本発明は、肺炎連鎖球菌の一つの新規の表面タンパク質(SpsA-タンパク質)、該表面タンパク質の欠失誘導体、該表面タンパク質とその誘導体用の一つの発現システム及びそれらを使用した一つのワクチンに関する。

【発明の詳細な説明】
肺炎連鎖球菌の新規の表面タンパク質(SpsA-タンパク質)、欠失した誘導体、該
タンパク質用の発現システム及び該タンパク質を含むワクチン
始めに:
肺炎連鎖球菌(Streptococcus pneumoniae)(肺炎球菌)は、一つの莢膜多糖に囲
まれたグラム陰性菌で、1881年初めて健康な保菌者から分離することができた(1
)。この肺炎球菌はヒトの呼吸器官の上部の自然の寄生菌の一種で、40%までの健
康な成人の鼻咽頭にコロニーを形成し、その際4種類までの種々の血清型が数ヶ
月にわたって検出できた(2)。肺炎連鎖球菌は米国その他の多くの国でしばしば
感染症を引き起こす。この感染症は普通内因的に発生し、特に2歳未満の幼児、
60歳を超える高齢者及び免疫反応の低下した人の死亡の原因となる(3)。
更に肺炎球菌は細菌性髄膜炎(ヘモフィルスインフルエンゼb型による)(4)及
び子供の中耳炎(5)を引き起こし易い第二の原因である。例えば米国だけで肺炎
球菌により年間50万の肺炎の事例、700万の中耳炎の事例とそのための4万の死
亡事例が発生している(6,7)。
しかも肺炎連鎖球菌による疾病の死亡率は、抗生物質の普及にもかかわらず一
定の高い水準を維持しており、例えば肺炎球菌による菌血症の死亡率はこの40年
間25乃至29%の間を上下している(8)。
莢膜多糖構造の組成により、肺炎球菌は90種の血清型に分類される(9)。この
莢膜多糖は、多形核白血球(PMN)による食作用から肺炎球菌を保護し、補体の第
二径路の活性化を妨げて(10,11,12)、この肺炎連鎖球菌の重要な病毒性(ビルレ
ンス)の因子である(13,14,15)。
この菌株の病毒性は、莢膜に関してはその多糖類の化学的組成に依存し、莢膜
多糖の大きさには関係しない(16)。
現在使用されている免疫強化用のワクチン(肺炎球菌ワクチン)には、肺炎連
鎖球菌の23種の莢膜多塘の非抱合型の形態が含まれている。即ちこのワクチンは
、菌血症感染の85-90%を引き起こす血清型の多糖類を含み、子どもや老人の場合
には多糖抗原によるヘルパーT-細胞の刺激が認められない(17)ので、このワクチ
ンの適用は成人に限られる。
肺炎球菌感染の治療のもう一つの問題は全世界に広がった抗生物質抵抗性菌種
(ペニシリン耐性菌)の増加である(18)。更にβ-ラクタム系抗生物質の使用は
、肺炎連鎖球菌の感受性菌株の場合に治療された子供の30%に死亡又は治癒でき
ない脳障害を引き起こす。細菌はβ-ラクタム系抗生物質により溶解し、その際
放出された細胞壁成分が脳の容積を増大し、この脳腫張により頭蓋内の圧力が異
常に高くなって不可逆的損傷を招くようになる(19)。
抗生物質抵抗性の増加と多糖類ワクチンの作用の限界のために、莢膜多糖用の
担体として機能できる肺炎球菌タンパク質、又は肺炎
連鎖球菌に於ける固有の強い免疫原性と遍在性とに基づいてその分離物を予防接
種として使用できる肺炎球菌タンパク質の発見に大きな関心が寄せられている。
現在ワクチンの候補に挙げられているタンパク質としては、自己溶菌酵素(20)
,ノイラミニダーセ(21)、肺炎球菌表面アドヘシンA(pneumococcal surface ad
hesin A,PsaA)(22)、肺炎球菌表面タンパク質A(pneumococcal surface prote
in A,PspA)(23)及びニューモリシン(Pneumolysin、一種の肺炎連鎖球菌タンパ
ク質)(24)がある。
このニューモリシンは細胞内タンパク質の一つで、チオール活性毒素属に所属
する(25)。この53kDaの大きさの細胞質のタンパク質は、肺炎球菌が自己溶菌酵
素の影響を受けて自発的に溶菌する時に放出される。高濃度の場合には哺乳動物
の細胞の上にニューモリシンのオリゴマーが形成され、これが膜を貫通する微孔
の増加により細胞の溶解を引き起こす。低濃度では、ニューモリシンが炎症性サ
イトカインの産生を刺激し(26)、試験管内で上部呼吸器官の上皮細胞の単層を破
壊し(27)、好中球の殺菌活性と移動とを低下する(28)。更にニューモリシンは抗
