科学文献は、そのような封入体を細菌から単離し、精製し、次いで組換えタンパク質を可溶化し、その天然の状態にリフォールディングすることができる多くの方法を開示する(用語「フォールディング」および「復元」は本明細書では同義に用いる)。
種々の戦略が組換えタンパク質を可溶化するために用いられた。イオン性および非イオン性界面活性剤(例えばドデシル硫酸ナトリウムもしくはN−ラウリルサルコシンン(サルコシル))に加え、カオトロピック試薬(例えば塩酸グアニジンまたは尿素)を用いて目的とするタンパク質を可溶化した。しばしば、可溶化は、還元剤、例えばジチオスレイトール(DTT)、ジチオエリスロール(DTE)、または2−メルカプトエタノール(ME)の存在下、アルカリ条件下(pH 8〜12.5)で行う(Marston 1986、Rudolph 1990、Rudolph 1996、Dietrich 2003)。典型的には、可溶化タンパク質は、最初に完全に還元および不活化され、次いでリフォールディングを受け、次いでクロマトグラフィ精製される。
本発明は、封入体から抽出した組換え果粒球コロニー刺激因子 (G−CSF)を生物活性分子にリフォールディングするための新規な方法に関する。該方法は、G−CSFを可溶化剤で可溶化し、酸化し、次いでそのような可溶化剤の存在下でG−CSFを部分的にリフォールディングし、該可溶化剤を効率的に除去し、可溶化剤の非存在下でリフォールディングを完結することを含む。部分的リフォールドG−CSFから可溶化剤を分離するための種々の方法が開示されている。

本発明は、果粒球コロニー刺激因子 (G−CSF)の新規リフォールディング法を提供する。特に、本発明は、精製G−CSFの産業的製造を可能にする、封入体から活性G−CSFを高収率で得るための新規方法を提供する。
本発明は従来技術の欠点を克服する。
a)可溶化剤を用いて封入体からG−CSFを可溶化し(所望により封入体を抽出および/または単離および/または洗浄する前に)、
b)酸化剤および可溶化剤存在下でインキュベーションすることにより可溶化G−CSFを酸化および部分的にリフォールディングし、
c)可溶化剤(および他の不純物)を除去し、そして
d)部分的にリフォールドしたG−CSFを希釈およびインキュベーションすることによりリフォールディングを完結する。
すなわち、該第2リフォールディング工程は可溶化剤非存在下で行う。
(a) 可溶化剤存在下でのG−CSFの可溶化、
(b)酸化剤および可溶化剤存在下で可溶化G−CSFをインキュベーションすることを含む酸化および第1リフォールディング工程、
(c)イオン交換樹脂吸着および/または酸沈殿および/またはイオン交換クロマトグラフィによる可溶化の除去、および
(d)可溶化剤非存在下で工程(c)のG−CSFを希釈およびインキュベーションすることを含む第2リフォールディング工程。
該封入体は微生物から得ることができる。該微生物は、例えば組換え的にG−CSFを産生することができる。したがって、該本発明は、所望によりさらに微生物細胞から該封入体を単離する工程を含みうる。
本発明は、封入体からG−CSFをリフォールディングする方法を提供する。過去に種々の異なる発現系が大量のG−CSFを製造するために試験されてきた。試験したすべての細菌株がG−CSFタンパク質を封入体 (IB)の形で発現する。封入体 (IB)はG−CSFを大量に、アンフォールドされた不活性形で含む。単離され、好ましくは洗浄された封入体の分画を本明細書に記載の方法の出発物質として用いることができる。そのような封入体調製物は、適切な発現系、発酵条件、回収および溶解手順、およびiBの単離および洗浄に適した方法により得ることができる。そのような方法は、従来技術に開示されている(例えば、Rudolph 1990、Rudolph 1996、Heidari 2001、Khalilzadeh 2008、Rao 2008、Vanz 2008、US5849883、EP0219874、EP1630173、またはWO2004001056参照)。宿主細胞から封入体を抽出する方法は、一般的に、細胞の溶解および破壊、次いで遠心分離を含む。該封入体は、分離器で細胞を回収し(例えば遠心分離(例えば11000g)により)、細胞を高圧ホモゲナイザー(例えば約1000bar)で機械的に破壊し、次いで分離器で細胞デブリから封入体を分離する(例えば遠心分離(例えば11000g)により)により得ることができる。大部分の古典的封入体を含むペレットを、通常、界面活性剤で洗浄する。G−CSFを可溶化する前に、該封入体を凍結保存することができる。分離器で細胞デブリから分離し、−80℃の還元緩衝液中で保存したIBは最高8カ月安定であることがわかった。

