を染色 したところ、2時間までで胞子構造が残っていないということが示された(図2A-2
C)。このように、非処理の対照では2時間の時点までに100%の胞子の発芽が起こ った。BCTPで処理された胞子調製物では、最初の2時間の間CFU/mlは増加を示さ ず、次いで2-4時間の時期の間に急速に減少した。2-4時間の間の基準線CFU/mlか
らの減少は約1000倍であった。同時点に調製し、胞子構造を染色した細菌塗抹標
本により8時間の実験の最後まで胞子構造が残っていることが示された。従って 、BCTP処理培養物では発芽過程の阻害または胞子が損傷を受け発芽できなくなっ
たためのいずれかによって胞子の発芽は起こらなかった。

した。BCTPで前処理されたB.セレウスを接種されたマウスでは壊死病変が小さい
ようであった。下記表は実験の結果を示す。

TP(1:5の滅菌生理食塩水で希釈)を2本目の試験管に加えて混合し、BCTPの最 終希釈を1:10とした。B.セレウス胞子懸濁液/BCTP(1:10)を、37℃にて混合 しつつ4時間インキュベートした。10匹のCD-1マウスにB.セレウス胞子懸濁液/BC
TP(1:10)0.1ccを皮下注射した。等量のBCTP(1:50の滅菌生理食塩水で希釈)を
3本目の試験管に加えて混合し、BCTPの最終希釈を1:100とした。B.セレウス胞 子懸濁液/BCTP(1:100) を、37℃にて混合しつつ4時間インキュベートした。10匹
のCD-1マウスにB.セレウス胞子懸濁液/BCTP(1:10)0.1ccを皮下注射した。等 量のBCTP(1:5の滅菌生理食塩水で希釈)およびTSBを混合し、BCTPの最終希釈 容量を1:10とした。10匹のCD-1マウスにBCTP/TSB0.1ccを皮下注射した。等量の
BCTP(1:50の滅菌生理食塩水で希釈)およびTSBを混合し、BCTPの最終希釈を1 :100とした。10匹のCD-1マウスにBCTP/TSB 0.1ccを皮下注射した。

た。病変の辺縁部を切開し、綿棒でぬぐった。胸部をベータジン、次いで70%滅
菌イソプロピルアルコールで洗浄した。心臓穿刺により血液を採取した。腹部を

筋を、個別の滅菌器具で切開した。個別の滅菌器具を用いて肝臓および脾臓の標
本を採取した。肝臓および脾臓の標本は、炎に短時間かざし、滅菌した器具で切
断した。新たに露出した表面を培養に用いた。BHI寒天(Difco)を加え、37℃に
て一晩通気培養した。

菌不活化用エマルションを接触させてグラム陽性菌を不活化する工程を含んでな
り、前記細菌不活化用エマルションが、界面活性剤安定剤により水相に分配され
た不連続な油相の形態にある水中油型エマルションからなり、前記油相が有機ホ
スフェート系溶媒および担体油を含む、前記方法。 - 【請求項2】 前記エマルションの油相が0.5〜5ミクロンの範囲の平均粒径
を有する油滴からなる、請求項1記載の方法。 - 【請求項3】 前記界面活性剤がTween20、Tween80およびTriton X-100から
なる群より選択される、請求項1記載の方法。 - 【請求項4】 前記界面活性剤がTritonX-100である、請求項3記載の方法。

不連続な油相の形態にある水中油型エマルションからなり、前記油相が有機ホス
