DESCRIZIONE
della domanda di brevetto per Invenzione Industriale dal titolo: "IMPIEGO DEL 2-CICLOPENTEN-1-ONE E SUOI DERIVATI COME INIBITORI. DEL FATTORE NF-kB"
CAMPO DELL'INVENZIONE
La presente invenzione riguarda l'impiego del 2-ciclopenten-ione e dei suoi derivati come inibitori del fattore NF-kB. In particolare l'invenzione si riferisce all'impiego del 2-ciclopenten-i-one e dei suoi conposti derivati contenenti il nucleo ciclopentenonico come inibitori del fattore NF-kB con attività anti-inf iammatoria, anti-prolif erativa, immunosoppressiva, citoprotettiva e antivirale.
TECNICA ANTERIORE NF-kB (Nuclear Factor - kB) è un fattore di trascrizione eucariotico della famiglia rel , che normalmente è localizzato nel citoplasma in un complesso inattivo, la cui forma predominante è un eterodimero composto delle subunità p50 e p65 legato a proteine inibitorie della famiglia ΙkΒ (D. Thanos e T. Maniatis, Celi 80:529-532, 1995).
NF-AB viene attivato in risposta a stimoli di vario tipo, tra cui citochine infiammatorie, radiazioni ultraviolette, infezioni batteriche e virali. La stimolazione causa il rilascio di NF-AB da IAB in seguito alla fosforilazione e conseguente degradazione della proteina ΐΑΒ-alfa (P.A. Baeuerle e T. Henkel, Annu. Rev. Immunol . 12: 141-179, 1994) . NF-AB, una volta attivato, transloca nel nucleo dove si lega al DNA in specifici siti "AB", e induce la trascrizione di una serie di geni codificanti per proteine coinvolte nella regolazione della risposta immunitaria e infiammatoria, tra cui vari tipi di interleuchine, il tumor necrosis factor alfa, la NO sintasi e la ciclo-ossigenasi 2 (S. Grimm e P.A. Baeuerle, Biochem. J. 290: 297-308, 1993). Di conseguenza NF-AB è considerato un mediatore precoce della risposta immunitaria e infiammatoria ed è stato coinvolto nel controllo della proliferazione cellulare e nella patogenesi di varie malattie nell'uomo, fra cui l'artrite reumatoide (H. Becker et al., Clin. Exp. Immunol. 99: 325, 1995), l'ischemia (A. Salminen et al Biochem. Biophys. Res. Commun., 212: 939, 1995), 1'arterosclerosi (A.S. Baldwin, Annals Rev. Immunol., 14: 649, 1996) e nella patogenesi della sindrome da immunodeficienza acquisita (AIDS), a causa dell'aumento della trascrizione del virus della immunodeficienza umana HIV-l in presenza di NF-AB attivato. L'aumento della trascrizione degli RNA del virus HIV-l da parte di NF-AB è dovuto alla presenza di siti "AB" nelle sequenze (LTR) (Long Terminal Repeats) del genoma del virus (M.J. Lenardo e D. Baltimore, Celi 58: 227-229, 1989). E' anche noto che le prostaglandine (PGs) appartengono alla classe degli eicosanoidi naturali, acidi grassi con 20 atomi di carbonio, sono sintetizzate da molti tipi di cellule eucariotiche in risposta a stimoli esterni e giocano un ruolo importante nelle risposte fisiologiche alla proliferazione e differenziazione delle cellule. Sin dalla loro scoperta esse hanno mostrato di fungere da ormoni microambientali' e mediatori del segnale intracellulare e di partecipare al controllo di una gran quantità di processi fisiologici e patologici, inclusa la proliferazione e differenziazione delle cellule, la risposta immunitaria, l'infiammazione, la citoprotezione e le patologie febbrili. In particolare le prostaglandine di tipo A e J, a struttura ciclopentenonica, sono potenti inibitori della replicazione di virus ("Stress Proteins: Induction and Function" Schlesinger MJ, Garaci E., Santoro M.G. ed.s, Springer-Verlag, Heidelberg-Berlin, 27-44, 1990). In particolare, è stato recentemente dimostrato che le prostaglandine ciclopentenoniche inibiscono la replicazione del virus Hiv-l bloccando la trascrizione degli RNA virali (C. Rozera et al., J. Clin. Invest. 97: 1795, 1996).
A loro volta, le proteine da stress, anche proteine da Shock Termico (Heat Shock Proteins - HSPs) {Proc. Nati. Acad. Sci. USA 86, 8407-8411, 1989) sono costituite da polipeptidi sintetizzati da cellule eucariotiche e procariotiche in risposta ad uno shock termico o ad altri tipi di stress ambientali. Le HSPs sono codificate da un sottogruppo di geni delle cellule, identificati come geni da stress.
