Claims (11)
1. Förfarande för framställning av xylitol, käiinetecknat därav, att man a) odlar i ett xyloshaltigt medium en jäststam transformerad med en DNA- 5 sekvens som kodar föt ett xylosreduktasenzym, vilken DNA-sekvens dä den transformerats i den nämnda jäststammen ger jäststammen förmäga att reduce-ra xylos tili xylitol, och b) ätervinner bildad xylitol frän mediet. 10
2. Förfarande enligt patentkravet 1, kännetecknat därav, att den nämnda DNA- sekvensen kodar för en polypeptid omfattande väsentligen aminosyrasekvensen enligt SEKV.ID-nr 2, eller ett fragment därav, väri den nämnda polypeptiden eller fragmentet uppvisar xylosreduktasaktivitet. 15
3. Förfarande enligt patentkravet 1 eller 2, kännetecknat därav, att den nämnda transformerade jäststammen tillhör arten Saccharomyces cerevisiae, Kluyveromy-ces spp., Schizosaccharomyces pombe eller Pichia spp.
4. Förfarande enligt nägot av patentkraven 1-3, kännetecknat därav, att den nämn-20 da transformerade jäststammen tillhör arten Saccharomyces cerevisiae.
5. Förfarande enligt nägot av patentkraven 1-4, kännetecknat därav, att den nämn-da transformerade jäststammen är Saccharomyces cerevisiae H475 eller H477, som erhälles genom transformation av Saccharomyces cerevisiae GPY55-15B<% 25 eller respektive S150-2B med plasmiden pUA103, som anges i fig. 1.
6. Förfarande enligt patentkravet 1, kännetecknat därav, att minst en ytterligare ‘ ' kolkalla finns närvarande i mediet för att öka regenereringen av en ko-faktor, som är NADPH eller NADH, som krävs för xylosreduktasaktivitet.
7. Förfarande enligt patentkravet 6, kännetecknat därav, att den ytterligare kolkäl-lan är etanol, glukos eller glycerol. 30 104636 31
8. Förfarande enligt patentkravet 1, kännetecknat därav, att den nämnda transfor-merade jäststammen vidare transformeras med en andra DNA-sekvens som styr uttryck i den nämnda jäststammen av ett xylitoldehydrogenasenzym, vilket xylitol-dehydrogenasenzym möjliggör regenerering av en ko-faktor som krävs för xylos- 5 reduktasaktivitet.
9. Förfarande enligt patentkravet 8, kännetecknat därav, att den transformerade jäststammen tillhör arten Saccharomyces cerevisiae, Kluyveromyces sppSchi-zosaccharomyces pombe eller Pichia spp. 10
10. Förfarande enligt patentkravet 9, kännetecknat därav, att den transformerade jäststammen är Saccharomyces cerevisiae VTT-C-91181 som bar plasmiden pMW22 (NCYC nr 2352) som dessutom är transformerad med plasmiden pUA103, som anges i Fig. 1. 15

Enzymatiskt förfarande för framställning av xylitol, kännetecknat därav, att man a) odlar en rekombinant jäststam transformerad med en DNA-sekvens som kodar för xylosreduktasenzym under betingelser som möjliggör uttryck av det 20 nämnda xylosreduktaset, b) ätervinner det nämnda xylosreduktasenzymet, c) använder det nämnda xylosreduktaset i en enzymreaktor med ett ko-faktor- * regenererande system, och d) isolerar och renar bildad xylitol. 25 ^ J l • · <
