Abstract
Un raton modificado geneticamente, que comprende: a. una inactivacion del gen RAG de raton; b. una inactivacion del gen II2rg de raton; c. una sustitucion de un gen de IL-3 de raton por un gen de IL-3 humana, en la que el gen de IL-3 humana esta en un locus del gen de la IL-3 de raton; d. una sustitucion de un gen de GM-CSF de raton por un gen de GM-CSF humano, en la que el gen de GM- CSF humano esta en un locus del gen de la GM-CSF de raton; e. un injerto de celulas madre y progenitoras hematopoyeticas humanas (HSPC); y f. una infeccion por un patogeno humano; y que carece sustancialmente de celulas hematopoyeticas de raton endogenas.
Description
La capacidad para estudiar los tejidos humanos en un contexto in vivo en ratones ha abierto una amplia gama de posibles vías de investigación. Las principales limitaciones han impedido la aplicación del enfoque y, de estos, una de las deficiencias más importantes ha sido la incapacidad de los factores de ratón para soportar células humanas. En efecto, en el sistema inmunológico, muchos factores esenciales necesarios para el desarrollo y función de células
5 inmunitarias humanas son específicos para cada especie y el ratón no los puede proporcionar de manera efectiva. Por lo tanto, se decidió seguir una estrategia de sustitución de los genes de ratón con sus homólogos humanos, lo que permite mejorar el desarrollo y la función de las células humanas e inactivar potencialmente los mismos de las correspondientes células de ratón. Mediante la aplicación de este concepto a ratones KI de citocinas humanas, en el presente documento se presenta la prueba de concepto de que la sustitución de los genes inmunes en el huésped ratón con genes humanos mejora los ratones HIS. En concreto, esta divulgación apoya la idea de que la reactividad cruzada de las citocinas adecuadas entre ratón y se humano y tener que competir con las células de ratón, de hecho, limita el prendimiento y la función de las células mieloides humanas en los ratones HIS actuales.
Las citocinas humanas pueden liberarse en los ratones HIS mediante inyección intravenosa, por ejemplo, para
15 reforzar la reconstitución de células NK y células T humanas por medio de inyecciones de complejos de IL–15/IL– 15Rα e IL-7, respectivamente. Otro enfoque es la inyección hidrodinámica del ADN plasmídico que expresa citocinas humanas, lo que conduce a la expresión transitoria en el hígado. Este enfoque ha sido muy utilizado recientemente para mejorar la reconstitución de CD humanas mediante la liberación hidrodinámica de GM-CSF e IL-4 (véase, Chen et al. (2009) Expression of human cytokines dramatically improves reconstitution of specific human–blood lineage cells in humanized mice, Proc Natl Acad Sci USA, 106(51):21783–21788). En contraste con la presente divulgación, no se han notificado respuestas funcionales de células mieloides o respuestas in vivo a patógenos en estos ratones. Por último, las citocinas humanas también pueden sobreexpresarse como transgenes en ratones HIS. Este enfoque se ha utilizado para generar ratones transgénicos (tg) de IL-3/GM-CSF/factor de células madre (SCF) (véase, Nicolini et al. (2004) NOD/SCID mice engineered to express human IL–3, GM–CSF and Steel factor constitutively
25 mobilize engrafted human progenitors and compromise human stem cell regeneration, Leukemia 18:341–347). En estos ratones, la expresión de citocinas humanas está dirigida por el promotor del citomegalovirus, que conduce a la expresión ubicua. Sin embargo, los ratones His tg para hIL–3/GM–CSF/SCF se ven obstaculizados por la reducción del mantenimiento de las células madre hematopoyéticas humanas en la médula ósea y la mielopoyesis terminal expandida. Una vez más, a diferencia de la presente divulgación, no se describieron la mejora de la función de las células mieloides ni las respuestas in vivo. Por el contrario, en el sistema descrito en el presente documento, la expresión fisiológica de los genes específicos en el estado de equilibrio y la inflamación permite el desarrollo y la función apropiada del tipo celular apropiado únicamente. Es importante destacar que el enfoque descrito en esta divulgación genera cepas de ratones que se pueden mantener y propagar en condiciones altamente reproducibles y ponerse a disposición de todo el mundo para los estudios.
