Claims (19)
1. Postupak presavijanja čimbenika stimuliranja kolonija granulocita (G-CSF) iz inkluzijskih tijela, naznačen time što se sastoji u:
a) solubiliziranju G-CSF u prisutnosti sredstva za solubiliziranje;
b) provođenju oksidiranja i koraka prvog presavijanja, koji se sastoji u inkubiranju solubiliziranog G-CSF u prisutnosti oksidacijskog sredstva i sredstva za solubiliziranje;
c) uklanjanju sredstva za solubiliziranje adsorpcijom na ionskoizmjenjivačku smolu i/ili ionskoizmjenjivačkom kromatografiju, uz moguće provođenje kiselog taloženja; i
d) provođenju koraka drugog presavijanja, koji se sastoji u razrjeđivanju niskovodljivim puferom, ili poželjnije vodom, uz inkubiranje G-CSF u koraku (c),
gdje se korak drugog presavijanja provodi u niskovodljivom puferu, s vodljivošću od u najmanju ruku ispod 2 mS/cm i u trajanju od više od 12 sati.
2. Postupak u skladu s patentnim zahtjevom 1, naznačen time što su inkluzijska tijela od mikroorganizma, po mogućnosti od bakterije E. coli.
3. Postupak u skladu s patentnim zahtjevom 1 ili 2, naznačen time što je G-CSF rekombinantni goveđi ili ljudski metionil-G-CSF.
4. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što je sredstvo za solubiliziranje N-lauroilsarkozin.
5. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što je oksidacijsko sredstvo CuSO4.
6. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što se solubiliziranje G-CSF provodi kod vrijednosti pH iznad pH 7.
7. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što je sredstvo za solubiliziranje N-lauroilsarkozin, u koncentraciji od 0,5% to 1,5%.
8. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što se oksidiranje i korak prvog presavijanja provodi u trajajnu od najmanje dva sata.
9. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što se oksidiranje i korak prvog presavijanja provodi pod strujom zraka i bez hlađenja.
10. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što se oksidiranje i korak prvog presavijanja provodi kod vrijednosti pH od 7-9 i/ili na temperaturi od 20-28 °C i/ili u trajanju od 15-25 sati.
11. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što uklanjanje sredstva za solubiliziranje u koraku (c) se sastoji u: AEX i CEX, moguće tim redoslijedom.
12. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što uklanjanje sredstva za solubiliziranje u koraku (c) se sastoji u:
a) vezanju na anionskoizmjenjivački smolni materijal miješanjem otopine G-CSF sa suspendiranim smolnim materijalom i uklanjanje smolnog materijala filtriranjem, i/ili
b) ionskoizmjenjivačkoj kromatografiji u uvjetima gdje se sredstvo za solubiliziranje veže na smolu, a G-CSF zaostaje u protoku i/ili,
c) ionskoizmjenjivačkoj kromatografiji u uvjetima gdje se G-CSF veže na smolu, a sredstvo za solubiliziranje zaostaje u protoku.
13. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što se sredstvo za solubiliziranje i druge nečistoće uklanja uzastopnom primjenom sljedećih koraka:
(i) AEX,
(ii) kiselog taloženja,
(iii) AEX, i
(iv) CEX.
14. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što se sredstvo za solubiliziranje i druge nečistoće uklanja uzastopnom primjenom sljedećih koraka:
a) vezanja sredstva za solubiliziranje na anionskoizmjenjivački smolni materijal miješanjem otopine G-CSF sa suspendiranim smolnim materijalom i uklanjanja smolnog materijala filtriranjem;
b) taloženja nečistoća snižavanjem pH ispod pH 5 i uklanjanjem taloga filtriranjem;
c) anionskoizmjenjivačku kromatografiju, koju se provodi u uvjetima u kojima se preostalo sredstvo za solubiliziranje veže na smolu, a G-CSF zaostaje u protoku;
d) kationskoizmjenjivačku kromatografiju, koju se provodi u uvjetima u kojima se G-CSF veže na smolu, a preostalo sredstvo za solubiliziranje zaostaje u protoku; i
e) eluiranja vezanog G-CSF s kationskoizmjenjivačke smole stupnjevitim ili gradijentnim eluiranjem upotrebom pufera za eluiranje uz povećani pH ili koncentraciju soli.
15. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što se korak drugog presavijanja provodi u uvjetima hlađenja.
16. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što se korak drugog presavijanja provodi na temperaturi od 2-8 °C i/ili u trajajnu od najmanje 24 sata.
17. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što se korak drugog presavijanja provodi kod vrijednosti pH iznad pH 7.
18. Postupak u skladu s bilo kojim od prethodnih patentnih zahtjeva, naznačen time što postupak dodatno uključuje korak poliranja, koji se sastoji u jednoj ili više ionskoizmjenjivačkih kromatografija, gdje jedna ili više ionskoizmjenjivačkih kromatografija u koraku poliranja može uključivati anionskoizmjenjivačku kromatografiju, za kojom slijedi kationskoizmjenjivačka kromatografija.
19. Postupak u skladu s patentnim zahtjevom 18, gdje se jedna ili više ionskoizmjenjivačkih kromatografija sastoje u sljedećim koracima:
a) anionskoizmjenjivačke kromatografije, koju se provodi u uvjetima gdje se G-CSF veže na smolu;
b) eluiranja vezanog G-CSF stupnjevitim ili gradijentnim eluiranjem upotrebom pufera za eluiranje uz smanjenu pH ili povećanu koncentraciju soli;
c) kationskoizmjenjivačku kromatografiju, koju se provodi u uvjetima gdje se G-CSF veže na smolu;
d) eluiranja vezanog G-CSF stupnjevitim ili gradijentnim eluiranjem upotrebom pufera za eluiranje uz povećani pH ili koncentraciju soli;
naznačen time što okosnični polimeri smola za anionsku i kationsku izmjenu oba sadrže metakrilatne derivate.