すなわち、本明細書において「G−CSF」に対する言及は、ヒトG−CSFの種相同分子種および変異体、すなわち機能的変異体に対する言及を含む。
IB分画のG−CSFは、可溶化剤の存在下で可溶化する。あらゆる適切な可溶化剤 (すなわち、本明細書に記載のG−CSFの可溶性をもたらすあらゆる物質)を用いることができる。そのような可溶化剤は、例えば(限定されるものではないが)変性剤またはカオトロピック剤の群、例えば (限定されるものではないが) GuHClまたは尿素、もしくは界面活性剤(detergent、tenside、またはsurfactant)の群、例えば、(限定されるものではないが) N−ラウロイルサルコシン(サルコシル)、ラウリン酸、ドデシル硫酸ナトリウム(SDS)、またはN−セチルトリメチルアンモニウムクロリドなどから選ぶことができる。
7〜10、または7〜9、または7.5〜8.5、または7.8〜8.2の範囲内のpHで行う。好ましい態様において、pHは約8または8である。ある態様において、可溶化は、室温、すなわち20〜25℃で行う。可溶化のために特定pH範囲で用いるのに適した緩衝液は当該分野で知られている。例えばTris−HClを用いることができる。好ましくは、可溶化は攪拌下で行う。
第1リフォールディング (酸化的フォールディング)
酸化的リフォールディングは連続気流下で行うことができる。

イオン交換樹脂吸着
可溶化剤を除去する第1工程として、イオン交換樹脂吸着を行うことができる。適切な方法が当該分野で知られている。上記のごとく、可溶化剤(例えばサルコシル)を除去するための方法には、Dowex 陰イオン交換樹脂 (Dow Chemicals、USA)、好ましくはDowex 1X4へのバッチ吸着がある(WO8910932、Lu1992、Heidari 2001)。結合した界面活性剤を捕捉する樹脂はろ過して除去する。Dowex樹脂と非常に似た製品に、同様に用いることができるBioRadのAG樹脂 (例えばAG−1X8)がある。
好ましい態様において、該イオン交換樹脂吸着工程は、該可溶化剤を90%以上、95%、より好ましくは96%以上、より好ましくは97%以上、より好ましくは98%以上、より好ましくは99%以上、より好ましくは99.2%以上、より好ましくは99.3%以上、より好ましくは99.4%以上、より好ましくは99.5%以上、より好ましくは99.6%以上、より好ましくは99.7%以上、より好ましくは99.8%以上、より好ましくは99.9%以上除去する。好ましい態様において、該可溶化剤はサルコシルである。ある態様において、該イオン交換樹脂吸着工程は、AG 1−X8バッチ吸着抗知恵によりサルコシルを95%以上、より好ましくは少なくとも99%除去する。ある態様において、捕捉されたサルコシルを有するAG 1−X8樹脂は、該溶液からろ過により除去される。好ましくは、該ろ過工程は、100μmのステンレススチールメッシュを用いる。より好ましくは、該ろ過工程は、100μmナイロンバッグフィルターメッシュを用いる。

イオン交換樹脂吸着 (酸沈殿を用いるかまたは用いない)を用いても可溶化剤が完全に除去されなかった場合は、1またはそれ以上のイオン交換クロマトグラフィ工程を行って可溶化剤を完全に除去することができる。該イオン交換工程は、AEXおよび/またはCEXをあらゆる順番で含みうる。あらゆる他の適切なイオン交換技術を用いることができる。

(表の簡単な説明)