Gli autori hanno recentemente dimostrato che la prostaglandina ciclopentenonica PGA inibisce l'attivazione del fattore NF-JcB in cellule umane attraverso l'inibizione della degradazione della proteina inibitoria IkB-alfa (A. Rossi, G. Elia e M.G. Santoro, Cold Spring Harbor, New York 1-5 maggio, 1996)
Gli autori hanno anche trovato che l'inibizione dell'attivazione di NF-kB è uno dei meccanismi molecolari utilizzati dalle prostaglandine ciclopentenoniche per esercitare un blocco selettivo e reversibile della trascrizione degli RNA del virus HIV- 1.
SOMMARIO DELL'INVENZIONE
E' stato ora sorprendentemente trovato che il 2-ciclopenten-lone, struttura che costituisce il nucleo centrale della PGA, risulta avere attività analoga ad essa, cioè è in grado di inibire l'attivazione del fattore NF-JcB, pur non contenendo la corrispondente funzione acida e le catene laterali alifatiche. Risulterebbe quindi che le catene laterali che sono presenti nella PGA, con i loro sostituenti e doppi legami, in particolare la funzione acida, che comporta la natura di acido grasso delle prostaglandine, possono essere soppresse senza modificare sostanzialmente l'attività specifica descritta.
E' stato inoltre trovato che l'inibizione di NF-kB da parte del gruppo ciclopentenonico è strutturalmente correlata all'attivazione del fattore di trascrizione HSF (Heat Shock Transcription Factor) che è responsabile della sintesi di HSPs (Heat Shock Proteins).
Visto che l'inibizione di NF-kB è associata all'attivazione del fattore HSF, si evince che le molecole contenenti il nucleo ciclopentenonico attivo nell'inibire NF-kB saranno induttori del fattore HSF e quindi saranno induttori di proteine da shock termico .
Costituisce pertanto oggetto della presente invenzione l'impiego di 2-ciclopenten-i-one e suoi composti derivati comprendenti il nucleo ciclopentenonico come inibitori di NF-kB, i sostituenti essendo scelti fra quelli che non modificano negativamente l'attività di inibizione.
Altro oggetto della presente invenzione è l'impiego di 2-ciclopenten-l-one e suoi composti derivati farmacologicamente accettabili come inibitori di NF-kB.
Ancora altro oggetto dell'invenzione è costituito dall'impiego del 2-ciclopenten-l-one e suoi derivati come inibitore di NF-kB con attività anti-infiammatoria, anti-prolif erativa, immunosoppressiva, citoprotettiva e antivirale.
Ulteriore oggetto dell'invenzione è costituito da composizioni farmaceutiche comprendenti il 2-ciclopenten-l-one e suoi derivati farmacologicamente accettabili per la realizzazione di medicamenti ad attività anti-infiammatoria, anti-proliferativa, immunosoppressiva, citoprotettiva e antivirale, in particolare attività antivirale nei confronti del virus HIV-l e virus la cui trascrizione è regolata da NF-kB.
Ancora altro oggetto dell'invenzione è costituito dall'impiego di derivati farmacologicamente accettabili del 2-ciclopenten-1-one come induttori del fattore HSF.
Ulteriori oggetti dell'invenzione risulteranno evidenti dalla descrizione dettagliata dell'invenzione.
BREVE DESCRIZIONE DELLE FIGURE

DESCRIZIONE DETTAGLIATA DELL'INVENZIONE
Il 2-ciclopenten-l-one è un prodotto noto, sintetizzabile ad esempio secondo il procedimento descritto in Beilstein (Daene, Eder, A. 539 [1939] 207, 211).
Secondo la presente invenzione il 2-ciclopenten-l-one, preferibilmente in concentrazione da 100 a 500 uM, è in grado di inibire l'attivazione del fattore NF-kB in cellule umane (Fig.
1A).
Prove di inibizione sono state in particolare condotte in cellule linfoidi di tipo T umane (linea Jurkat). inoltre l'inibizione del fattore NF-kB risulta essere associata all'attivazione del fattore HSF (Fig. 1B).
Risulta inoltre che l'inibizione di NF-kB si ottiene in maniera specifica con il 2-ciclopenten-i-one, mentre molecole simili, come il ciclopentanone o il ciclopentene non inibiscono NF-kB (Fig. 1C) e non attivano HSF (Fig. 1D).
Sulla base di questi risultati è possibile impiegare il 2-ciclopenten-i-one come principio attivo per realizzare farmaci, in particolare farmaci ad attività inibente l'attivazione di NF-kB e quindi: dato il ruolo di NF-kB come stimolatore della risposta infiammatoria e immunitaria, farmaci antinfiammatori e immunosoppressivi ; dato il ruolo di NF-kB nell'ischemìa e danno ossidativo, farmaci citoprotettivi; dato il ruolo di NF-kB nella proliferazione cellulare, farmaci antiproliferativi,· dato il ruolo di NF-kB nell'attivazione della trascrizione e di RNA virali, farmaci antivirali.
