Claims (1)
- 【特許請求の範囲】 1.式 (式中、 R1は、 (a) 水素、 (b) 低級アルキル、 (c) アルケニル、 (d) アルコキシ、 (e) チオアルコキシ、 (f) ハロ、 (g) ハロアルキル、 (h) アリール−L2 −(式中、L2は存在しないか、または −CH2−、 −CH2CH2−、 −CH(CH3)−、 −O−、 −S(O)q−(但し、qは0、1または2)、 −N(R)−(但し、Rは水素または低級アルキル)、お よび −C(O)− からなる群から選択され、 アリールは、 フェニル、 ナフチル、 テトラヒドロナフチル、 インダニル、および インデニル からなる群から選択され、かつアリール基は未置換であるかまたは置換されてい る)、 (i) 複素環式−L3 −(式中、L3は存在しないか、または −CH2−、 −CH2CH2−、 −CH(CH3)−、 −O−、 −S(O)q−(但し、qは0、1または2)、 −N(R)−(但し、Rは水素または低級アルキル)、お よび −C(O)− からなる群から選択され、 複素環は単環式複素環であって、複素環は未置換であるか または 低級アルキル、 ヒドロキシ、 ヒドロキシアルキル、 ハロ、 ニトロ、 オキソ(=O)、 アミノ、 N−保護されたアミノ、 アルコキシ、 チオアルコキシ、および ハロアルキル からなる群から独立して選択される1、2または3個の置換基で置換されている )、 からなる群から選択され、 R2は、 (a) (式中、 R12aは、水素、低級アルキル、または−C(O)OR13であり、但しR13は水 素またはカルボキシ−保護基であり、 R12bは、水素または低級アルキルであり、但しR12aおよびR12bは、両方とも 水素ではない)、 (b) −C(O)NH−CH(R14)−C(O)OR15であり、但しR14は、 低級アルキル、 シクロアルキル、 シクロアルキルアルキル、 アルコキシアルキル、 チオアルコキシアルキル、 ヒドロキシアルキル、 アミノアルキル、 カルボキシアルキル、 アルコキシカルボニルアルキル、 アリールアルキル、または アルキルスルホニルアルキル から選択され、 R15は水素またはカルボキシ−保護基である)、 (c)(d) −C(O)NH−CH(R14)−C(O)NHSO2R16であり、但し、 R14は前記定義の通りであり、R16は、 低級アルキル、 ハロアルキル、 置換または未置換アリール、 置換または未置換複素環 から選択される)、 (e) −C(O)NH−CH(R14)−テトラゾリルであり、但し、テトラゾー ル環は未置換であるか、または低級アリールまたはハロアルキルで置換されてい る)、 (f) −C(O)NH−複素環、および (g) −C(O)NH−CH(R14)−C(O)NR17R18であり、但し、R17 およびR18は、水素および低級アルキルから独立して選択される) からなる群から選択され、 R3は、ピリジル、置換ピリジル、イミダゾリル、または置換イミダゾリルであ り、 R4は、 水素、 低級アルキル、 ハロアルキル、 ハロゲン、 アリール、 アリールアルキル、 複素環、および (複素環)アルキル からなる群から選択され、 L1は、存在しないか、または (a) −L4−N(R4)−L5−であり、但し、 L4は存在しないか、または C1〜C10−アルキレン、および C2〜C10−アルケニレン から選択され、但し、アルキレン基またはアルケニレン基は未置換であるか、ま たは 低級アルキル、 アルケニル、 ヒドロキシ、 アミノ、 Nが保護されたアミノ、 カルボキシ、 アルコキシカルボニル、 オキソ、 チオキソ、 イミノ、 =N−OH、 =N−O−低級アルキル、 =N−O−アリール、および =N−O−複素環(但し、複素環は、未置換であるか、ま たは 低級アルキル、 ハロ、 ニトロ、 ハロアルキル、 オキソ、 ヒドロキシ、 ヒドロキシアルキル、 アミノ、 Nが保護されたアミノ、 アルコキシ、および チオアルコキシ からなる群から独立して選択される1、2または3個の置換基で置換されている )、 R4は、水素または低級アルキルであり、 L5は、存在しないか、または −CH2−、 −CH2CH2−、および −CH(CH3)− から選択され、 (b) −L4−O−L5−であり、但し、L4およびL5は前記定義の通りであり、 (c) −L4−S(O)n−L5−であり、但し、nは0、1または2であり、L4 およびL5は前記定義の通りであり、 (d) −L4−L6−C(W)−N(R5)−L5−であり、但し、L4、L5および L6は前記定義の通りであり、 WはOまたはSであり、 