G−CSF(フィルグラスチム)を組換えE. coli C2523 T7 pol pRG/GCSFaクローン (G−CSFを含む発現ベクターで形質転換したE. coli)を用いて製造した。無菌条件下で製造したシード培養液に、液体窒素で保存し、解凍した機能細胞バンクバイアルから得た0.10〜0.15cm3細胞サスペンジョンを接種した。接種したシード培養フラスコを、旋回振盪インキュベーター中で37℃、185rpmで24〜28時間インキュベーションした。6振盪フラスコ培養の600nm (OD)での光学密度の平均値が0.9〜1.1に達したら、フラスコの内容をシリコンチューブを装着した無菌5dm3ガラスフラスコに回収した。回収した3dm3容量のシード培養を、WM323U/Rポンプで、無菌の添加製造培地(GBA、炭素源としてグリセロールを含む合成培地)を最大75dm3充填した100dm3発酵槽に移した。37℃の水中(submerged)培養中、厳密な好気条件下で培養を行った。炭素源が培地中から枯渇したら、グリセロールフィーディング溶液を適切な速度で該培養に加えた。溶解酸素圧を全培養期間中30%以上のレベルに維持した。培養のOD値が30に達したら、温度を32℃に下げ、0.33mM IPTGを加えてG−CSFの発現を誘導した。該細菌をG−CSFを産生させるためにODが80〜95になるまで5時間さらに培養した。
攪拌、通気、および炭素源の供給を止め、培養を15℃以下に冷却し、細菌を11000gで分離して回収した。細胞をローターに沈殿させ、次いで水で洗い流した(排出)。細菌細胞濃縮物を回収し、水でその半分量に希釈し戻し、次いで0.5M NaH2PO4を最終濃度10mMとなるように加えた。湿細菌細胞(バイオマス)の総量は約10〜11.5kgであった。
分離し洗浄した細菌を、加圧下(100MPa)でホモゲナイザーを3回通して破壊した。封入体を、11000gで分離器中に沈殿させて細胞デブリから分離した。沈殿した封入体を、5mM DTT、10mM NaH2PO4、5mM EDTA、および2%Tween20(pH7.2)を含む洗浄用緩衝液中に排出させた。濃縮IBサスペンジョンを同じ緩衝液で2倍に希釈し、再度沈殿させた。この洗浄手順を、10mM NaH2PO4緩衝液を用いて2回反復し、該第2手順の終了時は希釈しなかった。IBの最終沈殿物を−80℃で凍結保存した。
凍結封入体 (湿質量650g)を解凍し、32.5dm3の総容量中40mM Tris−HClおよび1%(w/v) N−ラウロイルサルコシン(サルコシル)を含む可溶化剤(pH 8)で可溶化した。該サスペンジョンを連続攪拌下、室温でインキュベーションした。
可溶化IB懸濁液を水で2倍に希釈し、総容量65dm3中の最終濃度を0.5%サルコシルおよび20mM Tris−HClとした。CuSO4を最終濃度40μMで加えた。G−CSFを、室温で少なくとも20時間、連続的に攪拌および頭部スペースに気流を流して酸化し、部分的にリフォールドさせた。EDTAを最終濃度1mMで加えて酸化を止めた。
サルコシルをバッチモードで陰イオン交換 (AEX)樹脂に吸着させた。20g AG 1−X8樹脂 (BioRad、USA)/gサルコシルの量を該溶液に適用し、加えた。該サスペンジョンを2時間攪拌し、ほとんどのサルコシルを結合させた。該樹脂を、100μmポアサイズのナイロンバッグフィルターメッシュを通してろ過して除去した。濾液中の残存サルコシルを続く生成工程(実施例7および8)で生成物から完全に除去した。
pH 4.3〜4.5の酸性沈殿により、G−CSFは可溶性のままで、いくらかの不純物は容易に除去された。G−CSFのあらゆる潜在的な非特異的で望ましくない共沈殿を最終濃度1Mの尿素を加えて抑制した。尿素を6Mストック溶液で提供し、実施例6の濾液に1dm3/minの速度で徐々に加えた。次いで、pHを1/20容量の1M酢酸ナトリウム(pH 4.8)を加えて下げた。pHをさらに50%酢酸で滴定して 4.3〜4.5に下げた。少なくとも1時間沈殿させた。次に、沈殿した物質を深層ろ紙(Pall K700/KS50二重層)を通すろ過により除去した。
