Un parche dérmico para retener selectivamente uno o más analito(s) de un cuerpo de un sujeto que comprende: - una capa de membrana (101, 601) para retener uno o más analito(s) de un cuerpo de un sujeto, con un primer y un segundo lado, en donde el primer lado se adapta para estar en comunicación fluida con la piel del sujeto, en donde la membrana se configura de modo que no sea expandible; - opcionalmente medios de sujeción para asegurar la capa de membrana en su sitio; - una capa adhesiva (104); - una capa de refuerzo (606); y en donde entre la capa de refuerzo (606) y la capa de membrana (101) hay una capa expandible (103) para aplicar presión a la capa de membrana.

capa expandible se hincha tras entrar en contacto con agua, tal como, por ejemplo, celulosa comprimida. El líquido de activación es en algunas formas de realización acuoso, tal como agua, o agua isotónica. En otras formas de realización el líquido de activación es tampón PBS acuoso.
La figura 7 muestra un ejemplo de un kit. En el paso 1 el kit contiene una toalla de limpieza desechable para limpiar el área donde se van a aplicar los dos parches. El paso 2 puede incluir un envase con un ingrediente activo (por ejemplo, producto, fármaco en crema, etc.) para aplicación tópica y un anillo adhesivo para marcar el sitio donde se aplicaría el ingrediente activo sobre la piel. El ingrediente activo se deja funcionar durante algún tiempo, por ejemplo 60 minutos. El paso 3 incluye dos parches: un parche control que se coloca en el área sin tratar limpiada, y un parche de medida que se coloca en la piel tratada con el ingrediente activo del paso 2. El paso 3 también incluye un líquido de activación para la membrana expandible y para humedecer la membrana. Los parches se dejan para recoger muestras de la piel durante algún tiempo, por ejemplo, 15 minutos. El paso 4 implica pegar el parche control y el parche de medida en las áreas designadas y enviar los parches a análisis.
En algunas formas de realización los ingredientes activos aplicados por vía tópica a la piel podrían ser, por ejemplo, ácido retinoico y/o progesterona, y los analitos que se van a detectar son al menos un analito seleccionado de uno o más de los grupos que comprenden:
a) un primer conjunto de analitos indicativos de que el compuesto o composición afecta a la regeneración de la
piel, en donde dicho primer conjunto de analitos es al menos una quimioquina tal como CCL5, CCL27 y CXCL1
y/o al menos un PAM tal como DEFB1, DEFB103A, DEFB4A, CAMP, DCD, S100A7 y RNASE7, y/o al menos
una interleuquina tal como IL1. b) un segundo conjunto de análisis indicativo de que dicho compuesto o composición afecta a la elasticidad de la
piel y/o la resistencia de la piel, en donde dicho segundo conjunto de analitos es al menos un PAM tal como
DEFB1, DEFB103A, DEFB4A, CAMP, DCD, S100A7 y RNASE7.
El dispositivo de detección de la invención puede detectar uno o más PAM y/o una o más quimioquinas y/o interleuquinas de cada conjunto de biomarcadores. El conjunto específico de biomarcadores que se va a medir puede depender de qué compuesto o composición cosmética se va a evaluar. La razón es que la correlación entre una MEC y un PAM puede variar dependiendo del compuesto o composición aplicada a la piel.
El efecto de la capa expandible se puede ver en el ejemplo 4, que compara la intensidad cuantitativa y deviación estándar de los analitos unidos en un parche según la figura 6 con y sin la capa expandible.
La diferencia en intensidad también se puede determinar cualitativamente comparando el parche de la figura 6 con y sin capa expandible, los resultados mostrados en la figura 4, donde se ve claramente que la intensidad es mejor en las membranas con la capa expandible que sin la capa expandible.
Cuando se describen las formas de realización de la presente invención, las combinaciones y permutaciones de todas las formas de realización posibles no se han descrito explícitamente. No obstante, el solo hecho de que ciertas medidas se enumeran en reivindicaciones dependiente mutuamente diferentes o se describen en formas de realización diferentes no indica que una combinación de estas medidas no se pueda usar con ventaja. La presente invención prevé todas las posibles combinaciones y permutaciones de las formas de realización descritas.

Ejemplo 1 – Preparación del parche de análisis que contiene anticuerpos de captura hBD1 e IL1a
El parche de análisis se produjo usando anticuerpos de captura de un kit de desarrollo de ELISA IL-1α humana (no. de catálogo 900-K11 de PeproTech) a una concentración de 50 µg/ml; y kit de desarrollo de ELISA de HBD 1 humana (no. de catálogo 900-K202 de PeproTech) a una concentración de 50 µg/ml. Como control negativo se usó PBS 1x-glicerol al 20%, y como control positivo se usó anticuerpo de detección de hBD1 a 50 µg/ml. Los anticuerpos de captura se aplicaron en manchas en la membrana usando un sistema de dispersión sin contacto (BioDot AD 3400 con sistema de dispersión BioJet plus). En este ejemplo el volumen de mancha dispersado fue 30 nl.