毒素抗体の不存在下で古典的補体径路を活性化する(29)。
肺炎球菌表面タンパク質A(PspA)は、種々の肺炎球菌の菌株の間の構造的及び
抗原的変異性を備えた表面タンパク質で、これらの菌株の機能はこれまで解明さ
れていない。PspAは大抵の臨床分離物の中に存在し(30)、肺炎球菌の全病毒性の
展開にとっても重要である(31,32)。
その他の既知の病毒性因子には、その遺伝子がクローン化され特性が規定され
た(34,35)、IgA1-プロテアーゼ(33)、エラスターゼ(36)の一種のインヒビター
、及び同様に鼻咽頭に寄生する他の連鎖球菌のパーミアーゼに対して相同な、ペ
プチドパーミアーゼがある。ここで特に興味があるのは、その機能の喪失の結果
として真核細胞への肺炎球菌の付着が減少するパーミアーゼである(37)。これら
のパーミアーゼの内でAmi-A-パーミアーゼとパーミアーゼ様タンパク質A(PlpA,
permease-like protein A)とは、小さいペプチドの輸送を受け持つ、タンパク質
に依存するパーミアーゼの属の一員である(38,39)が、これらのパーミアーゼが
真核細胞への付着に直接関与しているかどうかはこれまで立証できなかった。そ
の他肺炎球菌の付着の調節因子として可能性があると見なされるのはピルビン酸
オキシダーゼSpxBで、これはリン酸アセチルの濃度に影響する(40)。
本発明の一つの実施態様は、分泌型免疫グロブリンA(sIgA)に結合することを
特徴とする肺炎連鎖球菌の表面タンパク質(SpsA-タンパク質)に関する。
本発明のもう一つの実施態様は、分泌型IgA(sIgA)に結合することを特徴とす
る肺炎連鎖球菌の分泌タンパク質に関する。
本発明の肺炎連鎖球菌の表面タンパク質又は分泌タンパク質は部分消化される
ことができ、分泌型IgA(sIgA)に結合することを特徴とする。
更に本発明は、分泌型IgA(sIgA)に結合することを特徴とする、
本発明の一つの表面タンパク質又は分泌タンパク質のC-末端が欠失した一つの誘
導体に関する。
更に本発明は、本発明の一つの表面タンパク質又は分泌タンパク質の一つの欠
失した誘導体に関し、且
― 少なくともシグナル配列及び/又は
― 表面タンパク質又は分泌タンパク質の少なくとも随意の反復が欠失し、しか
し
― 分泌型IgA(sIgA)に結合する領域が存在して、誘導体が分泌型IgA(sIgA)
に結合する
ことを特徴とする。
本発明の誘導体は、表面タンパク質又は分泌タンパク質が、分泌型IgA(sIgA)
に結合する領域の所まで欠失していることを特徴とすることができる。
本発明の表面タンパク質は第2図の523個のアミノ酸(部位1乃至523)を特徴と
することができる。
本発明の表面タンパク質のC-末端を欠失した誘導体は第2図の324個のアミノ
酸(部位1乃至324)及び
反復1乃至6(部位325乃至444)又は
反復1乃至7(部位325乃至464)又は
反復1乃至8(部位325乃至484又は乃至485)
を特徴とすることができる。
更に本発明は、表面タンパク質のN-末端を欠失した一つの誘導体に関し、これ
が
(i) 第2図の部位1乃至159の範囲に於いて1乃至最大159までのアミノ酸だけが
欠失し、
(ii) しかし部位160乃至523の範囲では欠失がない
ことを特徴とする。
更に本発明は、本発明の表面タンパク質のN-末端及びC-末端を欠失した一つの
誘導体に関し、これが
(i) 第2図の部位1乃至159の範囲に於いて1乃至最大159までのアミノ酸だけが
欠失し、
(ii) しかし部位160乃至324の範囲では欠失がなく、場合によっては
(iii) 反復1乃至8(部位325乃至484又は乃至485)又は
反復1乃至7(部位325乃至464)又は
反復1乃至6(部位325乃至444)を有する
ことを特徴とする。
本発明の誘導体は、部位174乃至285の範囲には欠失がないことを特徴とするこ
とができる。
本発明のもう一つの実施態様は、そのアミノ酸配列が本発明の表面タンパク質
の配列又はその一つの誘導体の配列と少なくとも80%まで同一である、分泌型IgA
(sIgA)に結合する一つのタンパク質に関する。
本発明のもう一つの実施態様は一つの発現システム、特に本発明の一つの表面
