Diferentes ensayos en las formulaciones
Se utilizaron los métodos estándar para:
-SE-HPLC,
-RP-HPLC,
-Humedad residual,
-pH,
-Osmolalidad (Hora 0, solamente),
-SDS-PAGE / SS, y
-Mapeo de péptidos / UPLC (formas oxidadas).
Contenido de proteínas
La cuantificación de la proteína, es decir, trFGF-18, en las diferentes formulaciones se realizó mediante Biacore. Se ensayaron muestras (25μL) que contienen trFGF-18 (adecuadamente diluido en presencia de 10 mg/ml de BSA) haciendo fluir, en 5μL/min a 25° C, sobre la superficie del sensor, previamente recubierto con el clon F5A2 del anticuerpo monoclonal anti-FGF-18, seguido por 5μl de glicina 10 mM a pH 2,0 como tampón regeneración. Se utilizó el patrón de referencia (IRS FGF-18 No. 051230), que va desde 125 hasta 2.000 ng/ml, en cada sesión de análisis. Los resultados se recogen como Unidad de Resonancia (RU) y los niveles de FGF-18 de cada muestra se extrapolaron a partir de la curva estándar ajustada utilizando el algoritmo cuadrático con transformación logarítmica de los datos.
Bioensayo
La actividad biológica de FGF-18 se mide como actividad de proliferación por medio de un bioensayo in vitro utilizando la línea celular estable BaF3 transfectada con el receptor 3c de FGF (FGFR3c). Las células BaF3 que expresan FGFR3c proliferan bajo el estímulo de FGF-18.
Las células BaF3/FGFR3c se cultivan en medio selectivo basado en RPMI 1640 en presencia de r-hIL-3 como factor de crecimiento. Con el fin de probar específicamente el efecto proliferativo de FGF-18, las células tienen que ser privadas de IL-3. Por lo tanto, las células se cultivaron a 37° C, 5% de C02, en ausencia de r-hIL-3 durante 26 horas antes del ensayo.
El estándar y las muestras se diluyeron 5 veces de forma seriada en un medio de ensayo con un intervalo de concentraciones de 0,002 U/ml hasta 177,5 U/ml. Las células BAF3/FGFR3c privadas de IL-3 (20.000 células/pocillo) se incuban a 37° C, 5% de C02 con FGF-18 en presencia de 1 µg/ ml de heparina y 10% de Suero de Ternero Recién Nacido, y después se evaluó la proliferación celular después de 48 horas de ensayo de luminiscencia "ATPIite 1 etapa", un sistema de control de ATP basado en luciferasa de luciérnaga.
La potencia de la muestra se mide aplicando el modelo de curva de dosis-respuesta extendido. Con este modelo, toda la curva dosis-respuesta del estándar y de las muestras se ajusta a la curva dosis-respuesta sigmoidal con un algoritmo de pendiente variable (4PL) que informa de los valores cps (es decir, cuentas por segundo) frente al logaritmo de los valores de las concentraciones de FGF-18. Se calcula automáticamente EC50 para cada curva por el software GraphPad. La potencia de la muestra se calcula sobre la base de la relación entre la EC50 de la preparación de referencia y la de la muestra desconocida (relación de potencia). La potencia de FGF-18 se expresa como U/ml.
Ejemplo 1: Formulaciones liofilizadas de trFGF-18
La composición de las formulaciones liofilizadas de FGF-18 se proporciona en la tabla 1 a continuación.

Las formulaciones liofilizadas se prepararon de la siguiente forma: Se disolvieron sacarosa y Poloxamer 188 en el tampón fosfato. Después, se añadió la cantidad necesaria de sustancia fármaco (trFGF-18), se comprobó el pH y la disolución se llevó hasta el volumen final. Después, la solución se filtró a través de una membrana de 0,22 μm (Durapore®) bajo presión de nitrógeno, manteniendo un coeficiente de filtración de aproximadamente 20 cc/cm2 o de aproximadamente 50 cc/cm2.
La disolución final se envasó de forma manual, en condiciones estériles en viales de vidrio (0,5 ml/vial) y se liofilizó de acuerdo con el siguiente ciclo:
Las formulaciones liofilizadas están listas para ser reconstituidas, en cualquier momento, con un disolvente, tal como el agua para inyección o una solución salina.
Ejemplo 2: Estudios preliminares de estabilidad de las formulaciones FGF-18
Ejemplo 2.1: Se utilizaron condiciones de estabilidad aceleradas (a 2-8° C, 25° C y 40° C con el fin de identificar las 15 formulaciones previas liofilizadas para su estudio posterior (datos no mostrados).
Se prepararon varias formulaciones preliminares liofilizadas, con el fin de identificar las mejores formulaciones candidatas. En el marco de este estudio preliminar, se han evaluado diversos tampones, incluyendo tampones fosfato y de histidina. También se probaron diversas concentraciones de sacarosa y poloxámero 188. Se ensayaron las formulaciones pre-candidatas para determinar la pureza (por SE-HPLC y RP-HPLC), el contenido de proteínas
20 (por Biacore), la actividad biológica (bioensayo in vitro), la humedad residual, el pH y la osmolalidad. Los datos de estabilidad de apoyo también se generaron por medio de SDS-PAGE/SS (pureza) y por mapeo de péptidos/UPLC (formas oxidadas).
