Abstract

The invention comprises sulphuric acid esters of dextranic acid and salts thereof. The dextranic acid sulphate may be made by reacting dextranic acid with chlorosulphonic acid or fuming sulphuric acid. In one method dextranic acid is reacted in pyridine with chlorosulphonic acid at temperatures of from 50 DEG -100 DEG C. Alternatively, the dextranic acid is first dissolved p in formamide and this solution is then added to the chlorosulphonic acid in pyridine solution. Also, thechlorosulphonic acid may be added at a temperature of -15 DEG -5 DEG C. to the dextranic acid in finely divided form cooled to the same temperature. Upon completion of esterification, the mixture is poured into cold water, then into alcohol, neutralized with an alkali metal or alkaline earth metal hydroxide to precipitate the salt which is recovered and dissolved in water, the solution is dialyzed and concentrated and the salt is precipitated with methyl or ethyl alcohol. The free acid of the ester is obtained by acidification of the salt with a mineral acid, followed by dialysis and concentration of the solution. Dextranic acid, used as starting material, is made dissolving native dextran in water together with the dextransplitting enzyme from Aspergillus Wentii and the solution digested at pH 4,3 at 70 DEG F. for 5 1/2 hours. Isopropanol is added and the supernatant liquid is separated from the precipitate so formed and is treated with sodium hydroxide. The precipitate now formed is dissolved in water, acidified, treated with isopropanol and finally with sodium hydroxide to precipitate dextranic acid.ALSO:A blood anti-coagulant composition consists essentially of an aqueous solution of the sodium salt of a dextranic acid sulphuric acid ester adjusted to pH 7.0.