酢酸ナトリウム50mM/pH4.5緩衝液を以下を平衡化するために用いた:1) DEAE Macro−Prep (Bio−Rad、USA) AEX樹脂を充填した4dm3カラム、および2)Toyopearl SP−650C (Tosoh、Japan) CEX樹脂を充填した8dm3カラム。両カラムを直列配列でAeKTAプロセスクロマトグラフィシステム(GE Healthcare、Sweden)と直接接続した。0.2μm無菌フィルターを通して浄化後、実施例7の濾液を第1カラムにロードした。残存サルコシルはDEAE樹脂に結合し、G−CSFは非結合(非結合モード)のままであり、第1カラムのフロースルーに出現した。このフロースルーを該第2カラム(SP樹脂)に直接ロードし、G−CSFを結合させた(結合モード)。. A simple step elution with 20mM Tris−HCl/pH 8を用いる簡単な段階溶出により該樹脂からG−CSFを脱離させた。残存サルコシルの除去に加えて、この方法では酢酸ナトリウム/pH 4.5からTris−HCl/pH 8への緩衝液交換も達成された。
この段階で該タンパク質分画の約半分のフォールディングが完結したが、残りのタンパク質は不完全にフォードするか、ミスフォールドした。G−CSF溶液を、Toyopearl SP−650Cから20mM Tris−HCl(pH 8)で溶出し、0.2μm無菌フィルターを通してステンレススチール容器に入れた。ろ過した溶液を水で2倍に希釈した。タンパク質フォールディングのための第2インキュベーション(第2リフォールディング)を、2〜8℃の冷却下で32〜42時間、pH8の低電導度環境(<1mS/cm)で行った。
カラムにDEAE Macro−Prep (Bio−Rad、USA)を充填し、10mM Tris−HCl/pH8で平衡化した。第2リフォールディングから得た溶液(実施例9)をDEAE カラムにロードした。正しくフォールドしたG−CSFを、10mM Tris−HCl/pH 8中の0mM〜200mMの増加する直線NaCl勾配により溶出させた。溶出したG−CSFをプール子、水で2倍希釈した。pHを50%酢酸で滴定して4.5に調節した。
最終ポリッシング工程では、正しくフォールドしたタンパク質からなるAEXクロマトグラフィから回収したG−CSFプール(実施例10)をToyopearl CM−650S樹脂を充填したCEX カラムに直接流した。該カラムを20mM酢酸ナトリウム(pH5.3)で平衡化した。結合G−CSFを24カラム容量以内で20mM〜400mM酢酸ナトリウム(pH 5.3)の増加する直線塩勾配により溶出した。精製G−CSFを含む分画を回収し、処方用にプールした。
CEXカラムから溶出した精製G−CSF(実施例11)を0.2μm無菌フィルターでろ過し、次いで処方用緩衝液(10mM酢酸ナトリウム、pH4.5%ソルビトール、および0.006%ポリソルベート80)で平衡化したSephadex G−25微粒子樹脂を充填した14dm3カラムを通した。同じ緩衝液をランニング緩衝液として用い、G−CSFを処方用緩衝液のボイド容積に溶出した。全バッチ(35〜48g G−CSF)について、Sephadex G−25を用いる3回連続処方操作(それぞれろ過CEX溶出液の3分の1)を行った。処方化G−CSFを処方用緩衝液で希釈して0.9〜1.0mg/mlの濃度に調整し、最終的に0.2μm無菌ろ過カプセルを通した。無菌溶液としての処方化G−CSFは非常に安定であり、何ヶ月間(数年ではないかもしれないが)にもわたり2〜8°で保存することができた。
よく知られた標準分析方法を、欧州薬局方(European Pharmacopoeia (Ph. Eur.):具体的分析方法を説明するフィルグラスチムのための文献を含む(European Directorate for the Quality of Medicines & Health Care (EDQM) (2010):Filgrastim concentrated solution. Claims (21)
- 以下の工程を含む封入体からの果粒球コロニー刺激因子 (G-CSF)のリフォールディング方法:
a) 可溶化剤の存在下で G-CSFを可溶化し、
b)酸化剤および可溶化剤の存在下で可溶化G-CSFをインキュベーションすることを含む酸化および第1リフォールディング工程を実施し、
c)イオン交換樹脂吸着および/またはイオン交換クロマトグラフィにより可溶化剤を除去し、
d)工程(c)のG-CSFを2mS/cm以下の低電導性緩衝液で希釈しインキュベーションすることを含む第2リフォールディング工程を実施する。 - 該封入体が、微生物由来である請求項1記載の方法。