35 Los ratones hIL–3/GM–CSF KI descritos en esta divulgación representan una mejora considerable sobre anteriores ratones HIS y los enfoques alternativos descritos anteriormente. En primer lugar, la liberación de IL-3 y GM-CSF humanos mediante la estrategia de KI descrita en el presente documento conduce a la expresión de citocinas a largo plazo, lo que evita la necesidad de inyecciones repetidas de citocinas caras. En segundo lugar, se consigue una expresión fiel en órganos en los que la IL-3 y el GM-CSF se expresan de forma normal. En condiciones fisiológicas, el GM-CSF se expresa principalmente en el pulmón (Fig. 5a). En contraste, la liberación hidrodinámica conduce a la expresión predominante en el hígado y en la circulación. En ambos órganos, el GM-CSF no se expresa en condiciones de equilibrio. En tercer lugar, las cantidades fisiológicas de IL-3 y GM-CSF se expresan en ratones KI en contraste con la liberación por inyección hidrodinámica o la sobreexpresión ubicua en ratones tg para hIL–3/GM–
45 CSF/SCF. Se ha demostrado que los niveles fisiológicos de GM-CSF son importantes para una respuesta inmunitaria protectora contra M. tuberculosis. Por lo tanto, los ratones transgénicos con sobreexpresión local de GM-CSF el pulmón muestra formación defectuosa de granulomas y aumento de la susceptibilidad a las M. tuberculosis. Del mismo modo, la administración intravenosa de GM-CSF también conduce un control alterado de la infección por
M. tuberculosis en ratones. En cuarto lugar, los ratones hIL–3/GM–CSF KI homocigotos permiten el deterioro simultáneo del compartimento mieloide de ratón, ya que la IL-3 y el GM-CSF de ratón no se expresan en ratones homocigotos. Esto conduce a una ventaja competitiva para las células mieloides humanas, como se muestra en la presente divulgación.
La tuberculosis causada por la infección con M. tuberculosis da lugar a 1,7 millones de muertes al año. Por lo tanto,
55 se necesitan con urgencia nuevas medidas preventivas y terapéuticas eficaces. Aunque los ratones pueden infectarse con M. tuberculosis, no representan un modelo ideal para la tuberculosis humana. Esto se debe a las diferencias específicas de especies en la respuesta inmunitaria a M. tuberculosis, por ejemplo, los ratones infectados no desarrollan granulomas bien organizados. Los granulomas son el sello distintivo de la respuesta inmunitaria en seres humanos con tuberculosis y contienen macrófagos activados que se fusionan para formar células epitelioides y gigantes multinucleadas y células T activadas. Los granulomas juegan un papel importante en la limitación de la replicación bacteriana y en el control de propagación de micobacterias. El GM-CSF promueve la diferenciación de MA en células gigantes multinucleadas in vitro. Los estudios en ratones transgénicos también revelaron un papel del GM-CSF en la fusión de los macrófagos para formar células gigantes multinucleadas in vivo. Además, el GM-CSF es esencial para la formación de granulomas después de la infección por micobacterias. La ausencia de granulomas en
65 los ratones inactivados para GM-CSF infectados con M. tuberculosis se asocia con un aumento de la replicación bacteriana y una reducción de la supervivencia reducida. Por último, los seres humanos con PAP, causada por la

plaquetas humanas) y una combinación de los mismos.
Aplicaciones no limitantes de ratones injertados modificados genéticamente
5 Un ratón modificado genéticamente injertado con células hematopoyéticas humanas es un animal útil en el que estudiar los patógenos que normalmente no infectan a ratones. Un ejemplo de ello es el agente causante de la fiebre tifoidea, S. typhi.
