発明の分野
本発明は、接触時に細菌を不活化する水中油型エマルションと細菌とを接触させることにより、胞子を含めた細菌を不活化する方法に関する。

発明の背景
水と不混和性の脂質相を、例えば超分散機による機械攪拌により水相に混合させれば、水中油型エマルションのような分散系が生じることが知られている。得られた分散系を安定にするには乳化剤の添加が必要であり、その分子は油/水界面に吸着して一種の連続した膜を形成し、これが2つの隣接する油滴間の接触を妨げる。水中油型エマルションの1つの利点は、水で希釈して容易に所望の組成物とすることができることである。

水相に分散した離散型の油滴の他、水中油型エマルションには、小さな脂質小胞(すなわち、水相層により互いから隔離された実質的に同心のいくつかの脂質二重層からなることが多い脂質球)、ミセル(すなわち、極性頭部基が水相に面し、非極性尾部が水相から離れて内部へ封鎖されるように並んだ50〜200分子の小塊にある両親媒性分子)、またはラメラ相(各々の粒子が、水の薄い膜によって隔離された両親媒性の平行した二重層からなる脂質分散系)といった他の脂質構造も含まれる。これらの脂質構造は非極性残基(すなわち、長鎖炭化水素)を水から遠ざける疎水力の結果として形成されるものである。

病原性細菌が侵入する入り口は主として皮膚と粘膜である。多くの感染で最初のステップは皮膚または粘膜への付着またはそこにおけるコロニー形成であり、次いで感染性の病原体の侵入および蔓延が起こる。従って、細菌不活化製剤およびかかる製剤を用いて細菌を不活化する方法を提供することが望まれている。

さらに、多種の細菌が好ましくない条件に応答して、高い抵抗性を持ち、厚い壁を有する内生胞子(胞子とも呼ばれる)を形成するが、これらは条件がよくなれば代謝活性を再開する。これらの脱水した菌体は、休眠状態で保持され、いつでも吸水して活性を再開できる細胞成分を含んでいる。従って、細菌胞子不活化製剤およびその製剤を用いて細菌胞子を不活化する方法を提供することが望まれている。

胞子を含む細菌は熱、圧力およびしばしば殺菌剤と呼ばれる化学薬剤により不活化できる。例えば、腐食性の組成物、例えばホルムアルデヒドや次亜塩素酸ナトリウム(漂白剤)が胞子を不活化するために用いられてきた。残念なことにかかる組成物は有毒であるか、または皮膚や粘膜に刺激を与える。従って、皮膚や粘膜に無毒な、細菌や胞子を含む細菌を不活化する組成物および方法を提供することが望まれている。また、in vivoで有効な、細菌および細菌胞子を不活化する組成物および方法を提供することも望ましい。

従って、本発明の目的は、細菌と細菌不活化用エマルションとを接触させることにより、胞子を含む細菌を不活化する方法を提供することである。

発明の概要
本発明は細菌を不活化する方法を提供し、その方法は細菌不活化用エマルションを準備し、細菌とそのエマルションとを接触させる工程を含む。このエマルションは、界面活性剤、有機ホスフェート系溶媒および担体油を含んでなる水中油型エマルションである。1つの実施形態では、細菌はグラム陽性菌、すなわちグラム染色と呼ばれる方法で紫色の色素(クリスタルバイオレット)を容易に吸収する密度の高いペプチドグリカン壁を有する細菌である。特定の好ましい実施形態では、グラム陽性菌または細菌胞子はバチルス属である。特に好ましい実施形態では、細菌または胞子は炭疸菌である。

もう1つの実施形態では、細菌はグラム陰性菌、すなわちグラム染色で紫色の色素を容易には吸収しない細菌である。この実施形態では、細菌不活化用エマルションは、細胞壁によるエマルションの取り込みを増強することができる化合物を予め混合してある。特定の好ましい実施形態では、この化合物はキレート剤(例えば、エチレンジアミン四酢酸(EDTA))、溶媒(例えば、ジメチルスルホキシド(DMSO))、洗剤(例えば、ドデジル硫酸ナトリウム(SDS))およびそれらの組み合わせである。その他の好ましい実施形態では、この化合物は、ペプチドと併用して、細胞壁によるエマルションの取り込みを増強するのに用いられ、例えば、ジペプチドおよびオリゴペプチドペルミアーゼ、ジグリシン、トリグリシン、それらの混合物または他のオリゴペプチドがある。

細菌不活化用エマルションは飲み込んだり、吸引したり、あるいは皮膚に塗布しても無毒で安全である。このことは、公知の刺激物である化学殺菌剤とは対照的である。細菌不活化用エマルションはまた植物に対しても無毒であると考えられる。