- 該微生物がE. coliである請求項2記載の方法。
- G-CSFが組換えウシまたはヒトメチオニル-G-CSFである請求項1〜3のいずれかに記載の方法。
- 可溶化剤がN-ラウロイルサルコシンである請求項1〜4のいずれかに記載の方法。
- 酸化剤がCuSO4である請求項1〜5のいずれかに記載の方法。
- G-CSFの可溶化がpH7より大きいpH値で行われる請求項1〜6のいずれかに記載の方法。
- 可溶化剤が0.5%〜1.5%の濃度のN-ラウロイルサルコシンである請求項1〜7のいずれかに記載の方法。
- 該酸化および第1リフォールディング工程が少なくとも2時間実施される請求項1〜8のいずれかに記載の方法。
- 該酸化および第1リフォールディング工程が気流下で冷却せずに実施される請求項1〜9のいずれかに記載の方法。
- 該酸化および第1リフォールディング工程が、7〜9のpH値および/または20〜28℃の温度で、および/または15〜25時間実施される請求項1〜10のいずれかに記載の方法。
- 工程(c)の該可溶化剤の除去が、AEXおよびCEXを含む請求項1〜11のいずれかに記載の方法。
- 工程(c)の該可溶化剤の除去が以下の工程を含む請求項1〜12のいずれかに記載の方法:
a)G-CSF溶液と懸濁樹脂物質を混合してイオン交換樹脂物質に結合させ、次いでろ過して該樹脂物質を除去し、および/または
b)可溶化剤が該樹脂と結合し、G-CSFがフロースルー中に止まる条件下でのイオン交換クロマトグラフィ、および/または
c)G-CSFが該樹脂と結合し、可溶化剤がフロースルー中に止まる条件下でのイオン交換クロマトグラフィ。 - 可溶化剤および他の不純物を下記工程を連続的に適用することにより除去する請求項1〜13のいずれかに記載の方法:
(i)AEX、
(ii)酸沈殿、
(iii)AEX、および
(iv)CEX。 - 該可溶化剤および他の不純物を下記工程を連続的に適用することにより除去する請求項1〜14のいずれかに記載の方法:
a)G-CSF溶液と懸濁樹脂物質を混合して該可溶化剤をイオン交換樹脂物質と結合させ、次いでろ過して該樹脂物質を除去し、
b)pHをpH5以下に下げて不純物を沈殿させ、次いでろ過して該沈殿物を除去し、
c)残存可溶化剤が該樹脂と結合し、G-CSFがフロースルー中に止まる条件下で行う陰イオン交換クロマトグラフィ、および/または
d)G-CSFが該樹脂と結合し、残存可溶化剤がフロースルー中に止まる条件下で行う陽イオン交換クロマトグラフィ、
e)増加したpHまたは塩濃度の溶出緩衝液を用いる工程または勾配溶出により陽イオン交換樹脂から結合G-CSFを溶出する。 - 該第2リフォールディング工程を2mS/cm以下の低電導性緩衝液中および/または冷却条件下で、および/または12時間以上実施する請求項1〜15のいずれかに記載の方法。
- 該第2リフォールディング工程を2.0 mS/cm以下の電導度および/または2〜8℃の温度で、および/または少なくとも24時間実施する請求項1〜16のいずれかに記載の方法。
- 該第2リフォールディング工程をpH7以上のpH値で実施する請求項1〜17のいずれかに記載の方法。
- さらに、1またはそれ以上のイオン交換クロマトグラフィを含むポリッシング工程を含む請求項1〜18のいずれかに記載の方法。
- 該ポリッシング工程中の1またはそれ以上のイオン交換クロマトグラフィが陰イオン交換クロマトグラフィ、次いで陽イオン交換クロマトグラフィを含む請求項19記載の方法。
- 1またはそれ以上のイオン交換クロマトグラフィが以下の工程を含む請求項19記載の方法:
a) G-CSFと該樹脂が結合する条件下で行う陰イオン交換クロマトグラフィ、
b)低下したpHまたは増加した塩濃度の溶出緩衝液を用いる工程または勾配溶出による結合G-CSFの溶出、
c) G-CSFが該樹脂と結合する条件下で行う陽イオン交換クロマトグラフィ、
d)増加したpHまたは塩濃度の溶出緩衝液を用いる工程または勾配溶出による結合G-CSFの溶出、
ここで、陰イオンおよび陽イオン交換樹脂の主鎖ポリマーが共にメタクリレート誘導体を含むことを特徴とする。
