Claims (1)
- 【特許請求の範囲】 1.前立腺癌抗原3(PCA3)をコードする単離された核酸分子。 2.次の(a)〜(e)よりなる群より選ばれる塩基配列と少なくとも90%の 相同性を有する塩基配列を包含することを特徴とする、請求項1の単離された核 酸分子。 (a)配列番号2の全長アミノ酸配列を包含するPCA3ポリペプチドをコー ドする塩基配列; (b)配列番号7の全長アミノ酸配列を包含するPCA3ポリペプチドをコー ドする塩基配列; (c)Centraal voor Schimmelculturesに受託番号CBS 682.97として寄託され ているポリヌクレオチドクローンがコードしている全長アミノ酸配列を包含する PCA3ポリペプチドをコードする塩基配列; (d)Centraal voor Schimmelculturesに受託番号CBS 100521として寄託され ているポリヌクレオチドクローンがコードしている全長アミノ酸配列を包含する PCA3ポリペプチドをコードする塩基配列;及び (e)上記(a)、(b)、(c)及び(d)の塩基配列のいずれかに相補的 な塩基配列。 3.配列番号1又は6のPCA3をコードする塩基配列を包含することを特徴と する、請求項1の単離された核酸分子。 4.配列番号1、3、4又は6の塩基配列を包含する、請求項1の単離された核 酸分子。 5.配列番号2又は7の全長アミノ酸配列を包含するポリペプチドをコードする ことを特徴とする、請求項1の単離された核酸分子。 6.Centraal voor Schimmelculturesに受託番号CBS 682.97として寄託されてい るポリヌクレオチドクローンがコードしている全長アミノ酸配列を包含するPC A3ポリペプチドをコードしていることを特徴とする、請求項1の単離された核 酸分子。 7.Centraal voor Schimmelculturesに受託番号CBS 100521として寄託されてい るポリヌクレオチドクローンがコードしている全長アミノ酸配列を包含するPC A3ポリペプチドをコードしていることを特徴とする、請求項1の単離された核 酸分子。 8.PCA3をコードするRNA又はDNAに特異的にハイ ブリダイズする10〜50塩基からなる単離された核酸分子であって、PCA3 のエクソン1、2、3、4a、4b、4c又は4dの塩基配列の中の少なくとも 10個の連続した塩基からなるエクソン塩基配列、又は該エクソン塩基配列に相 補的な塩基配列からなる核酸分子であり、該核酸分子は、配列番号1の511番 〜985番の配列、配列番号1の567番〜961番の配列、配列番号6の53 3番〜1007番の配列、又は配列番号6の589番〜983番の配列には特異 的にハイブリダイズしないことを特徴とする、単離された核酸分子。 9.サンプル材料中のPCA3核酸を検出する方法にして、 a)ハイブリダイゼーションの起こりうる条件下において該サンプル材料と請 求項8の核酸分子とを接触せしめ、 b)PCA3核酸に結合した該核酸分子の存在を検出する、ことを包含する方 法。 10.請求項8の核酸分子の入った少なくとも1つの容器手段を包含することを 特徴とする、サンプル材料中のPCA3核酸を検出するためのキット。 11.5’から3’への方向に、宿主細胞において転写を開始させるのに有効な プロモーター、及び請求項1の核酸分子 を包含することを特徴とする組換え核酸分子。 12.ベクター及び請求項1の核酸分子を包含することを特徴とする組換え核酸 分子。 13.請求項11の組換え核酸分子を含む細胞。 14.請求項11の組換え核酸分子を含むヒト以外の生物。 15.精製されたPCA3ポリペプチド、又はそのエピトープ含有領域。 16.次の(a)〜(e)よりなる群より選ばれるアミノ酸配列と少なくとも9 0%の相同性を有するアミノ酸配列を包含することを特徴とする、請求項14の 精製されたなPCA3ポリペプチド。 (a)配列番号2の全長アミノ酸配列を包含するPCA3ポリペプチドのアミ ノ酸配列; (b)配列番号7の全長アミノ酸配列を包含するPCA3ポリペプチドのアミ ノ酸配列; (c)Centraal voor Schimmelculturesに受託番号CBS 682.97として寄託され ているcDNAクローンがコードしている全長アミノ酸配列を包含するPCA3 ポリペプチドの アミノ酸配列; (d)Centraal voor Schimmelculturesに受託番号CBS 100521として寄託され ているcDNAクローンがコードしている全長アミノ酸配列を包含するPCA3 ポリペプチドのアミノ酸配列;及び (e)上記(a)、(b)、(c)及び(d)のいずれかのポリペプチドのエ ピトープ含有領域のアミノ酸配列。 17.請求項15のポリペプチド又はそのエピトープ含有領域に対して特異的な 結合親和性を有する抗体。 18.サンプル材料中のPCA3ポリペプチドを検出する方法にして、 a)免疫複合体が形成され得る条件下において、該サンプル材料と請求項17 の抗体とを接触せしめ、 b)PCA3ポリペプチドに結合した該抗体の存在を検出することを包含する 方法。 19.診断用キットであって、 a)請求項17の抗体の入った第1容器手段、及び b)該抗体に結合しうる物質と標識物質とからなる検出用結合物質の入った第 2容器手段を包含するキット。 20.請求項17の抗体を製造するハイブリドーマ。 21.治療に有効な量の、請求項17の抗体を哺乳類に投与することを包含する 、哺乳類の前立腺癌の治療方法。 22.治療に有効な量の、アンチセンスPCA3核酸分子を哺乳類に投与するこ とを包含する、哺乳類の前立腺癌の治療方法。 23.患者の有する前立腺癌又は前立腺癌の素因を診断するための方法であって 、 a)該患者からサンプルを採取し、 b)該サンプル中のPCA3RNA又はPCA3タンパク質の量を定量し、 c)該サンプル中の該PCA3RNA又は該PCA3タンパク質の量を、前立 腺癌を患っていない健常者の有するPCA3RNA又はPCA3タンパク質の量 と比較し、該サンプル中の該PCA3RNA又は該PCA3タンパク質の量が増 加している場合に、該患者は前立腺癌又は前立腺癌の素因を有すると診断するこ とを特徴とする診断方法。
