Claims (1)
- 【特許請求の範囲】 1.グロース・チャンバと、軟骨を生成させるのに適した細胞の付着のための基 質と、この基質に付着した細胞に約1〜約100ダイン/cm2の剪断流れ応力 をもたらすために液体培養媒質と基質との間の相対運動を与えるための手段とを 備えてなる軟骨生成用バイオリアクタ。 2.約1〜約50ダイン/cm2の剪断流れ応力を加えるための手段をさらに備 えている請求項1記載のバイオリアクタ。 3.剪断流れ応力を加えるための前記手段が、貯蔵器と、ポンプと、前記グロー ス・チャンバ、前記貯蔵器及び前記ポンプを相互接続するための管類とを備え、 前記ポンプによって与えられた駆動力に応じて、前記貯蔵器から前記グロース・ チャンバを通り再び前記貯蔵器へ戻る液体増殖媒質の連続流を生じさせるように されている請求項1記載のバイオリアクタ。 4.前記基質が、骨格である請求項1記載のバイオリアクタ。 5.前記骨格が、生体吸収性のものである請求項1記載のバイオリアクタ。 6.前記基質が、前記細胞の増殖を単層で支持する非多孔性面 である請求項1記載のバイオリアクタ。 7.前記非多孔性面が、回転可能なドラムの表面である請求項6記載のバイオリ アクタ。 8.前記非多孔性面が、回転可能なディスクの表面である請求項6記載のバイオ リアクタ。 9.前記非多孔性面が、静止プレートである請求項6記載のバイオリアクタ。 10.(a)細胞付着のための基質を備えたグロース・チャンバを用意するステッ プと、 (b)その基質を液体増殖媒質で被覆するステップと、 (c)軟骨を生成させるのに適した細胞を前記媒質に接種するステップと、 (d)前記細胞を前記基質に付着させるステップと、 (e)前記細胞に約1〜約100ダイン/cm2の剪断流れ応力をもたらす ために、液体増殖媒質と基質に付着した細胞との間に相対運動を加えてそれを維 持し、それによって剪断流れ応力の加えられた細胞を生成するステップと、 (f)前記剪断流れ応力の加えられた細胞を、軟骨を生成させるのに充分な 時間だけ培養するステップとを備えている軟骨生成方法。 11.前記剪断流れ応力が、約1〜約50ダイン/cm2である請求項10記載の方 法。 12.前記基質が、骨格である請求項10記載の方法。 13.前記骨格が、生体吸収性のものである請求項10記載の方法。 14.前記基質が、前記細胞の増殖を単層で支持する非多孔性面である請求項10記 載の方法。 15.前記非多孔性面が、回転可能なドラムの表面である請求項10記載の方法。 16.前記非多孔性面が、回転可能なディスクの表面である請求項10記載の方法。 17.前記非多孔性面が、静止プレートである請求項10記載の方法。 18.前記剪断流れ応力の加えられた細胞が、 (a)軟骨細胞の表現型の増進した維持を表し、かつ (b)II型コラーゲンのI型コラーゲンに対する増大した比を含む細胞外マ トリックスを生成させる請求項10記載の方法。 19.(a)細胞付着のための基質を備えたグロース・チャンバを用意するステッ プと、 (b)その基質を液体増殖媒質で被覆するステップと、 (c)前記媒質に哺乳類幹細胞を接種するステップと、 (d)前記細胞を前記基質に付着させるステップと、 (e)前記幹細胞に約1〜約100ダイン/cm2の剪断流れ応力をもたら すために、液体増殖媒質と基質に付着した細胞との間に相対運動を加えてそれを 維持し、それによって剪断流れ応力の加えられた幹細胞を生成するステップと、 (f)前記剪断流れ応力の加えられた幹細胞を、軟骨細胞に分化させるのに 充分な時間だけ培養するステップとを備えている 幹細胞の軟骨細胞への分化誘発方法。 20.前記剪断流れ応力が、約1〜約50ダイン/cm2である請求項19記載の方 法。 21.(a)細胞付着のための基質を備えたグロース・チャンバを用意するステッ プと、 (b)その基質を液体増殖媒質で被覆するステップと、 (c)前記媒質に、軟骨細胞または軟骨幹細胞以外の哺乳類細胞を接種する ステップと、 (d)前記細胞を前記基質に付着させるステップと、 (e)前記細胞に約1〜約100ダイン/cm2の剪断流れ応力をもたらす ために、液体増殖媒質と基質に付着した細胞との間に相対運動を加えてそれを維 持し、それによって剪断流れ応力の加えられた細胞を生成するステップと、 (f)前記剪断流れ応力の加えられた細胞を、軟骨細胞に完全分化させるの に充分な時間だけ培養するステップとを備えている 培養細胞の軟骨細胞への完全分化誘発方法。 22.前記剪断流れ応力が、約1〜約50ダイン/cm2である請求項21記載の方 法。 23.分化された前記哺乳類細胞が、繊維芽細胞及び筋細胞から構成される群から 選ばれる請求項21記載の方法。 24.細胞が、軟骨細胞、軟骨幹細胞、間葉系幹細胞、及び軟骨細胞の表現型へ完 全分化する細胞から構成される群から選ばれる請求項1記載のバイオリアクタ。 25.骨格が、生体吸収性のものである請求項4記載のバイオリアクタ。 26.骨格が、生分解性のものである請求項25記載のバイオリアクタ。 27.骨格が、非生分解性のものである請求項25記載のバイオリアクタ。 28.細胞が、軟骨細胞、軟骨幹細胞、間葉系幹細胞、及び軟骨細胞の表現型へ完 全分化する細胞から構成される群から選ばれる請求項10記載の方法。 29.骨格が、生体吸収性のものである請求項12記載の方法。 30.骨格が、生分解性のものである請求項29記載の方法。 31.骨格が、非生分解性のものである請求項29記載の方法。