水中油型エマルションの形成に有用な油には、ダイズ油、アボガド油、スクアレン油、その他の魚油、スクアラン油、セサミ油、オリーブ油、カノーラ油、コーン油、ナタネ油、ベニバナ油、ヒマワリ油、フレーバー油およびそれらの混合物など広範な水不混和性物質が含まれる。

本発明の方法で用いられるエマルションの形成に有用な界面活性剤には、種々の陰イオン界面活性剤および非イオン界面活性剤、ならびに水中油型エマルションの形成を促し得るその他の乳化剤が含まれる。一般には、乳化剤は比較的親水性のものであり、必要な品質を達成するため乳化剤混合物を使用することができる。非イオン界面活性剤はイオン乳化剤よりも有利であり、それらは広いpH範囲で適合し、イオン(例えば、ソープタイプの)乳化剤よりより安定なエマルションを形成することが多い。特に有用な界面活性剤としては、商標Tween20、Tween80およびTriton(すなわち、X-100)のようなフェノキシポリエトキシエタノールとして市販されている洗剤が挙げられる。最も好ましい界面活性剤はTritonX-100(t-オクチルフェノキシポリエトキシエタノール)である。

水中油型エマルションの形成に有用な有機ホスフェート系溶媒としては、ジアルキルホスフェートおよびトリアルキルホスフェートが挙げられる。好ましい実施形態では、ジまたはトリアルキルホスフェートの各アルキル基は1〜10個の炭素原子、より好ましくは2〜8個の炭素原子を有する。このジまたはトリアルキルホスフェートのアルキル基はすべて同じであっても異なっていてもよい。特に好ましいトリアルキルホスフェートは、可塑剤であるトリ-n-ブチルホスフェートである。異なるジアルキルおよびトリアルキルホスフェートの混合物も使用できる。さらに、溶媒としてアルコール類を使用してもよく、例えばオクタノールが挙げられる。

本発明の方法で使用される細菌不活化用水中油型エマルションは、当該技術分野で公知の標準的エマルション形成手法を用いて形成できる。便宜には、油相を比較的高い剪断力下で水相と混合すると、直径約1〜2ミクロンの油滴を含む水中油型エマルションが得られる。油性不連続相は(a)油担体;(b)界面活性剤;および(c)有機ホスフェート系溶媒を混合して形成する。エマルションは油相を水相(例えば水)と約1:4〜4:1の範囲の容量比で、好ましくは約1:4 油相:水相で混合して形成する。エマルションを形成するに十分な剪断力を発生させ得るいずれかの装置(例えばFrench Pressまたは工業用高剪断力ミキサー)を用いて油相および水相を混合できる。

本発明の方法で使用される細菌不活化用水中油型エマルションを用いて、種々の細菌および細菌胞子を接触時に不活化することができる。例えば現在、開示されているエマルションを用いて、バチルス・セレウス(Bacillus cereus)、バチルス・サーキュランス(B. circulans)、バチラス・メガテリウム(B. megaterium)および枯草菌(B. subtillus)をはじめとするバチルス属を不活化でき、またボツリヌス菌(C. botulinum)および破傷風菌(C. tetanil)をはじめとするクロストリジウム属も包含している。本発明の方法は特定の細菌戦病原体、例えば炭疽菌(B. anthracia)の不活化に特に有用である。

グラム陰性菌の細胞壁による細菌不活化用エマルションの取り込みを増強し、それによって耐性グラム陰性菌に対するエマルションの殺菌効果を増強するために、エマルションを予めEDTA(エチレンジアミン四酢酸)と混合した。EDTAは低濃度(50-250μM)で用い、種々のグラム陰性菌とともに混合物を15分間インキュベートした。次いで、混合物の殺菌効果をトリプチケースソイブロス上で評価した。結果を以下の表4に示す。1/100希釈液においてBCTPを用いて細菌総数に99%以上の減少が認められた。250μMのEDTA単独で15分で細菌総数を減少させることができなかった対照群からわかるように、この総数の減少はEDTA殺菌効果のみによるものではなかった。

実施例4
in vitro 殺菌力研究 III- 栄養および胞子形態
炭疽菌のモデル系としてバチルス・セレウス(B.セレウス、ATCC#14579)を用いた。BCTPで希釈した調製物を用いて実験を行い、栄養形態の(活発に増殖する)B.セレウスに対する本発明の化合物の殺菌力を研究した。37℃で10分間の培地中での処理を評価した。表5に要約されるように、BCTPエマルションはB.セレウスの栄養形態に対して効力を有する。1:100の高い希釈率の希釈液を含む試験したすべての濃度で、この調製物への10分間の曝露により、栄養形態のB.セレウスを実質的に完全に死滅させることができる。

