Description
Claims (1)
- 【特許請求の範囲】 1.実質的に均一な粒径を有する自己集合タンパク質粉砕物、生体適合性可塑剤 、及び水を含む、多孔性移植製品の表面上に体液密着障壁を形成する生体適合性 水性スラリー組成物であって、25℃で約8,000〜約60,000センチポアズの粘度及 び懸濁した前記タンパク質粉砕物を十分維持するだけのpHを有する、前記生体適 合性スラリー組成物。 2.前記自己集合タンパク質が約1〜約3重量%の濃度で前記スラリー中に存在 する、請求項1記載の生体適合性スラリー組成物。 3.前記自己集合タンパク質がコラーゲン、フィブロネクチン、ビトロネクチン 、プロテオグリカン、ラミニン、ヒアルロン酸、テナシン、インテグリン、カド ヘリン及びその混合物からなる群より選ばれる、請求項1記載の生体適合性スラ リー組成物。 4.前記自己集合タンパク質がウシコラーゲンI型である、請求項3記載の生体 適合性スラリー組成物。 5.透過性低下組成を更に含む、請求項1記載の生体適合性スラリー組成物。 6.前記透過性低下組成が約0〜約24%のエタノールである、請求項5記載の生 体適合性スラリー組成物。 7.前記粒径が約5〜約750μmである、請求項1記載の生体適合性スラリー組成 物。 8.前記粘度が約30,000〜約50,000センチポアズである、請求項1記載の生体適 合性スラリー組成物。 9.前記pHが前記タンパク質粉砕物の十分に等電点外の範囲にあり前記タンパク 質を水性状態で維持する、請求項1記載の生体適合性スラリー組成物。 10.前記pHが約3.5〜約3.9である、請求項9記載の生体適合性スラリー組成物。 11.前記可塑剤がグリセロール及びソルビトールの1種である、請求項1記載の 生体適合性スラリー組成物。 12.前記可塑剤が約8〜約30重量%で前記スラリー組成物中に存在する、請求項 1記載の生体適合性スラリー組成物。 13.前記自己集合タンパク質が架橋される、請求項1記載の生体適合性スラリー 組成物。 14.前記自己集合タンパク質がホルムアルデヒド及びグルタルアルデヒドの1種 で架橋される、請求項13記載の生体適合性スラリー組成物。 15.生物活性剤を更に含む、請求項1記載の生体適合性スラリー組成物。 16.前記生物活性剤が抗生物質、抗凝固剤、抗菌剤及びその混合物からなる群よ り選ばれる、請求項15記載の生体適合性スラリー組成物。 17.多孔性高分子移植製品と緊密に接触して配置され、前記製品を実質的に体液 密着にする、請求項1記載の生体適合性スラリー組成物。 18.可撓性の多孔性高分子基質;及び 下記成分を含む、前記多孔性基質と緊密に接触している水性シーラント組成物: 均一な粒径を有する自己集合タンパク質粉砕物のスラリー、 生体適合性可塑剤、及び 水 を含む体内で使用するための移植部材であって、前記シーラント組成物が25℃で 約8,000〜約60,000センチポアズの粘度及び懸濁した前記タンパク質粉砕物を十 分維持するだけのpHを有する、前記移植部材。 19.前記自己集合タンパク質が約1〜約3重量%の濃度で前記シーラント組成物 中に存在する、請求項18記載の移植部材。 20.前記自己集合タンパク質がコラーゲン、フィブロネクチン、ビトロネクチン 、プロテオグリカン、ラミニン、ヒアルロン酸、テナシン、インテグリン、カド ヘリン及びその混合物からなる群より選ばれる、請求項18記載の移植部材。 21.前記自己集合タンパク質がウシコラーゲンI型である、請求項20記載の移植 部材。 22.前記シーラント組成物が透過性低下組成を更に含む、請求項18記載の移植部 材。 23.前記透過性低下組成が約0〜約24%のエタノールである、請求項22記載の移 植部材。 24.前記可塑剤がグリセロール及びソルビトールの1種である、請求項18記載 の移植部材。 25.前記可塑剤が約8〜約30重量%で前記シーラント組成物中に存在する、請求 項24記載の移植部材。 26.前記集合タンパク質が架橋される、請求項18記載の移植部材。 27.前記タンパク質がホルムアルデヒド及びグルタルアルデヒドの1種で架橋さ れる、請求項26記載の移植部材。 28.前記シーラント組成物が生物活性剤を更に含む、請求項18記載の移植部材。 29.前記生物活性剤が抗生物質、抗凝固剤、抗菌剤及びその混合物からなる群よ り選ばれる、請求項28記載の移植部材。 30.前記粘度が約30,000〜約50,000センチポアズである、請求項18記載の移植部 材。 31.前記pHが前記タンパク質粉砕物の十分に等電点外の範囲にあり前記タンパク 質を水性状態で維持する、請求項18記載の移植部材。 32.前記pHが約3.5〜約3.9である、請求項18記載の移植部材。 33.移植製品を体液密着にする生体適合性水性シーラントスラリーの製造方法で あって、 a)生体吸収性自己集合タンパク質を含むペーストを供給する工程; b)前記ペーストを均一な粒径を有する粉末に低温ですりつぶす工程; c)前記粉末と可塑剤及び水とを混合して25℃で約8,000〜約60,000センチポアズ の粘度を有する前記水性シーラントスラリーを形成する工程;及び d)前記ペーストの十分に等電点外のpHで前記スラリーを維持して前記スラリー 中に懸濁した前記タンパク質を維持する工程 を含む、前記方法。 34.前記均一粒径が約5〜約750μmである、請求項33記載の方法。 35.前記pHが約3.5〜3.9である、請求項33記載の方法。 36.前記シーラントスラリーの粘度が約30,000〜約50,000センチポアズである、 請求項33記載の方法。 37.体液密着移植製品の調製方法であって、 a)均一な粒径を有する自己集合タンパク質粉砕物、生体適合性可塑剤及び水を 含む水性スラリーであって、約8,000〜約60,000センチポアズの粘度及び前記ス ラリー中に懸濁した前記タンパク質を十分維持するだけのpHを有する、前記スラ リーを供給する工程; b)前記スラリーを可撓性の多孔性高分子基質の表面に前記タンパク質と前記基 質の前記多孔性構造との密接な会合を十分行わせるだけの力で被覆する工程;及 び c)前記スラリーを前記基質表面上で乾燥させる工程 を含む、前記方法。 38.前記タンパク質が前記乾燥工程前に架橋される、請求項37記載の方法。 39.前記均一な粒径が約5〜約750μmである、請求項37記載の方法。 40.前記pHが約3.5〜3.9である、請求項37記載の方法。 41.前記シーラントスラリーの粘度が約30,000〜約50,000センチポアズである、 請求項37記載の方法。 42.前記力が圧力である、請求項37記載の方法。
