Abstract

1289607 2-Hydroxypropylamine derivatives PFIZER Ltd 14 May 1971 [14 May 1970] 23381/70 Heading C2C The invention comprises novel 2-hydroxypropylamine derivatives of the formula wherein R is acylamino-alkyl, R<SP>7</SP>, R<SP>8</SP>-N.CO- alkylene, or R<SP>7</SP>, R<SP>8</SP>-N.SO 2 -alkylene, in which R<SP>7</SP> and R<SP>8</SP> each are H, C 1-6 alkyl, phenyl or phenyl- C 1-6 alkyl ; R<SP>1</SP> is H, halogen or C 1-6 alkyl ; R<SP>2</SP> is H or C 1-6 alkyl; R<SP>3</SP> is H, C 1-6 ( alkyl or phenyl- C 1-6 alkyl ; R<SP>4</SP> is phenyl-C 1-6 alkyl or phenoxy- C 1-6 alkyl; and X is O or S; the esters of compounds in which R<SP>2</SP> is H; the aldehyde condensation products of compounds in which R<SP>2</SP> and R<SP>3</SP> are H with aldehydes of the formula R<SP>9</SP>CHO, where R<SP>9</SP> is H or C 1-6 alkyl; the N- oxides of compounds in which R<SP>3</SP> is not H; and the pharmaceutically acceptable acid addition salts thereof. The above compounds are prepared by reacting the appropriate 1,2-epoxy-3- phenoxy (or phenylthio) propanes or chlorohydrins or ethers thereof of the formula with amines of the formula HNR<SP>3</SP>R<SP>4</SP>. Alternatively the corresponding 3-phenoxy (or phenylthio) - 2 - hydroxypropylamines or ethers thereof may be reacted with aldehydes or ketones of the formula R<SP>10</SP>R<SP>11</SP>CO, where R<SP>10</SP> is phenyl- C 1-5 alkyl or phenoxy-C 1-5 alkyl and R<SP>11</SP> is H or C 1-4 alkyl, the alkyl groups of R<SP>10</SP> and R<SP>11</SP> together containing not more than 5 carbon atoms, and the resulting products reduced. Pharmaceutical compositions, suitable for oral or parenteral administration, contain the above novel compounds and pharmaceutically acceptable carriers. The compounds have anti-hypertensive activity.