Ad illustrazione dell'invenzione vengono appresso riportati gli esempi seguenti, che tuttavia non devono essere considerati limitativi dello scopo e della portata della medesima.
I reagenti usati, compreso il 2 -ciclopenten- 1-one, il ciclopentene, il ciclopentanone e il 12-o-tetradecanoil-forbol-13-acetato (TPA), erano prodotti dalla Sigma Aldrich. <32>P era prodotto dalla AMERSHAM. Siero fetale bovino e mezzi di coltura cellulare erano prodotti dalla GIBCO.
ESEMPIO 1
L'effetto del trattamento con 2-ciclopenten-1 -one sull'inibizione del fattore NF-JcB da parte di TPA è stato valutato in cellule Jurkat con le modalità appresso descritte e illustrate in Fig. 1.
Effetto della dose
La preparazione delle cellule fu effettuata secondo le modalità descritte in C. Amici et al. (Cancer. Res. 55:4452, 1995).
Le cellule venivano trattate con varie concentrazioni di 2-ciclopenten-l-one per 1 ora e poi trattate con TPA (25 ng/ml). Dopo 3 ore gli estratti di cellule intere furono preparati e sottoposti a EMSA (Electrophoretic Mobility Shift Assay) come descritto per NF-JcB in U. Zabel et al. (J. Biol. Chem. 266:252, 1991) e per HSF in C. Amici et al. (Cancer. Res. 55:4452, 1995). In Fig. 1A sono indicate le posizioni del complesso NF-kB-DNA (NF-kB) e il legame non specifico (ns).
In Fig. 1B sono indicate le posizioni del complesso HSF-DNA (HSF), dell'attività costitutiva HSF-DNA (CHBA) e le interazioni non specifiche di proteine-DNA (ns). La linea "control" rappresenta cellule non stimolate con TPA come controllo di NF-kB non attivato.
E' evidente che il ciclopent enone è in grado di inibire l'attivazione di NF-JcB da parte di TPA già a concentrazioni di 125 uM. Alla concentrazione di 500 uM la banda NF-kB è completamente assente (Fig. 1A). In corrispondenza dell'inibizione di NF-kΒ negli stessi campioni è evidente l'attivazione di HSF a partire dalla concentrazione di 125 uM {Fig. 1B).
Specificità dell'effetto inibente (Fig. 1C)
Le cellule venivano trattate con uguali concentrazioni (500 uM) di 2-ciclopenten-i-one (linea 3), ciclopentanone (linea 4) o ciclopentene (linea 5) per 1 ora e poi venivano stimolate con TPA (25 ng/ml). Dopo 3 ore gli estratti di cellule intere venivano preparati e sottoposti a EMSA per verificare l'attivazione di NF-kB (Fig. 1C) e l'attivazione di HSF (Fig. 1D) .
E' evidente che: (i) il TPA attiva NF-kB (linea 2); ( ii) il 2-ciclopenten-l-one inibisce l'attivazione di NF-kB (linea 3); (iii) il ciclopentanone (linea 4) e il ciclopentene (linea 5) non hanno effetto inibitorio su NF-JcB.
Nella Fig. 1D è inoltre evidente che negli stessi campioni il fattore HSF viene attivato dal 2-ciclopenten- 1-one in corrispondenza con l'inibizione di NF-kB.
Claims (7)
- RIVENDICAZIONI 1. Impiego del 2-ciclopente-l-one e suoi derivati comprendenti il nucleo ciclopentenonico come inibitori del fattore NF-kB, i sostituenti di detto nucleo essendo scelti fra quelli che non modificano negativamente l'attività di inibizione.
- 2. Impiego del 2-ciclopenten- 1 -one e suoi derivati farmacologicamente accettabili come inibitori del fattore NF-kB con attività anti-infiammatoria, anti-prolif erativa, immunosoppressiva, citoprotettiva e antivirale.
- 3. Impiego del 2-ciclopenten- 1 -one e suoi derivati farmacologicamente accettabili per la realizzazione di composizioni farmaceutiche ad attività anti-infiammatoria, antiproliferativa, immunosoppressiva, citoprotettiva e antivirale.
- 4. Impiego del 2-ciclopenten-l-one e suoi derivati farmacologicamente accettabili come induttori del fattore HSF.
- 5. Composizioni farmaceutiche comprendenti il 2-ciclopenten-lone e suoi derivati farmacologicamente accettabili per la realizzazione di medicamenti ad attività anti-infiammatoria, anti-proliferativa, immunosoppressiva, citoprotettiva e antivirale.
- 6. Composizioni farmaceutiche comprendenti il 2-ciclopenten-lone e suoi derivati farmacologicamente accettabili per la realizzazione di medicamenti ad attività antivirale nei confronti del virus Hiv-i e virus la cui trascrizione è re dal fattore NF-kB .
- 7. Composizioni farmaceutiche comprendenti il 2-ciclopenten-lone o suoi derivati farmacologicamente accettabili come induttori del fattore HSF.