La fiebre tifoidea afecta a más de 21 millones de personas en todo el mundo, principalmente en el mundo en vías de desarrollo, incluyendo aproximadamente 400 casos/año en Estados Unidos. La fiebre tifoidea se ha tratado con los fármacos amoxicilina, ampicilina, cefotaxima, ceftriaxona, ceftazidima, cloranfenicol, ciprofloxacino, cotrimoxazol, ertapenem, imipenem, fluoroquinolonas (por ejemplo, ciprofloxacino, gatifloxacino, ofloxacino), estreptomicina, sulfadiazina, sulfametoxazol, tetraciclina y combinaciones de los mismos. Las infecciones recurrentes son habituales, lo que limita el tratamiento de enfermedades mediante terapia con antibióticos. Además, la resistencia a
15 múltiples fármacos también prevalece con las infecciones por S. typhi.
Se necesitan nuevas terapias, nuevas vacunas y nuevas maneras de probar la eficacia de las terapias y vacunas. Un ratón que puede infectarse con S. typhi, por ejemplo, sería útil identificar nuevas terapias y nuevas vacunas. Nuevas terapias y nuevas vacunas se podrían probar en un ratón de este tipo mediante, por ejemplo, la determinación de la cantidad de S. typhi en el ratón (en sangre o en un tejido dado) en respuesta al tratamiento con un supuesto agente anti-S. typhi o mediante la inoculación del ratón con una supuesta vacuna, seguido de la exposición a una administración infectiva de S. typhi y la observación de cualquier cambio en la infectividad debido a la inoculación con la supuesta vacuna en comparación con un control al que no se le ha inoculado la vacuna, pero se ha infectado con S. typhi.
25 Un ratón modificado genéticamente e injertado de acuerdo con la invención es útil para hacer un ratón que se pueda infectar por un patógeno humano que no infecta a ratones. Por ejemplo, el ratón es útil como animal no humano infectable por S. typhi. En una realización, el ratón modificado genéticamente e injertado muestra un injerto mejorado de las células humanas en comparación con un ratón injertado que carece de la modificación o modificaciones genéticas, en el que la mejora es suficiente para mantener una infección por S. typhi. En una realización específica, el mantenimiento de una infección por S. typhi incluye la capacidad de S. typhi para reproducir en el ratón. En una realización específica, la infección por S. typhi incluye la capacidad del ratón infectado para reproducir S. typhi. En una realización específica, el ratón es capaz de reproducir S. typhi al menos una semana, 10 días, dos semanas, tres semanas o cuatro semanas después de la introducción inicial o la exposición infecciosa de S. typhi.
35 También se proporciona un método para identificar un agente anti-S. typhi, en el que el método emplea un ratón como se describe en el presente documento que es infectable por S. typhi. Los ratones de tipo silvestre, y otros ratones inmunodeprimidos conocidos (por ejemplo, ratones inactivados para los genes RAG1/RAG2) no son susceptibles de ser infectados por S. typhi.
Se proporciona un ratón modificado genéticamente que comprende una inactivación del gen II2rg y una inactivación del gen RAG (por ejemplo, una inactivación del gen RAG2) (primer tipo) y que comprende también un reemplazo del gen de IL-3 endógeno de ratón con un gen de IL-3 humana y el gen de GM-CSF endógeno de ratón con un gen de GM-CSF humano (segundo tipo), en el que el ratón modificado genéticamente cuando está injertado con células
45 hematopoyéticas humanas es capaz de infectarse con S. typhi.
Los datos mostrados en la figura 1 son representativos del primero y el segundo tipo de ratón. Las modificaciones genéticas de los ratones en la figura 1 comprenden: (a) una inactivación del gen RAG de ratón; y (b) una inactivación del gen II2rg de ratón. Los ratones de la figura 1 también comprenden un injerto de células hematopoyéticas humanas. Los ratones se pueden modificar adicionalmente mediante otras dos modificaciones para crear el segundo tipo de ratón, que son: (c) la sustitución de un gen de IL-3 de ratón endógeno con un gen de IL-3 humana; y (d) la sustitución de un gen de GM-CSF de ratón con un gen de GM-CSF humano.

