Abstract
A glucose derivative with the aldehyde group protected is oxidized to the corresponding glucuronic acid derivative by the action of a permanganate salt in aqueous alkaline solution. The permanganate is used in the theoretical amount to oxidize the primary alcohol group to carboxyl with production of manganese dioxide. A temperature of 10-25 DEG C. and a pH of 11-14 are suitable. The permanganate solution is preferably added incrementally or continuously to an aqueous solution of the glucose derivative, the latter being in a concentration of about 5-15 per cent. The manganese dioxide formed is filtered off and the glucuronic acid derivative hydrolysed (if desired) to free glucuronic acid. The examples shown: (1) oxidation of methyl a -D-glucoside with potassium permanganate to give the potassium salt of methyl glucuronoxide; and (2) similar oxidation of 1 : 2 - isopropylidene - glucose to 1 : 2 - isopropylidene-glucuronic acid. Ethyl glucoside and sodium permanganate are also specified as reactants.
