Abstract
Cristal disódico del ácido 4,5-dihidro-4,5-dioxo-1H-pirrolo[2,3-f]quinolina-2,7,9-tricarboxílico (forma cristalina 1) que tiene un contenido de agua del 4,6% en peso o menos y que tiene picos de ángulo 2q en 11,4 ± 0,4, 13,5 ± 0,4, 18,0 ± 0,4, 18,7 ± 0,4, 26,0 ± 0,4, y 28,5 ± 0,4° obtenido por difracción de rayos X en polvo utilizando una radiación Cu Ka.

[0037] Se investigó la forma cristalina del cristal a cada una de las temperaturas preestablecidas y se demostró que: cuando la temperatura establecida era de 30ºC, el cristal resultante era similar al cristal precursor; cuando la temperatura establecida era de 120ºC, el cristal resultante se produjo como una mezcla del cristal precursor y la forma cristalina 1, que es similar a la mezcla de cristales en el Ejemplo 4 que tiene un contenido de agua del 7%; cuando la temperatura establecida era de 150ºC, el cristal resultante era un cristal en forma cristalina 1; y cuando la temperatura establecida era de 180ºC, los cristales resultantes fueron un cristal disódico de PQQ en una nueva forma cristalina (Forma cristalina 2) que tenía picos de ángulo 2θ en 8,7 ± 0,4, 11,5 ± 0,4, 12,0 ± 0,4, 17,4 ± 0,4, y 18,7 ± 0,4°. Los resultados de la difracción de rayos X en polvo para el cristal tratado a cada temperatura se muestran en la figura 2. El análisis termogravimétrico para el cristal en forma cristalina 2 reveló que el contenido de agua del cristal en forma cristalina 2 era casi el 0%. Sin embargo, este cristal era inestable. Cuando se dejó el cristal reposar a 30ºC durante un tiempo (por ejemplo, medio día o más), absorbió agua y se volvió a un cristal en la forma cristalina similar a la del cristal precursor (Figura 2).
Ejemplo Comparativo 1: Cristal disódico de PPQ diferente del Ejemplo de Referencia 1
[0038] El cristal precursor en el Ejemplo de Referencia 1 se disolvió en agua, y se añadió etanol a la solución para precipitar un cristal disódico de PQQ rojo. La medición del contenido de agua demostró que el cristal resultante tenía un contenido de agua del 14,5%. Posteriormente, el cristal resultante se secó a una presión reducida durante 96 horas de una manera similar al Ejemplo 1 y se sometió a una medición de difracción de rayos X en polvo. Sin embargo, se encontró que el cristal preparado por este procedimiento tenía una estructura con un bajo grado de cristalinidad con picos de ángulo 2θ en 9,4 y 22,4°. (Figura 3). La medición del contenido de agua demostró que el cristal resultante tenía un contenido de agua del 9,6%, y una pureza del 90,4%.
Ejemplo 6: Prueba de Dispersabilidad para los cristales disódicos de PQQ en forma cristalina 1 y forma cristalina 2
[0039] En este ejemplo, el cristal disódico de PQQ en forma cristalina 1 y forma cristalina 2 se comparó en la dispersabilidad en aceites comestibles con los cristales convencionales.
[0040] En primer lugar, los cristales disódicos de PQQ en forma cristalina 1 y forma cristalina 2 se prepararon de la siguiente manera: Un cristal en forma cristalina 1 se preparó añadiendo el cristal precursor (3 g) de la sal disódico de PQQ obtenida en el Ejemplo de referencia 1 a una recipiente de vidrio, evacuando mediante una bomba de vacío de tipo diafragma (DRYFAST ULTRA producida por A Gardner Denver, número de producto: 2032) hasta el límite del dispositivo (2 Torr) y, a continuación, secando a 150ºC durante 4 horas a una reducida presión para producir un sólido marrón oscuro. Después del secado, la presión en el recipiente volvió a la presión atmosférica, e inmediatamente el recipiente de vidrio se cerró con una tapa. Un cristal en forma cristalina 2 se preparó añadiendo el cristal precursor (3 g) de la sal disódica de PQQ obtenida en el Ejemplo de Referencia 1 a un matraz en forma de berenjena de 200 ml, y calentando a 200ºC durante 4 horas bajo reflujo con gas nitrógeno a una velocidad de flujo de 100 ml/min para producir un sólido marrón oscuro. Después del secado, la temperatura dentro del matraz se redujo a temperatura ambiente con gas nitrógeno bajo reflujo. En este ejemplo, se utilizó el cristal disódico de PQQ (cristal precursor) en el Ejemplo de Referencia 1 como control.
[0041] Cada uno (5 mg) de los cristales resultantes (el cristal precursor, el cristal en forma cristalina 1 y el cristal en forma cristalina 2) se añadió a aceite de soja (200 µL), y las mezclas se transfirieron a un recipiente de polipropileno y se mezclaron vigorosamente. A continuación, se les permitió reposar a temperatura ambiente, y se midió el período de tiempo hasta que todos los cristales que flotaban en el aceite de soja precipitaron en la parte inferior del recipiente, lo que se usó como un índice de la dispersabilidad para cada cristal.