タンパク質、一つの分泌タンパク質、一つの誘導体又は一つのタンパク質、及び
表面タンパク質又は誘導体をコードする
一つのDNA-配列を発現するための大腸菌(Escherichia coli)用の発現システムに
関する。
最後に本発明は肺炎連鎖球菌による罹病予防用のワクチンで、本発明の表面タ
ンパク質、分泌タンパク質又は誘導体の何れか一つを用いて生産可能のワクチン
に関する。
次に本発明を図面、実験結果及び実例を基にして詳しく説明する。

結果:
放射能で標識したヒト分泌型免疫グロブリンA(sIgA)による結合研究とヒトsIg
Aによるウエスタンブロット試験とに於いて、肺炎連鎖球菌と分泌型IgAとの結合
が立証できた。sIgAの結合能を、臨床分離物のみならず、微生物寄託機関(ATCC
乃至NCTC)からの臨床分離物に最も頻繁に認められる血清型の菌株に就いても試
験した。ウェスタンブロットの場合、検討した肺炎球菌の内で73%がsIgAと結合
した(第1図)。更に結合研究では、肺炎球菌タンパク質をプロテアーゼで消化し
た後で、肺炎連鎖球菌の一つのタンパク質により分泌型IgAが結合することを示
すことができた。
sIgA-結合タンパク質をコードする遺伝子の検出は、肺炎連鎖球菌ATCC 33400(
血清1型)のλZAP-発現バンクを分泌型IgAによりスクリーニングして実施した。
選択された陽性のクローンpSHA1はファージミドpBK-CMVの中に肺炎連鎖球菌ATCC
33400の5085bpの大きさの挿入を有している。pSHA1の欠失クローンの構築によ
り、sIgA-結合タンパク質をコードする遺伝子がサブクローンpSHA2に存在するこ
とを立証できた。このサブクローンpSHA2は両方の他の
サブクローンpSHA3及びpSHA4とは異なりウエスタンブロットで分泌型IgAとの結
合を示した。2204bpの大きさの挿入の配列決定とその後の配列の分析の結果、pS
HA2の中にヌクレオチド282乃至1853の一つの読み取り枠が示された(第2図)。こ
の1572bpの大きさの読み取り枠は、血清1型の肺炎球菌の523個のアミノ酸の大
きさのsfgA−結合タンパク質をコードする。SpsA(Streptococcuspneumoniae sec
retory IgA結合タンパク質)と称するこのタンパク質の分子量は59151Daである(
第2図)。
spsAのヌクレオチド酸配列をEMBL-データバンクに登録された配列と比較して
、肺炎連鎖球菌のpspAに対して78.8%の同一性が得られた。タンパク質のレベル
では、PspAに対して64.1%の同一性が立証された。この場合、同一性は特に両方
のタンパク質のC-末端に限定される。PspAのC-末端は、それぞれアミノ酸20個の
長さの10個の反復で構成される(41)。9個の反復から成るSpsAのC-末端の同一性
は92.5%である。
PspAの反復は、肺炎球菌の細胞壁へのタンパク質の付着に関係がある。この機
構には、膜会合したリポテイコ酸とPspsAの反復領域との間にコリンが仲介する
相互作用が必要である(42)。細胞壁に付着するには、20個のアミノ酸の長さの反
復が少なくとも6個発現されることが必要で、さもないとPspAが分泌される(42,
43)。
SpsAのC-末端、即ちSpsAの反復の領域は、PspAの反復と92.5%まで同一である
ので、肺炎球菌の細胞壁への付着にSpsA-反復が関与していると見なすことがで
き、SpsAの二次構造のコンピュータ分析の際に、グラム陽性の細胞壁の中でのタ
ンパク質のアンカリン
グに必要と思われるらせん状の膜貫通構造が検出されなかったと言う事実がこの
SpsAの付着の機構を支持している。
更に以前の試験結果が、pspAの3'-末端は肺炎連鎖球菌の場合の他の配列に相
同な一つの保存された配列であることを示唆している。この相同な配列は肺炎球
菌表面タンパク質A(PspA)に類似の細胞壁への付着機構を有するタンパク質をコ
ードしている可能性がある(44)。
両方のタンパク質SpsAとPspAとのN-末端の同一性はわずか34.5%である。SpsA
のシグナル配列(リーダー)は37個のアミノ酸の長さで、ストレプトコッカスア
ガラクティエ(Streptococcusagalactiae)のIgA-受容体のシグナル配列に対して7
5.7%の同一性を示す(45、46)。
