Abstract

1,260,893. Urea derivatives. PFIZER Ltd. 13 Aug., 1970 [15 Aug., 1969], No. 40830/69. Heading C2C. The invention comprises novel urea derivatives of the formula wherein R is a ureido group; R<SP>1</SP> is H, halogen or C 1-6 -alkyl; R<SP>2</SP> is H or C 1-6 alkyl; R<SP>3</SP> is H, C 1-6 alkyl or aryl-C 1-6 alkyl ; R<SP>4</SP> is C 1-6 alkyl, hydroxy- C 1-6 alkyl, C 1-6 alkoxy-C 1-6 alkyl, aryloxy-C 1-6 alkyl, cycloalkyl; C 2-6 alkenyl or aryl-C 1-6 alkyl; and X is O or S ; the esters of compounds having free hydroxy groups; the aldehyde condensation products of compounds in which R<SP>2</SP> and R<SP>3</SP> are each hydrogen; the N-oxides of compounds in which R<SP>3</SP> is not hydrogen; and pharmaceutically acceptable acid addition salts thereof. The novel compounds are prepared by reacting the appropriate epoxides or chlorohydrins or ethers thereof of the formulµ with amines of the formula or by reacting propanolamines of the formula with aldehydes or ketones of the formula and reducing the products to give compounds in which R<SP>3</SP> is H and R<SP>4</SP> is CHR<SP>9</SP>R<SP>10</SP>, or a cycloalkyl group, R<SP>9</SP> being C 1-5 alkyl, which may be substituted by OH 1 C 1-6 alkoxy, aryloxy or aryl, and R<SP>10</SP> being H or C 1-4 alkyl, the alkyl groups of R<SP>9</SP> and R<SP>10</SP> together not containing more than S carbon atoms, or R<SP>4</SP> and R<SP>10</SP> together with carbon atom to which they are attached forming a cycloalkylidene group. Pharmaceutical compositions, suitable for oral or parenteral administration, contain the above novel compounds and pharmaceutically acceptable carriers.
