SE OBTIENE UN ACIDO NUCLEICO INMOVILIZADO A UN SOPORTE SOLIDO, SEGUN EL PROCEDIMIENTO DE LA FIGURA, PARTIENDO DE UNA SONDA DE ACIDO NUCLEICO, A LAS QUE SE UNE, UN NUCLEOTIDO MODIFICADO CON GRUPOS MERCURIO O MERCAPTO, EN EL EXTREMO 3''-HIDROXILO DEL DNA, MEDIANTE LA REACCION ENZIMATICA DE DESOXINUCLEOTIDIL-TRANSFERASA TERMINAL Y POSTERIOR INCUBACION A 15G DURANTE 60 MINUTOS, EXTRACCION CON FENOL Y PRECIPITACION CON ALCOHOL. PREVIAMENTE SE HABRA PREPARADO UN SOPORTE SOLIDO A BASE DE 0,5 GR DE CELULOSA, EN 5 ML DE AGUA DESTILADA, LLEVADA A PH 10,5-11 CON HIDROXIDO SODICO Y A LA QUE AÑADE CIANOGENO SOLIDO Y SE AMIDA CON 5 ML DE HEXAMETILENDIAMINA. ESTA CELULOSA AMIDADA SE LE PUEDE AÑADIR UNA SOLUCION DE SPDP (N-SUCCINIMIDIL-3)(2-PIRIDILTIO) PROPIONATO) O TIOLACTONA DE N-ACETILHOMOCISTEINA O ACETATO DE P-AMINOFENIL-MERCURIO SEGUN LA MODIFICACION DEL NUCLEOTIDO, QUE DA LUGAR A LAS TRES VARIANTES DE LA FIGURA.
