Abstract

PURPOSE:To detect even a gene having an amino acid arrangement by the synthesis of a few kinds of oligo nucleotide, by synthesizing oligouncleotide having basic arrangement corresponding to <=4 amino acids and using it as a probe. CONSTITUTION:Oligoliponuclueotide having basic arrangement CCXCCXCC, etc. [X shows cytidine (C), uridine (U) and adenosine (A)] whch is capable of forming strong hybrid (complex of DNA and RNA) such as guanosine (G), cytidine (C) and having the basic arrangement correspondig to 3-4 amino acids of a gene, is synthized arranging eight base at most by using an enzymatic method. The tip 5' of this synthesized oligolipouncleotide labeled by radioactive <32>P. Further, the gene coding specified amino acid arrangement i. e. specified protein, can be detected by using said labeled oligolipounucleotide. Thus, the useful gene is utilized by cloning.
