1508133 Immunological test method TECHNICON INSTRUMENTS CORP 20 May 1975 [20 May 1974] Divided out of 1508132 21619/75 Heading G1B A method of analyzing a biological fluid sample for antibody Ab, antigen Ag or Ab: Ag complexes comprises adding to the sample before or after adding other reagents a solution of rheumatoid factor (Rf) or a particular component (Clq) of complement to bind with the Ab: Ag complexes present. The Rf or Clq may carry as identifying label a radioactive atom, enzyme, co-enzyme, enzyme inhibitor or fluorescent group. When the label is an enzyme, for example, catalase or amylase, its activity may be reduced when the Rf or Clq is bound to an Ab: Ag complex. Direct or competitive methods may be used for the assay. In the determination of an Ab: Ag complex there may be added to the sample a material which agglutinates with Rf or Clq, for example, polystyrene particles coated with immunoglobulin, the absence of agglutination indicating the presence of Rf or Clq bound to Ab: Ag complex. Examples relate to determination of thyroxine (T4), human IgE, alpha-1-foetal protein, tetanus anotoxin. Parent Specification 1508132 is referred to.