Una diferencia importante entre los sistemas inmunológicos de ratón y humano es la fracción de granulocitos presente en la sangre. Los linfocitos son preponderantes en los ratones, mientras que la sangre humana es rica en granulocitos, una diferencia de especie poco clara en su significado. Curiosamente, la presencia de TPO humana mejoró la diferenciación de los granulocitos humanos (Figura 14). Por lo tanto, la presencia de TPO humana en
5 ratones receptores favorece un equilibrio entre los granulocitos y los linfocitos que refleja mejor el estado fisiológico humano, un hallazgo posiblemente debido a un mejor mantenimiento y/o diferenciación de las células progenitoras mieloides humanas.
Más importante aún, los resultados muestran que la humanización de TPO favorece el mantenimiento del receptor secundario, repoblando las células madre y progenitoras hematopoyéticas humanas en el medio ambiente del ratón (Figura 15). Por lo tanto, el ratón Rag2–/–γc–/–TPOh/h representa un nuevo modelo para estudiar varios aspectos de la función de las células madre y progenitoras humanas in vivo.
Sin embargo, aunque se observó un mejor equilibrio entre los linajes mieloide y linfoide en la sangre, no se
15 observaron efectos significativos de la humanización de TPO sobre los niveles generales de injerto en los tejidos linfoides periféricos (incluyendo el bazo, la sangre y el timo) (Figura 13(g)–(i)). Esto podría explicarse por diferentes factores. En primer lugar, aunque se irradia a niveles subletales a los ratones receptores antes del trasplante, todavía hay presente una gran población de células mieloides de ratón. Entre esas células, los macrófagos son capaces de fagocitar células humanas y limitar los niveles generales de injerto en la periferia. Por lo tanto, el agotamiento genético de los macrófagos de ratón, o su inactivación funcional, podría permitir niveles más altos de injerto periférico. En segundo lugar, las células humanas pueden requerir factores de crecimiento humanos adicionales para favorecer su diferenciación terminal, la salida de la médula ósea y/o su supervivencia en la periferia. Se puede considerar un panel diverso de citocinas para cada linaje. Por último, aunque los órganos linfoides secundarios se forman en ratones humanizados, su estructura no es óptima en comparación con los tejidos
25 humanos. Esta estructura parcialmente defectuosa podría representar un límite en el número de células humanas que pueden sobrevivir en estos órganos.
Se pueden usar reemplazos de genes adicionales para mejorar aún más los ratones receptores. Para lograr esto, la tecnología utilizada en este estudio, basada en la sustitución de activación de un gen de ratón por su homólogo humano, presenta dos ventajas principales en comparación con los enfoques transgénicos clásicos. En primer lugar, ya que mantiene la mayor parte de las secuencias reguladoras de origen murino, garantiza que el gen humanizado se exprese fielmente en el ratón huésped. En segundo lugar, como la estrategia de activación sustituye la citocina de ratón por su homólogo humano, puede afectar a la población o poblaciones de células de origen murino que dependen de esta citocina, en el caso de que la citocina humana no tenga reacción cruzada completa en el receptor
35 de ratón. Esto puede proporcionar una ventaja competitiva adicional a la población o poblaciones de células humanas después del trasplante. De hecho, este parece ser el caso para la TPO humana, ya que la sustitución homocigota de TPO conduce a una reducción de las plaquetas de ratón y las CMH en animales no injertados (Figuras 13(a), 15(a) y 15(b)).
Con ratones con activación de TPO humana, se proporciona un modelo mejorado que puede ser útil para estudiar la fisiología in vivo de la hematopoyesis humana en general y en células madre y progenitoras hematopoyéticas humanas en particular. Por otra parte, estos ratones sostienen in vivo neoplasias malignas hematopoyéticas humanas que se originan a partir de células hematopoyéticas tempranas, tales como, por ejemplo, leucemias mieloides y neoplasias mieloproliferativas.

