Claims (12)
1. Postupak detektiranja pojave HPV-om prouzročenih prekancerogenih lezija grlića materice visokog stupnja i/ili HPV-om prouzročenih invazivnih karcinoma grlića materice koji obuhvaćaju oglede cervikovaginalnog uzorka lavaže na prisutnost promjene u metilaciji proteina povezanog sa starenjem (MAL) T limfocita i hsa-miR124, pri čemu navedena promjena označava prisutnost HPV-om uzrokovanih prekursorskih lezija s invazivnim potencijalom i/ili HPV-om prouzročenim invazivnih karcinoma.
2. Postupak sukladno patentnom zahtjevu 1, pri tome je navedeni HPV-om prouzročeni karcionom HPV-om prouzročen invazivni karcinom visokog rizika.
3. Postupak iz bilo kojeg od prethodnih patentnih zahtjeva pri čemu je navedena promjena povećanje metilacije u usporedbi s normalnim kontrolnim uzorkom.
4. Postupak sukladno bilo kojem od prethodnih patentnih zahtjeva, pri čemu je nukleinska kiselina DNK.
5. Postupak sukladno patentnom zahtjevu 4, pri čemu se ovaj ogled izvodi s restriktivnom endonukleazom, preporučljivo restriktivnom endonukleazom osjetljivom na metilaciju.
6. Postupak sukladno patentnom zahtjevu 4, pri čemu je reagens funkcijska sonda nukleinske kiseline ili početnica koja se veže na nukleinsku kiselinu.
7. Postupak sukladno patentnom zahtjevu 6, pri čemu sukladno funkcijska sonda nukleinske kiseline ili početnica nukleinske kiseline ima detektibilnu oznaku.
8. Postupak sukladno patentnom zahtjevu 6 ili 7, pri čemu ta funkcijska sonda nukleinske kiseline ima sekvencu nukleotida odabranu iz skupine koja sadrži sekvence prikazane u Tablici 1.
9. Upotreba molekularnog dijagnostičkog markera podešenih za detektiranje HPV-om prouzročenih pretkancerogenih lezija visokog rizika ili HPV-om prouzročenih invazivnog karcinoma u uzorku cerviko-vaginalne lavaže, pri čemu navedeni komplet markera označava promjenu u metilaciji MAL gena ili promotoru i promjeni za metilaciju hsa-miR124 sekvence.
10. Upotreba kompleta dijelova u postupku detektiranja HPV-om prouzročene prekancerogene lezije visokog rizika ili HPV-om prouzročenog invazivnog karcinoma u testiranim stanicama iz uzorka cervikovaginalne lavaže ispitanika, navedeni komplet sadrži na metilaciju osjetljivu restriktivnu endonukleazu i
- sredstva za detekiranje metilacije MAL gena ili promotora pri tome navedena sredstva sadrže funkcijske sonde, početnice specifične za MAL ili specifične za MAL sekvencu nukleotida sa Slike 1, i
- sredstva za detektiranje hsa-miR124 metilacije, pri tome navedena sredstva sadrže funkcijske sonde, primere specifične za hsa-miR124.
11. Upotreba sukaldno patentnom zahtjevu 10, pri tome navedeni komplet dijelova još sadrži sredstva za detektiranje HPV infekcije, pri tome navedena sredstva sadrže funkcijske sonde i početnice specifične za HPV.
12. Upotreba kompleta dijelova kako je definirano u patentnim zahtjevima 10 ili 11, još sadrži sredstva za uzimanje cervikovaginalne lavaže.