結合領域を解明するために、i)spsA(pQSH 12)とii)N-末端(pQSH14)とiii)S
PSAの9個の反復をコードする核酸配列(pQSH30)とを発現ベクターpQE30の中でク
ローニングし、sIgA-結合をウエスタンブロットにより検討した。このウエスタ
ンブロットでの分析により、SpsAのN-末端における分泌型IgAの結合が認められ
た。更に、SpsAの結合領域を、SpsAのN-末端のアミノ酸1乃至159のみを発現しs
IgAを結合しないpSHA1のもう一つのサブクローン(pSHA3)によりアミノ酸160乃至
324まで狭めることができた(第3図)。
肺炎連鎖球菌ATCC 33400(血清1型)及び肺炎連鎖球菌ATCC 11733(血清2型,R
36A)のサザンブロット分析により、一つのpspA-
特異性,spsA-特異性及び一つの5'-spsA-特異性のDNA-プローブを用いてspsAがp
spAとは同一でないことを立証した(第4図)。
この検討により、spsAのC-末端配列が記載され保存された配列の一つであり、
この配列はpspAに相同的であり、又肺炎連鎖球菌の場合にタンパク質の細胞壁へ
の付着に関与することが立証できた。
従ってspsAは肺炎連鎖球菌の一つの新規の表面タンパク質をコードする。この
タンパク質の生物学的機能が即ちSpsAのN-末端における分泌型免疫グロブリンA
の結合である。
一つのIgA-J-IgA-SC(SC:分泌成分)複合体から成る分泌型IgAは、ヒトの呼吸器
官と消化器官との中の最も重要な免疫グロブリンである。この分泌型IgAの前段
階である、15.6kDaの大きさのJ鎖により結合されたIgA-二量体は、B-リンパ球
の中で合成され、上皮細胞の基底外側面の表面に局在したポリ免疫グロブリン受
容体に結合する(47)。このポリ-Ig-受容体は細胞によるトランスサイトーシスを
仲介する。その際受容体のC-末端の部分が分離し、(IgA)2-J-鎖複合体の分泌成
分になる。細胞の先端の膜への輸送の後で複合体は膜と融合して分泌型IgAとし
て放出される(48)。この分泌成分はジスルフィド結合により複合体に共有結合し
ているが、これが合成されたsIgAを外部の液体の中で変性とタンパク質分解とに
対して保護する。
肺炎球菌のヒト上皮細胞への付着の研究が示したように、結合研究に於いてsI
gAを結合した菌株に対しては上皮細胞当たり100個
の肺炎球菌よりも大きい一つの付着が見られたが、結合研究の結果がマイナスの
菌株の場合はμ2の肺炎球菌の付着しか認められなかった。
これらの結果は、前記SpsAが肺炎球菌の上皮細胞への付着と上皮細胞への浸潤
とに関与していることを示している。
従って肺炎連鎖球菌の分泌型IgA-結合タンパク質SpsAは、ワクチンの開発に対
する有望な新規の候補者である。
例1:
この例ではウエスタンブロットに於ける肺炎連鎖球菌の菌株の分泌型IgA-結合
を説明する。
肺炎連鎖球菌の溶解物のタンパク質[1.0のOD600に調整し、細菌を100μlの溶
解細胞溶液(グリセリン20%、SDS3%、β-メルカプトエタノール3%、ブロモフ
ェノールブルー0.05%)に採取後94℃にて10分煮沸]をSDS-PAGEによりほぐしてか
らニトロセルロース膜の上に移した。10%脱脂乳の0.1M PBS溶液で飽和後、ニト
ロセルロースフィルタに分泌型IgA[1μg/ml](Sigma,ミュンヘン、ドイツ)の0
.1M PBS溶液を加えて室温で1時間振盪しながらインキュベートした。0.1M PBS
にて3回洗浄後、前記フィルタにヤギ-抗ヒトIgA-HRP-抱合体抗体(IgA-HRP-Konj
ugat Antikoerper)(ICN,Eschwege、ドイツ)を加えて1時間インキュベートした
。1mgの4-クロロ-4-ナフトールとPBS1ml当たり0.1%のH2O2とにより3回洗浄後
発色が得られた。
例2:
この例では肺炎連鎖球菌の菌株の125ヨードで標識した分泌型IgAの結合を説明
する。
0.05Mリン酸塩緩衝液1ml(pH7.5)中の100ng sIgAを、20μgのクロラミンTを
添加後、350μCiの125ヨードを加えてインキュベートし、5分後に反応を20μg
のメタ重亜硫酸ナトリウムを加えて停止した。標識タンパク質を無標識タンパク
