1361863 Piperazine derivatives PFIZER Ltd 25 Sept 1972 [24 Sept 1971] 44615/71 Addition to 1325876 Heading C2C Novel piperazine derivatives of the formula wherein (1) R is thioureido, thiocarbamoyl, cyano, carboxy, N-aminocarbamoyl or C 1-6 alkoxycarbonylamino, R<SP>1</SP> is H, halogen or C 1-6 alkyl; R<SP>2</SP> is H or C 1-6 alkyl; R<SP>3</SP> is H, halogen, C 1-6 alkyl or C 1-6 alkoxy; n is 1 or 2; and X is O or S, and those wherein (2) R is ureido, thioureido, sulphamoylamino, carbamoyl, thiocarbamoyl, sulphamoyl, C 1-6 alkoxycarbonylamino, cyano, carboxy C 2-7 alkoxy carbonyl or N- aminocarbamoyl; R<SP>1</SP> is one or two methoxy groups; and R<SP>2</SP>, R<SP>3</SP>, n and X are as defined above; the esters of those compounds in which R<SP>2</SP> is H; and the N-oxides and pharmaceutically acceptable acid addition salts thereof are prepared by reacting the corresponding epoxides of the formula with the appropriate N-phenylpiperazines, and, if desired, converting the resulting compound, in which R<SP>2</SP> is H, into the corresponding ether or esters. Pharmaceutical compositions, suitable for oral or parenteral administration, contain the above novel compounds or esters, N-oxides or acid addition salts thereof and pharmaceutically acceptable carriers. The compounds possess hypotensive activity.
