Una molécula que tiene las características de la invención puede incluir uno o más conectores X de manera que una porción ácida A se puede conectar a las porciones B y C por medio de una o más conexiones. Tales conexiones que se conectan a la porción A pueden ser a la porción B, a la porción C, o a ambas porciones B y C. Cuando una molécula que tiene las características de la invención incluye múltiples conexiones X, la separación de la porción A de las otras porciones de la molécula requiere la escisión de todas las conexiones X. La escisión de múltiples conectores X puede ser simultánea o secuencial. El hecho de que los capilares tengan a menudo fugas alrededor de los tumores y otros sitios de trauma debería mejorar la capacidad de las moléculas de alto peso molecular (p. ej., peso molecular de aproximadamente 40 kDa o más) para alcanzar el compartimento intersticial. Dado que la escisión de un conector X es típicamente extracelular, se espera algún marcaje circunstante, es decir, las células que no expresan la proteasa relevante, pero que son inmediatamente adyacentes a las células que expresan son propensas a recoger un poco de la carga. Para los tumores, tal direccionamiento circunstante se considera beneficioso debido a la heterogeneidad de los fenotipos celulares y el deseo de eliminar el mayor porcentaje de células sospechosas.
El hecho de que un solo mecanismo pueda mediar la captación tanto de cargas de formación de imágenes y terapéuticas será especialmente valioso, puesto que la obtención de imágenes con cantidades de trazador no perjudiciales se puede utilizar para comprobar si es probable que una dosis terapéutica subsiguiente se concentre correctamente en el tejido diana.
Se pueden utilizar D-aminoácidos en moléculas de MTS que tienen las características de la invención. Por ejemplo, algunos o todos de los péptidos de las porciones A y B puede ser D-aminoácidos en algunas realizaciones preferidas de la invención. Ejemplo 4
Péptido escindido por metaloproteinasa de la matriz 2 (MMP-2):
La MMP-2 (5 µg en 88 µl) fue activada a partir de proenzima fibroblastos sinoviales reumatoides humanos (Invitrogen) en tampón Tris-HCl como describen Stricklin et al (1983) en Biochemistry 22: 61 y Marcy et al (1991) Biochemistry) 30: 6476), a continuación se incubó con 32 µl de solución de partida de péptido 0,5 mM durante una hora a temperatura ambiente. Ejemplo 5
Análisis FACS de la captación celular:
Se cultivaron células Jurkat de tipo salvaje de la línea de células T humanas en medio RPMI 1640 con suero de ternera fetal desactivado (FBS) al 10% (v/v) y alcanzaron una densidad de ~1x106 células/ml. El medio se cambió por medio de nueva aportación un día antes de ser utilizado. Antes del experimento, las células Jurkat se lavaron con tampón HBSS tres veces y se resuspendieron en HBSS a una densidad de (0,5 -1) x 106 células/ml. En el experimento de captación celular, las células se tiñeron con el péptido 1 µM o compuesto a temperatura ambiente durante 10 min, después se lavaron dos veces con HBSS frío y se sometieron a análisis FACS. La captación celular se controló mediante fluorescencia a 530 nm ejecutada en FACS y se registraron 5.000-10.000 eventos a partir de las células consideradas sanas por dispersión delantera y lateral. Los datos representan la fluorescencia media de la población celular registrada lo que indica la captación de los compuestos marcados con fluorescencia. En la mayoría de los experimentos, se incluyó F1-GGR10-CONH2 (abreviado como "R10" en los gráficos; SEQ ID NO: 49) como control positivo para la captación.
La fluorescencia media medida en las células Jurkat incubadas durante diez minutos con los péptidos indicados (cada uno con radicales de carga fluorescentes) se muestra en las Figuras 8, 9 y 10.
Como se muestra en la Figura 9, los compuestos 6-14 (SEQ ID NO: 7) y 6-16 (SEQ ID NO: 8) mostraron mucha más fluorescencia, lo que indica mucha más captación, de la forma escindida de los péptidos que los péptidos intactos. Del mismo modo, como se muestra en la Figura 10, los compuestos 7-2 (SEQ ID NO: 11) y 7-6 (SEQ ID NO: 13) también mostraron mucha más fluorescencia después de la escisión en comparación con la fluorescencia de los compuestos no escindidos. Por lo tanto, estos resultados demuestran la prevención de la captación celular de los compuestos que tienen aminoácidos alcalinos conectados a una porción ácida. Además, estos resultados demuestran una mayor captación celular de las porciones fluorescentes de estos péptidos (que tienen aminoácidos alcalinos) después de la escisión de las porciones ácidas.
Tal captación celular aumenta a medida que aumenta el tiempo de incubación. La Figura 11 ilustra la fluorescencia media medida en células Jurkat incubadas durante diez minutos con los péptidos indicados que tienen radicales de carga fluorescentes, porciones alcalinas y ácidas, y porciones conectoras escindibles. Como se muestra en la Figura 12, la fluorescencia media medida en células Jurkat incubadas durante una hora aumentó en comparación con la fluorescencia medida como se muestra en la Figura 11.