Obtención de ratones con IL–3/GM–CSF humano y TPO humana

Una construcción de direccionamiento para la sustitución de un gen de IL-3 de ratón por un gen de IL-3 humana y un gen de GM-CSF de ratón por un gen de GM-CSF humano en una sola etapa de direccionamiento se construyó utilizando la tecnología VELOCIGENE® (véase, por ejemplo, la patente de Estados Unidos 6.586.251 y Valenzuela et al. Se presentan los resultados de ELISA para IL-3 (f) de ratón y humana y la producción de GM-CSF (g) por esplenocitos activados. Los esplenocitos de los ratones m/m (barras abiertas) o h/m KI (barras rellenas) se estimularon con ConA e IL-2 durante 48 horas y los sobrenadantes se recogieron (cada n = 1). La IL-3 y el GM-CSF humanos no fueron detectables (ND) en ratones m/m.
Ejemplo 5
Ratones hIL–3/GM–CSF injertados: respuestas inflamatorias humanas aumentadas
15 Los ratones hIL–3/GM–CSF KI se generaron a partir de células madre embrionarias (ME) con un alelo de ambos Rag2 y II2rg ya eliminado. Se criaron en Rag2 KO II2rg KO, después se dejó el injerto con células hematopoyéticas CD34 + humanas. El quimerismo de las células hematopoyéticas CD45+ humanas y la distribución de células T, B, y natural killer (NK, asesinas naturales)) en la médula ósea, el timo, el bazo y la sangre no aumentó significativamente en los ratones/hIL–3/GM–CSF KI (datos no mostrados). También, las frecuencias de las células mieloides CD33+ humanas, los granulocitos CD66 +, los monocitos/macrófagos CD14+, monocitos no clásicos CD14IoCD16+, las células dendríticas (CD) CD11c + CD14IoCD16 (DC), y CD123 + CD11c- plasmocitoide de CC no se incrementaron significativamente en ratones hIL–3/GM–CSF KI (datos no mostrados). Esto se aplica a ratones tanto h/m como h/h en condiciones de equilibrio. Por último, las células de médula ósea humana de ratones hIL–3/GM–CSF KI injertados tenían una capacidad similar para formar colonias mieloides en metilcelulosa in vitro (datos no mostrados).
25 Estos resultados son consistentes con los resultados de estudios en ratones inactivados que muestran que tanto la IL-3 como el GM-CSF son en gran parte prescindibles para la mielopoyesis en estado en equilibrio en los órganos analizados en el presente documento.
Por el contrario, el GM-CSF desempeña un papel importante en la mediación de las respuestas inflamatorias. La expresión de GM-CSF es inducida por estímulos inflamatorios, lo que conduce a la producción de citocinas inflamatorias (tales como IL-6 y TNF) por los monocitos/macrófagos y a su reclutamiento a sitios de inflamación. Los monocitos CD14+ humanos procedentes de ratones hIL–3/GM–CSF KI injertados tenían la expresión más elevada de la cadena α del receptor de GM-CSF (CD116) (Figura 8 (d)). Por lo tanto, el análisis de ratones hIL–3/GM–CSF KI injertados se centró en los monocitos/macrófagos humanos. En primer lugar, se analizó la respuesta inflamatoria
35 de los monocitos humanos en ratones hIL–3/GM–CSF KI injertados. La inflamación sistémica se indujo mediante inyección intraperitoneal (i.p.) de lipopolisacárido (LPS). La frecuencia de monocitos CD14+ humanos en circulación aumentó significativamente en ratones h/m en comparación con los ratones control m/m después de la inyección de LPS (Figura 8 (e), (f)). La movilización potenciada de los monocitos humanos en ratones h/m se asoció con un aumento de las concentraciones séricas de la IL-6 humana después de una y dos inyecciones de LPS (Figura 8 (g)). La producción inducida por LPS de TNFα humano también se incrementó en ratones h/m, pero este resultado no alcanzó significación estadística. Estos datos indican que los ratones hIL–3/GM–CSF KI injertados con células hematopoyéticas humanas tienen respuestas inflamatorias mejoradas mediadas por las células mielo-monocíticas humanas.

La figura 8 (d) muestra el análisis por citometría de flujo de células de médula ósea humanas de ratones hIL–3/GM–CSF h/m KI injertados en estado estacionario; la gráfica de puntos (izquierda) está acotada en las células hCD45+ mCD45.