質からPD10-カラム(Pharmacia、フライブルク、ドイツ)を用いて分離し、-20
℃で凍結した。PBST(0.05%のトゥイーン20を含むPBS)中の肺炎球菌懸濁液(600nm
の透過率10%)250μlに0.023μCiの125ヨード標識sIgAを加えて45分インキュベ
ートし、1mlのPBSTの添加により反応を停止した。肺炎連鎖球菌へのsIgA-結合
の測定はγ線計数器(Packard,Dreieich,ドイツ)中の細菌の活性の測定により
実施した。
例3:
この例では、ベクターλZAP ExpressTM(商品名)における肺炎連鎖球菌ATCC334
00の染色体DNAのクローニングと肺炎連鎖球菌分泌型IgA結合タンパク質(SpsA)を
求める遺伝子バンクのスクリーニングとを説明する。
肺炎連鎖球菌ATCC33400の染色体DNAを分離し、Sau3Aで部分消化し、20%の塩化
ナトリウム溶液の冷凍と解凍とにより形成した塩化ナトリウム勾配の中でDNA-断
片の大きさにより分画した。BamHI切断λZAP ExpressへTMの染色体DNAの2.0kb乃
至6.0kbの大きさのDNA-断片の連結反応とインビトロ パッケージングとを市販
の
セット(Stratagene,ハイデルベルク、ドイツ)を用いてメーカーの記載に従って
実施した。ファージ遺伝子バンクをそれ以上増幅せずにプレートアウトし、分泌
型IgA-結合タンパク質の発現について組換えプラークを調べた。タンパク質をニ
トロセルロースフィルタに移し、10%脱脂乳の0.1M PBS溶液で飽和後、前記フィ
ルタに分泌型IgA[1μg/ml](Sigma,ミュンヘン、ドイツ)の0.1M PBS溶液を加
えて室温で1時間振盪しながらインキュベートした。0.1M PBSにて3回洗浄後、
上記フィルタにヤギ-抗ヒトIgA-HRP-抱合体抗体(IgA-HRP-Konjugat Antikoerper
)を加えて1時間インキュベートした。発色は、1mgの4-クロロ-1-ナフトールと
PBS1ml中の0.1%のH2O2とにより3回洗浄後に実現した。陽性のプラークを単離
し、増幅後pBK-CMV-ファージミドのインビボ切り出しをExassistヘルパーファー
ジとXLOLRシステムとを使用してメーカー(Stratagene,ハイデルベルク、ドイツ
)の記載に従って実施した。
例4:
この例にはDNA-配列決定とアミノ酸配列の誘導とを記載する。pSHA1の名称の
、ファージミドpBK-CMV中の肺炎連鎖球菌ATCC 33400の5085kbの挿入物の配列決
定は、ABI PRISMTMDye Terminator Cycle Sequencingによりメーカー(Perkin-El
mer、ドイツ)の記載に従って、ベクタープライマーT3X(5'-AATTAACCCTCACT AAAG
GG-3')とT7X(5'-TAATACGACTCACTAT CGGG-3')と得られた配列から誘導したプライ
マー(次表参照)とを用いて実施した。核酸配列のアミノ酸配列への翻訳にはプロ
グラムGene-Works,Version2.45(Intelligenetics,Montain View,カリフォル
ニア、米国)を使用した。
例5:
この例にはpSHA1のサブクローンの構築とウエスタンブロットでのその特性決
定とを記載する。
サブクローンpSHA2(pSHA1のnt1-nt2203)は2882bpの大きさのEcoRI-断片の欠失
により得られ、サブクローンpSHA3(ntl-nt757)はpSHA1の4328bpの大きさのHindI
II-断片の欠失により得られる。もう一つのpSHA4の名称のサブクローン(pSHA1の
nt2952-nt5085)は2133bpの大きさのSacI-断片の欠失により得られる。
クローンの特性決定には、組換えE.coli細胞の溶解細胞のタンパク質[1.0のO
D600に調整し、細菌を100μlの溶解細胞溶液(グリセリン20%、SDS3%、β-メ
ルカプトエタノール3%、ブロモフェノールブルー0.05%)に採取後94℃にて10分
煮沸]をSDS-PAGEによりほぐしてから、分泌型IgAを用いたウエスタンブロット
の中のニトロセルロース膜の上に移した。10%脱脂乳の0.1M PBS溶液で飽和後、