胞子形態の炭疽菌は、生物兵器として最も使用されそうな生物の1種である。ほとんどの殺菌剤に対し胞子が高い耐性を持つことは十分に知られている。前記のように、胞子を効果的に死滅させるには、通常、ホルムアルデヒドまたは次亜塩素酸ナトリウム(すなわち漂白剤)などの有毒で刺激性のある化学製品の使用が必要である。従って、胞子形態のB.セレウスを用いて同様の実験を行った。表6に示すように、両培地とも37℃での10分間の処理はB.セレウスの胞子を死滅させるのに十分ではなかった。

一定時間にわたる胞子形態のB.セレウスに対する本発明の化合物の効力を評価するために、BCTPを1:100希釈で固形寒天培地中に混合し、胞子を表面に均一に広げて37℃で96時間インキュベートした。BCTPが混合された固形寒天培地上では96時間の最終まで増殖は見られなかった(すなわち、>99%死滅、平均>99%死滅、3実験)。

BCTPによる胞子の死滅が起こる時間をより厳密に規定する試みとして、以下の実験を行った。便宜には、胞子調製物を1:100の希釈率でBCTPで処理し、非処理の対照と比較した。0.5、1、2、4、6および8時間後にミリリットルあたりのコロニー形成単位(CFU/ml)数を数量化した。図1に示すように、非処理の対照におけるCFU/mlは最初の4時間のインキュベーションの間増加し、次いでプラトーに達した。細菌塗抹標本を0、1、2、4および6時間の時点に調製し、胞子構造を染色したところ、2時間までで胞子構造が残っていないということが示された(図2A-2C)。このように、非処理の対照では2時間の時点までに100%の胞子の発芽が起こった。BCTPで処理された胞子調製物では、最初の2時間の間CFU/mlは増加を示さず、次いで2-4時間の時期の間に急速に減少した。2-4時間の間の基準線CFU/mlからの減少は約1000倍であった。同時点に調製し、胞子構造を染色した細菌塗抹標本により8時間の実験の最後まで胞子構造が残っていることが示された。従って、BCTP処理培養物では発芽過程の阻害または胞子が損傷を受け発芽できなくなったためのいずれかによって胞子の発芽は起こらなかった。

エマルションがB.セレウスに加え他のバチルス種の死滅に効果的であるかどうかを決定するために、前記のような同様の実験を行い、ここでは胞子調整物をエマルションで処理し、4時間のインキュベーション後、非処理の対照と比較した。下記表は結果を示しているが、数値は括弧内の平均値とともに数回の実験からの結果の範囲を示す。

実施例5
in vitro 殺菌力研究
バチルス・セレウスを血液寒天(5%ヒツジ血液を含むTSA、REMEL)で3回継代した。B.セレウスを3回目の継代プレートから掻き取ってトリプチケースソイブロス(TSB)(BBLから入手可能)に再懸濁した。B.セレウス懸濁液を2本の試験管に分注した。1本の試験管に等量の滅菌生理食塩水を加えて混合した。B.セレウス懸濁液/生理食塩水0.1ccを5CD-1マウスに皮下注射した。等量の(滅菌生理食塩水で1:5希釈した)BCTPを1本の試験管に加えて混合し、BCTPの最終希釈率を1:10とした。混合しつつB.セレウス懸濁液/BCTPを37℃で10分間インキュベートし、B.セレウス懸濁液/BCTP0.1ccを5CD-1マウスに皮下注射した。等量の(滅菌塩水で1:5希釈した)BCTPおよびTSBを混合し、BCTPの最終希釈率を1:10とし、BCTP/TSB0.1ccを5CD-1マウスに皮下注射した。

バチルス・セレウスは、胞子形成を誘導するために、0.1%酵母抽出分(Difco)および50μg/mlのMnSO4を含む栄養寒天(Difco)で増殖させた。プレートを掻き取って滅菌50%エタノールに懸濁し、室温で2時間攪拌しながらインキュベートして残存する栄養細菌を溶解した。懸濁液を2,500×gで20分間遠心分離し、上清を廃棄した。蒸留水中にペレットを再懸濁し、2,500×gで20分間遠心分離して上清を廃棄した。胞子懸濁液を分割した。ペレットをTSBに再懸濁し、生理食塩水で1:2希釈したB.セレウス胞子懸濁液0.1ccを3CD-1マウスに皮下注射した。等量の(滅菌生理食塩水で1:5希釈した)BCTPおよびB.セレウス胞子懸濁液を混合し、BCTPの最終希釈率を1:10(プレインキュベーション時)とし、BCTP/B.セレウス胞子懸濁液0.1ccを3CD-1マウスに皮下注射した。

