Abstract

Método para preparar un reactivo que se aglutina al ponerse en contacto con fluidos que contienen gonadotropina coriónica humana, consistente en: inyectar a un conejo una determinada cantidad de gonadotropina coriónica humana y dejar que el conejo produzca anti-cuerpos para la gonadotropina coriónica; retirar una parte de la sangre de dicho conejo y dejar que la sangre sufra las reacciones normales de coagulación; separar el suero del coagulo; precipitar la porción de globulina de dicho suero mediante la adición de una solución del sulfato amónico del 35 al 60% aproximadamente; purificar el precipitado para retirar todas las trazas de contaminantes flotantes; disolver el precipitado en una solución salina normal; mezclar la solución de globulina así obtenida con partículas de látex de poliestireno que tienen un tamaño comprendido entre 0,15 y 0,9 micras aproximadamente; incubar dicha mezcla a 56º aproximadamente; añadir una cantidad de 0,005 a 1M de agente tampón de dihidrocloruro de piperacina que tiene un pH de 5,0 a 8,6 y conteniendo de 0,0025 a 0,1M de ácido aminoacético y de 1 a 10% en peso de cloruro sódico; incubar dicha mezcla durante un tiempo adicional; y añadir suerosa a dicha mezcla en una cantidad aproximada del 0,5 al 20% y albúmina de suero bovino en una cantidad del 0,1 al 20% aproximadamente.