ニトロセルロースフィルタに分泌型IgA[1μg/ml](Sigma,ミュンヘン、ドイツ
)の0.1M PBS溶液を加えて室温で1時間振盪しながらインキュベートした。0.1M
PBSにて3回洗浄後、前記フィルタにヤギ-抗ヒトIgA-HRP-抱合体抗体(IgA-HRP-
Konjugat Antikoerper)を加えて1時間インキュベートした。1mgの4-クロロ-1-
ナフトールとPBS1ml当たり0.1%のH2O2とにより3回洗浄後発色が得られた。
例6:
この例には、発現ベクターpQE30(Pharma-cia)へのspsAとspsAの5'-範囲(nt1-n

spsA用とspsA-断片用とのPCRプライマーは、肺炎連鎖球菌ATCC 33400血清1型
のpSHA1の中で得られたspsA-配列から誘導された。5'-プライマーSH22(5'-GCGC
GCGCGCGGATCCTTGTTTGCATCAAAAAGCGAAAG-3')は39bpの長さでspsA-遺伝子の変化
した開始コドン(ATGの代りにTTG)で始まる。反復用の5'-プライマーSH24(5'-G
CGCGCGCGCGGATCCACAGGCTG GAAACAAGAAAAC-3')はspsA-遺伝子のヌクレオチド973
に於いて最初の反復の開始配列で始まる。spsASH23の3'-プライマー(CTCAGCTAA
TTAAGCTTGTTTAGTTTACCCATTCACCATTGGC-3')は停止コドンで始まり、N-末端の3'-
プライマーSH 25(5'-CTCAGCTAATTAAGCTTTTTTGGAGTAGATGGTTGTGCTGG-3')はspsA-
遺伝子のヌクレオチド972に於いて始まる。プライマーSH22-SH23はpQSH12の構築
に、プライマーSH22-SH25はpQSH14の構築に、プライマーSH24-SH23はpQSH30の構
築に使用された。5'-プライマーはクローニング用に一つのBamHI制限切断部位を
、3'-プライマーは一つのHindIII制限切断部位を含んでいた。
5'-及び3'-プライマー(各20pmol)によるゲノム肺炎球菌-DNAの増幅は、サー
モサイクラー(Thermocycler,MWG-Biotech,Ebersberg,ドイツ)の中の100μlの
容積の中でGoldstar Taq-ポリメラーゼの2.5ユニットをメーカー(Eurogentec,S
eraing,ベルギー)の記載通り用い、更に50ngの染色体-DNAを用いて実施した。
プローブを94℃で2分間変性し、増幅にはDNAの94℃での1分間の変性とプライ
マーの55℃での1分間のアニーリングと72℃での2分間の伸長とから成るサイク
ルを35回実施した。
プライマーSH22乃至SH23は又、肺炎連鎖球菌の血清2型(R36A滑面、ATCC 1173
3とD39,NCTC 7466)と血清47型(R36A粗面、NCTC 10319)との増幅とクローニン
グとに使用することができた。
プライマーSH22乃至SH25も又、肺炎連鎖球菌の血清47型(R36A粗面、NCTC 103
19)の5'-領域の増幅とクローニングとに使用することができた。
例7:
この例には肺炎連鎖球菌の菌株のヒト上皮細胞への付着の調査を記載する。
DMEM/5%コウシ胎児血清(FCS)中で生育した周密的HEp-2喉頭癌細胞又はA549肺
胞細胞(2x105)に、DMEM/1mM HEPESにより細胞を洗浄した後、DMEM/1mM HEPESの
中の107個の肺炎球菌を37℃で1時間感染させた。次に細胞をPBSで3回洗浄しメ
タノールで固定した(-20℃、30分)。細胞外の肺炎球菌を顕微鏡計数のためにギ
ムザによりメーカー(Sigma Diag-nostic、ミュンヘン、ドイツ)の記載に従って
染色した。付着した肺炎球菌の数を少なくとも100個の上皮細胞での計数により
測定した。

請求の範囲
1. 分泌型IgA(sIgA)の結合のための肺炎連鎖球菌の一つの表面タンパク質(SpsA
-タンパク質)の使用。
2. 分泌型IgA(sIgA)の結合のための請求の範囲第1項記載の肺炎連鎖球菌の部
分的に消化された表面タンパク質の使用。
3. 分泌型IgA(sIgA)の結合のための請求の範囲第1項記載の一つの表面タンパ
ク質の一つのC-末端欠失誘導体の使用。