皮膚病変、血液、肝臓および脾臓からの、B.セレウスの再分離を試みた。皮膚病変をベータジン(betadine)、次いで70%滅菌イソプロピルアルコールで洗浄した。病変の辺縁部を切開し、綿棒でぬぐった。胸部をベータジン、次いで70%滅菌イソプロピルアルコールで洗浄した。心臓穿刺により血液を採取した。腹部をベータジン、次いで70%滅菌イソプロピルアルコールで洗浄した。皮膚および腹筋を、個別の滅菌器具で切開した。個別の滅菌器具を用いて肝臓および脾臓の標本を採取した。肝臓および脾臓の標本は、炎に短時間かざし、滅菌した器具で切断した。新たに露出した表面を培養に用いた。BHI寒天(Difco)を加え、37℃にて一晩通気培養した。

栄養型B.セレウスおよびB.セレウス胞子の前処置により、実験動物に導入された際に疾患症状を引き起こすそれらの能力が低下した。これは皮膚病変のサイズの減少に反映され、一般に、その病変部から回収されるB.セレウス数も減少する。これに加え、血液、肝臓および脾臓からB.セレウスが再分離される頻度が低いことより、敗血症が予防できる可能性が示唆された。

In vivo 毒性試験I
CD-1マウスに0.1ccの本発明の化合物を皮下注射し、炎症および/または壊死の徴候を4日間観察した。滅菌生理食塩水でこの化合物を希釈した。
組織病理学的検査のためにマウスの組織標本を10%中性緩衝ホルマリン中に保存した。組織学的検査に供した(非希釈化合物を注射したマウスの)皮膚および筋肉の標本は組織壊死の兆候を示すことが報告された。希釈した化合物を注射したマウスの組織標本については、組織学的検査を行わなかった。以下の2つの表は、独立した2回の実験結果を示す。

In vivo 毒性試験II
各群5匹の雄および5匹の雌からなるSprague-Dawleyラットの1群を、個々のケージに収容して、投与前に5日間馴化させた。ラットには14日間毎日投薬した。0ないし13日目、すなわち連続14日間、第1群の各ラットにそれぞれ3mlのBCTP(濃度1:100)を経口投与した。ラットに対する最大許容経口用量を考慮して、容量を3mlと決定した。0日目と7日目の投与前に各ラットの体重を測定した。その後は、試験期間中、1週間に1回体重を測定した。毎日、動物の病気または死亡を観察した。14日間の休薬期間を設けた。28日目にラットの体重を測定し、安楽死させた。

経口毒性試験の平均体重の結果を表16に示した。0,7,14,21および28日目の雄雌の平均体重、ならびに0ないし28日目の平均体重増加量も、表16に示した。14日目の投与中に、栄養挿管操作による機械的外傷のため、1匹のラットが死亡した。生存したすべてのラットは、28日間の試験期間わたって体重が増加し、病気は認められなかった。

このように、トリブチルホスフェート単独では毒性を有し、粘膜に刺激を与える事が知られているが、本発明のエマルションに取り込まれた場合は、これらの特性は現れない。
BCTPエマルション(濃度1:100)については、16CFR§1500.3に規定されたプロトコールに従ってウサギにおける経皮毒性も試験した(データは示していない)。
本エマルションは、試験動物の皮膚に対して刺激性を示さなかった。

Claims (42)
- 水中油型エマルションを含む、グラム陽性菌を不活化するための組成物であって、該エマルションは水相に分配された不連続な油相の形態にあり、かつ、該エマルションは、
a) Tween20(商標)、Tween80(商標)、およびフェノキシポリエトキシエタノール界面活性剤からなる群より選択される界面活性剤安定剤;
b) ダイズ油、アボガド油、スクアラン油、スクアレン油、オリーブ油、カノーラ油、コーン油、ナタネ油、ベニバナ油、ヒマワリ油、魚油、フレーバー油、水不溶性ビタミン類およびそれらの混合物からなる群より選択される担体油;
c) ジアルキルホスフェートまたはトリアルキルホスフェートのいずれかより選択される有機ホスフェート系溶媒;
を含むが、グリセロールエステルは含まない、
上記組成物。 - 前記エマルションの油相が0.5〜5ミクロンの範囲の平均粒径を有する油滴からなる、請求項1記載の組成物。
- 前記界面活性剤がTween80(商標)である、請求項1記載の組成物。

a) Tween20(商標)、Tween80(商標)、およびフェノキシポリエトキシエタノール界面活性剤からなる群より選択される界面活性剤安定剤;
b) ダイズ油、アボガド油、スクアラン油、スクアレン油、オリーブ油、カノーラ油、コーン油、ナタネ油、ベニバナ油、ヒマワリ油、魚油、フレーバー油、水不溶性ビタミン類およびそれらの混合物からなる群より選択される担体油;
