Claims (1)
- 【特許請求の範囲】 1.a)陰イオン性界面活性剤の存在下、抗体、そのフラグメント、または抗原 を、酸性溶液中に存在する固相と接触させ、該抗体、そのフラグメント、または 該抗原を固相に付着させ、そして b)段階a)で得た、抗体、そのフラグメント、または抗原と、固相との組み合 わせ中に、塩基性溶液を添加してpHを塩基性に変え、それらの抗体、フラグメ ントまたは抗原の一級または二級アミノ基と、固相の官能基との間に共有結合を 形成させること、 を含んでなる、抗体分子、そのフラグメント、または抗原分子を固相と共有結合 させる方法。 2.酸性溶液が、塩酸溶液、好ましくは5mM HCl、pH2〜4、である、 請求項1に記載の方法。 3.塩基性溶液が、ホウ酸塩緩衝液、好ましくは0.1Mホウ酸塩緩衝液、pH 9.0、である、請求項1に記載の方法。 4.陰イオン性界面活性剤が、濃度0.1から0.01%のポリオキシ-1,2-エ タン-ジイル-α-ノニルフェニル-o-ヒドロキシ-ホスフェート 界面活性剤であ る、請求項1に記載の方法。 5.固相の共有結合基が、クロロメチル、エポキシ、アルデヒド、トシル、メシ ルおよびn-ヒドロキシスクシンイミド基、およびそれらの基の混合物から選択さ れる、好ましくはクロロメチルである、請求項1に記載の方法。 6.該方法中の酸性の段階に5から30分、好ましくは15分かける、請求項1 に記載の方法。 7.塩基性段階の結合を酸性段階の直後に開始させ、1から24時間、好ましく は18時間かける、請求項1に記載の方法。 8.固相が、ポリマープラスティック、マイクロタイタープレート、試験管、ア ッセイキュベット、免疫クロマトグラフィー物質、フィルター、試験ストリップ または金コロイド、好ましくはポリマープラスティックである、請求項1に記載 の方法。 9.ポリマー粒子が、ホモポリマーまたはコポリマーから形成されているか、ま たはコアーシェル粒子である、請求項8に記載の方法。 10.ポリマー粒子の構造成分が、ポリスチレン、ポリビニルナフタレン、ポリビ ニルカルバゾール、ポリビニルトルエン、ポリビニルブチルスチレン、ポリビニ ルベンゼンおよびポリビニルクロライド、およびそれらの混合物から選択される 、請求項9に記載の方法。 11.ポリマー粒子の構造成分が、ポリスチレンおよびポリビニルクロライドの共 重合体である、請求項10に記載の方法。 12.ポリマー粒子のサイズが、10nmから10000nm、好ましくは90nmであ る、請求項10または11に記載の方法。 13.請求項1に記載の方法を使用して、抗体分子、そのフラグメント、または抗 原分子を共有的に付着させた固相を含んでなる、固相-生体分子コンジュゲート 。 14.スチレンビニル-ベンジル-クロライドポリマー粒子と結合した、ヒツジ-抗- ヒト-CRP抗体のF(ab,)2フラグメントを含んでなる、請求項13に記載の固相- 生体分子コンジュゲート。 15.請求項13または14に記載の固相-生体分子コンジュゲートを含んでなる、診 断用試験キット。