4. 分泌型IgA(sIgA)の結合のための請求の範囲第1項記載の一つの表面タンパ
ク質の一つの欠失誘導体の使用であって、その際
― 少なくともシグナル配列及び/又は
― 前記表面タンパク質の少なくとも随意の反復が欠失し、しかし
― 分泌型IgA(sIgA)に結合する領域が存在して、前記誘導体が分泌型IgA(
sIgA)に結合する
前記誘導体の使用。
5. 前記表面タンパク質が分泌型IgA(sIgA)に結合する前記領域の所まで欠失し
ている請求の範囲第4項記載の一つの誘導体の使用。
6. 分泌型IgA(sIgA)の結合のための、第2図の523個のアミノ酸(部位1乃至523
)を特徴とする請求の範囲第1項記載の一つの表面タンパク質の使用。
7. 分泌型IgA(sIgA)の結合のための、第2図の324個のアミノ酸(部位1乃至324
)及び
反復1乃至6(部位325乃至444)又は
反復1乃至7(部位325乃至464)又は
反復1乃至8(部位325乃至484又は乃至485)
を特徴とする請求の範囲第1項又は第6項記載の表面タンパク質の一つのC-末
端欠失誘導体の使用
8. 分泌型IgA(sIgA)の結合のための、請求の範囲第1項又は第6項記載の表面
タンパク質の一つのN-末端欠失誘導体の使用であって、
(i) 第2図の部位1乃至159の範囲に於いて1乃至最大159までのアミノ酸だけ
が欠失し、
(ii) しかし部位160乃至523の範囲では欠失がない
前記誘導体の使用。
9. 分泌型IgA(sIgA)の結合のための、請求の範囲第1項又は第6項記載の表面
タンパク質のN-末端及びC-末端を欠失した一つの誘導体の使用であって、
(i) 第2図の部位1乃至159の範囲で1乃至最大159までのアミノ酸だけが欠失
し、
(ii) しかし部位160乃至324の範囲では欠失がなく、場合によっては
(iii) 反復1乃至8(部位325乃至484又は乃至485)又は
反復1乃至7(部位325乃至464)又は
反復1乃至6(部位325乃至444)を有する
前記誘導体の使用。
10.分泌型IgA(sIgA)の結合のための、部位174乃至285の範囲には欠失がない請
求の範囲第9項記載の一つの誘導体の使用。
11.分泌型IgA(sIgA)に結合する肺炎連鎖球菌の一つの表面タンパク質(SpsA-タ
ンパク質)を使用して生産可能な、肺炎連鎖球菌による罹病予防用のワクチン。
12.分泌型IgA(sIgA)に結合する請求の範囲第1項記載の肺炎連鎖球菌の一つの
部分的に消化された表面タンパク質を使用して生産可能な、肺炎連鎖球菌による
罹病予防用のワクチン。
13.分泌型IgA(sIgA)にを結合する請求の範囲第1項記載の一つの表面タンパク
質の一つのC-末端欠失誘導体を使用して生産可能な、肺炎連
鎖球菌による罹病予防用のワクチン。
14.請求の範囲第1項記載の一つの表面タンパク質の一つの欠失誘導体であって
、その際
― 少なくともシグナル配列及び/又は
― 前記表面タンパク質の少なくとも随意の反復が欠失し、しかし
― 分泌型IgA(sIgA)に結合する領域が存在して、前記誘導体が分泌型IgA(
sIgA)に結合する
前記誘導体を使用して生産可能な、肺炎連鎖球菌による罹病予防用のワクチン
。
15.前記表面タンパク質が分泌型IgA(sIgA)に結合する領域の所まで欠失してい
る請求の範囲第5項記載の一つの誘導体を使用して生産可能な、肺炎連鎖球菌に
よる罹病予防用のワクチン。
16.第2図の523個のアミノ酸(部位1乃至523)を特徴とする請求の範囲第1項記
載の一つの表面タンパク質を使用して生産可能な、肺炎連鎖球菌による罹病予防
用のワクチン。
17.第2図の324個のアミノ酸(部位1乃至324)及び
反復1乃至6(部位325乃至444)又は
反復1乃至7(部位325乃至464)又は
反復1乃至8(部位325乃至484又は乃至485)
を特徴とする請求の範囲第1項又は第16項記載の表面タンパク質の一つのC-末
端欠失誘導体を使用して生産可能な、肺炎連鎖球菌による罹病予防用のワクチン
。
18.請求の範囲第1項又は第16項記載の表面タンパク質の一つのN-末端欠失誘導
体であって、
(i) 第2図の部位1乃至159の範囲に於いて1乃至最大159までのアミノ酸だけ
が欠失し、