Ejemplo 2 – Exposición del parche de análisis a la piel
Colocar el parche de análisis sobre la piel (control para unión estrecha, la membrana tiene que tener un buen contacto con la piel) y mojar el parche (capa de expansión, membrana de análisis) con 150 µl de PBS 1x. Mantener el parche en contacto con la piel durante 15 min. Retirar el parche y separar la membrana del resto del parche. Colocar la membrana en una placa de 48 pocillos con el lado de la proteína hacia arriba.
Ejemplo 3 – Visualización del analito unido a la membrana
Para visualizar el analito unido a la membrana después de la exposición a la piel la membrana se incuba con anticuerpos de detección de la siguiente manera. Lavar las membranas 3x5 min en 200 µl de tampón de lavado. Preparar la mezcla de solución de anticuerpo de detección, hBD-1, 1:2000 e IL-1a, 1:1000 en tampón diluyente. Incubar las membranas con la solución del anticuerpo de detección 45 min a temperatura ambiente RT en un agitador. Lavar las membranas 5x5 min con 200 µl de tampón de lavado cada vez.
La señal se amplifica usando el sistema de amplificación de señal catalizada (CSA) (Dako) de la siguiente manera. Preparar el complejo estreptavidina-biotina: 1 gota (una gota es ~ 40 µl) de reactivo A del complejo estreptavidina biotina, 1 gota de reactivo B del complejo de estreptavidina biotina y hasta 1 ml con solución de reactivo del complejo estreptavidina biotina. Diluir el reactivo del complejo de estreptavidina biotina preparado para 1/9 en PBS 1x-Tween al 0,05%. Pipetear en cada pocillo 150 µl de solución e incubar durante 15 min a temperatura ambiente (RT). Lavar las membranas 3x5 min en 200 µl de tampón de lavado. Incubar las membranas con la solución de reactivo de amplificación: 1/8 de reactivo de amplificación, 7/8 de PBS 1x-Tween al 0,05%. Pipetear en cada pocillo 150 µl de solución. Incubar durante 15 min a temperatura ambiente (RT) seguido por lavar 3x5 min en 200 µl de tampón de lavado. Incubar con solución estreptavidina-HRP diluida: 1/8 de estreptavidina-HRP; 7/8 de PBS 1x-Tween al 0,05%. Pipetear en cada pocillo 150 µl de solución e incubar durante 15 min a RT.
Lavar las membranas dos veces con 200 µl de tampón de lavado seguido por un lavado con PBS 1x/Tween al 0,05%. Incubar con 100 µl de DAB (tetracloruro de 3,3’-diabencidina (H2O2 al 0,8% añadido; 20 µl de H2O2 al 0,8% para 1 ml de DAB) durante 15 min. Para la reacción con agua MilliQ (~200 µl por pocillo): añadir agua MilliQ en los pocillos y aspirar instantáneamente. Lavar las membranas con agua MilliQ 10x2 min (100 µl de una vez).
Las membranas se dejan secar al aire y después la intensidad del color, proporcional a la concentración de los analitos se determinó usando software de análisis de imagen (Dige Imager, GE Healthcare) según el protocolo en el software para cuantificar la cantidad de analito unido. Las membranas se escanearon usando un escáner de documentos a 1200 dpi (Canon Scan LiDE 600F). Las imágenes obtenidas se analizaron usando software de análisis de micromatriz y transferencia en mancha/ranura Image Quant TL V2005 (GE Healthcare). La cantidad de analitos unidos a los anticuerpos de captura se calculó usando una curva de calibración estándar. En el procedimiento de calibración se incubaron membranas con anticuerpos de captura en soluciones que contenían diferentes concentraciones de analitos seguido por el procesamiento usando el protocolo estándar. Basado en los resultados obtenidos se construyó la curva de calibración y se usó para determinar la cantidad de analitos unidos al anticuerpo de captura.
Ejemplo 4 – Comparación de los resultados de parches con y sin material expandible
La comparación de resultados de análisis obtenidos usando el análisis de parches de piel con y sin capa expandible muestra que la capa expandible mejora la reproducibilidad y calidad de los resultados del análisis, como es evidente de la figura 6 y la tabla posterior.
La tabla muestra el efecto de la capa expandible en el nivel de detección y variación de los resultados del análisis del parche.
