Claims (10)
1. Probe för bestämning av bakterier i ett prov; eller lokalisering av DNA-fragment, vilka inne- 5 halier bakteriers 16S RNA kodande gener, ur elektrofo-retiskt eller kromatografiskt separerade DNA-fragment, kännetecknad därav, att proben bestär av antingen pentadekadeoxiribonukleotider, med sekvensen: (5')d - AAG GAG GTG ATC CAG (3'), 10 väri (5’) och (3') är polymerens 5'- och 3’- ändar och bokstäverna betecknar följande rester (nukleotider): A, adenyl-rest; C, cytidyl-rest; T, tymidyl-rest; och G, guanidyl-rest; eller pentadekadeoxiribonukleotider, med den komplementära sekvensen: 15 (5')d - CTG GAT CAC CTC CTT (3’)·
2. Probe för bestämning av bakterier i ett prov; eller lokalisering av DNA-fragment, vilka inne-häller bakteriers 16S RNA kodande gener, ur elektrofo-retiskt eller kromatografiskt separerade DNA-fragment, 20 kännetecknad därav, att proben bestär av sekvensen: (3') ACCTAGTGGAGGAAA (5'), eller motsvarande komplementära sekvens.
3. Probe för bestämning av bakterier i ett 25 prov; eller lokalisering av DNA-fragment, vilka inne- häller bakteriers 16S RNA kodande gener, ur elektrofo-retiskt eller kromatografiskt separerade DNA-fragment, kännetecknad därav, att proben bestär av en sekvens, vilken väljs ur följande grupp av sekvenser: 30 (5')d - AAGGAGGT (3‘); (5')d - AAAGGAGGT (3'); (5')d - AAGGAGGTG ( 3 ' ) ; (5’)d - AAGGAGGTGA (3'); (5' )d - AAGGAGGTGAT (3'); (5')d - AAGGAGGTGATC (3'); (5' )d - AAGGAGGTGATCC (3'); (5')d - AAGGAGGTGATCCA (3') eller motsvarande komplementära sekvens. .35
4. Probe för bestämning av eukaryotiska eller prokaryotiska organismers celler, bakterierna medräk-nade, ur ett prov; eller lokalisering av DNA-fragment, 20 891 85 vilka innehäller dessa cellers 16S serier RNA kodande gener, ur elektroforetiskt eller gaskromatografiskt separerade DNA-fragment, kännetecknad därav, att proben bestar av antingen pentadekadeoxiri-5 bonukleotider med sekvensen (5 * )d - ACC TTG TTA CGA CTT (3'), vari (5') och (3’) är polymerens 5’ och 3' ändar och bokstäverna betecknar följande rester (nukleoti-der) : A, adenyl-rest; C, cytidyl-rest; T, tymidyl-rest; och G guanidyl-rest; eller pentadekadeoxiribonukleoti-10 der med den komplementära sekvensen: (5')d - AAG TCG TAA CAA GGT (3').
5. Probe för bestämning av arkebakterier (ar-chaebacteria) ur ett prov; eller lokalisering av DNA-fragment, vilka innehäller arkebakteriers 16S RNA ko- 15 dande gener, ur elektroforetiskt eller gaskromatogra-fiskt separerade DNA-fragment, kännetecknad därav, att proben bestär av en sekvens med ätminstone 8 eftervarandra varande nukleotider, vilka ingär i följande sekvens:
20 AAGCTGCTCR'ACCCCGCG, och av högst nämnda 18 nukleotiders sekvens, väri R* är T eller C; eller ovan nämnda komplementära sekvens.
6. Probe enligt nägot av patentkraven 1-5, kännetecknad därav, att en oligonukleotid- 25 probe, med en i patentkraven 1-5 nämnd sekvens, är radioaktivt märkt, med i synnerhet en eller tvä radio-aktiva nukleotider av vilken typ som heist, i ena eller i vardera ändan, eller i synnerhet märkt med enzym-, ligand-, luminicerande eller fluorescerande märken. 30
7. Förfarande för bestämning av bakterier i ett prov, varvid efter att bakteriernas nukleinsyror bringats tili behandling och gjorts ensträngiga bringas nukleinsyrorna i kontakt med proben under förhällanden, vilka gynnar hybridisation, och de möljigen bildade 35 hybriderna bestäms, kännetecknat därav, att säsom probe används en probe enligt patentkrav 1, 2 eller 3. 2i 89185
8. Förfarande för bestämning av eukaryotista eller prokaryotiska organismers, bakterierna medräkna-de, celler ur ett prov, varvid efter att cellernas nukleinsyror bringats tili behandling och gjorts en-5 strängiga bringas nukleinsyrorna i kontakt med proben under förhällanden, vilka gynnar hybridisation, och de möjligen bildade hybriderna bestäms, kanne-t e c k n a t därav, att säsom probe används proben enligt patentkrav 4. 10
9. Förfarande för bestämning av arkeabakterier (archaebacteria) ur ett prov, varvid efter att ar-keabakteriernas nukleinsyror bringats tili behandling och gjorts ensträngiga bringas nukleinsyrorna i kontakt med proben under förhällanden, vilka gynnar hybridisa-15 tion och alla möjliga bildade hybrider bestäms, k ä n-netecknat därav, att säsom probe används proben enligt patentkrav 5.
10. Förfarande för lokalisering av DNA-frag-ment, vilka innehäller 16S seriers RNA kodande gener, 20 ur elektroforetiskt eller gaskromatografiskt separerade DNA-fragment, varvid de separerade och denaturerade DNA-fragmenten bringas i kontakt med en probe under förhällanden vilka tilläter en möjlig hybridisation och de möjligen bildade hybriderna bestäms, k ä n n e -25 tecknat därav, att säsom probe används vilken som heist probe enligt patentkraven 1 - 5 .