L6は存在しないか、または −O−、 −S−および −N(R6)−(式中、R6は水素または低級アルキルであ る)から選択され、 (e) −L4−L6−S(O)m−N(R7)−L5−であり、但し、L4、L5およ びL6は前記定義の通りであり、 mは1または2であり、 R7は、水素または低級アルキルである、 (f) −L4−N(R7)−C(W)−L7−L5−であり、但し、W、R7、L4、 およびL6は前記定義の通りであり、 L7は存在しないか、または−O−、および−S−から選 択され、 (g) −L4−N(R7)−S(O)p−L7−L5−であり、但し、pは1または 2であり、L4、R7、L5およびL7は前記定義の通りであり、 (h) 場合によっては1または2個のヒドロキシ基で置換されたC2〜C4−アル キレン、 (i) C2〜C4−アルケニレン、および (j) C2〜C4−アルキニレン からなる群から選択される)を有する化合物、またはその薬学上許容可能な塩。 2.R1がアリール−L2−であり、但し L2が存在しないか、または−CH2−および−O−から選択され、 アリールは、フェニルおよびナフチルから選択され、 アリール基は未置換であるか、または置換されている、 請求の範囲第1項に記載の化合物、または薬学上許容可能なその塩。 3.R1が置換または未置換フェニルである、請求の範囲第2項に記載の化合 物またはその薬学上許容可能な塩。 4.R2が、 (a) −C(O)NH−CH(R14)−C(O)OR15であり、但しR14は、 低級アルキル、 シクロアルキル、 シクロアルキルアルキル、 アルコキシアルキル、 チオアルコキシアルキル、 ヒドロキシアルキル、 アミノアルキル、 カルボキシアルキル、 アルコキシカルボニルアルキル、 アリールアルキル、または アルキルスルホニルアルキル から選択され、 R15は水素またはカルボキシ−保護基である、 (b) −C(O)NH−CH(R14)−C(O)NHSO2R16であり、但し、 R14は前記定義の通りであり、R16は、 低級アルキル、 ハロアルキル、 置換または未置換アリール、 置換または未置換複素環 から選択される、 からなる群から選択される、請求の範囲第3項に記載の化合物、またはその薬学 上許容可能な塩。 5.R2が、 (a) (b) (c)(d) および (e) からなる群から選択される、請求の範囲第3項に記載の化合物、またはその薬学 上許容可能な塩。 6.R3が、ピリジル、または 低級アルキル、 ハロゲン、 ハロアルキル、 ヒドロキシ、 アルコキシ、 アルコキシカルボニル、 アリール、および アリールアルキル から独立して選択される1または2個の置換基で置換されたピリジルである、請 求の範囲第5項の化合物、またはその薬学上許容可能な塩。 7.R3が、イミダゾリル、または低級アルキルで置換されたイミダゾリルで ある、請求の範囲第1項に記載の化合物、またはその薬学上許容可能な塩。 8.哺乳類に請求の範囲第1項に記載の化合物の治療上有効量を投与すること を含んでなる、治療を必要とする哺乳類におけるタンパク質イソプレニルトラン スフェラーゼの阻害法。 9.薬学担体および請求の範囲第1項に記載の化合物の治療上有効量を含んで なる、タンパク質イソプレニルトランスフェラーゼを阻害する組成物。 10.哺乳類における癌の阻害または治療法であって、哺乳類に請求の範囲第1 項に記載の化合物の治療上有効量を単独で、または他の化学療法剤と組み合わせ て投与することを含んでなる、方法。 11.請求の範囲第1項に記載の化合物を他の化学療法剤および薬学上許容可能 な担体と組み合わせて含んでなる、癌の治療のための組成物。 12.哺乳類におけるタンパク質ファメシルトランスフェラーゼ、タンパク質ゲ ラニルゲラニルトランスフェラーゼ、または両方による発癌性Rasタンパク質 の翻訳後修飾の方法であって、哺乳類に請求の範囲第1項に記載の化合物の治療 上有効量を投与することを特徴とする、方法。 13.哺乳類におけるタンパク質ファメシルトランスフェラーゼ、タンパク質ゲ ラニルゲラニルトランスフェラーゼ、または両方による発癌性Rasタンパク質 の翻訳後修飾のための組成物であって、請求の範囲第1項に記載の化合物を薬学 上許容可能な担体と組み合わせて含んでなる、組成物。 14.哺乳類における再狭窄を治療または予防する方法であって、哺乳類に請求 の範囲第1項に記載の化合物の治療上有効量を投与することを特徴とする、方法 。 15.請求の範囲第1項に記載の化合物を薬学上許容可能な担体と組み合わせて 含んでなる、哺乳類における再狭窄を治療または予防するための組成物。