(ii) しかし部位160乃至523の範囲では欠失がない
前記誘導体を使用して生産可能な、肺炎連鎖球菌による罹病予防用のワクチン
。
19.請求の範囲第1項又は第16項記載の表面タンパク質のN-末端及びC-末端を欠
失した一つの誘導体であって、
(i) 第2図の部位1乃至159の範囲に於いて1乃至最大159までのアミノ酸だけ
が欠失し、
(ii) しかし部位160乃至324の範囲では欠失がなく、場合によっては
(iii) 反復1乃至8(部位325乃至484又は乃至485)又は
反復1乃至7(部位325乃至464)又は
反復1乃至6(部位325乃至444)を有する
前記誘導体を使用して生産可能な、肺炎連鎖球菌による罹病予防用のワクチン
。
20.部位174乃至285の範囲には欠失がない請求の範囲第19項記載の一つの誘導体
を使用して生産可能な、肺炎連鎖球菌による罹病予防用のワクチン。

- 【特許請求の範囲】 1. 分泌型IgA(sIgA)に結合することを特徴とする肺炎連鎖球菌の表面タンパク 質(SpsA-タンパク質)。 2. 分泌型IgA(sIgA)に結合することを特徴とする肺炎連鎖球菌の分泌タンパク 質。 3. 分泌型IgA(sIgA)に結合することを特徴とする部分的に消化された請求の範 囲第1項又は第2項記載の表面タンパク質又は分泌タンパク質。 4. 分泌型IgA(sIgA)に結合することを特徴とする請求の範囲第1項又は第2項 記載の表面タンパク質又は分泌タンパク質のC-末端欠失誘導体。 5. 請求の範囲第1項又は第2項記載の表面タンパク質又は分泌タンパク質の欠 失誘導体であって、 ― 少なくともシグナル配列及び/又は ― 前記表面タンパク質又は分泌タンパク質の少なくとも随意の反復が欠失し 、しかし ― 分泌型IgA(sIgA)に結合する領域が存在して、前記誘導体が分泌型IgA( sIgA)に結合する ことを特徴とする前記誘導体 6. 前記表面タンパク質又は前記分泌タンパク質が、分泌型IgA(sIgA)に結合す る前記領域の所まで欠失していることを特徴とする請求の範囲第5項記載の誘導 体。 7. 第2図の523個のアミノ酸(部位1乃至523)を特徴とする請求の範囲第1項記 載の表面タンパク質。 8. 第2図の324個のアミノ酸(部位1乃至324)及び 反復1乃至6(部位325乃至444)又は 反復1乃至7(部位325乃至464)又は 反復1乃至8(部位325乃至484又は乃至485) を特徴とする請求の範囲第1項又は第7項記載の表面タンパク質のC-末端欠失 誘導体 9. 請求の範囲第1項又は第7項記載の表面タンパク質のN-末端欠失誘導体であ って、 (i) 第2図の部位1乃至159の範囲に於いて1乃至最大159までのアミノ酸だけ が欠失し、 (ii) しかし部位160乃至523の範囲では欠失がない ことを特徴とする前記誘導体。 10.請求の範囲第1項又は第7項記載の表面タンパク質のN-末端及びC-末端を欠 失した一つの誘導体であって、 (i) 第2図の部位1乃至159の範囲に於いて1乃至最大159までのアミノ酸だけ が欠失し、 (ii) しかし部位160乃至324の範囲では欠失がなく、場合によっては (iii) 反復1乃至8(部位325乃至484又は乃至485)又は 反復1乃至7(部位325乃至464)又は 反復1乃至6(部位325乃至444)を有する ことを特徴とする前記誘導体。 11.部位174乃至285の範囲には欠失がないことを特徴とする請求の範囲第10項記 載の誘導体。 12.そのアミノ酸配列が請求の範囲第7項記載の表面タンパク質の配列又は請求 の範囲第8項、第9項、第10項又は第11項記載の誘導体の配列と少なくとも80% まで同一であることを特徴とする分泌型IgA(sIgA)を結合する一つのタンパク質 。 13.一つの発現システム、特に前記請求の範囲の何れか1項記載の 一つの表面タンパク質、一つの分泌タンパク質、一つの誘導体又は一つのタンパ ク質、及び表面タンパク質又は誘導体をコードする一つのDNA-配列を発現するた めの大腸菌(Escherichia coli)用の発現システム。 14.肺炎連鎖球菌による罹病予防用のワクチンであって、請求の範囲第1項乃至 第12項の何れか1項記載の一つの分泌タンパク質、又は一つの表面タンパク質又 は一つの誘導体を用いて生産可能の前記ワクチン。
